Inhibition of mIGF-1 and mGHR Gene Expression using Tetracycline-Inducible RNAi System in Mouse Liver Cell (Tetracycline 유도적인 RNAi System을 이용한 생쥐 성장 관련 유전자의 발현 억제)

  • In this study, to further understand the mechanism of animal growth and to develop a miniature transgenic animal model, we constructed and tested tetracycline-inducible RNAi system using shRNA targeting the mRNA of mouse insulin-like growth factor (mIGF-1) or mouse growth hormone receptor (mGHR) gene. Quantitative real-time PCR analysis of mouse liver cell (Hepa1c1c7) cells transfected with these vectors showed 85% or 90% of expression inhibition effect of IGF-1 or GHR, respectively. In ELISA analysis, the protein level of IGF-1 in the cells expressing the shRNA targeting IGF-1 mRNA was reduced to 26% of non-transformed control cells. Unexpectedly, in case of using shRNA targeting GHR, the IGF-1 protein level was decreased to 75% of control cells. Further experiments are needed to explain the lower interference effect of GHR shRNA in IGF-1 protein. Accumulated knowledge of this approach could be applicable to a variety of related biological area including gene function study, gene therapy, development of miniature animals, etc.

A Dynamic Inferential Framework for Learning Geometry Problem Solving (기하 문제 학습을 위한 동적 추론 체계)

  • In spite that the main contents of mathematical and scientific learning are understanding principles and their applications, most of existing educational softwares are based on rote learning, thus resulting in limited educational effects. In the artificial intelligence research, some progress has been made in developing automatic tutors based on proving and simulation, by adapting the techniques of knowledge representation, search and inference to the design of tutors. However, these tutors still fall short of being practical and the turor, even a prototype model, for learning problem solving is yet to come out. The geometry problem-solving tutor proposed by this research involves dynamic inference performed in parallel with learning. As an ontology for composing the problem space within a real-time setting, we have employed the notions of propositions, hypotheses and operators. Then we investigated the mechanism of interactive learning of problem solving in which the main target of inference involves the generation and the test of these components. Major accomplishment from this research is a practical model of a problem tutor embedded with a series of inference techniques for algebraic manipulation, which is indispensable in geometry problem solving but overlooked by previous research. The proposed model is expected to be applicable to the design of problem tutors in other scientific areas such as physics and electric circuitry.

Identification of Actinobacillus pleuropneumoniae Genes Preferentially Expressed During Infection Using In Vivo-Induced Antigen Technology (IVIAT)

  • Porcine pleuropneumonia is an infectious disease caused by Actinobacillus pleuropneumoniae. The identification of A. pleuropneumoniae genes, specially expressed in vivo, is a useful tool to reveal the mechanism of infection. IVIAT was used in this work to identify antigens expressed in vivo during A. pleuropneumoniae infection, using sera from individuals with chronic porcine pleuropneumonia. Sequencing of DNA inserts from positive clones showed 11 open reading frames with high homology to A. pleuropneumoniae genes. Based on sequence analysis, proteins encoded by these genes were involved in metabolism, replication, transcription regulation, and signal transduction. Moreover, three function-unknown proteins were also indentified in this work. Expression analysis using quantitative real-time PCR showed that most of the genes tested were up-regulated in vivo relative to their expression levels in vitro. IVI (in vivo-induced) genes that were amplified by PCR in different A. pleuropneumoniae strains showed that these genes could be detected in almost all of the strains. It is demonstrated that the identified IVI antigen may have important roles in the infection of A. pleuropneumoniae.

Inhibition of melanogenesis by sodium 2-mercaptoethanesulfonate

  • Sodium 2-mercaptoethanesulfonate (mesna) is a protective agent that is widely used in medicine because of its antioxidant effects. Recently, reactive oxygen species (ROS) were shown to increase pigmentation. Thus, ROS scavengers and inhibitors of ROS production may suppress melanogenesis. Forkhead box-O3a (FoxO3a) is an antimelanogenic factor that mediates ROS-induced skin pigmentation. In this study, we aimed to investigate the whitening effect of mesna and the signaling mechanism mediating this effect. Human melanoma (MNT-1) cells were used in this study. mRNA and protein expression were measured by real-time quantitative PCR and Western blotting analysis to track changes in FoxO3a-related signals induced by mesna. An immunofluorescence assay was performed to determine the nuclear translocation of FoxO3a. When MNT-1 melanoma cells were treated with mesna, melanin production and secretion decreased. These effects were accompanied by increases in FoxO3a activation and nuclear translocation, resulting in downregulation of four master genes of melanogenesis: MITF, TYR, TRP1, and TRP2. We found that mesna, an antioxidant and radical scavenger, suppresses melanin production and may therefore be a useful agent for the clinical treatment of hyperpigmentation disorders.

Epigallocatechin-3-Gallate (EGCG) Attenuates Traumatic Brain Injury by Inhibition of Edema Formation and Oxidative Stress

  • Traumatic brain injury (TBI) is a major cause of mortality and long-term disability, which can decrease quality of life. In spite of numerous studies suggesting that Epigallocatechin-3- gallate (EGCG) has been used as a therapeutic agent for a broad range of disorders, the effect of EGCG on TBI remains unknown. In this study, a weight drop model was established to evaluate the therapeutic potential of EGCG on TBI. Rats were administered with 100 mg/kg EGCG or PBS intraperitoneally. At different times following trauma, rats were sacrificed for analysis. It was found that EGCG (100 mg/kg, i.p.) treatment significantly reduced brain water content and vascular permeability at 12, 24, 48, 72 hour after TBI. Real-time PCR results revealed that EGCG inhibited TBI-induced IL-$1{\beta}$ and TNF-${\alpha}$ mRNA expression. Importantly, CD68 mRNA expression decreasing in the brain suggested that EGCG inhibited microglia activation. Western blotting and immunohistochemistry results showed that administering of EGCG significantly inhibited the levels of aquaporin-4 (AQP4) and glial fibrillary acidic protein (GFAP) expression. TBI-induced oxidative stress was remarkably impaired by EGCG treatment, which elevated the activities of SOD and GSH-PX. Conversely, EGCG significantly reduced the contents of MDA after TBI. In addition, EGCG decreased TBI-induced NADPH oxidase activation through inhibition of $p47^{phox}$ translocation from cytoplasm to plasma membrane. These data demonstrate that EGCG treatment may be an effective therapeutic strategy for TBI and the underlying mechanism involves inhibition of oxidative stress.

  • In the paper, we divide the ATM cells generated by multimedia systems that want to transmit multimedia informations over B-ISDN into two categories:i)a media cell with a media information and ii) a synchronization cell with a synchronization information. We induce a media cell loss equation and a synchroni- zation cell loss equation based on probability distribution functions with on-off source as an input traffic model. In order to meet the requirements of real-time and integrity of multimedia informations, multimedia synchronization cells should provide the delay-sensitive and the loss-sensitive requirements. A traffic control mechanism needs to satisfy above requirements. According to the performance evaluation by a traffic control model simulation, we describe both spatial priority for minimizing the synchronization cell loss and temporal priority for reducing the synchronization cell delay must be applied simultaneously.

  • A 2U cube satellite called SNUGLITE has been developed by GNSS Research Laboratory in Seoul National University. Its main mission is to perform actual operation by mounting dual-frequency global positioning system (GPS) receivers. Its scientific mission aims to observe space environments and collect data. It is essential for a cube satellite to control an Earth-oriented attitude for reliable and successful data transmission and reception. To this end, an attitude estimation and control algorithm, Attitude Determination and Control System (ADCS), has been implemented in the on-board computer (OBC) processor in real time. In this paper, the Extended Kalman Filter (EKF) was employed as the attitude estimation algorithm. For the attitude control technique, the Linear Quadratic Gaussian (LQG) was utilized. The algorithm was verified through the processor in the loop simulation (PILS) procedure. To validate the ADCS algorithm in the ground, the experimental verification via a single axis Hardware-in-the-loop simulation (HILS) was used due to the simplicity and cost effectiveness, rather than using the 3-axis HILS verification (Schwartz et al. 2003) with complex air-bearing mechanism design and high cost.

MiR-675 Promotes the Growth of Hepatocellular Carcinoma Cells Through Cdc25A Pathway

  • Background: MicroRNAs (miRNAs) have fundamental roles in tumorigenesis. MiR-675 is upregulated in hepatocellular carcinoma(HCC) cells. However, the roles of miR-675 in hepatocellular carcinogenesis are still not fully elucidated. In this study, we focus on investigating the effect and mechanism of miR-675 in proliferation of HCC cells. Materials and Methods: The cell proliferation was measured by MTT assays after transfection with miR-675 inhibitor and miR-675 mimics in HCC cells. The expression level of miR-675 was detected by real-time quantitative reverse transcription polymerase chain reaction. Protein expression of Cdc25A was measured by western blotting analysis. Results: In MTT assays, overexpression of miR-675 promoted the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675mimics control group. Downexpression of miR-675 inhibited the proliferation of HCC cells(P<0.05. at 48 hours, P<0.01. at 72 hours) compared with the miR-675inhibitor control group. In western blotting analysis, the expression level of Cdc25A was significantly increased (p<0.05) after treatment with miR-675 mimics. The expression level of Cdc25A was significantly decreased (p<0.05) after treatment with miR-675 inhibitor. Conclusions: Our results indicate that miR-675 promotes the proliferation in human hepatocellular carcinoma cells by associating with Cdc25A signaling pathway.

Nonlinear Seismic Estimates of Recorded and Simulated Ground Motions Normalized by the Seismic Design Spectrum (설계용 탄성응답스펙트럼으로 규준화된 인공지진동과 기록지진동의 비선형 지진응답)

  • In the nonlinear response history analysis of building structures, the input ground accelerations have considerable effect on the nonlinear response characteristics of structural systems. As the properties of the ground motion, using time history analysis, are interrelated with many factors such as the fault mechanism, the seismic wave propagation from source to site, and the amplification characteristics of the soil, it is difficult to properly select the input ground motions for seismic response analysis. In this paper, the most unfavourable real seismic design ground motions were selected as input motions. The artificial earthquake waves were generated according to these earthquake events. The artificial waves have identical phase angles to the recorded earthquake waves, and their overall response spectra are compatible with the seismic design spectrum with 5% of critical viscous damping. It is concluded that the artificial earthquake waves simulated in this paper are applicable as input ground motions for a seismic response analysis of building structures.

Therefore, this paper will realize disaster recovery system, which is suspended because of disaster, based on cloud computing system. Realized High Available Replicate System applied a method of multi thread target database to improve Replicate performance, and real time synchronize technology can improve efficiency of network. From Active to Active operation, it maximizes use of backup system, and it has a effect to disperse load of source database system. Also, High Available Replicate System realized consistency verification mechanism and monitoring technique. For Performance evaluation, High Available Replicate System used multi thread method, which shows more than threefold of replicate performance than single thread method.
