The Effects of Treadmill Training on Neurotrophins and Immediately Early Protein in Obese Rats (트레드밀 트레이닝이 비만 쥐의 neurotrophins와 초기발현 단백질에 미치는 영향)

  • The purpose of this study was to investigate the biological effect of obesity-induced oxidative damage on neurogenesis and early protein expression. Obesity was induced I thirty 4-week old male Sprague-Dawley rats through a high fat diet for 15 weeks. After one week of environmental adaptation, the rats were divided into 2 groups: high fat diet sedentary group (HDS, n=15) and high fat diet training group (HDT, n=15). Exercise training was performed 5 times a week for 8 weeks, with mild-intensity treadmill running for weeks 1-4 and moderate-intensity treadmill running for weeks 5-8. After the 8 week training period, we analyzed lipid profiles, serum 8-hydroxyguanosine (8-OHdG), liver tissue malondialdehyde (MDA) related to oxidative damage factors, nerve growth factor (NGF), brain derived neurotrophic factor (BDNF), c-fos, c-jun, and extracellular signal regulated kinase (Erk) in the hippocampus. The results of this study are as follows. There were differences between HDS and HDT in triglyceride (TG) and total cholesterol (TC) (p<0.05). In high density lipoprotein (HDL-c), the HDT was higher than HDS after treadmill training (p<0.05). In 8-OHdG, the HDT was lower than HDS after treadmill training (p<0.05). Genetic expressions of c-jun, BDNF and MDA in the HDT were higher than in the HDS after treadmill training in hippocampus (p<0.05). Therefore, we conclude that 8 weeks of treadmill training can improve imbalanced lipid profiles, reduce oxidative damage, and activate neurogenesis in obese rats.

  • 1. Objectives This study was carried out to investigate the effects of Gunyuljejo-tang on the $CCl_4$-induced Liver Damage in Rats. 2. Methods Sprague-Dawley rats were devided into 5 experimental groups : Normal, $NS+CCl_4$(Solid extract of $CCl_4$ injection group after Normal Saline feed), $GYJJT+CCl_4$(Solid extract of $CCl_4$ injection group after Gunyuljejo-tang feed), $CCl_4+NS$(Normal Saline feed group after $CCl_4$ injection), $CCl_4+GYJJT$(Solid extract of Gunyuljejo-tang feed group after $CCl_4$ injection). Biochemical assays for serum enzyme activities such as AST, ALT, ALP, BUN, Creatinine, Uric Acid, Total Protein, Albumin, Total Cholesterol, Triglyceride, Glucose, and mRNA Revelation of Cytochrome p450 and activities such as LPO, GSH, GST, Glutathione Reductase, Glutathione Peroxidase, SOD, Catalase, Hydroxyproline, and ${\beta}$-Glucuronidase were performed. 3. Results (1) $GYJJT+CCl_4$ showed lower revelation of Cytochrome p450. (2) $GYJJT+CCl_4$ showed higher GSH activity than $NS+CCl_4$, $CCl_4+GYJJT$ showed higher GSH activity than $CCl_4+NS$ injection significantly. (3) $GYJJT+CCl_4$ showed higher GST activity than $NS+CCl_4$. $CCl_4+GYJJT$ showed higher GST activity than $CCl_4+NS$ significantly. (4) $GYJJT+CCl_4$ showed higher Glutathione Peroxidase activity than $NS+CCl_4$, $CCl_4+GYJJT$ showed higher Glutathione Peroxidase activity than $CCl_4+NS$ significantly. (5) $CCl_4+GYJJT$ showed higher SOD activity than $CCl_4+NS$ significantly. (6) $CCl_4+GYJJT$ showed higher Catalase activity than $CCl_4+NS$ significantly. (7) $GYJJT+CCl_4$ showed lower Hydroxyproline than $NS+CCl_4$ significantly, $CCl_4+GYJJT$ showed higher Hydroxyproline than $CCl_4+NS$ significantly. (8) $GYJJT+CCl_4$ showed higher ${\beta}$-Glucuronidase activity than $NS+CCl_4$, $CCl_4+GYJJT$ showed higher ${\beta}$-Glucuronidase activity than $CCl_4+NS$ significantly. 4. Conclusions Gunyuljejo-tang has the recovering effects on the $CCl_4$-induced Liver Damage significantly.

Effect of Cassia tora Ethanol Extracts on Carbon Tetrachloride-Induced Liver Injury in Rats (흰쥐에 있어서 사염화탄소 유발 간손상에 미치는 결명자 에탄올 추출물의 영향)

  • This study was conducted to investigate the effect of Cassia tora ethanol extracts on carbon tetrachloride($CCl_4$)-induced hepatotoxicity in rats. Male Sprague-Dawley rats were divided into the following 4 groups: normal group, $CCl_4$, treated group, $CCl_4-0.25%$ Cassia tora ethanol extracts group and $CCl_4-0.5%$ Cassia tora ethanol extracts group. Rats were fed with each experimental diet and water for 5 weeks. Liver weights of rats treated only with $CCl_4$, were significantly increased compared to normal group, but not in rats fed diet containing Cassia tora ethanol extracts. Cholesterol and triglyceride contents in serum and liver were also not influenced by either $CCl_4$, treatment or the supplementation of Cassia tora ethanol extracts. $CCl_4$, treatment significantly increased ALP activities, however the supplementation of Cassia tora ethanol extracts significantly decreased the activities of serum ALT, AST, ${\gamma}-GTP$ in dose-dependent manner. Cassia tora ethanol extracts significantly reduced $CCl_4-induced$ elevation of liver TBARS contents. Activities of superoxide dismutase and catalase were decreased by $CCl_4$, treatment, however by the supplement of Cassia tora ethanol extracts slightly increased activities of SOD and catalase. The activity of glutathione peroxidase in groups fed diets containing Cassia tora ethanol extracts was significantly decreased compared to that of the control group. These results suggest that Cassia tora ethanol extracts may exert protective effect against $CCl_4-induced$ liver injury through the prevention of lipid peroxidation.

A Study on the Effects of Magnetic Field and BEP on Hyperlipidemia and Enzymatic Activities in Rats (자기장(磁氣場) 및 BEP 조사(照射)가 식이성(食餌性) 고지혈증(高脂血症)이 유발(誘發)된 흰쥐에 미치는 영향(影響)에 대(對)한 연구(硏究))

  • It has been known that Ki(氣) energy is very effective on many adult diseases. Oriental Medicine has acknowledged Ki as an existing reality and investigated its effects on the body. However, the existence of Ki has not been fully explained. In order to find a conclusive evidence on the existence of Ki, this experiment was done to study the mutual relationship of Ki with a magnetic field and BEP (biological energy projector). The BEP apparatus was irradiated under the magnetic field on rats in the hyperlipidemic induced state. Following criterias were measured in this experiment: weight change, weight of the visceral organs, serum, hepatic lipid peroxide, bleeding time, tissue factor, and etc. The following results were obtained in this study: 1. The weight of rat significantly decreased in the magnetic field treated group and radically reduced in the group treated with both magnetic field and BEP. 2. The weight of liver, heart, and kidney increased in both the magnetic field treated group and magnetic field+BEP group compared to the normal group, but decreased in comparison to the control group. No changes were witnessed in the weight of spleen. 3. Serum and hepatic total cholesterol, total lipid, and lipid peroxide level significantly decreased in both magnetic field treated group and magnetic field+BEP treated group, while lipase activity has increased noticeably. 4. Serum HDL showed a significant increase in both magnetic field treated group and magnetic field+BEP treated group compared to the control group, while LDL and VLDL level decreased significantly. 5. A bleeding time significantly increased in both magnetic field treated group and magnetic field+BEP treated group compared to the control group. A tissue factor value of the lung decreased in the magnetic field treated group and magnetic field+BEP treated groups while increased in the control group. 6. Serum and hepatic lipid peroxide and glutathione level were significantly decreased in the magnetic field treated group and magnetic field+BEP treated group, while hepatic glutathione level was significantly increased compared to the control group. 7. A significant increase was found in the serum hydroxyl radical and SOD activity in the dietary hyperlipidemic rats, and significant decrease was found in the serum lipid peroxide content and superoxidase activity. 8. Hepatic cytosolic enzyme xanthine oxidase and aldehyde oxidase showed a significant decrease in the magnetic field treated group and magnetic field+BEP treated group. Through the above experimental results, one can suggest that the magnetic field with BEP can suppress hyperlipidemia and boost lipid metabolism and restructuring a lipid in liver, which increases the function of liver. To conclude, BEP is considered to show more potent effects under the exposure of magnetic field because magnetic field seems to increase the flow of Ki in the body.

Results 1) $CP+CCI_4$ showed significantly lower relation of Cytochrome p450 than $NS+CCI_4$. 2) As to LPO Hydroxyproline, $CCI_4+CP$ showed significantly lower activity than $CCI_4+NS$. 3) As to GSH GST Glutathione Peroxidase Catalase, $CP+CCI_4$ showed higher activity than $NS+CCI_4$, $CCI_4+CP$ showed significantly higher activity than $CCI_4+NS$. 4) As to Glutathione Reductase SOD, $CCI_4+CP$ showed significantly higher activity than $CCI_4+NS$. 5) As to ${\beta}-Glucuronidase$, $CP+CCI_4$ showed signficantly lower activity than $NS+CCI_4$. 4. Conclusions Chungpyesagantang has the recovering effects on the $CCI_4-induced$ Liver Damage.

The Effect of Polyphenols from Safflower Seed on HMG-CoA Reductase (HMGR) Activity, LDL Oxidation and Apo A1 Secretion (홍화씨 폴리페놀이 HMG-CoA reductase, LDL 산화 및 Apo A1 분비에 미치는 영향)

  • This study was conducted to examine the effect of polyphenols from safflower seed on HMG-CoA reductase (HMGR) activity, LDL oxidation and Apo A1 secretion from Hep3B cell. The safflower seed polyphenols were matairesinol (Iignan), enterolactone (lignan metabolite), acacetin (flavone) and serotonin derivative. In addition to safflower polyphenols, mevastatin, ${\alpha}-estradiol,\;{\alpha}-tocopherol$ and soy genistein were tested as reference compounds depending on the type of the test. HMGR source was liver microsome obtained from rat fed 2% cholestyramine for 10 days. Inhibition of HMGR activity was greater with mevastatin (53%) than safflower serotonin derivatives (45%), followed by genistein (35%), but was very small with matairesinol, enterolactone and acacetin. LDL oxidation induced by $CuSO_4$ was suppressed by all the test material used in the present study and in the order of safflower serotonin derivatives> matairesinol > ${\beta}-estradiol$ > genistein > acacetin > enterolactone. Apo A1 secretion from Hep3B cell was significantly stimulated by mevastatin, but moderately (p<0.1) by ${\beta}-estradiol$ and genistein as well as enterolactone. These results suggest that the safflower polyphenols improve body lipid status via inhibition of cholesterol synthesis and suppression of LDL oxidation.

The Effects of Simvastatin on Bone Healing in Mandible Fractured Rats. (백서의 하악골 골절 치유에서 Simvastatin이 미치는 영향)

  • Purpose: The hydroxymethylglutaryl coenzyme A reductase inhibitors (statins) are widely used in the treatment of dyslipidemia for the lowering of cholesterol. And studies about simvastatins have been shown to enhance bone formation in vitro and in vivo in rodents. But some other researchers have reported that there was no anabolic effect abouts simvastatins on bone. The peripheral distribution beyond the liver represents a small fraction of an orally administered dose. We hypothesize that this poor peripheral distribution is the likely reason that simvastatins, yield ambiguous results as anabolic agents. We therefore investigated whether the effects of simvastatins on bone may be enhanced by subcutaneous administration, providing better peripheral delivery of these drugs. Methods: 36 rat unilaterally mandible fractured models were prepared and divided into two groups. The simvastatin treated group where 1 mg/kg of simvastatin was daily injected subcutaneously. The same dose of normal saline was injected on the control group. And 3 rats in each group were sacrificed and taken bone samples in each week. Bone sample was evaluated with tensile strength and histological morphology after 1, 2, 3, 4, 5 and 6 weeks. Results: In simvastatin treated group, the fracture healing process, chondrocyte aggregation, collagen formation and trabecular bone formation was rapidly proceeded than the control group in histologically. The tensile strength of the simvastatin treated group was 1.02, 2.25, 3.95, 4.42, 5.49 and $6.00N/mm^2$ by weeks. The control group data was 0.60, 1.05, 2.17, 3.75, 4.15 and $5.17N/mm^2$ by weeks. The average tensile strength was higher by $1.04N/mm^2$ in simvastatin treated group. Conclusion: The currently available data on the effects of simvastatin on bone has done to confirm the finding that simvastatin helps fracture healing. And the potential for simvastatin to be used as anabolic agents for bone when delivered by the subcutaneous route.

Inhibitory Effects of Ethanol Extract of Purple Solanum tuberosum L. on Obesity and Hyperlipidemia (자색 마령서 에탄올 추출물의 비만 및 고지혈증 억제효능)

  • This experimental study was performed to evaluate the effects of ehtanol extract of Purple Solanum tuberosum L(EPSTL) on obesity and hyperlipidemia in rats induced by high fat diet. Sprague-Dawley rats were divided into normal group, control group, EPSTL (100 mg/kg and 200 mg/kg) treated group. EPSTL was orally administered to the obese rats by high fat diet for 6 weeks. The effect of EPSTL on obesity and hyperlipidemia were examined in vitro and in vivo as follows : EPSTL effectively inhibited the differentiation of 3T3-L1 adipocytes in a concentration dependent manner. EPSTL significantly lowered body weight of rats fe high fat diet for 6 weeks from 4 weeks treatment compared with untreated control. EPSTL significantly reduced the weight of retroperitoneal fat only at 100 mg/kg and epididymal fat pad in rats fed high fat diet for 4 weeks at 100 and 200 mg/kg. EPSTL significantly reduced the level of triglyceride at 200 mg/kg, while EPSTL tended to lower the levels of total lipid and phospholipid in rats fed high fat diet for 4 weeks with no significance. EPSTL significantly attenuated the level of total cholesterol, LDL and atherosclerosis index (Al) as well as increased the level of HDL at 200 mg/kg. EPSTL significantly reduced the levels of total lipid and triglyceride of hepatic tissues in rats fed high fat diet for 4 weeks at 200 mg/kg. EPSTL significantly lowered the level of insulin, while it did not affect leptin compared with untreated control. Taken together, these results suggest that EPSTL can be used for the treatment or prevention of hyperlipidemia and obesity.

Hepatoprotective Effects of Lycium chinense Mill Fruit Extracts and Fresh Fruit Juice (구기자 추출물과 생과즙의 간독성 보호효과)

  • Hepatoprotective effects of Bulro Kugi (Lycium chinense Mill) fruit extracts on $CCl_4-administered$ rats were investigated in vivo. Administration of $CCl_4$ increased plasma glutamic oxalacetic transaminase (GOT), glutamic pyruvic transaminase (GPT), and lactate dehydrogenase (LDH) activities, induced lipid peroxidation as measured by malondialdehyde (MDA) content of rat liver, and significantly increased liver weight. Feeding of B. Kugi (Lycium chinense Mill) slightly increased body weight gain, although not significantly different from normal group. B. Kugi (Lycium chinense Mill) fruit extracts reduced blood cholesterol level and inhibited $CCl_4-induced$ increases of plasma GPT, GOT, and LDH activities, whereas increased contents of MDA and cytochrome P-450, and GST activity in liver tissue of $CCl_4-administered$ rats. Roasted B. Kugi (Lycium chinense Mill) fruit extract showed highest hepatoprotective effect among samples tested. These results suggest water extracts of B. Kugi (Lycium chinense Mill) fruit possess promising hepatoprotective activity against $CCl_4-induced$ hepatic damage in rats.

Effect of Artemisia capillaris and Paecilomyces japonica on the Reduction of Hepatotoxicity and Lipid Metabolism Induced by Ethanol (에탄올 투여 흰쥐의 지질대사 및 간 독성 저하에 미치는 인진쑥 및 눈꽃동충하초의 영향)

  • The purpose of this study was to identify the inhibitory effect of hepatic toxicity and liver lipid metabolism after administration of Artemisia capillaris and Paecilomyces japonica. Along with the control, SD rats were divided into ethanol treated group with subgroups of 6% Artemisia capillaries (6A), 4% Artemisia capillaris+2% Paecilomyces japonica (4A2P), 3% Artemisia capillaris+3% Paecilomyces japonica (3A3P), 2% Artemisia capillaris+4% Paecilomyces japonica (2A4P) and 6% Paecilomyces japonica (6P). In this study we also intended to verify the optimum ratio of Artemisia capillaris and Paecilomyces japonica which can reduce hepatotoxicity. Artemisia capillaris and Paecilomyces japonica reduced cholesterol and triglyceride levels which were increased by ethanol. Total-cholesterol level was decreased the most in the groups of 4A2P and 3A3P. On the other hand, activity of superoxide dismutase (SOD) was enhanced significantly (p<0.05). Malondialdehyde (MDA) activity was decreased significantly (p<0.05) in the subgroup of 6A and 4A2P. When the ratio of Artemisia capillaris and Paecilomyces japonica was 2:1, the improvement of the rat serum and liver lipid metabolism and the alleviation of hepatic damage by ethanol were the most effective in this study. Therefore, it can be considered that the symptoms of severe chemically induced hepatotoxicity could be reduced by Artemisia capillaris and Paecilomyces japonica administration.
