Anti-hyperlipidemic Effect of Sophorae Fructus on Hyperlipidemic Rat Induced by High Fat-rich Diet (회화나무 열매의 고지혈증 개선 효능)

  • Anti-hyperlipidemic effect of Sophorae Fructus in male Spague-Dawley rats induced by high-rich diet were studied. Oral administration of Sophorae Fructus extract significantly decreased serum total cholesterol (TC), LDL-cholesterol, triglyceride (TG), GOT and GPT levels, and incresed serum HDL-cholesterol level and also decreased the level of liver TC, TG, GOT and GPT. These results suggest that Sophorae Fructus extract is effective for the hyperlipidemia.

  • Water and ethanol extracts of A. koreanum were prepared and tested for their improving effects on the rat's dry skin induced by household detergent. Rats treated with ethanol extracts of A. koreanum had higher serum HDL-cholesterol and lower LDL-cholesterol levels, when compared to the ones treated with water extracts. Animals treated with ethanol extracts showed that improved protein recovery patterns over time, when it compared to the ones treated with water extracts.

Serum Cholesterol Lowering Effect of Triterpene Acetate Obtained from Lactuca indica (왕고들빼기로부터 얻은 Triterpene Acetate의 혈청 콜레스테롤 저하효과)

  • The diets with three types of triterpenoid constituents, which were isolated from Lactuca indica, provoked significant changes of serum lipoprotein-cholesterol meta bolisms of hypercholesterolemic rats induced by high-cholesterol diet, together with the reduction of atherogenic index. Especially, triterpene acetates which have triterpene moieties such as ${\beta}-amyrin$, ${\alpha}-amyrin$, lupeol, pseudotaraxasterol, taraxasterol and germanicol showed a considerable hypocholesterolemic activity. The rat given orally with triterpene acetates did not exhibit a significantly higher value of atherogenic index than that of normal rats.

Effect of Glutinous Barley Intake on Lipid Metabolism in Middle-Aged Rats Fed a High-Fat Diet

  • This study was designed to determine whether dietary glutinous barley (GB) affects lipid metabolism in middle-aged rats previously fed a high-fat diet. To induce obesity, 20 male 9-month-old Sprague Dawley rats were raised for 1 month on a diet containing 20%(w/w) lipid. The rats were allocated to 1 of 2 groups of 10 rats each and for the subsequent 2 months were fed an 8%(w/w) lipid diet containing well-milled rice (WMR) or GB powder. Rats fed the GB diet had significantly lower concentrations of plasma triglyceride, plasma total cholesterol, and liver cholesterol than rats fed the WMR diet. Fecal excretions of triglyceride and bile acids were significantly greater for the GB group than for the WMR group. In conclusion, dietary GB has positive effects on lipid metabolism: it decreases plasma cholesterol concentration by increasing fecal excretion of bile acids.

A Study on the Safety of Korean Domestic Fermented Meju (한국산 재래식 발효메주의 안전성에 관한 연구)

  • For this experiment frist the fermented Soybean Meju was prepared by domestic ordinary method of Korean and concentrated Meju suspensoidal extract was made for toxicological test sample on mice and rat. Even the oral dose of 10g extract per kg of mice had not any acute toxicity. So the median lethal dose(LOD50) is lilely to be much greatr or not to be. In comparison of the average weight gain rate for 4 weeks the control and test groups of 0.308g Ex/rat kg and 9.24g Ex/rat kg daily were the much the same as about 19% but 3.08g Ex/rat kg group waas exceptionally 2.7% higher than control For this period all test groups did not show any abnormality in appearance and anatomic findings. In serum GPT GOT alkaline phosphatase and blood urea nitrogen value there were no difference significantly between control and the test groups of 0.308g/rat kg and 3.08g/rat kg but 9.24g/rat kg group was some what higher than control. Especially in cholesterol alkaline phosphatase and blood urea nitrogen value 9.24g/rat kg group or 28days had 100mg/이 37.5u 32mg/이 respectively which were increased significantly. The pathological findings of all test groups especially 9.24g/kg group did not showed any characterized sign of acute or subacute liver damage or intestine lesion. So it is supposed that there is not any tocixants originated from some molds in fermented Korean domestic Meju which is prepared traditionally in winter.

Plasma Cholesterol-Lowering Effects of Cinnamomi cortex Extract as an Inhibitor of Pancreatic Cholesterol Esterase (췌장 콜레스테롤 에스터레이즈 저해제로서의 계피 추출물레 혈중 콜레스테롤 농도에 미치는 영향)

  • Ethanol extract of Cinnamomi cortex inhibited potently cholesterol esterase activity in vitro. Chloroform fraction of ethanol extract showed the stronger inhibitory effect than other solvent fractions - ethylacetate fraction, butanol fraction, and aqueous fraction. The chloroform fraction of Cinnamomi cortex was studied as a candidator of plasma cholesterol-lowering material using high cholesterol-fed rats. In high cholesterol-fed rats, the diet with chloroform fraction of 150 mg/kg lowered not only plasma neutral lipids contents 25.1% but also plasma total cholesterol level 49.6% than only high cholesterol diet. Plasma HDL-cholesterol level in Cinnamomi cortex chloroform fraction-fed rats was recovered as those level of normal rats. LD$_{50}$ of Cinnamomi chloroform extract was calculated as 1,300 mg/kg.

Effect of Dietary Eicosapentaenoic Acid on Serum and Liver Lipids Patterns of Male Rat (어유의 Eicosapentaenoic Acid의 식이섭취가 성숙쥐의 혈청 및 간의 지질조성에 미치는 영향)

  • The dietry effects of marine n-3, plant n-3 and plant n-6 fatty acid on serum lipids levels, liver phospholipid fatty acid composition in rat were investigated. Four groups of male Sprague-Dawley rats, 30 weeks old, were fed on one of 4 different experimental diets for 4 weeks. The diets were composed of 15% fat(w/w) of either concentrated EPA oil(20:5, n-3 : 65%), fish oil(20:5, n-3 : 19%, 22:6, n-3 : 18%), perilla oil(18:3, n-3 : 60%) or corn oil(18:2, n-6 : 49%). Blood was initially taken before experimental feeding and also taken after 2 weeks and 4 weeks feeding the diet respectively and then examined for the levels of serum lipids. Rats were sacrificed at 4 weeks after the diet for the analysis of liver phospholipid fatty acid. EPA feeding remarkably decreased the serum levels of triglyceride, total cholesterol, HDL-cholesterol and total phospholipid than any other oil feeding. Fish oil feeding decreased serum HDL-cholesterol level comparable to the effect of EPA feeding and decreased total cholesterol and phospholipid less than but close to the effect of EPA feeding. Perilla oil feeding did not change serum levels of triglyceride and phospholipid, but it decreased serum total cholesterol a lot and HDL-cholesterol a little. Corn oil feeding did not affect triglyceride and total cholesterol while it increased serum level of HDL-cholesterol and total phospholipid. Serum HDL-cholesterol level was increased only in corn oil group. But contrary to the result of serum total phospholipid, liver phospholipid level found to be higher in fish oil and EPA groups than in perilla oil and corn groups. The fatty acid composition of liver phospholipid, phosphatidylcholine(PC) and phosphatidyl ethanolamine(PE) turned out to be affected by dietary fatty acid. 18:2 of liver PC was the lowest in FO group following CO group. The ratio of 20:4/18:2 was lower in PO group than in EPA group in consequence of higher 18:2 and lower 20:4 in PO group and vise versa in EPA group. In the liver PC and PE, similar trends in the ratios of n-6/n-3 and 20:4/18 were found showing higher ratios with CO and EPA group over FO and PO group. EPA group showed the lowest level of 20:5 and lower level of 20:6 than group. Fish oil was more efficient than EPA oil and PO in lowering the ratio of n-6/n-3 in consequence of the highest 22:6, and the lowest 18:2 in liver phospholipid. But PO lowers the ratio or 20:4/18 more than FO. In conclusion, EPA oil was more effective in lowering serum lipids than FO and PO. Reviewing the dietary effect of fatty acid on eicosanoids composition in rats, it is considered that more possibility was with FO than PO in the effectiveness of atherosclerosis prevention and more with PO than with EPA oil. It was also found that FO showed more effective than EPA oil for atherosclerosis prevention. It was hardly found that CO had any effect on lowering serum lipids and on eicosanoids composition in liver phospholipid for the prevention of atherosclerosis.

The Effects of Lycii fructus and Lycii folium on the Liver in Ovariectomized Rat (구기자와 구기엽이 난소적출 흰쥐의 간에 미치는 영향)

  • We aimed to investigate whether the extracts of Lycii fructus, Lycii folium, and estradiol can attenuate on the level of cholesterol on serum and ultrastructure alteration of hepatocytes isolated from ovariectomized rats. Experimental groups are divided into five: Sham (sham-operated), OVX (ovariectomized animal), LCF (OVX, Lycii fructus), LCL (OVX, Lycii folium), and $E_2$ (OVX, estradiol). The body weights of OVX group showed increase compared with Sham, LCF, and LCL groups. The levels of total cholesterol and low-density lipoprotein (LDL) cholesterol were significantly decreased in the LCF and LCL groups compared with those of OVX group. Moreover, the High-density lipoprotein (HDL) cholesterol level was increased in the LCL group. As the results of electron microscopical observation, lipid droplets in the OVX group showed increase compared with Sham group. But in the LCF group not showed lipid droplets. In conclusion, administration of Lycii fructus or Lycii folium lowers the serum cholesterol content of the ovariectomized rats and also the damage of the hepatocytes and formation of lipid droplets.

Effect of Chitosan Oligosaccharides on Cholesterol Level and Antioxidant Enzyme Activities in Hypercholesterolemic Rat (고콜레스테롤 식이에 있어 키토산 올리고당이 체내 콜레스테롤농도 및 항산화효소 활성에 미치는 영향)

  • Effect of chitosan oligo saccharides (COS) on the level of serum lipids, antioxidant enzyme activities and lipid peroxidation was investigated in rats fed with high cholesterol diet for 4 weeks, The rats were divided into three experimental groups that is, high cholesterol diet group (0.5% cholesterol; control). high cholesterol diet and 1.0% or 2.0% COS-supplemented groups (COS I , COS II). Serum total cholesterol, LDL-cholesterol and triglyceride level were significantly decreased and relative HDL-cholesterol level in total cholesterol significantly increased in COS II group. Liver TBARS level and activities of SOD and catalase of COS I were also significantly reduced. These results suggest that supplement of chitosan oligosaccharides reduce levels of serum cholesterol and reduce oxidative damage by activating hepatic antioxidative defense system in rats fed with high cholesterol diets.

Antiobesity and Cholesterol-Lowering Effects of Bifidobacteria animalis DY-64 in Rats Fed a High-Fat/High-Cholesterol Diet

  • The present study was carried out to investigate the antiobesity and hypocholesterolemic effects of Bifidobacteria animalis DY-64 (B. animalis DY-64), a lactic acid bacterium isolated from the human intestine, in rats fed a high-fat/high-cholesterol diet for 4 weeks. Forty male Sprague-Dawley rats were divided into four groups and fed either a normal (N) or high-fat/high-cholesterol (HFC) diet without or with oral administration of B. animalis DY-64 (N-BA, HFC-BA). The gain in body weight, and liver and adipose tissue weights of the HFC group were heavier than that of the HFC-BA group. Serum total cholesterol (TC), LDL-cholesterol, and leptin levels of the HFC group, which were significantly elevated compared to those of the N group, dropped by 19, 18, 21, and 13% in the HFC-BA group, respectively, whereas the serum HDL-cholesterol level markedly increased. However, serum TG, LDL-cholesterol, HDL-cholesterol, and leptin levels were not significantly different among the N groups (N, N-BA) with or without B. animalis DY-64 administration. TC and TG levels of the liver as well as the TG level of the adipose tissue were significantly reduced in the HFC-BA group. In addition, HR-LPL activity in adipose tissue was also lower in the HFC-BA group than in the HFC group. These results suggest that B. animalis DY-64 isolated from the human intestine exerts hypocholesterolemic effects by reducing serum and liver cholesterol levels and plays a role in the prevention of obesity induced by HFC diet.
