Extraction and analysis conditions of di-(2-ethylhexyl) adipate (DOA) and its migration contents into foods wrapped with plasticized polyvinyl chloride (PVC) film during storage and cooking with microwave were examined using gas chromatography. Experiments on the extraction solvent and method showed that the method of reflux using carbon tetrachloride for longer than one hour allowed the highest extraction of DOA. Analysis of four PVC wrap films from different factories in domestic market showed the DOA contents of 18.9% for home-use and 24.6% for retail-use on the average. The highest migration during storage was observed in pork belly, which were 21% and 26% after one and three days respectively. During microwave cooking; smaller than 5.5% of DOA were migrated into foods when the film was not contacted with foods, whereas about 49% migration was observed in pork belly contacted with film directly.

Microwave assisted extraction(MAE) is known as a more environmental friendly process with economic advantages in terms of less time, less solvent, less energy and less wastes than the current time consuming reflux method. It was applied to develop a rapid extraction method for soluble ginseng components that are major materials used for the processing of ginseng products. In a comparative study between pre established MAE(ethanol 60%, power 80 W, process time 4 min$\times$5) and current extraction method(ethanol 80%, temp. 85oC, time 8 hr$\times$5), MAE was more efficient than the current method to obtain an extract yield(soluble solid), but it was insufficient to extract individual ginsenosides, total phenols, reducing components and acidic polysaccharides. MAE with 80% ethanol by 5 times showed, however, that its extraction efficiency on soluble solid, crude saponin, major ginsenosides, and the other components was equal or superior to that of the current method, indicating that ethanol concentration is one of the critical parameters influencing the MAE process. The quality of ginseng extracts from MAE was assured by evaluating the corresponding standards and by comparing TLC and HPLC patterns with the control.
Extraction methods of medicinal plants were evaluated for growth enhancing effects of human neuroblastoma SK-N-SH cells. Hot-water extraction (reflux for 5 hr), hot-water extraction post treatment (100$^{\circ}C$ or 120$^{\circ}C$, 90 min) and ethanol extraction (reflux for 5 hr) methods were applied to Angelica gigas, Rhemania glutinosa, Paeonia lactiflora and Cnidium officinale samples to extract their constituents. Cells were treated for 2 hr with various concentrations of extracts (0, 0.5, 1, 2, 4 ${\mu}g/{\mu}l}$ media) prior to $H_2O_2$ (250 ${\mu}M$) treatment for 2 hr to provide oxidative stress. Cell viability, caspase-3 expression and apoptosis were measured for cells treated with sample extracts. Hot-water extract exhibited a stronger growth enhancing and apoptosis protecting ability than other extracts. These activities were shown at less than 1 ${\mu}g/{\mu}l}$ concentration, and not greater than 2 ${\mu}g/{\mu}l}$ concentration. Hot-water extract contained more polyphenolic compounds than other extracts coming along with stronger antioxidant activity. The efficacy of antioxidant activity was stronger in the hot-water extract of Angelica gigas than other hot-water extracts of medicinal plants. These results suggest that hot-water extraction is an appropriate method to extract materials for growth enhancing and apoptosis protection of SK-N-SH cells, and hot-water extracts of Angelica gigas might be useful materials for protection from aging brain cells.

Aster glehni was extracted by microwave-assisted extraction(MAE) and reflux extraction(RE) methods and their extraction efficiencies were compared. A considerable reduction in extraction time was achieved by MAE. When 70% methanol, 50% methanol, 70% ethanol, or 50% ethanol was used, MAE extract contained nearly same amounts of soluble solid and total polyphenol contents as obtained by RE. The optimum MAE conditions for the extraction of Aster glehni were $120{\sim}150$ watts of microwave energy and $4{\sim}8$ minutes of extraction time. No significant changes were found in electron donating ability(EDA) over the variation of microwave energy or extraction time. The use of diluted methanol or ethanol resulted in improving extraction yield(24%), total polyphenol content(2.6%) and EDA(60%).

In this paper, optimum extraction condition for berberine which is the indan companent of Coptidis rhizoma was examined at various conditions in the aspect of temperature, solvent and time followed by quantitation with reversed phase HPLC. The efficiency for extraction has been shown the best at $60^{\circ}C$ with 2~3 hours reflux time in methanol or acetic acid. Any significant effect can not be shown from the addition of SLS to extraction solvent.
This study prepared extracts of Angelica dahurica leaves using reflux water extraction (RW), reflux ethanol extraction (RE) and pressure heating water extraction (PW). The extracts were extraction for levels of polyphenol compounds, antioxidant activities, and inhibitory potencies for xanthine oxidase and tyrosinase. The PW extraction method yielded the highest content of polyphenol compounds (95.23 mg/g). The electron donating abilities (EDAs) of RE and PW extracts were 76.02% and 70.08% respectively. The superoxide dismutase (SOD)-like activities were 13.45 19.00%, when extracts were assayed at 1 mg/mL. The nitrite scavenging ability (pH 1.2) of the PW extract was 54.33% higher than levels shown (44.24%) by the RE and RW extracts. The inhibition of xanthine oxidase by the RW extract was highest (99.71% at 5 mg/mL) while that of the PW extract was over 97% at 500 g/mL. Tyrosinase inhibition was highest in the RE extract (46.25% at 5 mg/mL). All extracts showed dose-dependent inhibitory activities. The results indicated that the PW extract had the highest polyphenol content, the RW and RE extracts had the best nitrite scavenging ability, and the RE extract showed the most pronounced effect on EDA, SOD-like activity and tyrosinase inhibition.
To develope more effective extraction methods for paraquat in soil, some modification methods were accomplished in two different types of soil. For extraction of tightly bound-paraquat, conc. HCl 70ml were added with different shaking times, and then $H_2SO_4$ reflux were performed for an hour. In this case, 60 minutes shaking were optimum and recovery were increased more $1.09{\sim}1.50$ folds(84.0% in high clay contents soil, but 96.7% in low clay contents soil) and the long-time consuming step, filtration were easily done, with decreasing filtration time were shorter 4.6 folds(ca. $11{\sim}14min.$). than general paraquat analytical method(ca. $55{\sim}65min.$). And only $H_2O_2$ digestion with different volume and refluxing time resulted in recovery increasing. Nevertheless, considering analyst's safety, 30ml of $H_2O_2$ addition and 30 minutes reflux were regarded as optimum condition. Although, Kjeldahl digestion with $H_2O_2$ showed relatively high recovery, it is not significant statistically. For extraction of loosely bound-paraquat, 0.01, 0.1, 1.0, 10.0M of $NH_4Cl$ and of $CaCl_2$ compared with $1.5{\sim}24hr$ of different shaking time. There were no loosely bound residues of paraquat.
Top parts of $Dendranthema$$zawaskii$ var. $yezoense$ (DZY) and $Cosmos$$bipinnatus$ (CB) are believed to contain substances with antioxidant effects and activity. Present experiments were undertaken to investigate an adequate and efficient extraction method which ensures the highest yield of antioxidant components. Extraction yield was 1.11-1.92 times higher by ultrasonic method than other methods in both species. By 45 minute ultrasonic extraction, total polyphenol contents reached the highest level, $80.70mg{\cdot}g^{-1}$ tannic acid equiv./DW in $Cosmos$$bipinnatus$ and total flavonoid contents $50.41mg{\cdot}g^{-1}$ naringin equiv./DW in DZY. DPPH radical scavenging activity was almost the same among extraction methods or slightly higher in reflux extraction. The highest DPPH radical scavenging was observed by reflux extract in CB ($RC_{50}=0.135mg{\cdot}mL^{-1}$). ABTS radical scavenging activity was the highest by 15 minute ultrasonic wave in DZY and CB, $RC_{50}=0.159mg{\cdot}mL^{-1}$ and $RC_{50}=0.139mg{\cdot}mL^{-1}$, respectively. High ferrous ion chelating effects were obtained by 30-minute ultrasonic wave in DZY ($RC_{50}=0.803mg{\cdot}mL^{-1}$). Extracts of top parts of DZY obtained by reflux method showed the highest inhibition effect against peroxidation of linoleic acid, and extract of CB obtained by ultrasonic for 45 minutes showed the highest inhibition effect. In conclusion, optimum extraction method should be adjusted according to plant species and antioxidant substances.
New HPLC method was developed for determination of some flavonoids such as naringin, hesperidin, neohesperidin, rutin, quercitrin, naringenin, hesperetin and apigenin and their contents in citrus juice and citrus peel from citrus varieties Brown in Cheju. Detection was at 280nm and reverse phase ${\mu}$-Bondapak C-18 column was used. Water/methanol/acetic acid as the mobile phase was better than water/acetonitrile/acetic acid. Flavonoids were more stable in 20% n,n-dimethylformamide in methanol(20% DMF) than methanol and pH 12 adjusted by 1N-sodium hydroxide solution. Standard flavonoid solutions were injected three times consecutively and the reproduciability was 0.236 to 3.550%, Correlation coefficient of the calibration curve was 0.9946 to 0.9999. The exiraction efficiency of hesperidin from citrus peel was evaluated with different extraction method such as reflux, ultra-sonicating method, using three solvents (aqueous solutions with pH12 adjusted by 1N-sodium hydroxide, methanol and 20% DMF), respectively. The reflux for 4 hour in 20% DMF was the most efficient of the tested methods and solvents, and recovery percentage were 78.0∼130.0%. Flavonoids were determined in citrus juice. Naringin was 68.2mg/100$m\ell$ in Natsudaidai, Hesperidin were 85.6mg/100$m\ell$ in Sankyool and Neohesperidin was 25.3mg/100$m\ell$ in Dangyooja. Flavonoids were determined in citrus peel. Naringin was 110mg/g in Dangyooja, Hesperidin was 242mg/g in Hungjin and Neohesperidin was 87.9mg/g in Dangyooja.
A high performance liquid chromatographic method was designed for the quantitative analysis of oxymatrine in Sophora Radix. The separation of oxymatrine was performed by reversed-phase chromatography with a $C_{18}$ column and a buffered aqueous solution containing acetonitrile, and monitored by UV absorption at 215 nm. Extraction of oxymatrine in Sophora Radix was carried out using various solvents and extraction methods. The optimum extraction efficiency for the crushed Sophora Radix was achieved by reflux at $80^{\circ}C$ in 50% ethanol for five hours. Most extraction methods used to complicate pretreatments. In this study, sublimation was employed for a extraction method without going through complicate pretreatments. Sublimation was carried out under high vacuum ($1{\times}10^{-3}$ torr) and at high temperature ($200^{\circ}C$). Extraction efficiency using Sublimation was found to be inferior to other extraction methods.
