PSEUDOHYPOPARATHYROIDISM : CASE REPORT (가성부갑상선 기능저하증 환아의 구강 증상)

  • The parathyroid hormone plays a major role in the metabolism of calcium and phosphorus. In hypoparathyroidism the parathyroid glands are atrophied or absent associated with autoantibodies against parathyroid tissue. Pseudohypoparathyroidism is a metabolic disease caused by the disturbance in peripheral action of parathormone, but parathormone level is normal. In general, patients with pseudohypoparathyroidism have short stature, round face, brachydactylia, obesity, mental retardation, cataracts & ectopic calcifications on soft tissues. Dental manifestations are enamel hypoplasia, delayed eruption, blunting of root apex, hypodontia, pulp calcification, thickened lamina dura, excessive caries & malocclusion. In this case, intraoral examination showed enamel hypoplasia on the erupted permanent teeth & hypermobility on the remaining deciduous teeth. From the radiographic view severe dental anomalies were observed on canines and shortening and blunting of root apex was observed on mandibular incisors. Pathologic root resorption was also observed on deciduous teeth.

New Multi-herb Mixture, HT005, Induces Longitudinal Bone Growth in Male Rats (신한약복합물 HT005의 성장기 흰쥐 장골길이 성장에 미치는 효과)

  • Objectives : It was investigated new herbal prescription HT005 supported by traditional Korean medicine has a activity on the longitudinal bone growth of rats. HT005 were consisted of the root of Angelica sinensis, the fruit of Alpinia oxyphylla, the root of Astragalus membranaceus, the stem of Eleutherococcus senticosus, and the root of Dioscorea japonica, and Poria cocos. Method : To investigate the effect on longitudinal bone growth in adolescent male Sprague-Dawley rats, the longitudinal length of growth plate was measured directly by the tetracycline fluorescent labeling method and chondrocyte staining method. HT005 administered orally for four days, and the tetracycline was injected twice on the growth plate of the animals for fluorescent dying. The rate of longitudinal bone growth was measured the length between the tetracycline bands which fixed on the 3rd day and 5th day after injection. Cresyl violet was also used to stain the chondrocytes in the growth plate. The length of growth plate after administration was compared. Expression of IGF-1 and BMP-2 in the growth plate was investigated by immunohistochemistry. Results : HT005 showed a significant longitudinal bone growth which was $301.0{\pm}6.1\;{\mu}m/day$ at the dose of 100 mg/kg and $283.8{\pm}1.25\;{\mu}m/day$ (p < 0.001)at the dose of 10 mg/kg of HT005, compared to control group by the tetracycline fluorescent labeling method. HT005 showed a significant chondrocyte length on growth plate which was measured $797.19{\pm}3.31\;{\mu}m$ (p < 0.001) at the dose of 100 mg/kg and $720.14{\pm}2.19$ (p < 0.001) ${\mu}m$ at the dose of 10 mg/kg compared to the control group by cresyl-violet staining method. Both the number and intensity of BMP-2 and IGF-1 positive cells were increased in the hypertrophic zone of growth plate. There was a significant correlation between BMP-2 and IGF-1 expression and heights of chondrocytes in growth plate. Conclusions : Treatment of HT005 on Sprague-Dawley rats markedly increased the longitudinal bone growth. Therefore, HT005 may be an alternative herbal source to growth hormone as it promotes bone growth in children afflicted by growth retardation.

The Effects of Pretreatment on the Lipid Oxidation of Korean Geoduck Muscle during Frozen Storage (코끼리 조개의 동결저장 중 지질의 변화에 미치는 전처리의 효과)

  • The effects of pretreatment on the lipid oxidation of korean geoduck muscle during storage at $-20^{\circ}C$ were investigated. In the lipid compositions of geoduck muscle, neutral, glycolipid, and phospholipid contents were $46.1\%,\;4.5\%\;and\;49.5\%$, respectively. And the content of phospholipid decreased to $39.9\%$ by blanching. In the fatty acids composition, $C_{16:0},\;C_{20:5}\;and\;C_{22:6}$ fatty acids were major portions and their contents showed, respectively, $10\%$ of total fatty acids amount or more. $C_{18:0},\;C_{16:1},\;C_{18:1},\;C_{14:0}\;and\;C_{20:1}$ fatty acids were also important fatty acids and their amounts were so much as $5\%$ or more. The polyunsaturated fatty acid contents were relatively high as $39.25\%{\sim}42.60\%$. POV in RM and VM samples increased gradually and showed 3.72meq/kg, or 3.00meq/kg after 90 days storage, respectively. However, in case of samples pretreated by blanching, there was slightly increase of POV throughout storage. From the results above, it was suggested that the pretreatment of blanching was effective in retardation of lipid oxidation for the longterm storage.

Effect of Soy Protein Film Packaging on the Qualities and the Microbial Growth of Beef during Storage (대두 단백 필름 포장 방법에 따른 저장 중 쇠고기의 품질 및 미생물 변화)

  • To investigate the effect of soy protein film packaging on the quality of beef, the rate of weight loss, pH, 2-thiobarbituric acid reactive substance (TBARS), and microbial (total bacterial count) Salmonella spp., E. coli changes were determined during storage. Beef samples were packaged with soy protein film containing rosemary extract and then stored at $4^{\circ}C$. Soy protein film was effective on prevention of weight loss, compared with the control, regardless of addition of rosemary extract. Change in pH of soy protein films containing the rosemary extract (RPF) was less during storage, compared with the control. Packaging of beef products with RPF inhibited lipid oxidation by $86\%$ at day 8 of storage, compared to the control. RPF packaging also affected the microbial growth, resulting in retardation of total bacteria by more than 1 log cycle. These results indicate that storage of beef packaged with RPF should be appropriate in terms of quality as well as shelf-life.

  • Growth and anatomical responses of rice and barnyardgrass to molinate were examined under various cultural patterns in a greenhouse in which molinate had been applied at 3kg ai/ha 10 days after sowing or transplanting of rice. Molinate severely reduced growth of broadcast-sown rice compared to drill-seeded or transplanted rice. Molinate also significantly reduced the growth of barnyardgrass. The growth reduction of both species by molinate was more apparent under flooded than under dry conditions. Following the treatment of molinate, the leaf primordia of barnyardgrass were found to develop abnormally, resulting in a zigzag pattern. The effect of molinate on causing the symptom was more severe under flooded than under dry conditions.

Effects of Dietary Germanium Supplementation on the Meat Quality of Duck (게르마늄 급여가 오리의 육질에 미치는 영향)

  • This study was carried out to investigate the influence of dietary germanium supplementation on the meat quality of duck. Ducks raised for 42 days were slaughtered and the ground meats were stored at 3$^{\circ}C$ for 12 days under 1200 lux. Experimental treatments were divided into 3 kinds of meat from ducks fed germanium as follows; 1) Control(natural water + commercial feed), 2) T1(1~10 days: natural water + commercial feed, 11-42 days: natural water + commercial feed supplemented with 1 % germanium), 3) T2(1~10 days: germanium submersion water + commercial feed, 11~42 days: natural water + commercial feed supplemented with 1 % germanium). The pH of duck meat was not different among the treatments(p>0.05). The germanium treatments exhibited significantly higher crude fat content in both breast and thigh meat(p<0.05). In the fatty acid composition, T2 contained more unsaturated fatty acid than control or Tl. TPA(textural profile analysis) value such as hardness, gumminess, chewiness and adhesiveness decreased in meat from duck fed supplemental germanium. Cholesterol contents of duck meat decreased in dietary supplemental germanium treatment compared to control, but were not significantly different among them(p>0.05). Dietary germanium supplementation affected on lipid oxidation of meat during storage. TBARS of meat from duck fed germanium was lower than that of control(po.05). The CIE L*, b* and h$\^$0/ values of germanium treatments were significantly(p.o5) higher than those of control. Also a* value showed more stable in germanium diet treatment during storage. Therefore, dietary supplemental germanium to duck resulted in light brownish color formation, improvement tenderness and retardation of lipid oxidation of meat during refrigerated storage.

Transient neonatal diabetes mellitus with macroglossia diagnosed by methylation specific PCR (MS-PCR) (메틸화 특이 PCR로 진단된 거설증을 동반한 일과성 신생아 당뇨병)

  • Transient neonatal diabetes mellitus (TNDM) has been associated with paternal uniparental isodisomy of chromosome 6, paternally inherited duplication of 6q24, or a methylation defect at a CpG island of the ZAC or HYMAI gene. We experienced a case of TNDM in which the patient presented with hyperglycemia, macroglossia, and intrauterine growth retardation, caused by a paternally derived HYMAI. An 18-day-old female infant was admitted to the hospital because of macroglossia and recurrent hyperglycemia. In addition to the macroglossia, she also presented with large fontanelles, micrognathia, and prominent eyes. Serum glucose levels were 200-00 mg/dL and they improved spontaneously 2 days after admission. To identify the presence of a maternal methylated allele, bisulfite-treated genomic DNA from peripheral blood was prepared and digested with BssHII after polymerase chain reaction (PCR) amplification with methylation-specific HYMAI primers. PCR and restriction fragment length polymorphism analysis showed that the patient had only the paternal origin of the HYMA1 gene. TNDM is associated with a methylation defect in chromosome 6, suggesting that an imprinted gene on chromosome 6 is responsible for this phenotype.

Effect of Uniconazole and Silver Thiosulfate Treatment on Reduction of Ozone Injury in Snap Bean Plants (Uniconazole과 Silver Thiosulfate 처리(處理)가 강남콩의 오존피해(被害) 경감(輕減)에 미치는 효과(效果))

  • Studies were conducted to examine the effects of single or combined treatment of uniconazole [(E)-1-(4-chlorophenyl)-4, 4-dimethyl 2(1, 2,-4-triazol-1-yl)-1-penten-3-ol)] and silver thiosulfate (STS) on reducing ozone injury to snap beans (Phaseolus vulgaris L. 'Strike'). Two weeks after seeding, plants were given a soil drench of uniconazole(XE-1019) solution at concentrations of 0.001, 0.005 and 0.025 mg/pot, and then two days prior to ozone fumigation, 0.3 and 0.6 mM STS containing 0.01% Tween-20 were also sprayed. Uniconazole was effective in providing protection against ozone injury through increase activities of free radical scavengers such as superoxide dismutase (SOD) and peroxidase (POD) as well as the increase of chlorophyll content and stomatal resistance resulted from plant growth retardation. The phytoprotective effects of STS seemed to be related to its properly of blocking the ethylene action and increasing activities of SOD and POD. Even at low concentrations, a combined treatment with uniconazole drench, STS spray significantly reduced ozone injury compared to single application.

A Study on UV Degradation of SBR and NR Containing Polymeric UV Stabilizer (고분자형 자외선 안정제를 함유한 SBR과 NR의 UV 노화 거동에 관한 연구)

  • The retardation effects of a new polymeric UV stabilizer, DGEBA-HALS, on the UV degradation of SBR and NR were investigated in this study. The UV degradation behaviors of three different rubber compounds, without any photostabilizer and with commercial Cyabsorb UV-3529 and DGEBA-HALS, were compared. Also, the physical characteristics such as Young's modulus, tensile strength, blow-off deformation and crosslink density were examined. The Young's modulus was increased by UV irradiation for all samples, and the tensile strength, after UV irradiation, of the compound with UV stabilizer was better than that of the compound without stabilizer. Especially, the tensile strength of UV irradiated NR was improved by adding DGEBA-HALS. The results of blow-off deformation were in accord with those of Young's modulus. The degree of transmission of SBR compounds obtained from UV-vis transmission spectra was improved about 4% by adding the UV stabilizer. Commercial Cyabsorb UJV-3529 and DGEBA-HALS showed similar trends of transmission. The SEM photographs of SBR surface showed that the size of cracks created by UV irradiation became smaller by adding UV stabilizer. In particular, it is observed that the stabilization effect of DGEBA-HALS was better than that of commercial Cyabsorb UV-3529.

Regulation of sfs1 gene expression by the cAMP-cAMP receptor protein (sfs1 유전자의 cAMP-cAMP receptor protein에 의한 발현 조절)

  • We have cloned several E. coli sfs genes which stimulate mal gene expression with $crp^{{\ast}1}$). One the genes (pPVC2) was sequenced and potential CRP binding site is located in the upstream of the putative promoter in the regulatory region. In order to investigate the regulation of the sfs1 gene by the cAMP-CRP complex, we have constructed the sfs-lacZ fusion gene in this research. The overall transcriptional stimulations of sfs1 gene in the presence cAMP were confirmed by ${\beta}-galactosidase$ activity and Western blot analysis of sfs1-lacZ fusion gene. Transcriptional regulation by cAMP-CRP was also confirmed by Northern blot analysis. End-labelled DNA of the DNA fragment in sfs1 regulation region were used for gel retardation assay to examine the CRP-DNA complex in the presence of cAMP. Results here indicate that CRP binding site in the regulatory region of sfs1 gene is positive regulator for the expression of sfs1 gene.
