Chitosan Oligosaccharides Inhibit Adipogenesis in 3T3-L1 Adipocytes

  • The 3T3-L1 cell line is a well-established and commonly used in vitro model to assess adipocyte differentiation. Over the course of several days, confluent 3T3-L1 cells can be converted to adipocytes in the presence of an adipogenic cocktail. In this study, the effects of chitosan oligosaccharides (CO) on adipocyte differentiation of 3T3-L1 cells were studied. The CO significantly decreased lipid accumulation, a marker of adipogenesis, in a dose-dependent manner. The low molecular mass CO (1-3 kDa) were the most effective at inhibiting adipocyte differentiation. Moreover, mRNA expression levels of both CCAAT/enhancer-binding protein (C/EBP) ${\alpha}$ and peroxisome proliferator-activated receptor (PPAR) ${\gamma}$, the key adipogenic transcription factors, were markedly decreased by CO treatments. CO also significantly down regulated adipogenic marker proteins such as leptin, adiponectin, and resistin. Our results suggest a role for CO as antiobesity agents by inhibiting adipocyte differentiation mediated through the down regulated expression of adipogenic transcription factors and other specific genes.

  • Purpose: The Bio-energy metabolism control hormone by Adipocytokine is composed with Leptin, Adiponectin, resistin, TNF-a, IL-6. Adiponectin become known to participating in Insulin sensitivity exasperation, Fat metabolism accomodation and inducing metabolic disease such as diabetes mellitus, arteriosclerosis, hyperlipemia. When we accomplished the test with purpose of Research, we observed significance with a result of test related to diabetes mellitus with reference setting by way of suggestion. Methods: Result of normal group (n=100) which is committed in the SCL from September 2006 to December 2006 and result of control group (n=50) relationship examination item that is decided diabetes measures themselves against each other. Also, we measured the normal group against the control group for the reference range of adiponectin. Results: Result in normal group (n=100) appeared by Glucose (reference 70~120 mg/dl) Mean and the SD 96.99 (${\pm}24.35$), HbA1c (reference 4.0~6.0%) Mean and the SD 5.64 (${\pm}0.90$), Insulin (reference 2.0~25.0 uIU/ml) Mean and the SD 7.80 (${\pm}4.42$), the Adiponectin the Mean and the SD 9,861.23 (${\pm}4,977.0$). Result in control group(n=50) appeared by Glucose (reference 70-120 mg/dl) Mean and the SD 224.95 (5.30), the HbA1c (reference 4.0~6.0%) Mean and the SD 8.22 (1.63), Insulin (reference 2.0~25.0 uIU/ml) Mean and the SD 17.02 (3.01), C-peptide (reference 0.48~3.30 ng/ml) Mean and the SD 7.92 (${\pm}7.40$), the Adiponectin Mean and the SD 18,110.03 (${\pm}12,843.29$). Conclusions: Therefore, it seems that test results are significant and we consider that it can be apply to useful diagnosis of diabetes mellitus, arteriosclerosis, hyperlipemia patients throughout the reference range setting of Adiponectin, Leptin is one of the Bio-energy metabolism control hormone.

Effects of herbal mixture on blood glucose and lipid metabolism in type 2 diabetic mellitus mouse (천연물 복합제가 제 2형 당뇨 마우스에서 혈당 및 지질대사에 미치는 영향)

  • Objectives: This study aims to investigate the effects of diabetes mellitus care mixture (DCM) on blood glucose and lipid metabolism in type 2 diabetic mellitus mice. DCM consisted of lagerstroemia speciose, allium hookeri, momordica charantia, amaranthus tricolor, and boesenbergia rotunda, which have been proven to have antidiabetic properties. Methods: In this study, we researched the effects of DCM in type 2 diabetic mellitus mice. C57BLKS/J mouse groups had no treatment, db/db mouse randomly assigned to 2 groups, and treated with distilled water and DCM (200 mg/kg/day). Blood glucose levels and body weight were checked every week. After 4 weeks of treatment, liver function indicators (AST, ALT, and LDH) and lipid metabolites (triglyceride, total cholesterol, LDL-cholesterol, HDL-cholesterol) were measured with a biochemistry analyzer. Diabetic factors (insulin, resistin, and leptin) were measured with ELISA. Results: DCM was decreased blood glucose, diabetic factors, liver function indicators, triglyceride, total cholesterol, and LDL-cholesterol significantly. Also, HDL-cholesterol was significantly increased in DCM group. The bodyweight of DCM group decreased but, no significant difference with the control group. DCM may have the potential to improved diabetes mellitus by regulating blood glucose levels and diabetic factors. Also protecting from diabetic complications by adjusting liver function indicators and lipid metabolites. Conclusions: These results suggest that DCM to be used as an oriental medicine for diabetes, the results of clinical trials are needed.

Differential Expression of Adipocyte-Related Molecules in the Distal Epididymal Fat of Mouse during Postnatal Period

  • The epididymal fat of mouse is a part of visceral fat deposit and is divided into the distal or proximal part based on its histochemical characteristics. Even though the formation of the epididymal fat pad begins before the birth, a detailed adipogenic procedure of the epididymal fat has not been revealed. The epididymal fat pad becomes enlarged and expended with age, and expressional changes of numerous genes are associated with the maturation of fat tissues. In the present research, expressional patterns of adipose tissue-related genes in the distal epididymal fat of mouse at 2, 5, 8, and 12 months of postnatal age were determined by a quantitative real-time polymerase chain reaction (PCR) analysis. The lowest transcript levels of fatty acid binding protein 4 (Fabp4), lipoprotein lipase (Lpl), delta like non-canonical Notch ligand 1 (Dlk1), peroxisome proliferator-activated receptor gamma (Pparg), leptin (Lep), adiponectin (Adipoq), and resistin (Retn) were detected at 2 months of age, except fatty acid synthase (Fasn) showing the lowest level at 5 months of age. Even though expression of Lep and Fabp4 were gradually increased until 12 months of age, significant increases of Pparg and Adipoq transcript levels were continued until 8 months of age. The transcript levels of Lpl, Rent, Dlk1, and Fasn were significantly increased at 8 months of age, compared with those at 2 months of age. The current findings suggest that the expansion of the distal epididymal fat of mouse during postnatal period would be companied with differential expression of various adipocyte-associated molecules.

Assessment of Adipocyte Differentiation and Maturation-related Gene Expression in the Epididymal Fat of Estrogen Receptor α Knockout (ERαKO) Mouse during Postnatal Development Period

  • The absence of functional estrogen receptor α (Esr1) results in an overgrowth of the epididymal fat, as observed in estrogen receptor α knockout (ERαKO) mouse. The present research was aimed to evaluate expression of various molecules associated with adipocyte differentiation and maturation in the epididymal fat of ERαKO mouse at several postnatal ages by using quantitative real-time polymerase chain reaction. The highest transcript levels of all molecules were detected at 12 months of postnatal age, except leptin which the mRNA level was increased at 5 months of age and was unchanged until 12 months of age. The expression levels of CCAAT enhancer binding protein (Cebp) alpha, androgen receptor, and lipoprotein lipase were decreased at 5 months of age but increased at about 8 months of age. The mRNA levels of Cebp gamma and sterol regulatory element binding transcription factor 1 remained steady until 8 months of age. Continuous increases of transcript levels during postnatal period were found in Cebp beta, estrogen receptor (ER) beta, fatty acid binding protein 4, and delta like non-canonical Notch ligand 1. The increases of peroxisome proliferator-activated receptor gamma and adiponectin mRNA levels were detected as early as 8 months of age. The levels of fatty acid synthase and resistin transcript at 5 and 8 months of age were lower than that at 2 months of age. These findings show the aberrant expression patterns of genes related to adipocyte differentiation and maturation in the postnatal epididymal fat pad by the disruption of ER alpha function.

Circulating Levels of Adipokines Predict the Occurrence of Acute Graft-versus-host Disease

  • Currently, detecting biochemical differences before and after allogeneic stem cell transplantation (SCT) for improved prediction of acute graft-versus-host disease (aGVHD) is a major clinical challenge. In this pilot study, we analyzed the kinetics of circulating adipokine levels in patients with or without aGVHD before and after allogeneic SCT. Serum samples were obtained and stored at $-80^{\circ}C$ within 3 hours after collection, prior to conditioning and at engraftment after transplantation. A protein array system was used to measure the levels of 7 adipokines of patients with aGVHD (n=20) and without aGVHD (n=20). The resistin level at engraftment was significantly increased (p<0.001) after transplantation, regardless of aGVHD occurrence. In the non-aGVHD group, the concentrations of the hepatocyte growth factor (HGF) (mean values${\pm}$SD; $206.6{\pm}34.3$ vs. $432.3{\pm}108.9pg/ml$, p=0.040) and angiopoietin-2 (ANG-2) (mean values${\pm}$SD; $3,197.2{\pm}328.3$ vs. $4,471.8{\pm}568.4pg/ml$, p=0.037) at engraftment were significantly higher than those of the pre-transplant period, whereas in the aGVHD group, the levels of adipokines did not change after transplantation. Our study suggests that changes in serum HGF and ANG-2 levels could be considered helpful markers for the subsequent occurrence of aGVHD.

Anti-obesity effect of Amomum taso-ko ethanol extract in 3T3-L1 adipocytes (3T3-L1 지방세포에서 초과 에탄올 추출물의 항비만 효과)

  • Amomum tsao-ko used as a traditional oriental herbal medicine, is indigenous to several Asia countries. In this study, we investigated anti-obesity activity of the ethanol extract of Amomum Taso-ko (A. tsao-ko). The ethanol extract of A. tsao-ko inhibited adipocyte differentiation using Oil Red O assay in 3T3-L1 cells. Inhibitory effect of the ethanol extract of A. tsao-ko on adipogenesis was modulated by down-regulation adipogenic transcriptional factor such as peroxisome proliferator-activated receptor ${\gamma}$ ($PPAR{\gamma}$), CCAAT-enhancer-binding protein ${\alpha}$ ($C/EBP{\alpha}$) and suppressed expression of fatty acid synthase, aP2, and resistin. We demonstrated that A. tsao-ko significantly inhibited adipogenesis and reduced $PPAR{\gamma}$ and $C/EBP{\alpha}$ expression in a dose-dependent manner. These results suggest that A. tsao-ko has an anti-obesity effect by inhibition of adipogenic transcription factor and adipocyte-specific genes in 3T3-L1 cells.

Anti-obesity Effects of Galgeun-tang in High Fat Diet Induced Obese Mice Model (갈근탕이 고지방 식이 유발 백서 모델에 미치는 항비만 효과)

  • Objectives To investigate anti-obesity effects of Galgeun-tang, an herbal formula, in high fat diet induced obese mice model. Methods 24 Male C57Bl/6J mice were randomly assigned to normal group fed with normal research diet (NOR, n=6), high fat diet control group treated with water (HFD, n=6), high fat diet group treated with Orlistat (ORL, n=6, Orlistat 10 mg/kg), and high fat diet group treated with Galgeun-tang (GGT, n=6, Galgeun-tang 700 mg/kg). 12 weeks later, body weight, fat weight, liver weight, blood glucose, total cholesterol, triglyceride, HDL, ALT, AST, obesity related neuropeptides and adipokines, ratio of gut microbiota, and histopathology of liver were evaluated. Results In the GGT group, 1. body weight gain, liver weight gain, and total fat weight gain were significantly less than those in the HFD group. 2. blood glucose level was significantly lower and insulin level was significantly higher than in the HFD group. 3. total cholesterol level and triglyceride (TG) level were significantly lower and high density lipoprotein (HDL) level was significantly higher than in the HFD group. 4. appetite-promoting ARC neuropeptides such as Agrp and Npy were significantly less and appetite-inhibiting ARC neuropeptide, Cart was significantly more than in the HFD group in qRT-PCR analysis. 5. adiponectin level and visfatin level were significantly higher, and resistin level and leptin level was significantly lower than in the HFD group. 6. the relative level of Bacteroidetes was significantly higher, and the relative level of Firmicutes was significantly lower than in the HFD group. 7. the increase of adipose tissue was significantly more inhibited than in the HFD group. Conclusions The present study showed that Glageun-tang exerts anti-obesity effects in that it. 1. inhibited the increase in body weight, liver weight, and total fat weight. 2. decreased the level of TG, and increased the level of HDL. 3. influenced neuropeptides and adipokines that are important in regulating food intake and changes of body weight. 4. modified the beneficial quantitative changes in gut microbiota suppressing the tendency toward obesity.

Synergic Effect on Adipocytic Differentiation by a Combination of Thyroxine andGlucocorticoid in A549 Cells (A549 폐암세포주의 지방세포 분화에 미치는 티록신 및 당질 코르티코이드 호르몬의 상승 효과)

  • The present study examined the rate of cell growth and differentiation potential into adipocytes in A549 lung adenocarcinoma cells exposed to each adipogenic medium containing glucose metabolism hormones, such as thyroxine (T4) thyroid hormone and glucocorticoid (GC) adrenal steroid hormone, as well as pioglitazone (PGZ), a PPARγ agonist. Following each adipogenic treatment for 2 weeks, the rate of cell growth was significantly (p<0.05) inhibited, and the level of telomerase activity was significantly (p<0.05) decreased in the PGZ-based adipogenic medium containing both T4 and GC hormone compared with those containing each T4 or GC hormone. Moreover, the adiposome-like vesicles were highly reacted with Oil-Red O staining solution, and the levels of transcripts expressed in the differentiating adipocytes for adipogenesis, including adinopectin, leptin, and resistin, were significantly (p<0.05) increased in the PGZ-based adipogenic medium containing both T4 and GC hormone compared with those of the adipogenic medium containing each T4 or GC hormone, implying that adipocytic differentiation has fully occurred in the A549 cancer cells. Based on present observations, the PGZ-based adipogenic medium containing both T4 and GC efficiently induces inhibition of cell growth and cellular differentiation into adipocytes in A549 cancer cells rather than in the adipogenic medium containing only T4 or GC hormone. Adipogenic treatment could provide potential probability in cancer chemotherapy.

Antiadipogenic Effect of Korean Glasswort (Salicornia herbacea L.) Water Extract on 3T3-L1 Adipocytes (세포배양 모델을 통한 함초(Salicornia herbacea L.) 물 추출물의 항비만 효과 탐색)

  • As a natural plant ingredients, glasswort (Salicornia herbacea L.) contains various physiological activities, mainly anti-oxidative and anti-diabetic activities. However, only a few studies have been carried out on its anti-adipogenic effect. This study investigated the anti-obesity effects of Salicornia herbacea L. on 3T3-L1 adipocytes. As adipogenesis of preadipocytes to adipocytes involves proliferation and differentiation of cells, we treated three concentrations (125, 250, and $500{\mu}g/mL$) of Salicornia herbacea L. water extracts (SLW) in both pre-processing and post-processing stages. When 3T3-L1 adipocytes were differentiated and dyed with Oil Red O, adipocytes size and the value of relative Oil Red O staining were reduced by all concentrations of SLW in post-processing stage. Following adipogenic differentiation, the concentration of tumor necrosis factor-${\alpha}$ (TNF-${\alpha}$) in the cell supernatant significantly increased upon treatment with $125{\mu}g/mL$ of SLW and further rose at concentrations of 250 and $500{\mu}g/mL$ during post-processing stage. There was no significant difference in glycerol production upon SLW treatment. Leptin production significantly decreased at all SLW concentrations during post-processing stage, whereas peroxisome proliferator activated receptor-${\gamma}$ (PPAR-${\gamma}$) and adiponectin secretions were significantly enhanced. Overall results showed that SLW might have an anti-adipogenic effect via enhancement of TNF-${\alpha}$ production, which causes dedifferentiation and inhibits lipid accumulations in adipocyte. Furthermore, SLW might prevent diabetes and cardiovascular disease, as it reduces leptin secretion and enhances production of both PPAR-${\gamma}$ and adiponectin. However, further research is needed to elucidate the exact mechanism and bioactive compounds of glasswort.
