Isolation and Properties of Cytotoxic Antibiotics Produced by Myxococcus stipitatus JW150 (Myxococcus stipitatus JW150이 생산하는 세포독성 물질의 분리 및 특성)

  • Drug resistance is one of the most significant impediments to successful chemotherapy of cancer. Multidrug-resistance (MDR) is characterized by decreased cellular sensitivity to anticancer agents due to the overexpression of P-glycoprotein. By employing a resistant subline of HCT15 to adriamycin (CL02), we undertook the screening for agents which were effective to multidrug-resistant cancer cells. As a result, a myxobacterial strain JW150 was selected for study since an activity against CL02 cells was discovered in the strain. Cytotoxicity-guided fractionation of the culture broth led to the isolation of cystothiazole A and melithiazole F. The producing organism was identified as Myxococcus stipitatus by taxonomic comparison with type strains of Myxococcus sp. as well as its morphological and physiological characteristics. Cystothiazole A and melithiazole F demonstrated potent cytotoxicity against certain human cancer cells with $IC_{50}$ values ranging from 0.03~ $0.72{\mu}{\textrm{g}}$/ml. Both compounds were interestingly as active against drug-resistant sublines CL02 and CP70 as against the corresponding parental cells.

  • Antibiotic resistance patterns of 21 antibiotics were studied for 50 strains of Staphylococcus aureus isolated from a hospital in Busan from July 2005 to December 2006. All strains showed antibiotic resistance to more than one antibiotic and 3 strains showed resistance to 17 different antibiotics. The strains isolated between 2005 and 2006 had lower resistance rate to 12 antibiotics (other than vancomycin and ampicilin) than the strains isolated between 1989 and 1990. In particular, no chlorarmphenicol resistant strain was found in this study which is contrasted with 34.8% resistant rate obtained in the study conducted between 1989 and 1990. In respect of vancomycin, no resistant strain was found in this study which is the same result obtained in the 1989 to 1990 study; All strains investigated in this study showed 100% resistance rate to ampicillin compared to 69.6% in the previous study.

  • Two soybean cultivars, Kwanggyo and Hwanggeum (soybean mosaic potyvirus (SMV)-resistant cultivars), that had been inoculated with a virulent strain (G-5H, 4) of soybean mosaic potyvirus produced necrotic lesions on inoculated leaves as well as on upper trifoliate leaves. Cells in the lesion area contained sparse numbers of virus particles and very few characteristic pinwheel inclusions. Although a hypersensitive-like cellular response occurred in the two resistant cultivars, this response did not prevent the virus from spreading systemically in these resistant hosts, indicating a different mechanism from the general hypersensitive reaction in relation to host resistance.

  • Colistin is one of the last-resort antibiotics used to treat multidrug-resistant Gram-negative bacterial infection in both human and animals. Here, we report the complete genome sequence of colistin-resistant Salmonella enterica serotype Enteritidis strain CRSE-01 isolated from poultry carcass in South Korea. The assembled genome consists of a 4,783,907-bp circular chromosome containing numerous antimicrobial resistance genes and a 59,372-bp plasmid.

  • We report the whole genome sequence of a linezolid-resistant and vancomycin-susceptible Enterococcus faecalis strain, ES-2-1, which was isolated from a pig stool in South Korea. The assembled genome of ES-2-1 consists of a 2,648,168-bp circular chromosome containing the optrA gene (encoding the ABC-F type ribosomal protection protein), an 84,891-bp plasmid containing numerous antimicrobial resistance genes, and an 82,106-bp cryptic plasmid. The ES-2-1 strain belongs to sequence type 1024 (ST1024) and carries multidrug resistant genes including the optrA (oxazolidinone phenicol transferable resistance A) gene, which confers linezolid resistance.

  • Methicillin-resistant Staphylococcus aureus (MRSA) represents antimicrobial-resistant bacteria that can cause a wide range of illnesses both in humans and animals. Multidrug resistance phenotype is common, especially in healthcare-associated (HA) MRSA strains. Currently, one of the most prevalent HA-MRSA clonal lineages in Korean hospitals is sequence type (ST) 5 carrying staphylococcal cassette chromosome mec type II (ST5-SCCmec II). Here, we report the complete genome sequence of an ST5 HA-MRSA strain (SA492) originated from a patient in Korea.

  • The neutralization epitopes of the outer capsid protein VP4 of a porcine rotavirus, Gottfried strain, were studied using neutralizing monocolonal antibodies(N-MAbs). Eight N-MAbs which are specific for the VP4 of Gottfried strain were used for analyzing the antigenic sites of VP4. Three different approaches were used for this analysis; i)testing the serological reactivity of each N-MAb against different G and P types of human and animal rotavirusese ii) analyzing N-MAb-resistant viral escape mutants and iii) performing nucleotide sequence analysis of the VP4 gene of each N-MAb-resistant viral escape mutant. From experimental results, at least four antigenic sites(I, II, III, and IV) were identified. Antigenic site I recognized by N-MAbs 24B9, 23G10, and 26A2 was separated from antigenic site II recognized by N-MAbs 30H5, 32B3, and 29B3. However, these antigenic sites were overlapped with antigenic site III recognized by N-MAb 21A1. The other antigenic site IV recognized by N-MAb 16D2 was separated from antigenic sites I, II, and III.

  • During the screening for the antimicrobial agents against multidrug-resistant Staphylococcus aureus, we isolated an active compound produced by strain CNU30122. The active compound was purified from culture broth by HP-20 column chromatography, ethylacetate extraction. silica gel column and Sephadex LH-20 column chromatographies and HPLC. Based on various NMR studies including $^1H-^1H\;COSY$, $^1H-^{13}C\;COSY$ and HMBC experiments. the active compound was identified as fusidic acid.

  • Recently, we isolated a multidrug-resistant Escherichia coli strain KBN10P04869 from a patient with acute myeloid leukemia. We report the complete genome of this strain which consists of 5,104,264 bp with 4,457 protein-coding genes, 88 tRNAs, and 22 rRNAs, and the co-occurrence of multidrug- resistant genes including $^{bla}CMY-2$, $^{bla}TEM-1$, $^{bla}CTX-M-15$, $^{bla}NDM-5$, and $^{bla}OXA-18$.

  • Research to obtain molecular markers related to the major histocompatibility complex (MHC) gene in both strains of gourami is essential to increase the success of the selection program of disease resistance traits. Using a completely randomized design (CRD), the challenge test consists of four treatments and seven replications. The treatment was Jambi gourami injected with PBS (KJ), Kalimantan gourami injected with PBS (KK), Jambi strain injected with Aeromonas hydrophila (GJ), and Kalimantan strain injected with A. hydrophila (GK). The GJ population was more resistant to A. hydrophila than the GK population. The MHC II gene was detected in both test strains (GJ and GK), both resistant and susceptible fish. However, there were differences in the results of amplifying the MHC II gene in susceptible and resistant fish. Two DNA fragments approximately 400 and 585 bp were detected in the genome of susceptible fish, while in the genome of susceptible fish, only one DNA fragment was detected (400 bp). Therefore, the MHC II gene fragment with a size of about 585 bp can be used as a potential candidate for specific molecular markers to obtain resistance to A. hydrophila bacteria in the giant gourami.
