It has been known that retinoids are intrinsically of critical importance for control of premalignant epithelial cell differentiation. In the absence of retinoids, normal cellular differentiation and growth does not occur in epithelia such as those of trachea and bronchi. Furthermore, it was also reported that retinoid deficiency enhanced susceptibility to chemical carcinogenesis in the respiratory system, in the bladder, and in the colon of the experimental animal. In 1974, Bollag examined the effects of synthetic retinoids in prevention of development of cancer and demonstrated synthetic retinoids to have more favorable therapeutic index than retinoic acid for causing regression of skin papilloma in mice. Therefore, it was assumed that this anticarcinogenic effect of vitamin A derivatives could be due to modification of the metabolism of the carcinogenic polycyclic hydrocarbon, which must first be activated to exert their effect. Hill and Shih reported that vitamin A compounds and analogs had inhibitory effect on drug metabolizing enzyme from liver and lung tissue of mouse and hamster. Lucy suggested that the chemoprevention effect of vitamin A derivatives is due to reaction with molecular oxygen, and it is possible that inhibition of hydroxybenzpyrene formation is a result of this property. On the other hand, butylated hydroxytoluene which is a potent antioxidant strongly inhibited the formation of mammary tumor induced by dimethylbenranthracene. Also, it was observed that this antioxidant inhibited cancer induction in rats by N-2-fluo-renylacetamide. The purpose of this experiment was to investigate the effect of vitamin A derivatives such as retinoic acid and retinoid on drug-metabolizing enzyme and to determine whether riboflavin tetrabutylate or vitamin E could prevent of modify any changes induced by vitamin A delivatives in the rats. The results obtained were as followings. 1) Body weight was significantly reduced by retinoic acid, but not by retinoid. 2) Retinoic acid markedly increased liver weight while retincid showed no effect on liver weight. Treatment of riboflavin tetrabutylate did not affect retinoic acid-induced change in both body weight and liver weight. 3) Both retinoic acid and retinoid remarkably decreased the activity of aminopyrine demethylase. Pretreatment of riboflavin tetrabutylate, however, prevented inhibitory effect of retinoic acid on the enzyme activity. 4) No significant effect of vitamin E on aminopyrine demethylase was observed in both groups treated with retinoic acid and retinoid.
본 연구는 retinoic acid(RA)가 돼지지방전구세포의 분화와 유전자 발현에 미치는 영향을 구명하기 위해서 수행하였다. 지방전구세포는 갓난 돼지의 등지방에서 분리했으며 RA는 배양중인 세포에 4일 동안 처리하였다. 배양중인 세포에서 RNA를 추출한 후 14,688개의 유전자가 부착되어 있는 cDNA microarray와 혼성화 하여 유전자 발현 양상을 분석하였다. 지방전구세포의 분화는 GPDH의 활성도에 의해 측정했다. RA는 돼지지방전구세포의 분화를 78% 억제했다. Retinoic acid 처리에 의해 지질 대사에 관계된 유전자를 포함하여 특히 sphin- gomyelin phosphodiesterase, apolipoprotein R precursor, growth factor receptor-bound protein 14, retinoic receptor RXR gamma의 발현이 증가 되었다. 그리고 세포 성장에 중요 역할을 하는 vascular endothelial growth factor D precursor, growth hormone receptor precursor의 유전자의 발현이 감소되었다. 이러한 결과는, RA가 성장촉진인자 또는 성장호르몬 수용체의 조절을 통해서 돼지 지방전구세포의 분화를 억제함을 나타낸다.
The experiment was conducted to study the effects of water soluble vitamins and retinoic acid on the differentiation of preadipocyte from omental, subcutaneous, intermuscular and intramuscular adipose tissue of Hanwoo. Differentiation was assessed by the change in enzyme activity, glycerol-3 phosphate dehydrogenase in serum free cell culture system. Preadipocytes treated with biotin ($10{\mu}M$) and pantothenic acid ($100{\mu}M$) were significantly (p<0.05) less differentiated than those from the control in all adipose tissue depots except intramuscular tissue. Although there was no significance, vitamin C was shown to stimulate the adipocyte conversion in omental and subcutaneous, but not in intermuscular and intramuscular adipose tissues. Lower values of GPDH activity in intermuscular preadipocyte were interpreted to be caused by relatively higher amounts of protein. In this experiment vitamin C did not stimulate fat deposition in intramuscular adipose tissue but further experiments are needed on the role of vitamin C in preadipocyte differentiation. When treated with different levels of retinoic acids, differentiation of preadipocytes was significantly (p<0.05) reduced from the level of $0.5{\mu}g/ml$ in omental and intermuscular, from $50{\mu}g/ml$ in subcutaneous, and in intramuscular at $500{\mu}g/ml$, thus showing that intramuscular preadipocytes were least responsive to retinoic acid in differentiation. All-trans retinoic acid appeared to inhibit the differentiation in a dose dependent manner, regardless of adipose tissues type.
The structure-activity relationships of (E)-chalcone-4-carboxylic acids, flavone-4'-carboxylic acids, two types of aromatic amides, terephthalic monoanilides, and (arylcarboxamido)benzoic acids, which were made by Shudo group, are discussed by conformation analysis(AM1) of retinoic acid and those compounds. Conformer of each compound is superimposed on the conformationally restricted compound, 4-(6,7,8,9-tetrahydro-6,6,9,9-tetramethyl-4H-4-oxonaphto[ 2,3-b]pyran-2-yl) benzoic acid(Fv80), possessing the strongest differentiation-inducing activity on human promyelocytic leukemia cells HL-60. The results indicated that the lengths between the carboxylic carbon and the two 6, 9 carbons binding to dimethyl, 1.20 nm and 1.09 nm, as well as the planarity of molecule are very important factors for the activity, especially 1.20 nm. In the case of the recently synthesized azulenic retinoic acids by Sato, et al. in 1993, the distance probably is also important, resulted from superimposing them on a Ch55 conformer and Fv80. The distance 1.0 nm is also important in Ch55. Several conformers of all-trans retinoic acid (RA) are well superimposed on the almost non-flexible Fv80, RA, 9-cis RA, and, specifically s-10,12 cis RA. And a simple hexangular model of RA is suggested to draw RA conformers easily without computer drawing model or molecular model.
본 실험은 레티노익산에 의해 F9 Teratocarcinoma Stem Cell의 분화를 유도하고 이분화과정에서 세포형태의 변화와 라미닌유전자의 발현을 조사 하였다. 분화되지 않은 F9 Stem Cell은 지속적으로 증식을 하며 세포간의 간격을 구분하기 어령루 뿐만 아니라 불규칙적인 모양을 하고 있으나,레티노익산과 dibutyryl cyclic AMP처리후의 분화된 F9세포는 둥글고 평평한 모양을 나타내며 세포성장은 중지되었다. Northern blot분석에 의하여 레티노익산과 cyclic AMP처리 후의 F9세포내에서 라미닌 유전자의 발현은 현저하게 증가하였다. 즉, 라미닌 B1유전자 발현은 분화과정 동안 최소한 30배, 라미닌 B2 유전자의 발현은 약 20배 증가하였다. 또한 라미닌 항체를 이용한 면역형광 분석결과는 Northern 분석결과와 일치하게 분화 후에 라미닌 단백질 합성이 크게 증가되었으며, 생성된 라미닌 단백질은 거의 세포표면에 분포된 것으로 나타났다. 이러한 결과로부터, 레티노익산에 의해 F9 Stem Cell의 분화가 유도되며 이 분화과정에서의 형태적인 변화와 진행은 라미닌의 생성과 밀접한 관련이 있다고 추측된다.
본 연구에서는 수용액상에서 자기조립 나노입자를 형성할 수 있는 레티노익산이 접목된 양친성 폴리아미노산 유도체를 합성하였다. 합성한 양친성 폴리아미노산은 레티노익산의 접목도가 각각 5, 10, 30 mol%가 되도록 조절하였다. 수용액 상에서 양친성 폴리아미노산은 소수성 레티노익산의 분자 결합에 의해 안정한 자기조립 나노입자를 형성하였다. 자기조립 나노입자는 레티노익산의 접목도가 증가할수록 크기는 작아지고 형태는 구형에서 이중층 구조로 전이되었다. 또한 접목도가 10%일 때, 자기조립 입자의 구조 붕괴 없이 레티놀의 포집 및 전달이 가장 효과적인 것을 확인하였다. 접목도가 제어된 자기조립입자는 레티놀을 안정적으로 포집할 수 있기 때문에 주름개선제 및 다양한 기능성 화장품 전달체로 활용될 수 있다.
Peroxisome proliferator-activated receptor-gamma$(PPAR{\gamma})$ 는 DNA와 결합하기 위해 retinoid-X receptor(RXR)와 heterodimer를 형성해야만 한다. 그리고 전사에 대한 최대활성은 수용체에 대한 리간드 특이성에 의하는 것으로 생각되고 있다. 활성화된 $(PPAR{\gamma})$ 와 $(PPAR{\gamma})$ 리간드는 종양억제 PTEN의 조절을 통해 종양세포의 성장에 영향을 끼치게 된다. 본 연구의 목적은 $(PPAR{\gamma})$ ligand, ciglitazone그리고 RXR ligand로 동시에 자극하였을 때 급성전골수성백혈병(APL) 세포에 대해 이들이 함께 PTEN upregulate를 조절할 수 있는지를 결정하기 위함이다. 그리고 이들 세포의 성장과 분화주기에 대해 강력한 억제 능이 있는지를 결정하고자 하였다. 즉, 사람의 백혈병세포주인 HL-60세포에 all-trans-retinol과 ciglutazone을 노출시킨 뒤 PTEN 발현에 대한 측정을 위해 RT-PCR법으로 PTEN mRNA 발현 정도를 확인하고 western blot으로 분석하였다 세포주기의 분석은 propidium iodide(PI) 염색법과 FACScan으로 분석하였고, HL-60 cells에서 $(PPAR{\gamma})$ ligand, ciglitazone, 그리고 RXR ligand, retinoic acid 그리고 upregulated PTEN 발현에 대한 time- and dose-dependent방법으로 각각 확인하였던 바 ciglitazone과 retinoic acid를 동시 조합하여 처치하였을 때 유의적인 효과를 인정할 수 있었다. 더욱이 이들 혼합 물질은 세포의 성장과 G, phase를 동시 억제하는 능력이 있었다. 그러므로 $(PPAR{\gamma})$의 활성에 있어 RXR heterodimer가 사람의 백혈병세포에 대한 조절 경로로서 존재하며, PTEN의 upregulation을 통해 백혈병을 조절하기 때문에 백혈병의 예방 및 치료 접근에 $(PPAR{\gamma})$와 RXR ligands가 중요한 역할을 할 것이다.
The differentiation of mouse embryonic stem(ES) cell into smooth muscle cells(SMC) may play a major role in cardiovascular development and under pathophysiological conditions. Therefore, in the present study, we have examined the differentiation of ES cells and its related gene expression. SMC differentiation was indicated by cellular morphology and time-dependent induction of dibutyryl adenosine 3,5-cyclic monophosphate(DBcAMP)and retinoic acid(RA) on smooth muscle ${\alpha}$-actin($SM{\alpha}A$), smooth muscle myosin heavy chain(SMMHC) gene expression. The control was undifferentiated ES cells(protein expressions represent 50-60kDaOct-4). The results of this study show that morphology of embryoid body and confirmation of $SM{\alpha}A$ expression by immunocytochemistry. Moreover, SMMHC and desmin expression was significantly increased by time dependent manner(5, 7, 15 days), in contrast to $SM{\alpha}A$ expression was slightly decreased on 15days. In conclusion, DBcAMP and RA stimulate mouse ES cells differentiation into SMC and enhanced $SM{\alpha}A$, SMMHC and desmin expression.
