Effect of Deer Blood on Aplastic Anemia Induced Mouse (녹혈이 빈혈유발 Mouse에 미치는 영향)

  • Hematopoietic stem cells in bone marrow form all kinds of blood cells. In traditional medicine, functions of bone marrow cells are very similar to those of Essence(精) which is a fundamental factor of physical development and reproduction. Our experiment examined the effect of deer blood on aplastic anemia induced mouse using cyclophosphamide 150 mg/kg i.p injection before experiment and then another cyclophosphamide 120 mg/kg i.p injection on day 10. Then we administrated dried deer blood in distilled water for 5 days, 9 days and 10 days. We examined blood and marrow samples. In results, deer blood showed a trend of effectiveness on recovery of red blood cells and erythropoietin although they were not statistical significant. And deer blood did not show changes in CD34.

A Clinical Study of the Live Blood Condition of Women′s Shoulder Measurement (여성 견비통 환자의 생혈액 형태에 대한 임상적인 연구)

  • Objectives : To study the Live Blood Condition of chronic shoulder measurement. Methods : Twenty-one patients were studied. The observation items of the Live Blood Condition are the form's abnormality and cohesion of red blood cells, the abnormality of hemoglobin and the toxicity in plasma. Results : 1. The form's abnormality and cohesion of red blood cells observed were erythrocyte aggregation (8 persons), Rouleau (3 persons), target cells (7 persons), ovalocytes (3 persons), poikilocytes (2 persons). Double conditions observed were erythrocyte aggregation & target cells (3 persons), erythrocyte aggregation & poikilocytes (1 person), target cells & ovalocytes (l person). 2. The abnormality of hemoglobin and the toxicity in plasma observed were cholesterol crystals (5 persons), atherosclerotic plaque (3 persons), chylous (6 persons). 3. The form normality of red blood was generally observed in one woman. The normality of hemoglobin and the non-toxicity in plasma were generally observed in seven women. Conclusions : According to this study of the Live Blood Condition of women's shoulder measurement, these results suggest that the twenty-one patients evidence the conditions of extravasated blood and phlegm.

Morphological Variation Classification of Red Blood Cells using Neural Network Model in the Peripheral Blood Images (말초혈액영상에서 신경망 모델을 이용한 적혈구의 형태학적 변이 분류)

  • Recently, there have been researches to automate processing and analysing images in the medical field using image processing technique, a fast communication network, and high performance hardware. In this paper, we propose a system to be able to analyze morphological abnormality of red-blood cells for peripheral blood image using image processing techniques. To do this, we segment red-blood cells in the blood image acquired from microscope with CCD camera and then extract UNL fourier features to classify them into 15 classes. We reduce the number of multi-variate features using PCA to construct a more efficient classifier. Our system has the best performance in recognition rate, compared with two other algorithms, LVQ3 and k-NN. So, we show that it can be applied to a pathological guided system.

  • This paper presents the development of a microsystem for whole blood purification and electrophysiological analysis of the purified cells. Magnetophoresis using continuous diamagnetic capture (DMC) was utilized for whole cell purification and electrical impedance spectroscopy (EIS) was utilized for electrophysiological analysis of the purified cells. The system was developed on silicon and plastic substrates utilizing conventional microfabrication technologies and plastic microfabrication technologies. Using the magnetophoretic microseparator, white blood cells were purified from a sample of whole blood. The experimental results of the DMC microseparator show that 89.7% of the red blood cells (RBCs) and 72.7% of the white blood cells (WBCs) could be continuously separated out from a whole blood using an external magnetic flux of 0.2 T. EIS was used as a downstream whole cell analysis tool to study the electrophysiological characteristics of purified cells. In this work, primary cultured bovine chromaffin cells and human red blood cells were characterized using EIS. Further analysis capabilities of the EIS were demonstrated by successfully obtaining unique impedance signatures for chromaffin cells based on the whole cell ion channel activity.

  • Detailed knowledge on the motion of blood cells flowing in micro-channels under simple shear flow and the influence of blood flow is essential to provide a better understanding on the blood rheological properties and blood cell aggregation. The microscopic behavior of red blood cell (RBCs) is numerically investigated using a fluid-structure interaction (FSI) method based on the Arbitrary-Lagrangian-Eulerian (ALE) approach and the dynamic mesh method (smoothing and remeshing) in FLUENT (ANSYS Inc., USA). The employed FSI method could be applied to the motions and deformations of a single blood cell and multiple blood cells, and the primary thrombogenesis caused by platelet aggregation. It is expected that, combined with a sophisticated large-scale computational technique, the simulation method will be useful for understanding the overall properties of blood flow from blood cellular level (microscopic) to the resulting rheological properties of blood as a mass (macroscopic).

  • Background: Fc receptor-mediated phagocytosis is a complex process involving the activation of kinases and phosphatases. FcgammaRIIB has been known to transduces inhibitory signals through an immunoreceptor tyrosine-based inhibitory motif (ITIM) in cytoplasmic domains. In this study, we examined the involvement of inositol-phosphatase in the Fc receptor-mediated phagocytosis. Methods: J774 cells were infected using vaccinia viral vector containing SH2 domain-containing inositol-phosphatase (SHIP) cDNA and stimulated with the sensitized sheep red blood cells. Results: Stimulation of J774 cells induced the tyrosine phosphorylation of SHIP which was maximal at 5 minutes. Phosphatidylinositol-3 (PI-3) kinase inhibitor (wortmannin) inhibits J774 cell phagocytosis of sensitized sheep red blood cells in a dose-dependent manner. Heterologious expression of SHIP in J774 cells inhibits phagocytosis of sensitized sheep red blood cells in a dose-dependency manner, but catalytically dead mutants of SHIP has no effect on phagocytosis. Conclusion: These results strongly suggest that the active signals mediated by PI-3 kinase are opposed by inhibitory signals through SHIP in the regulation of Fc receptor-mediated phagocytosis.

Comparison of Oxidative Stress in Red Blood Cells Induced by Hair Dyeing Application to Young Women (염모제 도포방법에 따른 인체 적혈구의 산화스트레스 비교)

  • To ascertain the effects of hair dyeing application on oxidative stress in human, a mixture of permanent black colored hair dye with the same amount of oxidant containing 6% hydrogen peroxide was used. A hair dyeing with contacting the scalp (conventional dyeing) and a hair dyeing with 3 to 4mm away from the scalp (alternative dyeing) were applied to each 15 young healthy women. Blood was taken from the brachial vein at two sampling times, just before and 6 hours alter the hair dyeing, and antioxidant enzyme activities and antioxidant contents were measured in red blood cells. After dyeing, malondialdehyde(MDA) contents for conventional dyeing group was shown to a tendence of more increased than alternative dyeing group. After dyeing, reduced glutathione (GSH) contents for conventional dyeing group was shown to a tendence of more decreased than alternative dyeing group. After dyeing, superoxide dismutase (SOD) and catalase (CAT) activities were significantly decreased in conventional dyeing group (p < 0.01), however, SOD and CAT activities were not significantly decreased in alternative dyeing group. After dyeing, there was no significant decrease in glutathione peroxidase (GPX) activity both for conventional dyeing group and alternative dyeing group. Therefore, after dyeing, the degree of oxidative stress in red blood cells for alternative dyeing group was appeared to be lower than conventional dyeing group.

Clinical Study on the Effect of Exterior Vascular Laser Irradiation Therapy by Live Blood Analysis

  • Objectives: The purpose of this study was to investigate the effect of exterior vascular laser irradiation therapy by live blood analysis. Methods: We analysed the changing forms of the live blood sample with a microscope before and after exterior vascular laser irradiation therapy of the blood. Results: Rouleau of red cells, erythrocyte aggregation of red cells, thrombocyte aggregation, uric acid crystals, red crystals, and protoplasts in blood were decreased significantly after exterior vascular laser irradiation therapy. Conclusions: This study suggests that exterior vascular laser irradiation will have positive effects in eliminating various effete matters in blood and will have efficacy in the treatment and prevention of cardiovascular system disorders and hyperlipidemia, caused by effete matters, or numbness and arthralgia caused by blood stagnancy and blood circulation disorder.

  • Conventional drug carriers such as liposomes, nanoparticles, polymer micelles, polymeric conjugate and lipid microemulsion for cancer chemotherapy shield normal tissues from toxic drugs to treat cancer cells in tumors. However, inaccurate tumor targeting uncontrolled drug release from the carriers and unwanted accumulation in healthy sites can limit treatment efficacy with current conventional drug carriers with insufficient concentrations of drugs in the tumors and unexpected side effects as a result. Sickle red blood cells show natural tumor preferential accumulation without any manipulation due to the adhesive interaction between molecular receptors on the membrane surface and counter-receptor on endothelial cells. In addition, structural changes of microvascular in tumor sites enhances polymerization of sickle red blood cells. In this research, we examined the use of sickle red blood cells as a new drug carrier with novel tumor targeting and controlled release properties to quantify its therapeutic effects.

Effect of He-Ne laser intravascular irradiation by live blood analysis (생혈구분석을 통한 He-Ne laser 정맥혈관내 조사의 효과)

  • Objectives : The purpose of this study was to investigate the effect of Intravascular Laser Irradiation of Blood(ILlB) by the live blood analysis. Methods : We had analysed the changing forms of the live blood samples with Ultra Darkfield Microscope before and after Intravascular Laser Irradiation of Blood. Results : 1. Somatid did not showed significant change. 2. In the rouleau of red blood cells was decreased significantly. 3. In the morphological change of red blood cells, Burr cell, Ovalocyte and Poikilocyte were decreased significantly, but Acanthocyte and Target cell were increased significantly. 4. In the abnormal matters in plasma, the Cholesterol cristal did not showed significant change, but the Aggregation of platelet, Lipids, Spicule, Leucocyte, Uric acid cristal did showed a little significant decrease. Conclusion : These findings suggest that live blood analysis is useful to judge the effect of treatment and diagnosis in oriental medicine, and with the effect of Intravascular Laser Irradiation of Blood, it had showed significant effect on rouleau of red blood cells, morphological change of red blood cells and abnormal matters in plasma.
