Development and Validation of Multiplex PCR Method for the Identification of Pagrus major and Sciaenops ocellatus (참돔과 홍민어 판별을 위한 Multiplex PCR 검사법의 개발과 검증)

  • Nowadays, with increase of seafood consumption, there have been increasing reports of defective seafood products. There have been incidents of red drum (Sciaenops ocellatus) being sold as red seabream (Pagrus major). In this study, we sought to develop and validate species-specific PCRs to differentiate between P. major and S. ocellatus to prevent the sale of S. ocellatus as P. major. Primers for P. major were designed to bind 12s rRNA and those for S. ocellatus were designed to bind 16s rRNA. Multiplex PCR showed a 468 bp amplicon for P. major and a 181 bp amplicon for S. ocellatus. The limit of detection of P. major and S. ocellatus was present at 1 ng each. The developed primers were validated with 19 P. major samples of food items purchased through the internet. Using this monitoring method, the experimental results and tested species were in agreement. Hence, the developed multiplex PCR method is considered reliable to authenticate P. major and S. ocellatus.

A Refutation on the view of Parhae Marek's Origin as from Kokuryo (발해 말액(抹額)의 고구려 기원설 재검토)

  • I scrutinized the idea that Parhae Marek was inherited from Kokuryo which was presented in "The study on the Marek" published in the journal of the Korean Society of Costume 55-5, and concluded the following results: 1. The Marek which Samguk sagi recorded is the red head scarf used for dancers who do Koguryo dance in the Babarian Music System in Tang dynasty. Since its original record Tong dian tells that Kokuryo performers' costumes then had strikingly changed and Quichi and GaoChang dancers also wore Marek, so the idea that Marek was originated from Koguryo should be reconsidered. 2. Considering the Yaksoori mural's indistinguishable condition as well as the same scenes from other Kokuryo mural paintings, I can't evidently tell that the first person who carries shouldering drum puts Marek on. 3. should be pronounced [Mal] for the meaning of a head scarf and its definition can't be limited only for the sash type. 4. Diverse historical data on head scarves deny the assumption that the sash type of hairband would have been succeeded from Kokuryo to the Parhae Marek. 5. The Marek of Princess Junghyo's mural painting can be an example that shows the costume of Tang influenced Parhae's. But the similarity in styles of costume between contemporary countries doesn't mean their reciprocal racial or political identity.

Development of the Air-lifting & Suction-pumping System to Remove the Noxious Deposit in the Underwater (수중 유해성 유기퇴적물의 수거를 위한 Air-lifting & Suction-pumping System 개발)

  • Eutrophic matters accumulated on the bottom of sea, river and lake cause red tide phenomenon in ocean and outbreak green algae in river and lake. Systems are developed to remove the noxious deposit. But the existing systems remove not only the eutrophic matters but also natural materials, sand, pebbles etc. that should remain at the bottom. This paper describes a new system that can safely, and economically take away the noxious deposit in underwater. High pressure water jet is used to induce vortices in the triangular suction section, and air-lifting pump to lift up the deposit. The mixture of the water and deposit is filtered through the drum filters. An under camera shows the under water situation along the moving direction of the system that is controlled by a remote operator. This remote controlled moving system obliterate the necessity of the diver that usually costs high. The experimental results show the effectiveness of the suggested system.

Study on Prediction of Internal Quality of Cherry Tomato using Vis/NIR Spectroscopy (가시광 및 근적외선 분광기법을 이용한 방울토마토의 내부품질 예측에 관한 연구)

  • Although cherry tomato is one of major vegetables consumed in fresh vegetable market, the quality grading method is mostly dependant on size measurement using drum shape sorting machines. Using Visible/Near-infrared spectroscopy, apparatus to be able to acquire transmittance spectrum data was made and used to estimate firmness, sugar content, and acidity of cherry tomatoes grown at hydroponic and soil culture. Partial least square (PLS) models were performed to predict firmness, sugar content, and acidity for the acquired transmittance spectra. To enhance accuracy of the PLS models, several preprocessing methods were carried out, such as normalization, multiplicative scatter correction (MSC), standard normal variate (SNV), and derivatives, etc. The coefficient of determination ($R^2_p$) and standard error of prediction (SEP) for the prediction of firmness, sugar, and acidity of cherry tomatoes from green to red ripening stages were 0.859 and 1.899 kgf, with a preprocessing of normalization, 0.790 and $0.434^{\circ}Brix$ with a preprocessing of the 1st derivative of Savitzky Golay, and 0.518 and 0.229% with a preprocessing normalization, respectively.

A Study on the Dancing Costumes for the Buddhist Ceremony of 'Spirit Vulture Peak' (한국 불교 영산재 -무복에 관한 연구-)

  • The Buddhist costumes, unlike the ordinary ones, have not been studied comprehensively due to their religious, non-popular and non-social features. In this regard, this paper aimed at reviewing the styles and symbolic Characters of the monks' dancing costumes for the traditional Buddhist ceremony of 'Spirit Vulture Peak'(The Intangible Cultural Asset No.50.) Here, the 'Spirit Vulture Peak Ceremony' is a kind of Buddhist ceremony commemorating the Buddha's preaches on the peak of Mt. Grdhrakuta by means of symbolized ritual and reformation procedures. The methodology of this study depended on the research of literatures, personal observation of the actual ceremony and the discussion with Priest Park Song-am. The dancing style of the Ceremony can be divided into four categories; the cymbal dancing., the drum dancing , the butterfly dancing and the column-beating dancing. And for the dancing costumes, the loose ritual mantle, the long-sleeve robe and the buddhist costume are used; the loose mantle and the long-sleeve robe are for the drum dancing. The style of the loose mantle is rectangular, sewn every odd knot between 5 and 25 ones. According to the record, its color was initially grey or red-yellow, but varied depending on the local rules. The four corners of the loose mantle have a rectangular-cloth embroidery of sky & king patterns, and the center is embroidered with the Sun'Moon designs or 'Om/Nan' letters in the Korean spells. The light of Sun/ Moon designs symbolizing the sky and the earth respectively are indigenous to a particular school in Korea. The long-sleeve robe has a traditional over-coat style. and is called otherwise of if some design is added. The total clothing or dancing costumes for the Ceremony include 'the Dae-ryung (meaning great peake)loose mantle', 'the White Paldad(meaning Chinese Character eight) long-sleeve robe and the peaked hat. The Dae-ryung mantle is a kind of ritual costumes with the shorter length than the ordinary mantles, and has a character 'Ryung' in Chinese spell attached with three to six colors. The White Paldae robe is shorter than the ordinary robes in the total length but longer in the length of sleeves. It is put on over the mantle from the left shoulder, and used often for the butterfly and column-beating dancings. It is hoped that this study will promote the study on the customs and rites of the Buddhism rather than on the thoughts, and contribute to the further studies.

The Epidemiological Study on fish Diseases in the Southern Area of Kyeognam (경남 남부지역 양식어류 질병에 관한 역학적 연구)

  • An epidemiological study was performed to evaluate the state of fish diseases between aquaculture fishes in the area of southern Kyeongnam, especially Tongyeong-si, Geoje-si, and Goseong-gun. Examination for a total of 91 cases was carried out for a variety of fishes, including Japanese flounder(Paralichthys olivaceus), rock fish (Sebastes schlegeli), sea bass (Lateolabrax japonicus), sea breams(Pagrus major), and red drum (Sciaenops ocellatus) from November, 1999 to October, 2001. The investigation resulted in exploring 155 infectious diseases, including 62 Vibriosis, 16 Edwasiella infection, 11 Streptococcosis, 7 Pseudotuberculosis, 15 Scuticociliatida infection, 20 Gill flukes, and 9 Trichodiniasis. When the infections were classified according to fish species, Japanese flounder was infected by 30 Vibrio spp., 16 Edwardsiella tarda, 6 Streptococcus spp., 12 Scuticociliatida infection. In case of sea bass, 10 Vibrio spp. was able to isolate out of 10 outbreaks, on the other hand, rock fish was dominantly infected by gill flukes since it was observed 12 out of a total of 20 cases. With reference to seasonal fluctuation, Vibrio spp. infection was continually observed through all seasons but Edwardsiella septicaemia, Streptococcosis and Pseudotuberculosis was mainly isolated in summer and autumn when the water temperature was raised high. Scuticociliatida infection was frequently occurred from late autumn to early spring but gill flukes were watched all the year regardless of temperature. As regards, multiple infection, 47 out of 91 cases was Identified to have involved in more than 2 pathogens, especially Vibrio spp. was highly mixed with other pathogens since 42 out of 47 cases was mixed with Vibrio spp.
