Quantification of urea in serum by isotope dilution HPLC/MS (동위원소 희석 HPLC/MS에 의한 혈청 내 urea의 정량)

  • Urea in blood has been measured as an effective marker for diagnosis of renal function. Urea which is e end-product of nitrogen containing metabolites such as proteins is filtered through glomeruli of kidneys and then excreted as urine. If the renal function is deteriorated, the urea concentration in blood will be increased, from which the healthiness of renal function is judged. In order to improve the confidence of diagnosis results, the results must keep traceability chain to certified reference materials, which was certified by primary reference method. In this study, we proposed isotope dilution-liquid chromatography/mass spectrometry (ID-LC/MS) as a candidate primary method, in which $15^N_2$-urea is used as an internal reference material. The developed method is highly accurate in principle and is convenient as it does not require cumbersome derivatization. 0.1 mmol/L ammonium chloride was selected as a mobile phase for HPLC because it provided low interference in MS analysis of relatively low molecular weighted urea. HPLC and MS were connected with an electrospray ionization (ESI) interface of positive mode, which provided high sensitivity and reproducibility. The developed method was validated with internationally recognized reference materials, and we have obtained satisfactory results in an international ring trial. The expanded uncertainty calculated according to ISO guide was 1.8% at 95% confidence interval. The developed method is being used as a primary reference measurement method such as for certification of serum certified reference materials (CRMs).

Development of a Data Reference Model for Joint Utilization of Biological Resource Research Data (생물자원 연구데이터의 공동 활용을 위한 데이터 참조모델 개발)

  • The biological resources research data around the world are not only very critical themselves but should be shared and utilized. Up to now, the biological resources have been compiled and managed individually depending on the purpose and characteristics of the study without any clear standard. So, in this study, the data reference model would be suggested which is applicable in the phase ranging from the start of the construction of the information system and which can be commonly used. For this purpose, the data model of the related information system would be expanded based on the domestic and foreign standards and data control policy so that the data reference model which can be commonly applicable to individual information system would be developed and its application procedure would be suggested. In addition, for the purpose of proving the excellence of the suggested data reference model, the quality level would be verified by applying the Korgstie's data model evaluation model and its level of data sharing with the domestic and foreign standards would be compared. The test results of this model showed that this model is better than the conventional data model in classifying the data into 4 levels of resources, target, activities and performances and that it has higher quality and sharing level of data in the data reference model which defines the derivation and relation of entity.

APMP (Asia Pacific Metrology Programme) Regional Intercomparison Results of Acoustic Calibrators (음향교정기의 아시아.태평양 지역 국제비교 결과)

  • The results of the APMP (Asia Pacific Metrology Programme) regional intercomparison of acoustic calibrators were reviewed and analyzed. The artefacts used in intercomparison are a sound level calibrator and a pistonphone. The microphones used to measure the output pressure level are 1" and 1/2" standard microphones (LS1P, LS2P) as well as 1" and 1/2" reference microphones (WS1P/F, WS2P/F). The results obtained using standard microphones are satisfactory, but those obtained by the reference microphones, even though E/sub n/ values are within ±1.0, showed great deviations. Such results had come from the inaccurate calibration of reference microphones. By using the correct calibration results which were obtained by the recently established international standards, the new results were very similar to those of the foreign standard institutes.

Development of Isotope Dilution-Liquid Chromatography/Tandem Mass Spectrometry as a Candidate Reference Method for the Determination of Acrylamide in Potato Chips

  • An isotope dilution-liquid chromatography/tandem mass spectrometric method was developed as a candidate reference method for the accurate determination of acrylamide in potato chips, starch-rich foodstuff cooked at high temperature. Sample was spiked with 13C3-acrylamide and then extracted with water. The extract was further cleaned up with an Oasis HLB solid-phase extraction (SPE) cartridge and an Oasis mixed-phase cation exchange (MCX) SPE cartridge. The extract was analyzed by using LC/ESI/Tandem MS in positive ion mode. LC with a medium reversed-phase (C4) column was optimized to obtain adequate chromatographic retention and separation of acrylamide. MS was operated to selectively monitor [M+H]+ ions of the analyte and its isotope analogue at m/z 72 and m/z 75, respectively. Sample was also analyzed by the LC/MS with selectively monitoring the collisionally induced dissociation channels of m/z 72 → m/z 55 and m/z 75 → 58. Compared to the LC/MS chromatograms, the LC/MS/MS chromatograms showed substantially reduced background chemical noises coming from solvent clusters formed during ESI spray processes and interferences from sample matrix. Repeatability and reproducibility studies showed that the LC/MS/MS method is a reliable and reproducible method which can provide a typical method precision of 1.0% while the LC/MS results are influenced by chemical interferences.

Development of the calibration procedure of the reference sound source and case study on the uncertainty evaluation (기준음원의 교정 절차 개발 및 불확도 평가 사례)

  • A Reference Sound Source (RSS) is an important standard device employed in measuring sound power. The specifications of RSS is specified in international standards, and it is classified as a major calibration item in the field of acoustic metrology. Since the output power of RSS is affected by the supply voltage, each country needs to secure its own calibration service system. In this study, a procedure for calibrating a RSS is established based on the reverberant room conditions and uncertainty evaluation is conducted. Basically, the calibration procedure can apply a precision measurement process of acoustic power, and here, the measurement method using the reverberation chamber of ISO 3741 is applied. For this purpose, a measurement system is constructed, measurements are conducted with two types of RSS, and measurement uncertainty is evaluated. Through measurement examples, it is confirmed that the non-uniformity of the sound pressure distribution in the reverberation room and the volume measurement uncertainty contributed significantly to the overall uncertainty. Additionally, the influence of input voltage is experimentally examined to examine the uncertainty contribution that can be reflected in acoustic power measurements.

Measurements of the rheological properties of standard reference material 2490 using an in-line micro-Fourier rheometer

  • The control of the rheological properties of a fluid during processing is important and can determine the efficiency of the production in addition to the performance of the final product. The vast majority of process fluids are viscoelastic, hence an instrument that measures both the viscous and elastic properties of the material during processing would be of great practical use. However, most in-line instruments commercially available to date are only capable of measuring viscosity at a single shear rate. An in-line rheometer that measures both the viscous and elastic properties of fluids over a wide range of shear rates simultaneously has been described in a previous publication (Glasscock et at., 2003) in which the results of measurements on flowing sunflower oil were presented. Before this instrument can be used in an industrial situation, it must be demonstrated that the generated results are the same as, or bear some fixed relationship to, the results obtained by conventional off-line rheometers. To this end, the present investigation describes the measurements of a standard reference fluid, polyisobutylene dissolved in 2,6,10,14-tetramethylpentadecane, labelled SRM2490 by the National Institute of Standards and Technology (NIST) in the USA. The results obtained using the in-line rheometer show remarkably good correlation for viscosity, using a modified Cox­Merz rule, with the results supplied with the reference material from NIST.

Influence of Microstructure on Reference Target on Ultrasonic Backscattering (기준표적상의 미세구조가 초음파 후방산란에 미치는 영향)

  • This paper is based on our comments and proposed amendments to the documents, Annex A, Phantom for determining Maximum Depth of Penetration, and Annex B, Local Dynamic Range Using Acoustical Test Objects 87/400/CDV. IEC 61391-2 Ed. 1.0 200X, prepared by IEC technical Committee 87; Ultrasonics. The documents are concerned with the influence of microstructure of reference target material on the ultrasonic backscattering. Previous works on the attenuation due to backreflection and backscattering of reference target materials are reviewed. The drawback to the use of ungraded stainless steel and metallic materials without microstructural data such as, crystal structure, basic acoustic data of sound velocity and attenuation, grain size, roughness and elastic constants has been discussed. The analysis suggested that the insightful conclusion can be made by differentiating the influence arising from target size and microstructure on the backscattering measurements. The microstructural parameters are associated with physical, geometrical, acoustical and mechanical origins of variation with frequency. Further clarification of such a diverse source mechanisms for ultrasonic backscattering would make the target material and its application for medical diagnosis and therapy simpler and more reliable.

On the Ideology and the Standard of Reference Service (참고봉사의 이념과 기준설정을 위한 고찰 -미국에서의 이론과 전개과정을 중심으로-)

  • This study has two aims. one is to devide the ideology of reference service into three groups such as conservative, moderative, and progressive theories, and then investigate the validity of each theory as well as any problems to be suggested. The other is to establish appropriate standards of reference service according to the types of libraries and users, based on the analysis of such theories. Each theory has its own merits and demerits. Accordingly, it seems to be difficult to apply uniformly and theory into users who have different types of question and educational levels. And also it is difficult to apply into various libraries which have different aims of establishment, characteristics, and the users of each library. Consequently, it is suggested to set up a norm of reference service according to the types of libraries and users respectively. The conservative theory is applicable to small and medium sized public libraries, school libraries, and college and university libraries of which the users are mainly students and citizens. The progressive theory is desirable for special and academic libraries as well as large public libraries where the users are mainly specialists. and scholars.

Revision of Geomorphological Terms in the Reference Guide for Geography Textbook Compilation (지리교과 지형 편수용어 개선을 위한 모색)

  • This study has analyzed and revised the geomorphological terms in the Reference Guide for Geography Textbook Compilation issued by the Ministry of Education in 2017. The current geography glossary is composed of 1,150 terms of which 24.4 percent are geomorphological one. They also amount to 52.2 percent of the terms registered in the physical geography glossary. The number of terms in coastal landforms is the maximum of fifty-eight, while the one in weathering landforms is the minimum of six. This suggests a large difference in the number of terms among landform types originating from varying geomorphic processes. A lot of ordinary words are included in the geomorphology glossary. Since they have been already registered in the Korean Standards Dictionary which is another reference guide for textbook compilation, it is appropriate to delete them out of the geomorphological terms. The geomorphology glossary also contains difficult terms written in Chinese characters, inappropriate terms and even incorrect terms which are likely to lead to misconception on landforms. The revised glossary consists of 284 geomorphological terms in which 101 terms are newly added after taking all the problems into consideration.

Development a System to Manage the Code of Construction Specifications and Design Standards (국가건설기준 코드 관리시스템 구축)

  • The construction standards in Korea consist of design standards and specifications. They are provided by the national transportation electronic information center in file format, not in a DB, which may lead to their incomplete utilization and versatility, such as a lack of history management and reference functions. This also makes it difficult to cross-refer to standards and manage revision history. In this study, we developed a code management system for the national construction standards by analyzing domestic and overseas related systems, in order to pinpoint the problems in the current system and its development direction and, thus, to suggest a detailed plan for the design and implementation of a code management system. The developed system comprises four subsystems, namely a construction standards making system, construction standards service system, construction standards maintenance system and construction standards portal system. The results of this study are expected to improve the user's convenience by facilitating the revision, operation and history management of the complex and various construction standards, and to provide high-quality national construction standards by effectively supporting the revision and operation of the national construction standards with minimum duplication and conflicts.
