Effects of Xingjian(LR2),Shaofu(HT8) by Reinforcing and Reducing on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Normal Rats (행간(行間)·소부(少府) 보사침법(補瀉鍼法)이 국소뇌혈류량(局所腦血流量) 및 평균혈압(平均血壓)에 미치는 영향(影響))

  • Objective: The purpose of this study is to examine influences by the order, Reduction and Reinforcement in Acupuncture on cerebral hemodynamics [regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP)] in normal rats. Methods: This experiments was to to investigate eath other changes of rCBF and MABP at Xingjian(LR2)(1st) Shaofu(HT8)(2nd) Reduction, Xingjian(LR2)(1st) Shaofu(HT8)(2nd) Reinforcement, Shaofu(HT8)(1st) Xingjian(LR2)(2st) Reduction and Shaofu(HT8)(1st) Xingjian(LR2)(2st) Reduction in Acupuncture. Results: 1. LR2(1st) HT8(2nd) Reduction in Acupuncture was decreased rCBF and MABP in compared with normal condition. 2. LR2(1st) HT8(2nd) Reinforcement in Acupuncture was significantly decreased rCBF, and was decreased MABP in compared with normal condition. 3. HT8(1st) LR2(2st) Reduction in Acupuncture was decreased rCBF during acupuncture but was recovered rCBF after with-drawing of the neddle. 4. HT8(1st) LR2(2st) Reduction in Acupuncture was decreased MABP during acupuncture and after withdrawing of the neddle. 5. HT8(1st) LR2(2st) Reinforcement in Acupuncture was significantly increased rCBF during acupuncture and 30min after withdrawing of the neddle. 6. HT8(1st) LR2(2st) Reinforcement in Acupuncture was decreased MABP during acupuncture, but was recovered MABP after withdrawing of the neddle. Conclusions: I suggested that LR2 HT8 Reduction in Acupuncture and LR2 HT8 Reinforcement in Acupuncture cause a diverse response of cerebral hemodynas.

  • This study was penonned to provide basic data that predict the usefulness of Gastrodiae rhizoma as food materials for oriental medicinal cuisine(YakSun). We measured the changes of regional cerebral blood flow(rCBF) and blood pressure(BP) in rats, following the intravenous injection of Gastrodiae rhizoma water extract The measurement was continually monitored by laser-doppler flowmeter and pressure transducer in anesthetized adult Sprague-Dawley rats for about two to two and half hours through the data acquisition system composed of MacLab and Macintosh computer. The results of this experiment were as follows. Gastrodiae rhizoma increased the changes of rCBF in rats significantly. The rCBF of Gastrodiae rhizoma did not change by pretreated propranolol, atropin, methylene blue and indomethacin. But the rCBF of Gastrodiae rhizoma was increased by pretreated L-NNA. Gastrodiae rhizaoma decreased the changes of BP significantly. The BP of Gastrodiae rhizoma did not change by pretreated propranolol, atropin, methylene blue and indomethacin. But the BP of Gastrodiae rhizoma was decreased by pretreated L-NNA. These results indicated that Gastrodiae rhizoma might increase the rCBF and the BP which related to nitric oxide synthesis. Also these results indicate that Gastrodiae rhizoma can be used as a safe and clinically applicable to diet therapy of cerebral cardiovascular disease.

  • Yuldahansotang(YH) has been used in Sasang(四象) constitution medicine for many years as a therapeutic agent for cerebral disease. The effect of YH on the vascular system is not known. The purpose of this study was to determine the effect of YH on blood pressure, regional cerebral blood flow(rCBF) and pial arterial diameter of rats. 1. Blood pressure decreased by YH in rats. 2. rCBF was increased by YH in a dose-dependent manner. 3. Pretreatment with propranolol, methylene blue and indomethacin significantly inhibited YH induced increase in rCBF. 5. Blood pressure increased by Radix Puerariae(RP) and Radix Ligustici Tenuissimae(RLT) but Radix Scutellariae(RC) decreased blood pressure in rats. 6. rCBF was increased by RP and RLT in a dose-dependent manner but RC decreased low dosage, and RC increased high dosage. 7. Pial arterial diameter was increased by YH in a dose-dependent manner. 8. Pretreatment with propranolol significantly inhibited the increased in pial arterial diameter induced by YH. These results suggest that YH causes a diverse response of blood pressure, regional cerebral blood flow(rCBF) and pial arterial diameter. The increase in rCBF is also mediated by prostaglandins, cyclic GMP and adrenergic ${\beta}$ receptor and the increase in pial arteral diameter is mediated by adrenergic ${\beta}$ receptor.

  • As an attempt to develop new functional health beverage by using medicinal herb, green ginger(Zingiber officinale Roscoe), we investigated the effect of Zingiber officinale on blood pressure and regional cerebral blood flow (rCBF) of rats with Zingiber officinale extracts. Zingiber officinale extract increase rCBF significantly. The drink produced consisted of Zingiber officinale extract 1.825%, maltitol syrup 17.0%, citric aicd 0.06%, ascorbic acid 0.02%, stevioside 0.001%, ginger flavor 0.11% and water. Brix, pH and acidity of product were 13.7, 4.4 and 0.09, respectively. This drink had good score by the sensory evaluation. The above results showed that development of such functional beverage using Zingiber officinale can used as a functional material improving blood circulation in beverage industry.

Effects of Geopungjeseub-tang(Gufengchushi-tang) on the Changes of Cerebral Blood Flow in Rats (거풍제습탕이 뇌허혈이 유발된 백서의 뇌혈류 변화에 미치는 영향)

  • Objectives : Geopungjeseub-tang(Gufengchushi-tang) has been used in oriental medicine for many centuries as a therapeutic agent for hemiplegia caused by deficiency of qi(氣虛) and damp phlegm(濕痰). This study was performed to evaluate effects of Geopungjeseub-tang extract(GJT) on hemodynamics[regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP), heart rate(HR)] in normal rats and in rats with cerebral ischemia by middle cerebral artery(MCA) occlusion. Also, effects of adrenergic ${\beta}-receptor$, cyclooxygenase on response to GJT were evaluated. Methods : Laser-doppler flowmetry(LDF) measured changes of rCBF, MABP and HR. Video microscope and width analyzer measured changes in PAD. Results : rCBF and PAD increased after treatment with GJT(10mg/kg, i.v.) during the period of cerebral reperfusion, and pretreatment with indomethacin raised rCBF and PAD increased after treatment with GJT during the same period as above. Pretreatment with propranolol decreased rCBF, but increased after GJT treatment, but raised PAD increased after GJT treatment during this period of reperfusion. Conclusion : CR caused diverse responses were observed in rCBF and PAD after treatment with GJT. ACF action is mediated by adrenergic ${\beta}-receptor$ and cyclooxygenase. Result suggest that GJT has an anti-ischemic effect through the improvement of cerebral hemodynamics and has theraputic potential for cerebral apoplexy.

  • In this study, Artemisia capillaris, which has been used as oriental medicine area and folks remedy, was investigated for effective substance (scopoletin) analysis. As an attempt to develop new functional beverage by using medicinal herb, Artemisia capillaris. Therefore we investigate the effect of scopoletin in Artemisia capillaris on vessel and regional cerebral blood flow (rCBF) of rats. Artemisia capillaris extract was significantly decreased vessel constraction and significantly increased rCBF. Artemisia capillaris has often been cited in medical literature for its medicinal effects. Although the Korean Food and Drug Administration indicated Artemisia capillaris as the In-Jin-Sook which was allowed as a food stuff. From the view point of the subjects who eat In-Jin-Sook products, most of them ingested it for a certain pharmacological efficacy rather than as a beverage or a food. According to this facts, we need develop functional foods using In-Jin-Sook. Also scopoletin, get known to smooth muscle relaxation content was 326.13 ug/g. These results demonstrate that Artemisia capillaris has pharmaceutical effects and balanced nutritional contents. In according with Artemisia capillaris has a sufficiency of the terms use of food stuff in functional foods.

  • Kyungisan (KGS) has been used in oriental medicine for many centuries as a therapeutic agent for treatment of stroke caused by deficiency of qi(氣虛). This study was performed to evaluate effects of KGS extract on the regional cerebral blood flow(rCBF) and mean arterial blood pressure(MABP) in rats. The result of this study were as follow ; 1. KGS significantly increased rCBF irrelevant to MABP in normal rats, 2. To prescribe KGS after pretreatment with indomethacin(IDN) decreased rCBF as compared with control group to administered only KGS in normal rats. But the change of MABP is not significantly as compared with control group. 3. To prescribe KGS after pretreatment with methylen blue( MTB) decreased MABP and rCBF as compared with control group to administered only KGS in normal rats. Especially, it significantly decreased rCBF. These results suggest that KGS increase rCBF by enlargement diameter of pial artery in brain. The active mechanism of KGS is related with prostaglandin activated by cyclooxygenase. So, I suggest that KGS has an anti-ischemic effect through the improvement of cerebral blood flow and can be used for stroke.

Effects of FOENICULI FRUCTUS on the Regional Cerebral Blood Flow and Mean Arterial Blood Pressure in Rats (소회향이 흰쥐의 국소뇌혈류량 및 평균혈압에 미치는 영향)

  • This experimental study was designed to investigate the effects of FOENICULI FRUCTUS freeze dry powder (FF) on the change of cerebral hemodynamics [regional cerebral blood flow (rCBF) and mean arterial blood pressure (MABP)] in normal and further to determine the mechanism of action of FF. The results in normal rats were as follows ; FF significantly increased rCBF in a dose-dependent, but decreased MABP, This results were suggested that FF significantly increased rCBF by dilating PAD. The FF-induced increase in rCBF was significantly inhibited by pretreatment with 1H[1,2,4]oxadizolo[4,3-a]quinoxalin-1-one (ODQ, 10 ${\mu}$g/kg, i.p.), an inhibitor of guanylate cyclase and indomethacin (IDN, 1 mg/kg, i.p.), an inhibitor of cyclooxygenase and propranolol (PPN, 3 mg/kg, i.p.), a blocker of adrenalic f receptor and Lu-Nitro-L-Arginine (L-NNA, 1 m9/kB, i.p.), an inhibitor of nitric oxide synthase. The FFE-induced decrease in MABP was significantly increased by pretreatment with L-NNA and was increased by pretreatment with PPN, Dut was inhibited by pretreatment with ODQ and IDN, This results were suggested that the mechanism of FF was mediated by nitric oxide synthase and adrenalic ${\beta}$ receptor.

Effects of Jaeumgenby-tang adding Aurantii FructusㆍGastrodae Rhizoma on the Brain Cell and Changes of Cerebral Hemodynamics (자음건비탕가지각ㆍ천마가 뇌세포 및 뇌혈류역학 변동에 미치는 영향)

  • Jaeumgenby-tang(JGT) have been used in oriental medicine for many centuries as a therapeutic agent of vertigo caused by deficiency of qi(氣) and blood(血). Effect of Aurantii Fructus(AF) take off the phlegm by promoting the circulation of qi, Gastrodae Rhizoma(GR) has effects treating for headache, vertigo by calming the liver and suppressing hyperactivity of the liver-yang(陽). And, I designed to investigate whether injection of JGT adding AFㆍGR extract(JGTAG) affects cytotoxicity in vitro, cerebral hemodynamics [regional cerebral blood flow(rCBF), pial arterial diameter(PAD), mean arterial blood pressure(MABP)] in normal and cerebral ischemia rats by MCA occlusion method. The changes of rCBF and MABP were determinated by laser-doppler flowmetry(LDF), and the change of PAD was determinated by video microscope and width analyzer. The results were as follows in normal rats; JGTAG was not cytotoxicity in brain cells. And JGTAG was significantly increased rCBF, PAD and MABP. This results suggest that JGTAG increased significantly rCBF by dilating PAD. And the results were as follows in cerebral ischemic rats; The changes of rCBF and PAD were increased stably by treatment with JGTAG(10mg/kg, i.v.) during the period of cerebral reperfusion, and pretreatment with propranolol and indomethacin were increased JGT AG induced increase of rCBF and PAD during the period of cerebral reperfusion. We suggest that JGTAG has an anti-ischemic effect through the improvement of cerebral hemodynamics.
