MoRBP9 Encoding a Ran-Binding Protein Microtubule-Organizing Center Is Required for Asexual Reproduction and Infection in the Rice Blast Pathogen Magnaporthe oryzae

  • Like many fungal pathogens, the conidium and appressorium play key roles during polycyclic dissemination and infection of Magnaporthe oryzae. Ran-binding protein microtubule-organizing center (RanBPM) is a highly conserved nucleocytoplasmic protein. In animalia, RanBPM has been implicated in apoptosis, cell morphology, and transcription. However, the functional roles of RanBPM, encoded by MGG_00753 (named MoRBP9) in M. oryzae, have not been elucidated. Here, the deletion mutant ΔMorbp9 for MoRBP9 was generated via homologous recombination to investigate the functions of this gene. The ΔMorbp9 exhibited normal conidial germination and vegetative growth but dramatically reduced conidiation compared with the wild type, suggesting that MoRBP9 is involved in conidial production. ΔMorbp9 conidia failed to produce appressoria on hydrophobic surfaces, whereas ΔMorbp9 still developed aberrantly shaped appressorium-like structures at hyphal tips on the same surface, suggesting that MoRBP9 is involved in the morphology of appressorium-like structures from hyphal tips and is critical for development of appressorium from germ tubes. Taken together, our results indicated that MoRBP9 played a pleiotropic role in polycyclic dissemination and infection-related morphogenesis of M. oryzae.

Bioconversion of Ginsenoside Rb1 to Compound K using Leuconostoc lactis DC201

  • Ginseng (Panax ginseng) is frequently used in Asian countries as a traditional medicine. The major components of ginseng are ginsenosides. Among these, ginsenoside compound K has been reported to prevent the formation of malignancy and metastasis of cancer by blocking the formation of tumor and suppressing the invasion of cancer cells. In this study, ginsenoside $Rb_1$ was converted into compound K, via secreted ${\beta}$-glucosidase enzyme from the Leuconostoc lactis DC201 isolated, which was extracted from Kimchi. The strain DC201 was suspended and cultured in MRS broth at $37^{\circ}C$. Subsequently, the residue from the cultured broth supernatant was precipitated with EtOH and then dissolved in 20 mM sodium phosphate buffer (pH 6.0) to obtain an enzyme liquid. Meanwhile, the crude enzyme solution was mixed with ginsenoside $Rb_1$ at a ratio of 1:4 (v/v).The reaction was carried out at $30^{\circ}C$ and 190 rpm for 72 hours, and then analyzed by TLC and HPLC. The result showed that ginsenoside Rb1 was transformed into compound K after 72 hours post reaction.

Synbiotics (mixture of probiotics and prebiotics) ameliorates DSS-induced ulcerative colitis in vivo.

  • Ulcerative colitis (UC) is one of inflammatory bowel disease (IBD), characterized by chronic inflammatory response and dysregulation of immune function. The severity of US has been influenced by environmental factors and food habit. The immune modulatory, anti-inflammatory and steroidal medicine have been used for the treatment of UC. However, long-term administration of those medicine is accompanied with side-effect. So, it is necessary to develop the non side-effect medicine using natural product. Prebiotics influences intestinal condition and food consumption. The heredity, immunity and environmental condition are related with occurrence of UC. In recent study, UC patients had lower level of prebiotics such as Lactobacillus and Bifidobacterium compared with healthy people. Also, previous study announced that imbalance of enteric flora aggravates the severity of UC. The effectiveness of probiotics might affect colon ability and viable bacteria also could promote the proliferation of beneficial intestinal bacteria. Prebiotics, such as herbal medicine, could lead to balance of intestinal bacteria or increase beneficial bacteria. So, proper choice of herbal medicine could control the intestinal condition. This study aimed to investigate the effect of mixture of probiotics and prebiotics (synbiotics) on dextran sulfate sodium (DSS)-induced UC in vivo. The synbiotics consist of Lactobacillus buchneri, Polymnia sonchifolia and Glycine max Merr. in this study. To evaluate the effect of synbiotics, 3% DSS was administered in BALB/c mice and synbiotics was daily administered for experimental days. The administration of synbiotics regulated colon length shortening, body weight change and disease activity index effectively. Also, extract of synbiotics upregulated survival ability of Lactobacillus buchneri in gut condition. These results suggest that mixture of probiotics and prebiotics, called as synbiotics, could influence intestinal condition also regulate the colon disease. Synbiotics might be a therapeutic agent for treatment of UC.

High plant regeneration and ectopic expression of OsMADS1 gene in root chicory (Cichorium intybus L. var. sativus)

  • Optimal shoot regeneration and transformation conditions of root type chicory (Cichorium intybus L. var. sativus cv Cesare) were studied. Leaf explants were co-cultured with Agrobacterium tumefaciens, which contained NPTII as a selectable marker and a rice homeotic gene, OsMADS1, that encodes a MADS-domain-containing transcription factor. After one day of co-cultivation, explants were transferred to selection media consisting of MS basal medium supplemented with 0.5 mg/L BAP, 0.1 mg/L IAA, 70 mg/L kanamycin, and 250 mg/L cefotaxime. PCR and Southern blot analyses revealed stable integration of the OsMADS1 gene in the chicory genome. Four-teen original transgenic plants ($T_o$ plants) were acclimatized in the greenhouse and examined for their morphological characters. Most of the transgenic plants showed altered morphologies, such as short, bushy, and early-flowering phenotypes with reduced apical dominance. Additionally, half of the transgenic plants exhibited altered leaf shapes, and 4 out of 14 plants were sterile. These phenotypes were inherited by the next generation. Northern blot analysis confirmed expression of the OsMADS1 gene in both floral and vegetative organs.

Anti-Oxidant and Anti-Inflammatory Complex Health Functional Analysis of Chinese Resource Plant Boehmeria, Carlemannia and Clausena (중국 자원식물 보메리아, 칼라마니아, 클로세나의 항노화 항염 복합 건강관리 기능성 연구)

  • Ageing has been known to be deeply related to oxidative activities. Furthermore, inflammation is a response initiated by conditions such as infection and injury. It has been known that complex connections occur between oxidative stress and the inflammatory response, however underlying mechanism needs to be investigated. Chinese resource plants have been widely used as functional food and medicine for a long time, however it is not studied well how many of these resource plants work. We first decided testing anti-oxidant and anti-inflammatory activities with 95% ethanol extracts of the chinese resource plants, Boehmeria pilosinscula (Blume) Hassk., Carlemannia tetragona Hook f. and Clausena emarginata C.C.Huang. To measure anti-oxidant activity, we performed DPPH assay in Raw264.7 cells with 95% ethanol extracts. Clausena extracts showed significantly higher anti-oxidant activity than those of Boehmeria and Carlemannia. We then performed Nitric Oxide assay to measure the inflammation suppression levels with 95% ethanol extracts of these plants. Clausena extracts showed significantly higher suppression of nitric oxide production than those of Boehmeria and Carlemannia. This indicates that inflammation levels are significantly reduced by Clausena. After measuring anti-oxidant and anti-inflammatory activities, we then performed MTT assay to measure the cell survival rate of Raw264.7 cells treated with these extracts. Boehmeria showed much more cell survival rate than Carlemannia and Clausena. Taken together, this result suggests that Clausena extracts have more anti-oxidant and antiinflammatory activities than Boehmeria and Carlemannia, while Boehmeria extracts have more cell survival rate than Carlemannia and Clausena.

Survey on Wild Edible Plant Resources in Korea and Its Germination Characteristics (우리나라 야생식용 자원식물의 종류 및 발아 특성에 관한 연구)

  • This study was conducted to obtain the basic information of edible resources among plant species. Potentials of resource plants were important in various usages, healthy food, medicinal materials, and breeding materials. Through our investigation 609 species of resources plants were considered as edible resources. These species belong to 74 families in which Asteraceae, Poaceae, Fabaceae, and Rosaceae were major families. Brassicaceae, Liliaceae, and Asteraceae contained large numbers of edible plants because plants belonged to these families were mainly used as vegetable. But Cyperaceae and Poaceae had relatively small numbers of species in the category of edible plant because plants belonged to these families have not been used directly as vegetables, common usage of edible plant. But Poaceae have a potentials as genetic donor for resistance-breeding strategies in major cereals. Although Poaceae can not be used as food directly, Poaceae should be investigated intensely in future for improvement of major cereals or application as fodder plants. The beneficial traits of edible plants as breeding materials have not been studied yet and development of new crop from plant resources requires the information about current situation in occurrence of resources plants genetically related to current crop species. Our results do not cover all plant species in Korea but this classification and identification about edible resources will provide primary information about plant resources. The collected seeds of resource plants showed wide spectrum in germination rate among plant species. The germination rate would probably be affected by collecting times, collected stage, and stored period. The proper methods about improving germination rate have to be elucidated to propagate the resource plants.

Overexpression of a Pathogenesis-Related Protein 10 Enhances Biotic and Abiotic Stress Tolerance in Rice

  • Pathogenesis-related proteins play multiple roles in plant development and biotic and abiotic stress tolerance. Here, we characterize a rice defense related gene named "jasmonic acid inducible pathogenesis-related class 10" (JIOsPR10) to gain an insight into its functional properties. Semi-quantitative RT-PCR analysis showed up-regulation of JIOsPR10 under salt and drought stress conditions. Constitutive over-expression JIOsPR10 in rice promoted shoot and root development in transgenic plants, however, their productivity was unaltered. Further experiments exhibited that the transgenic plants showed reduced susceptibility to rice blast fungus, and enhanced salt and drought stress tolerance as compared to the wild type. A comparative proteomic profiling of wild type and transgenic plants showed that overexpression of JIOsPR10 led to the differential modulation of several proteins mainly related with oxidative stresses, carbohydrate metabolism, and plant defense. Taken together, our findings suggest that JIOsPR10 plays important roles in biotic and abiotic stresses tolerance probably by activation of stress related proteins.

Floristic Study of County Pyeong-chang and Yeong-wol including Limestone Regions (Prov. Gangwon-do) from Korea (평창, 영월(강원도)의 석회암 지역 식물상)

  • Located in the middle part of the Korean peninsula, Pyeong-chang and Yeong-wol (Gangwon-do) areas are limestone regions that are important biogeographically showing the peculiar flora. In order to manage efficiently endangered and rare plant species, we have investigated flora of these above regions from March to October, 2010. The result of the investigation showed a total of vascular plants of 1,145 taxa; 134 families, 503 genera, and 1,010 species, 11 subspecies, 110 varieties, 10 forms and four hybrids. Among taxa investigated, endangered species designated by the law called 'Protection Law for Endangered wild fauna and flora' were seven taxa, and 50 taxa were considered to be endemic to Korea. Based on the floristic degrees categorized by the specific distribution of plant species, 24 taxa revealed the floristic degree (FD) V, 30 for FD IV, and 60 for FD III, 45 for FD II and 79 taxa for FD I. The 82 taxa were identified to be naturalized. Of those species investigated in this survey, 53 taxa were identified as specific ones biogeographically as compared to flora of other terrestrial regions; 35 of calcicolous plants, 15 with distribution in the northern part of the Korean peninsula, three with coastal habitats.
