Copper is an essential transition metal involving in various biogeochemical processes. With the recent advances in analytical techniques and mass spectrometry, such as MC-ICP-MS, it is possible to measure Cu isotopes, which allows us to understand various biogeochemical processes in detail. Nonetheless, few studies have been performed in South Korea. In this study, we compared two purification methods previously reported using an anion exchange resin ($AG^{(R)}$ MP-1M), developed the best method in our lab environment, and then verified it by measuring Cu isotopic compositions in two USGS geological reference materials (BHVO-2 and BIR-1a). Although all matrix cations causing mass bias were effectively removed through both two methods with the yield of better than 95%, the method using the mixture of HCl and $H_2O_2$ only displays Cu isotopic compositions, in excellent agreement with reported values within the error. The method developed in this study is expected to be commonly applied to earth and environmental sciences.

The great economic losses caused by Cucumber mosaic virus (CMV) infection of peppers has led to the development of genetically modified (GM) CMV-resistant peppers. We developed virus-resistant pepper plants using Agrobacterium tumefaciens -mediated transformation. The expressed recombinant protein was purified using nickel-nitrilotriacetic acid resin and immunoaffinity chromatography, and purity was assessed by sodium dodecyl sulfate polyacrylamide gel electrophoresis. Immunoblot analysis revealed the purified CMV coat protein (CMV-CP) had a molecular mass of 25 kDa. After in-gel digestion and desalting, the internal peptide fragments of CMV-CP were sequenced by matrix-assisted laser desorption/ionization-time of flight. Most GM pepper and Escherichia coli BL21 internal peptides had identical peptide sequences and contained 137 of 183 whole peptides in CMV-CP. A quantitative enzyme-linked immunosorbent assay was performed to detect CMV-resistant GM peppers. We also provide basic information about the expressed protein in GM peppers for further safety assessment. The contents of soluble protein and CMV-CP were measured in GM and control peppers cultivated in three different areas of Korea. Statistical significance in terms of cultivation areas, harvest times, generations, and plant tissue origin were determined based on a P value of 0.05. The highest amount of CMV-CP was detected at the seedling stage from plant grown in each region. T3 and T5 showed significantly different levels of CMV-CP from T4 in leaves in the whorl stage. No statistical differences were observed among GM peppers at different stages of maturity in any cultivation area. The results from this study contribute to the safety evaluation of newly designed CMV-resistant GM peppers and provide a standard against which to compare other virus-resistant GM peppers.

Carbon nanotubes (CNT) are considered as one of ideal nano-fillers in the field of composites with their excellent electrical, mechanical, and thermal properties. Therefore CNT composites are increasingly used in fabricating conductive materials, structural materials with high strength and low weight, and multifunctional materials. The main problem of the CNT composites is difficulty in the dispersion of CNT in the polymer matrix. In this study multi-walled carbon nanotubes (MWNT) were pretreated by the physical process utilizing a wrapping method. After the pretreatment polymer/MWNT nanocomposites were prepared by melt processing. The effect of functionalization MWNT by wrapping with styrene acrylonitrile (SAN) on the mechanical and electrical properties of acrylonitrile butadiene styrene resin (ABS)/MWNT composites was studied by comparing the properties of ABS mixed with the neat MWNT. Electrical and mechanical properties of ABS/MWNT nanocomposites were studied as a function of the functionalization and content of MWNT. The tensile strength of the ABS/MWNT nanocomposites increased, but the impact strength decreased. The polymer wrapping in ABS system has little effect on the improvement of electrical properties.

Heterogeneous cation exchange membrane(HCEM) was prepared with LLDPE(Linear Low Density Poly-ethylene) as binder, powdered cation exchange resins($diameter{\leq}149{\mu}m$) as ion-exchange material and glycerol as additive for electrodialysis and electrodeionization system. The weight ratio of (binder/ion exchange)/glycerol was (60%/40%)/5%, (55%/45%)/5%, (50%/50%)/5% and (40%/60%)/5%. The characterization of prepared HCEM was evaluated on mechanical, electrochemical, morphology and ion permeable properties. It was compared with commercial membrane. Electrochemical properties of HCEM of (50%/50% )/5% were very similar to value of IONPURE(commercial membrane), in which ion exchange capacity, ion transfer number and membrane resistance were to be 1.733meq/g, 0.96 and $16.08{\Omega}/cm^2$, respectively. Ion permeability of the membrane was better than that of IONPURE membrane. Compared with IONPURE membrane, the HCEM had a higher tensile strength and lower elongation and modulus, in which HCEM had tensile strength of $62.33kg/cm^2$, elongation of 87.42% and modulus of $658.53kg/cm^2$. The HCEM of (50%/50% )15% was optimum combination.
Mechanical properties and microleakage of two composites [conventional hybrid type DenFil (VERICOM Co., Anyang, Korea) / micro matrix hybrid type Esthet X (Dentsply Caulk, Milford, DE, U.S.A.)] were evaluated to assess whether variable light intensity curing is better than conventional curing technique. Curing was done for 40 seconds in two ways of 2 step soft-start technique and 5 step ramping technique. Three kinds of light intensities of 50, 100, $200{\;}mW/\textrm{cm}^2$ were initially used for 10, 20, 30 seconds each and the maximum intensity of $600 {\;}mW/\textrm{cm}^2$ was used for the rest of curing time in a soft-start curing tech nique. In a ramping technique, curing was done with the same initial intensities and the light intensity was increased 5 times with the same rate to the maximum intensity of $600{\;}mW/\textrm{cm}^2$. After determining conditions that showed no different mechanical properties with conventional technique, Esthet X composite was filled in a class V cavity, which dimension was $4{\times}3{\times}1.5{\;}mm$ and cured under those conditions. Microleakage was evaluated in two ways of dye penetration and maximum gap estimation through SEM observation. ANOVA and Spearman's rho test were used to confirm any statistical significance among groups. The results were as follows : 1 Several curing conditions of variable light intensities resulted in the similar mechanical properties with a conventional continuous curing technique, except conditions that start curing with an initial light intensity of $50{\;}mW/\textrm{cm}^2$. 2. Conventional and ramping techniques were better than soft-start technique in mechanical properties of microhardness and compressive strength. 3. Soft-start group that started curing with an initial light intensity of $100{\;}mW/\textrm{cm}^2$ for 10 seconds showed the least dye penetration. Soft-start group that started curing with an initial light intensity of $200{\;}mW/\textrm{cm}^2$ for 10 seconds showed the smallest marginal gap, if there was no difference among groups. 4. Soft-start technique resulted in better dye-proof margin than conventional technique(p=0.014) and ramping technique(p = 0.002). 5. There was a very low relationship(p=0.157) between the methods of dye penetration and marginal gap determination through SEM evaluation. From the results of this study, it was revealed that ramping technique would be better than conventional technique in mechanical properties, however, soft-start technique might be better than conventional one in microleakage. It was concluded that much endeavor should be made to find out the curing conditions, which have advantages of both aspects or to solve these kinds of problems through a novel idea of polymerization.

This study was performed to evaluate wear characteristics of light-cured composites when opposed by human enamel. Seven light-cured restorative composites were selected and enamel cusps sectioned from premolars. All samples were stored in distilled water at $37^{\circ}C$ for 10 days. 68.6 N of weight was loaded during the test. The measurements of vertical loss of enamel cusps, weight loss and volume loss of composites, and SEM observations of the polished and abraded surfaces were made after 30,000 cycles. The results obtained were summarized as follows; 1. The highest hardness value of 70.4 was observed in the Spectrum group and the lowest value of 19.8 was observed in the Heliomolar group. Results of Tukey test showed that an overall significant difference was indicated except the Spectrum, Z100 and Clearfil AP-X groups(p<0.05). 2. Enamel showed the good abrasion resistance against the Heliomolar group of microfilled composite and the Palpique Toughwell group containing the submicron hybrid type spherical fillers. 3. The abrasive wear resistance of hybrid composites was improved with the decrease of mean particle size and hybrid of submicron particle fillers. 4. SEM observation of worn surfaces revealed the protrusion, attrition and missing of fillers, cracks developing and delamination in the matrix.

Purpose: This study was performed to investigate the influence of object shape and distance from the center of the image on the volumetric accuracy of cone-beam computed tomography (CBCT) scans, according to different parameters of tube voltage and current. Materials and Methods: Four geometric objects(cylinder, cube, pyramid, and hexagon) with predefined dimensions were fabricated. The objects consisted of Teflon-perfluoroalkoxy embedded in a hydrocolloid matrix (Dupli-Coe-Loid TM; GC America Inc., Alsip, IL, USA), encased in an acrylic resin cylinder assembly. An Alphard Vega Dental CT system (Asahi Roentgen Ind. Co., Ltd, Kyoto, Japan) was used to acquire CBCT images. OnDemand 3D (CyberMed Inc., Seoul, Korea) software was used for object segmentation and image analysis. The accuracy was expressed by the volume error (VE). The VE was calculated under 3 different exposure settings. The measured volumes of the objects were compared to the true volumes for statistical analysis. Results: The mean VE ranged from -4.47% to 2.35%. There was no significant relationship between an object's shape and the VE. A significant correlation was found between the distance of the object to the center of the image and the VE. Tube voltage affected the volume measurements and the VE, but tube current did not. Conclusion: The evaluated CBCT device provided satisfactory volume measurements. To assess volume measurements, it might be sufficient to use serial scans with a high resolution, but a low dose. This information may provide useful guidance for assessing volume measurements.
In 'solid-phase' PEGylation, the conjugation reaction occurs as the proteins are attached to a solid matrix, and thus it can have distinct advantages over the conventional, solution-phase process. We report a case study: rhIFN-${\alpha}$-2a was first adsorbed to cation exchange resin and then N-terminally PEGylated by aldehyde mPEG of 5, 10, and 20 kD through reductive alkylation. After the PEGylation, salt gradient elution efficiently recovered the mono-PEGylate in a purified form from the unwanted species such as unmodified IFN, unreacted PEG, and others. The mono-PEGylation and its purification were integrated in a single chromatographic step. Depending on the molecular weight of the mPEG aldehyde used, the mono-PEGylation yield ranged 50-64%. We could overcome the major problems of random, or uncontrollable, multi-PEGylation and the post-PEGylation purification difficulties associated with the solution-phase process. N-terminal sequencing and MALDI-TOF MS confirmed that a PEG molecule was conjugated only to the N-terminus. Compared with the unmodified IFN, the mono-PEGylate showed the reduced anti-viral activity as measured by the cell proliferation assay. The bioactivity was reduced more as the higher molecular weight PEG was conjugated. Immunoreactivity, evaluated indirectly by antibody binding activity using a surface plasmon resonance biosensor, also decreased. Nevertheless, trypsin resistance as well as thermal stability was considerably improved.
To analyze the interfacial property between the fiber and the matrix, work of adhesion was used generally that was calculated by surface energies. In this paper, it was determined what types of contact angle measurement methods were more accurate between static and dynamic contact angle measurements. 4 types of glass fiber and epoxy resin were used each other to measure the contact angle. The contact angle was measured using two types, static and dynamic contact angle methods, and work of adhesion, Wa was calculated to compare interfacial properties. The interfacial property was evaluated using microdroplet pull-out test. Generally, the interfacial property was proportional to work of adhesion. In the case of static contact angle, however, work of adhesion was not consistent with interfacial property. It is because that dynamic contact angle measurement comparing to static contact angle could delete the error due to microdroplet size to minimize the surface area as well as the meniscus measuring error.
Antarctic Environmental Monitoring Handbook' was published by COMNAP/SCAR in 2000. The standardized method described in this handbook is recommended for monitoring of antarctic environment. High pressure bomb technique in this guide was used to decompose soil samples. In compliance with this guide book, high pressure bomb technique was applied to decompose the antarctic soil sampled at the King Sejong Station. An Isotope Dilution-Inductively Coupled Plasma-Mass Spectrometry (ID-ICP-MS) was applied to determine mass concentrations of Pb, Cu and Zn in the soil. The accuracy in this method was verified by the analysis of certified reference materials (CRM) of NIST 2702 (marine sediment). The analytical results agreed with certified value within the range from 99.5~100.8%. Matrix separation was necessitated for the determination of Cu and Zn by Chelex 100 ion exchange resin. As a result, the average mass concentrations of Pb, Cu and Zn which are suspected to be caused by anthropogenic pollution were 332.9 mg/kg, 95.6 mg/kg and 115.3 mg/kg, respectively. Those for the metals sampled in the soils of the remote regions from the station were 28.1 mg/kg, 101.8 mg/kg and 115.6 mg/kg, respectively.
