미생물 유래의 Epoxide Hydrolase를 이용한 Chiral Styrene Oxide 생산용 비대칭 광학분할시스템개발 (Development of Asymmetric Resolution System for the Production of Chiral Styrene Oxide by Microbial Epoxide Hydrolase)

  • Apergillus niger LK의 epoxide hydrolase 활성을 이용하여 chiral styrene oxide를 제조할 수 있는 hollow-fiber 반응기 기반의 비대칭 분할 시스템을 개발하였다. 라세믹 styrene oxide 기질을 dodecane 유기용매에 용해시켜 hollow-fiber 반응기의 lumen 부위로 공급하였으며, 생촉매인 A. niger LK 미세분말은 shell 부위로 공급함으로써 막 표면에서 비대칭 분할 반응을 수행하였다. 반응 산물로 생성되는 phenyl-1,2-ethandiol에 의한 epoxide hydrolase 활성 저해효과를 감소시키기 위하여 2번째 hollow-fiber 반응기에서 완충용액을 이용하여 diol을 추출하여 제거시켰다. 2성분 용매를 사용한 cascade형 hollow-fiber 반응기 시스템을 이용하여 광학적으로 순수한 (ee > 99%) (5)-styrene oxide를 19.5% (이론 수율 대비 39%)의 수율로 얻을 수 있었다.

Design and performance prediction of large-area hybrid gamma imaging system (LAHGIS) for localization of low-level radioactive material

  • In the present study, a large-area hybrid gamma imaging system was designed by adopting coded aperture imaging on the basis of a large-area Compton camera to achieve high imaging performance throughout a broad energy range (100-2000 keV). The system consisting of a tungsten coded aperture mask and monolithic NaI(Tl) scintillation detectors was designed through a series of Geant4 Monte Carlo radiation transport simulations, in consideration of both imaging sensitivity and imaging resolution. Then, the performance of the system was predicted by Geant4 Monte Carlo simulations for point sources under various conditions. Our simulation results show that the system provides very high imaging sensitivity (i.e., low values for minimum detectable activity, MDA), thus allowing for imaging of low-activity sources at distances impossible with coded aperture imaging or Compton imaging alone. In addition, the imaging resolution of the system was found to be high (i.e., around 6°) over the broad energy range of 59.5-1330 keV.

  • V711 Tau의 채층활동을 조사하기 위하여 자외선 영역의 Mg II 방출선의 세기와 광도 변화의 상관 관계를 유추하였다. 첫째, 가시광선 영역의 광도곡선의 형태와 자외선 영역의 광도곡선의 형태를 비교하고, 둘째, IUE 고분산 스펙트럼에서 Mg II 방출선의 세기를 위상별로 측정하고 방출선 부근의 연속선을 이용하여 자외선 광도곡선을 완성하였다. 방출선의 세기와 광도곡선을 비교한 결과 방출선이 강하게 나타나는 위상, $0.^{p}4$에서는 광도가 어둡게 나타나 최소가 되고, 방출선의 세기가 약할 때는 광도가 밝아져서 최대가 되었다. 이는 자기장이 강한 위상에서는 채층활동과 흑점의 크기가 증가하고 반대로 자기장이 약한 위상에서는 채층활동과 흑점의 크기가 작다는 것을 뜻한다. 그러므로 자기장, 채층활동, 별 표면의 흑점과의 상관 관계가 밝혀지고, V711 Tau의 변광은 표면의 채층활동과 흑점에 의한 것이다.

Resolution of L-Carnitine from DL-Carnitine by Resting Cells of the Enterobacter sp. NH-104

  • For the resolution of L-carnitine from DL-carnitine, resting cells of Enterobacter sp. NH-104, which had a higher capacity of D-carnitine decomposition, were harvested at maximal specific activity of D-carnitine decomposition of 47.05 unit/mg cell. The cells were frozen at $-80^{\circ}C$ to assess functions as enzyme sources. Optimal concentration of cells and DL-carnitine were 17 g/$\ell \; and \; 20 g/\ell$, respectively, and reaction buffer was best at 75 mM of Tris. HCl. Optimal temperature and pH were $36^{\circ}C$ and 8.2, respectively. When the reaction at optimal conditions was carried out for 14 h, the optical purity was 98.21 %, and the quantity and yield of remaining L-carnitine were 4.432 g/$\ell$ and 44.32%, respectively.

  • 최근 베트남은 한국의 3대 교역국으로 부상하고 있다. 우리나라는 베트남과 교역규모가 크게 신장하고 있으며, 베트남에 대한 해외직접투자 역시 증가하고 있다. 한국과 베트남의 교역이 신장됨에 따라서 우리나라 기업과 베트남 기업 간의 분쟁 역시 증가할 소지가 크다. 본 연구는 첫째, 베트남의 문화와 제도적 특징을 분석하고자 한다. 둘째, 베트남의 고유한 분쟁해결방식 및 분쟁해결문화를 연구한다. 셋째, 베트남인과의 비즈니스에 있어서 협상전략을 연구한다. 지역연구방법론을 활용하여 연구한 결과 베트남은 여성 및 명예중시문화, 집단주의 문화적 특징을 보이는 것으로 나타났다. 베트남은 협상전략에 있어서 반띤 시스템 등, 중개인을 활용하는 간접적 의사소통방식을 선호하며, 합의에 의한 의사결정, 장기적 협상방식, ADR을 통한 분쟁해결 및 서면합의를 중시하고 있는 것으로 나타나고 있다.

  • 방향족 에폭사이드 기질에 대한 입체선택적 가수분해 활성이 우수한 Aspergillus niger LK를 이용하여 광학활성 (S)-para-nitrostyrene oxide를 제조하였다. 수용액에서 기질 의 낮은 용해도를 극복하기 위하여 유기용매에서 광학분할 반응을 수행하였으며, 생촉매 활성 저하가 가장 적은 dodecane을 유기용매로 선정하였다. 반응온도 3$0^{\circ}C$ 및 최적 water content 2% (v/v)의 조건에서 약 10시간 정도의 반응을 통해 ee 값이 100 %인 광학적으로 순수한 (S)-para-nitrostyrene oxide를 37% 정도(이론수율 : 50 %)의 높은 수율로 얻을 수 있었다.

Small-molecule probes elucidate global enzyme activity in a proteomic context

  • The recent dramatic improvements in high-resolution mass spectrometry (MS) have revolutionized the speed and scope of proteomic studies. Conventional MS-based proteomics methodologies allow global protein profiling based on expression levels. Although these techniques are promising, there are numerous biological activities yet to be unveiled, such as the dynamic regulation of enzyme activity. Chemical proteomics is an emerging field that extends these types proteomic profiling. In particular, activity-based protein profiling (ABPP) utilizes small-molecule probes to monitor enzyme activity directly in living intact subjects. In this mini-review, we summarize the unique roles of smallmolecule probes in proteomics studies and highlight some recent examples in which this principle has been applied.

Examining the star formation properties of Virgo galaxies undergoing ram pressure stripping

  • Understanding how ram pressure stripping (RPS) affects the star formation activity of cluster galaxies is one of the important issues in astrophysics. To examine whether we can identify any discernible trend in the star formation activity of galaxies undergoing ram pressure stripping, we study the star formation properties of galaxies in the Virgo cluster for which high-resolution HI images are available. We first classify galaxies in the Extended Virgo Cluster Catalog into different stages of RPS based on their HI morphology, HI deficiency, and location in phase space. We then examine various star formation activity indicators of these galaxies, which include starburstiness, g - r color, and WISE [3.4]-[12] color. No noticeable enhancement in star formation was identified for galaxies undergoing early or active stripping. Our results suggest that star formation activity at best seems to be enhanced locally in such galaxies, making it challenging to detect with integrated photometry. With the combination of HI deficiencies and locations in phase space, we were instead able to capture the overall quenching of star formation activity with increasing degree of ram pressure stripping, which agree with previous studies.

The Role of Contradictions in the Development of Technology-Supported Constructivist Classroom Practices: A Cultural-Historical Activity Theory Perspective

  • The notion of contradiction from Cultural-Historical Activity Theory (CHAT) perspectives is known as an "engine" for the development of human practices because participants attempt to adjust their practices to resolve contractions. This study examines two middle school teachers' classroom practices from CHAT, focusing on the role of contradictions that emerged between their existing teaching practices and constructivist activities in the development of a student-centered technology-integrated learning environment. Findings indicated that teachers' awareness and resolution of contradictions played a large role in the development of a technology-supported student-centered learning environment, a culturally more advanced activity system: students displayed greater responsibilities for their learning and were guided to make effective decisions for their learning activity.
