Development of a Rapid PCR Test for Identification of Streptococcus agalactiae in Milk Samples Collected on Filter Paper Disks

  • Streptococcus (Strep.) agalactiae is one of the major pathogens of bovine mastitis and is the main cause of subclinical infection. This study attempted to develop a rapid PCR diagnosis procedure using milk samples collected on filter paper disks. Chromatographic filter paper was employed as the preservation media and kept at room temperature for one to four weeks. The revival rate of Strep. agalactiae kept on dried filter paper disks was affected by the pretreatment preservation time. The revival test suggested that not all the bacteria in artificially contaminated milk samples on the filter paper disks could be recovered. After that, a PCR based on the 16-23S intergenic spacer region of Strep agalactiae was performed. The results distinguished the strep. agalactiae from major pathogens of bovine mastitis at a $2{\times}10^2$ colony forming units (CFU)/ml level, which showed similar sensitivity to the results from liquid milk samples. The results also showed that milk samples collected on filter paper disks could be kept at room temperature for one to four weeks with little negative effect on sensitivity and specificity. The field test showed that the diagnostic sensitivity and specificity was 96.15% and 98.60%, respectively. In conclusion, the protocol will provide a rapid and economic procedure for the detection of bovine mastitis.

  • When medicine is unable to cure, and the end becomes imminent, or when the patient is tired of the side effects associated with chronic use of drugs, the search for alternative and new ways of healing is begun. Coincidentally, sometimes the alternative is the origin, as is the case for traditional Arab medicine and traditional American Indian healing. Traditional healing is the first healing that all people have used for 200,000 years, since the beginning of Homo sapiens. The sources and elements of traditional Arab medicine have been examined in books and by consulting with traditional Arab healers. Arabic medicine is a career combining both elements of science and philosophy based on religion and traditions, and includes a diversity of healing approaches: spiritual, physical, and using natural products. These approaches are discussed with emphasis on wet cupping (Alhijamah), a practice that is undergoing a revival nowadays in Arab countries. American Indian healing is a career based on religion, tradition, an innate healing gift and extensive training, both in a medical school setting and as an apprentice. Arabic healing approaches are compared to American Indian healing approaches.

  • The regional economic crises in the late 1990s and the global economic crisis in the late 2000s had reduced the differences in housing policies among the major Asian economies. This paper attempts to explain and compare housing welfare policy shifts between subsidizing home owning and subsidized renting from the perspectives of the economic and social roles of housing, the lock-in effect of policy processes, and the welfare provision strategy of the East Asian economies. It argues that the impact of economic crises on housing welfare policy in East Asia depended on the duration and the intensity of the crisis and the length and severity of the subsequent economic depression. Another important factor was the role of housing in the economic and social development, especially whether housing market development was considered as an engine of economic growth or revival, and whether the tools of housing policy caused the economic crisis. The loss of impetus for home ownership drive and the new emphasis on rental subsidy provision are new policy trends. Nonetheless, the economic revival since mid-2009 has caused the re-introduction of home ownership subsidies for quenching the housing affordability problems and enhancing home ownership making use of the strong economic conditions.

A Study on School Uniform and Ornaments Behaviors of High School Girls (여고생의 교복 및 장식 행동에 관한 연구 -경상남도 마산.창원 지역을 중심으로-)

  • The purpose of this study was to examine how high school girl students used their school uniforms and what types of ornaments they preferred, in an effort to identify the image of school uniform. The result of this study were as follow: First, 52.6 percent of the students investigated altered their uniforms, and as fur the major alteration style, revival one to take them in to be well-fitting was dominant. But concerning skirts, hip-hop style was also found. The students viewed ornaments more favorably, as the larger group of the students perceived that it was necessary to restrict the school-uniform alteration yet not to control ornaments. Concerning how much they wore school uniforms, most of them continued to wear them outside school in many cases. Specifically, those who engaged in pastime or went shopping wearing school uniforms enjoyed altering them, and they also enjoyed using ornaments that weren't allowed by the school dress code. Second, as to school-uniform image, elegant image was most preferred, followed by practical, innocent and fashion-conscious ones. They were most preferred with its elegant image. Third, as to ornaments image, they were most preferred with its positive image.

  • In order to satisfy customers' desire that is becoming more diverse, and to create the unique character of a product in the fast changing social structure and living environment, today's designers have to make efforts to provide new ideas continuously. In the twentieth century, the development of design in various aspects could be possible through an epoch-making progress of technology and industrialization, and thus the meaning and style of design has changed differently according to the current of the times. The retro design, which borrows the designs and styles which were popular in the past and recreates the design of modern meaning and concept, is widely applied now and in the spotlight as a latest design trend. This phenomenon of revival style appears as an image that makes us recollect the past in various fields while being recognized as an another value of new sense. Considered from the history of style changes for the past 100 years, the automobile design, the most representative in product design fields, has gradually changed in its types according to continuous historical, social, and cultural changes and technological progress, while a variety of designs have been continuously presented. The recent current of automobile styles, according to the rapid progress of advanced scientific technology, shows the fast and various changes and the retro style is becoming a new trend arousing people's interests and reintroducing the past designs. The revival trend in automobile designs is expected to stimulate human sensitivity and restore humanity on a different level before the trend, and also expected to be recreated as a new design trend in modern sense, connecting us with the past culture and expanding the realms of future designs.

Effects of Skimmed Goat Milk as a Semen Extender on Preservation of Bull Spermatozoa (탈지산양유(脫脂山羊乳)가 우정자보존(牛精子保存)에 미치는 영향(影響))

  • Skimmed goat milk heated at $92^{\circ}C$ for 10 minutes was used as a basal extender for bull semen. The extenders for liquid semen were prepared by adding simultaneously at various ratio of 5% dextrose solution and egg yolk to skimmed goat milk. After bull seven was diluted with the extenders at the rate of 20 million spermatozoa per ml of the extenders. The extenders were stored at $5^{\circ}C$ and the survival rates of spermatozoa were examined at 4 and 24 hours, and 3, 5 and 7 days after dilution. The extenders for frozen semen were prepared by adding various ratlo of glycerol to skimmed goat milk containing 20 parts of 5% dextrose solution and 3 parts of egg folk to 77 parts of skimmed goat milk. After bull semen was diluted with the extenders at the rate of 40 million spermatozoa per ml of the extenders, the extenders were frozen in liquid nitrogen tank. The frozen extenders were thawed at $40^{\circ}C$ for 2 minutes, and the revival rates of the spermatozoa in the extenders were examined. These thawed extenders were stored at $5^{\circ}C$ and the survival rates of the spermatozoa were examined at 10 minutes and 24 hours and 3 and 5 days after thawing. The results obtained were as follows: 1. Among the extenders stored at $5^{\circ}C$, the survival rate of the sperm was the highest in the extender including 20 parts of 5% dextrose solution and 3 parts of egg yolk to 77 parts of skimmed goat milk, and the survival rate was significantly higher that of the spermatozoa in egg folk-2.9% sodium citrate (1 : 4) extender. (P<0.05) 2.Among the extenders frozen in liquid nitrogen tank, the revival rate of the spermatozoa was the highest in the extender containing 7ml of glycerol per 100ml of the extender with consisted of 77 parts of skimmed goat milk, 20hparts of 5% dextrose solution and 3 parts of egg yolk, and the revival rate was significantly higher than that of the spermatozoa in egg yolk-2.9% sodium citrate (1 : 4) extender containing 8ml of glycerol per 100ml of the extender (p<0.01). 3. Among the extenders stored at $5^{\circ}C$ after thawing, the survival rate of the spermatozoa was the highest in the extender containing 7ml of glycerol per 100ml of extender which consisted of 77 parts of skimmed goat milk, 20 parts of 5% dextrose solution and 3 parts of egg yolk, and the survival rate was significantly higher than that of the spermatozoa in egg yolk -2.9% sodium citrate (1 : 4) extender containing 8ml of glycerol per 100ml of the extender (p<0.01).

  • The wireless communication revolution has spawned a revival of interest in the design and optimization of radio transceivers. Radio transmit modules continue to shrink in die size and cost, requiring novel approaches for integration of the numerous passive elements of the radio front-end. A 3 dB Wilkinson power divider based on GaAs substrates, for DCS 1710-1880 MHz band was designed and fabricated showing excellent performance.

  • It is expected to confront numerous technological advancements in ter railroad technology with an opening of state-of-art Kyung-Bu High Speed Railroad that permits traveling speed in excess of 300 Km/hr. Expansions of railroad facility such as revival of silk railroad in Eurasia due to the connection of South-North Korea and modernization of rolling stock and ancillary equipment have certainly contributed to the technological development of Korean Railroad. Such technological advancements will provide of opportunities to participate in foreign projects that have been offered in past years. Thus, a comprehensive plan and R&D policy is necessary to continuously raise and upgrade national competitiveness in the railroad industry.

  • 밀러-유리 실험은 1950년대 처음으로 초기 지구의 대기 방전 실험으로 아미노산을 합성한 실험이다. 생명의 기원에 대한 내용이 교과서나 책에 실릴 때마다 빠지지 않고 등장하는 실험으로, 학생들의 탐구 활동 연구주제로 실험을 설계하여 진행하였다. 실험실 설계부터 시작하여 실험 과정인 유리기구 세척, 진공 만들기, 기체주입과 방전실험을 소개한다. 실험 장치가 완성 된 후, 실험 시간 23시간 중 약 4시간 동안 방전하여 갈색 용액을 얻는데 성공하였다. 아민기와 반응하는 ninhydrin 반응에서 양성반응을 보여 아미노산 수용액임을 확인하였다.
