Aspergillus awamori와 Zymomonas mobilis로 구성된 혼합고정화 배양계의 최적 조건 (Optimal Conditions of Co-Immobilized Mixed Culture System with Aspergillus awamori and Zymomonas mobilis)

  • Gel bead의 효율적인 이용을 목적으로 산소요구성이 전혀 다른 두 균주로 구성된 혼합고정화 배양계의 개발을 시도하였다. 호기성 균으로서 A. awamori, 혐기성균으로서 Z. mobilis를 사용하여 생전분으로 부터 에탄올 생산을 행한 결과는 다음과 같다. 두 균주의 최적 혼합비율은 A. awamori $1.25{\times}10^{9}\;spore/L-gel$, Z. mobilis 0.5g cell/L-gel이었다. 배양이 완료된 후의 gel bead 내의 균주 분포는 각각 혐기부와 호기부, 즉 gel bead의 표면부와 중심부로 생육장소가 구분되어 있었다. A-Z계의 배양에서는 에탄올의 수율이 낮았고, 균사가 gel bead로 부터 누출되었다. 배양 개시 후 36시간째에 배양기의 면전을 특수 제조한 check valve가 부착된 실리콘 plug로 교환하여 혐기적 배양을 행한 A-Z 36계에서는 gel bead로 부터 균사의 누출이 상당히 억제되었고, pH가 4.3을 유지하면서, 에탄올의 수율이 대조구 보다 약2배 높게 나타났다.

펄프에 함유된 $\alpha$-셀룰로오스의 함량이 셀룰로오스의 에스테르화반응에 미치는 영향 (Effect of $\alpha$-cellulose content in pulps on esterification of cellulose)

  • Cellulose triacetate (CTA) was prepared from cotton linter and pulps which contain various contents of $\alpha$-cellulose. CTA which contains 2.8 of degree of substitution (DS) and 222 of degree of polymerization (DP) was obtained from V-81 pulp under the heterogeneous system. The DS was measured by the titration method, and the DP was obtained by measurement of viscosity. FT-IR spectometer (FT-IR 6300, JASCO) was used to analyze the chemical structure of raw materials and cellulose triacetate, and X-ray diffractometer (X-pert MPD PW3040, Philips) was used to confirm the crystal structure and to calculate the relative crystallinity index (RCI). As $\alpha$-cellulose content in pulp increased, the acetylation yield increased. Besides with a kind of pulp, it contains insoluble residue which was mainly formed due to the formation of glucomannan triacetate and xylan diacetate during the esterification.

  • 본보는 Polyethylene대에 저견한 고치와 면대에 저견한 것을 수개월간에 걸쳐서 조사시험한 것으로서 그 결과는 다음과 같다. 1. 살용견은 재건시까지 Polyethylen대에 저장하여 옥외에 방치할 수 있다. 2. Polyethylene대에 저견한 것은 면대저견한 것의 생계량비율보다 약 0.5% 증가되었다. 3. 본 대장으로 곰팡이 해를 방지할 수 있었다. 그러나 서해는 방지할 수 없었다. 4. 본 대장견은 면포대장견보다 수시렁이 유인이 다소 있으나 나프타린 방충제를 대대직전에 견대에 넣으므로서 방지할 수 있다.

Evaluation of effects of textile wastewater on the quality of cotton fabric dye

  • In this study, reuse of biologically treated wastewater of denim washing and dyeing industry has been evaluated by membrane technologies. After that experiments were carried out at laboratory scale in textile dyeing unit by using obtained permeate water samples on 100% cotton based raw fabric belonging to examined industry. During membrane experiments, two different UF (UC100 and UC030) and two different NF (NP010 and NP030) were evaluated under alternative membrane pressures. In permeate water obtained on selected samples, conductivity at the range of $1860-2205{\mu}S/cm$, hardness at the range of 60 to 80 mg/L, total color at the range of 2.4 to 7.6 m-1 and COD at the range of 25-32 mg/L was determined. The following analyzes were performed for the dyed fabrics: perspiration fastness, rub fastness, wash fastness, color fastness to water, color fastness to artificial light, color measurement through the fabric. According to analysis results, selected permeate water have no negative impact on dyeing quality. The study showed that membrane filtration gave good performance for biologically treated textile wastewater, and NF treatment with UF pre-treatment was suitable option for reuse of the effluents.

출토복식을 통해본 17세기 봉제구성 -임경백.임계백 묘 중심- (Sewing Construction of the 17th Century-Clothing Through the Excavated Costume - Based on Tombs in O chang -)

  • This study is on the sewing construction of the 17th century clothing through the Excavated Costumes in O-chang, ChungBuk in 1998. The followings are the results of this study. There are various sewing constructions in this excavates costume : a running stitch.a back stitch.a half back stitch.a french seam.a gobsol(korean traditional seam sewing). We can see that the sewing construction of the 17th century-clothing depends on construction styles : one layer, two layers, clothing padded with cotton(include quilt). We can see that sewing construction in the one taler clothing consisted of a running stitch, a french seam, a gobsol seam. In the two layer clothing the construction included a running stitch and half back stitch. a running switch and a decorated top stitch(3stitch) were used in the clothing padded with cotton. We can see that the sewing construction enhanced the function of keeping the shape of clothing and the raw edges. We can see that the shape of clothing is important in presentation of aesthetic of the clothing. Such a kind of sewing construction lasted from the early Chosen Dynasty through the 17th century.

  • Effect of the pH condition was examined on extraction and dyeing of the yellow and red dyes of safflower. Absorption of dye solution extracted from safflower was changed by pH of solvent. In the case of yellow dyes, cotton and ramie weren't dyed regardless of pH of dye bath, but silks were significantly dyed at pH4. Raw silk showed better dyeability than refined silk. In the case of red dyes, cotton and ramie absorbed red dyes selectively, but silks absorbed yellow dyes more significantly than red dyes. Efficiency of extraction and dyeing of yellow dyes in acidic range were find out higher than that in neutral range. Red dyes was effectively extracted at pH11 and dyed at pH6.

외나로도지역의 의생활 (Dressing Practices of Residents at the Woinarodo Region)

  • The purpose of this study is to look into dressing practices at the Woinarodo region in terms of ordinary and ritual clothes. Men wore Bqji(trousers) and Jeokori(jackets) as their plain clothes and sometimes Jangsam mid Durumaki(topcoat). During the period of Japanese colony, men wore Western-style clothes. For women it was basic to wear Chima(skirts) and Jeokori. And they preferred Momppe rather when in Japanese rule. In arrangements for their head, men put on gut, and had their hair cut during Japanese nile. Women laid a bundle of their braided hairs on the head or braided their hair, while married women did their hair up in a chignon during the ruling period People of the region put on straw and leather shoes, and then rubber ones since the late 1930s. Hand weaving was a major means of living for women at the region. Ramie, hemp and cotton were mainly weaved by hand. Starching was applied mainly to ramie and cotton. Glues for starching were made of raw rices, cooked rices, wheat flour or gloiopeltis tenax. For ritual clothes, especially in wedding, bridegrooms arranged themselves with Samokwandae and then Put on Baji, Jeokori, Durumaki and Danryung. But they Put on Western-style dresses as the liners of Danryung, and wear Nambawi Rather than the Samo after korean independence from Japanese rule. Bridges wore Chima, Jeokori and Wonsam and Chokdoori and covered their face with Hansam Wonsam did not be worn any longer after Korean independence from the rule. Shrouds for funeral ceremony were manufactured with silks, cotton and hemp, when the chief mourner wore hempen hoods and funeral robes, while women, Chima, made of hemp. and any type of Jeokori.

시판 쪽 분말염료의 색소 함량에 따른 면직물의 색상 및 항균성 비교연구 (Analysis of the Pigment Contents of Commercial Indigo Powders and Their Effect on the Color and the Antimicrobial Function of Dyed Cotton Fabrics)

  • Market available fermented indigo powders of Indian origin (FI1, FI2), Chinese origin (FC1, FC2), and raw indigo powders of Indian origin (R1, R2) were examined using TLC and HPLC analyses to investigate their pigment contents. TLC analysis gave $R_f$ values of 0.81 and 0.72 for blue and red pigments, respectively. All the powder products and the synthetic and natural indigo standards eluted at 6.9 min and 8.3 min in the HPLC chromatograms, and the peaks showed the ${\lambda}_{max}$ at 610nm and 542nm, representing indigotin and indirubin, respectively. The pigment content calculated based on the area of indigotin and indirubin peaks in the HPLC chromatograms showed that the indigotin content was higher in FC1 and FC2, while FI1 and FI2 had a higher indirubin content. The relative percentage of indirubin was the highest in R2, but the HPLC peak intensity was quite low. Despite the higher indigotin content in FC1 and FC2, cotton dyed with FI1 and FI2 (versus cotton dyed with FC1 and FC2) showed a higher blue (B) hue, the highest K/S values, and the highest antimicrobial effect.

A Nutritional Evaluation on Whole Cottonseed Removed Germination Ability by Heat-treatment

  • In Korea, wide spread use of whole cottonseed, which is primarily a GMO plant imported from foreign countries and being fed to animals as raw state, has aroused concern that it may disturb the existing ecology of the country unless dispersion of the seed is under proper control. The objective of this study was to elucidate the changes in various nutritive parameters due to heat treatment and to determine the effective condition for removing germination ability of whole cottonseed (WCS). Of the various temperatures applied (76, 78, 80, 85, $100^{\circ}C$/30 min) $85^{\circ}C$ for 30 min was confirmed to be the lowest temperature treatment which resulted in a complete removal of the germination ability of WCS. Therefore, based on the determined temperature condition ($85^{\circ}C$ 30 min) we tried to examine the changes of various nutritional parameters, including nutrient composition, in vitro digestibilities and ruminal protein degradabilities, comparing raw whole cotton seed (RWCS) and heated whole cotton seed (HWCS). Some changes in amino acid composition were observed with heat treatment of WCS, but these were regarded to originate from the variation in plant quality and seed morphology, which are usually affected by different environmental factors during the vegetation period. As for fatty acid composition, no significant differences were observed to occur during heat treatment. However, WCS heated at $85^{\circ}C$ for 30 min in a circulating oven showed a significant decrease (p<0.05) of in situ rumen degradability in both dry matter (DM) and crude protein (CP), as compared to raw WCS. Overall results obtained in the study indicate that the heating condition used in this study, which was proven to be the most appropriate and economic to remove germination ability of WCS, may also improve the nutritional value of the ruminant with regard to reducing its protein degradability within the rumen.

Relative Performance of Fattening Lambs on Raw and Processed Cottonseed Meal Incorporated Diets

  • In order to find the feasibility of feeding raw or processed cotton (Gossypium) seed meal (CSM), 30 male crossbred lambs were randomly assigned to 5 dietary treatments with 6 animals each. Lambs were fed each of the isonitrogenous and isocaloric concentrate mixtures containing 30% deoiled groundnut cake (DGNC) (reference diet), 40% raw, 45 minute cooked, 1% $Ca(OH)_2$ or iron (1 part free gossypol:0.3 parts iron) treated CSM (replacing about 50% nitrogen moiety of reference concentrate mixture). The concentrate mixture was fed to meet 80% of CP requirements (NRC, 1985) along with ad libitum maize (Zea mays) hay for 180 days. Incorporation of raw or processed CSM did not affect the palatability of diets as evidenced by higher (p<0.01) or comparable overall daily intakes of DM and protein (CP and Digestible CP) per kg $W^{0.75}$ by lambs in comparison to reference group. Intakes of DE and ME increased (p<0.01) due to inclusion $Ca(OH)_2$ treated CSM in diets, while the intakes were lower (p<0.01) on iron treated CSM based diet when compared with reference diet. The intakes of DM, CP, TDN, DE and ME by reference and experimental lambs were higher than the requirements stipulated by NRC (1985). The growth rate was highest (p<0.01) in lambs fed on diets with cooked CSM followed by those fed raw or $Ca(OH)_2$ treated CSM based diets. The growth of lambs fed on iron treated CSM diets was depressed (p<0.01) in comparison to that of other experimental lambs probably due to increased iron intake (889 mg/kg DM of diet) which was much higher than the toxic levels of 500 ppm (NRC, 1980). The lambs on various diets utilized DM with similar efficiency while the utilization of energy (TDN, DE and ME) was more efficient (p<0.01) when the diets contained raw or cooked CSM in comparison to that of reference diet. The lambs fed raw and $Ca(OH)_2$ treated CSM incorporated diets utilized DCP with higher (p<0.01) efficiency than the lambs fed reference diet. The feed cost per kg weight gain was lower (p<0.01) on raw, cooked and $Ca(OH)_2$ treated CSM based diets in comparison to reference diet. Cooking of CSM for 45 minutes further reduced the feed cost of weight gain.
