On the Influence of Susceptible and Resistant Soybean Varieties to the Development of the Soybean Cyst Nematode, Heterodera glycines (Heteroderidae, Nematoda) (저항성 및 감수성 콩 품종이 콩씨스트선충 (Heterodera glycines)의 발육에 미치는 영향에 관한 연구)

  • Influence of susceptible and resistant soybean varieties to the development of the soybean cyst nematode(Heterodera glycines) was examined. It does not seem likly that there are any differences between varieties in attracting the Second Stage larvae of the nematode. In susceptible varieties, there were large populations and three generations during a ten·week examination period, while the resistant varieties maintained very low populations and had only two generations.

  • Domestic rail industry for the development of Korea High Speed Rail paid for high technology, imported to Korea. However, If technology based on conditions that are not equipped with relevant skills is received, there are limits to what it used to. so team of high speed railway began technical development. In this study, through the motor block resistant plan, the current block resistant identifies problems related to it, in order to improve the quality of the parts, secure the standards and guarantee their life span. The localization of the resistant is really possible thanks to the technical skills.

Screening and Histopathological Characterization of Korean Carrot Lines for Resistance to the Root-Knot Nematode Meloidogyne incognita

  • In total, 170 carrot lines developed in Korea were screened for resistance to Meloidogyne incognita race 1 to select parental genetic resources useful for the development of nematode-resistant carrot cultivars. Using the gall index (GI), gall formation was examined on carrot roots inoculated with approximately 1,000 second-stage juveniles of the nematode 7 weeks after inoculation. Sixty-one carrot lines were resistant (GI ${\leq}1.0$), while the other 109 were susceptible (GI > 1.0) with coefficient of variance (CV) of GI for total carrot lines 0.68, indicating low-variation of GI within the lines examined. The histopathological responses of two carrot plants from resistant and susceptible lines were examined after nematode infection. In susceptible carrots, giant cells formed with no discernible necrosis around the infecting nematodes. In the resistant carrot line, however, no giant cells formed, although modified cells were observed with extensive formation of necrotic layers through their middle lamella and around the infecting nematodes. This suggested that these structural modifications were related to hypersensitive responses governed by the expression of true resistance genes. Therefore, the Korean carrot lines resistant to the nematode infection are potential genetic resources for the development of quality carrot cultivars resistant to M. incognita race 1.

Study of Crease Resistant Finish on Hemp Fabrics(Andongpo) (대마직물(안동포)의 방추가공에 관한 연구)

  • Andongpo, 100% Korean hemp fabric was treated with the glyoxale resin type finishing agent and/or the soluble urethane type finishing agent to determine the optimum process condition of the crease resistant finish and the crease recovery of treated sample fabrics was evaluated for the study. The treatment conditions for the study were 6 conditions, such as, A-1~A-6, in which A-1 was the condition of treatment glyoxale resin type finishing agent only and A-2~A-6 were the condition of treatment both glyoxale resin type finishing agent and soluble urethane type finishing agent. Among the 6 conditions, the crease recovery of the sample treated with A-4 condition was $148^{\circ}$(angle of recovery method) and grade 3.2(appearance method) and so, these samples showed the excellent crease recovery. From the result, 15g/l of the catalyst conc., 50g/l of the glyoxale resin type finishing agent cone., and 40g/l of the softner were the optimum treatment condition for the crease resistant finish of the andongpo.

  • Recently, antibiotic resistant genes (ARGs) in the environment are emerging as pollutants, since these genetic contaminants can eventually be transferred to human pathogens. The aim of this study was to develop the experimental model of antibiotic resistant gene (ARG) plasmid transfer as a function of various environmental conditions. For this purpose, the multi drug resistant plasmid pB10, which is known to be originally isolated from a wastewater treatment plant, was selected as a model transfer plasmid and Escherichia coli $DH5{\alpha}$ containing pB10 was used as a model donor. Pseudomonas aeruginosa, an opportunistic pathogen, was selected as the recipient for the conjugation experiment. When the donor and recipient were exposed to various stressors including antibiotics and heavy metal as a function of the concentrations (10, 100 and, 1000 ppb), statistically increased plasmid transfer rate was observed at a concentration of 10 ppb of tetracycline and sulfamethoxazole compared to control (no antibiotic exposure). Accordingly, the developed experimental ARG model by various stressor is a promising tool for evaluating the dissemination of ARGs by micro-contaminants in aquatic environment.

Evaluation of leaf rust resistance and characteristics of Korean wheats

  • Leaf rust is the most widespread and destructive fungal disease, and outbreaks have always caused considerable losses in wheat yields. Thus, worldwide increases in wheat production depend on the development of leaf rust-resistant wheat varieties. In this study, we evaluated the resistance of forty Korean wheat cultivars to leaf rust at the seedling stage. Only two Korean wheats, Ol and Jonong, were resistant to leaf rust, whereas the remaining thirty-eight Korean wheats were susceptible to leaf rust. The Ol and Jonong varieties presented larger dry seed weights and higher antioxidant activity in response to leaf rust than the susceptible wheat varieties. No differences in ${\beta}$-1,3-glucanase activity or chlorophyll content between resistant and susceptible wheat varieties were observed. Overall, these results are important for the development of wheat varieties that are highly resistant to leaf rust and to understand the underlying mechanisms that confer leaf rust resistance.

  • This study was conducted to investigate the herbicidal activity against Scirpus planiculmis (SCPPL) which has been reported as resistant biotype on sulfonylurea (SU) herbicides at the west coast reclaimed area first and proliferated continuously at Hwasung and Pyeongtaek in Gyeonggi province, and some Chungnam province. In order to investigate resistance breakability against SCPPL, new rice herbicides containing tefuryltrione [p-hydrophenyl pyruvate dioxygenase (HPPD) inhibitor], pyrimisulfan and triafamone[actolactate synthethase (ALS) inhibitor] were tested. In both lab and field experiments, tefuryltrione GR, 4-HPPD inhibitor showed growth retardation with a bleaching on the young leaves of SCPPL at early time after treatment. However, it restored and regerminated with insufficient control effect. On the other hand, pyrimisulfan+mefenacet SC, triafamone+tefuryltrione GR, and ALS inhibitor showed excellent control effect on SU-resistant SCPPL with growth retardation and necrosis. In particular, pyrimisulfan+mefenacet SC showed excellent controlling effect on SU-resistant SCPPL in regional field experiments.

  • Heat resistant putrefactive microorganisms causing spoilage of canned and processed foods were surveyed in the compost on mushroom growing bed, casing soil, raw mushrooms and canned products before sterilization at canneries located at 8 places including Buyo in Chung-Cheung-Do and monitored the total count and spore formers from the sample taken. The 9 strains of most severe heat resistant among the selected 140 spore formers were selected and determined D and Z value by TDT method. The most strong heat resistant strain was No. F-10, facultative thermophile, which was isolated from raw mushroom in Buyo area and it's Z value was $21.1^{\circ}F$ (M/15 phosphate buffer solution) and $D^{250}$ was 6.6 min.

Acute Toxicity Evaluation to Daphnia magna of Disease Resistant(OsCK1) Rice (병저항성 GM(OsCK1)벼의 물벼룩(Daphnia magna)에 대한 급성독성 평가)

  • BACKGROUND: The disease resistant (OsCK1) rice was generated by inserting choline kinase (CK1) and phosphinothricin acetyltransferase (PAT) genes isolated from Oryza sativa and Streptomyces hygroscopicus into the genome of the rice, Nakdongbyeo. With the potential problems of safeties, the evaluations on non-target organisms are essentially required for the environmental risk assessment of genetically modified (GM) crops. In the present study, we conducted the evaluation of acute toxicity on Daphnia magna that commonly used as a model organism in ecotoxicological studies for non-target organism evaluation. METHODS AND RESULTS: Effect of acute toxicity to Daphnia magna by each concentration were investigated in the disease resistant (OsCK1) rice and non-genetically modified (non-GM) rice, Nakdongbyeo, as concentration (0, 1,000, 1,800, 3,240, 5,830, 10,500 and 20,000 mg/L). The OsCK1 rice used for the test was confirmed to express the OsCK1/PAT gene by the PCR(Polymerase chain reaction) and western blot analysis. Feeding test showed that no significant differences in cumulative immobility and abnormal response of Daphnia magna fed on OsCK1 rice or non-GM rice. The 48hr-$EC_{50}$ values showed no difference between OsCK1 rice (3,147.18 mg/L) and non-GM rice (3,596.27 mg/L). CONCLUSION: This result suggested that there was no significant difference in toxicity to Daphnia magna between OsCK1 rice and non-GM counterpart.
