  • A simple. specific and sensitive method for the determination of cyclosporine A (CsA) in human serum has been developed by a high performance liquid chromatography/diode array detector (DAD) and applied to pharmacokinetic study of CsA. This method involves the use of solid phase extraction procedure following rapid protein precipitation with zinc sulphate from 1 $m\ell$ of human serum, using a disposable $C_{18}$ extraction cartridge.   • Simultaneous determination of crop protection agents(CPAs) in food are done with multi-residue methods, which are composed of sample clean-up, concentration, chromatographic separation and detection. Stir Bar Sorptive Extraction(SBSE) technique is used for sample preparation of various analytes in several fields. The aim of this study was to develop a sensitive and fast method based on SBSE followed by thermal desorption - gas chromatography - mass spectrometry(TD - GC/MS) to determine CPAs in tobacco sample. For the analysis of tobacco sample prior to the SBSE method, solvent extraction or ultrasound-assisted solvent extraction was performed. methanol was used as the extraction solvent. The extract was then diluted with water. Finally, the sample was subjected to SBSE. A method for fast screening of crop protection agents in tobacco using SBSE-TD - GC/MS has been developed. About 17 CPAs including organochlorine, organophosphorous and others were identified and quantified. This method showed good linearity and high sensitivity for most of the target CPAs. The method was applied to the determination of CPAs at ng/mL levels in tobacco sample. This method is simple, rapid and may be applied in detection of other components.

  • The influence of extraction temperature, powder concentration, and extraction time on the antioxidant properties of aqueous ginger extract was investigated. The possibility of estimating the antioxidant properties of the extract from its absorbance and colour properties was also investigated. Results indicated that powder concentration was the most significant factor to consider in optimizing antioxidant extraction. However, temperature and time still influenced the 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) radical scavenging activity while extraction temperature influenced the 2,2-diphenyl-1-picrylhydrazyl (DPPH) radical scavenging activity of the extract. Using the total phenol content, total flavonoid content, ABTS radical scavenging activity, and DPPH radical scavenging activity of the extract, the multiresponse optimization condition for extraction of antioxidant based on the experimental range studied is $96^{\circ}C$, 2.10 g/100 mL, and 90 min. The absorbance of the ginger extract at 610 nm could be exploited for rapid estimation of its total flavonoid and polyphenol with a $R^2$ of 0.713 and 0.753, respectively.

  • Microwave assisted extraction(MAE) is known as a more environmental friendly process with economic advantages in terms of less time, less solvent, less energy and less wastes than the current time consuming reflux method. It was applied to develop a rapid extraction method for soluble ginseng components that are major materials used for the processing of ginseng products. In a comparative study between pre established MAE(ethanol 60%, power 80 W, process time 4 min$\times$5) and current extraction method(ethanol 80%, temp. 85oC, time 8 hr$\times$5), MAE was more efficient than the current method to obtain an extract yield(soluble solid), but it was insufficient to extract individual ginsenosides, total phenols, reducing components and acidic polysaccharides. MAE with 80% ethanol by 5 times showed, however, that its extraction efficiency on soluble solid, crude saponin, major ginsenosides, and the other components was equal or superior to that of the current method, indicating that ethanol concentration is one of the critical parameters influencing the MAE process. The quality of ginseng extracts from MAE was assured by evaluating the corresponding standards and by comparing TLC and HPLC patterns with the control.

Rapid Detection of Pathogens Associated with Dental Caries and Periodontitis by PCR Using a Modified DNA Extraction Method (PCR을 이용한 치아우식증 및 치주염 연관 병원체의 빠른 검출)

  • DNA extraction is a prerequisite for the identification of pathogens in clinical samples. Commercial DNA extraction kits generally involve time-consuming and laborious multi-step procedures. In the present study, our modified DNA isolation method for saliva samples allows for the quick detection of pathogens associated with dental caries or periodontitis by PCR within 1 h. To release DNA from the bacteria, 1 min of boiling was adequate, and the resulting isolated DNA can be used many times and is suitable for long term storage of at least 13 months at $4^{\circ}C$, and even longer at $-20^{\circ}C$. In conclusion, our modified DNA extraction method is simple, rapid, and cost-effective, and suitable for preparing DNA from clinical samples for PCR for the rapid detection of oral pathogens from saliva.

Extraction of paddy field in Jaeryeong, North Korea by object-oriented classification with RapidEye NDVI imagery (RapidEye 위성영상의 시계열 NDVI 및 객체기반 분류를 이용한 북한 재령군의 논벼 재배지역 추출 기법 연구)

  • While utilizing high resolution satellite image for land use classification has been popularized, object-oriented classification has been adapted as an affordable classification method rather than conventional statistical classification. The aim of this study is to extract the paddy field area using object-oriented classification with time series NDVI from high-resolution satellite images, and the RapidEye satellite images of Jaeryung-gun in North Korea were used. For the implementation of object-oriented classification, creating objects by setting of scale and color factors was conducted, then 3 different land use categories including paddy field, forest and water bodies were extracted from the objects applying the variation of time-series NDVI. The unclassified objects which were not involved into the previous extraction classified into 6 categories using unsupervised classification by clustering analysis. Finally, the unsuitable paddy field area were assorted from the topographic factors such as elevation and slope. As the results, about 33.6 % of the total area (32313.1 ha) were classified to the paddy field (10847.9 ha) and 851.0 ha was classified to the unsuitable paddy field based on the topographic factors. The user accuracy of paddy field classification was calculated to 83.3 %, and among those, about 60.0 % of total paddy fields were classified from the time-series NDVI before the unsupervised classification. Other land covers were classified as to upland(5255.2 ha), forest (10961.0 ha), residential area and bare land (3309.6 ha), and lake and river (1784.4 ha) from this object-oriented classification.

Establishing of a rapid analytical method on uranium isotopic ratios for the environmental monitoring around nuclear facilities (원자력 시설 주변 환경 감시를 위한 토양 중 우라늄 동위원소 신속 분석법 확립)

  • The uranium isotopic ratio in environmental samples around nuclear facilities is important because it reveals information regarding illegal activities or anthropogenic pollution. Determination of uranium isotopes, however, is a challenging task requiring much labor and time because of the complex separation procedures and lengthy process. In this study, a rapid determination method for uranium isotopes in environmental samples was developed using. The sample was completely decomposed using the alkali fusion method. The separation procedure using extraction chromatography (UTEVA) was simplified in a single step without any further removal process for Si and major matrix elements. The established method can be completed within 3 h from sample dissolution to ICP-MS measurement. Most matrix elements and uranium isotopes in the soil samples were well separated and purified. Five types of were used to assess the method's accuracy and precision for a rapid uranium analysis method. The analytical accuracy for all CRM samples ranged from 95.1 % to 97.8 %, and the relative standard deviation was below 3.9 %. From the analytical results, one may draw conclusions that the evaluated method for uranium isotopes using alkali-fusion, the extraction chromatography process, and ICP-MS measurements is fast and fairly reliable owing to its recovering efficiencies. Thus, it is expected that the evaluated method can contribute to the improvement of environmental monitoring ability.

Simultaneous Determination of Vitamin A and E in Infant Formula by Rapid Extraction and HPLC with Photodiode Array Detection (신속추출법 및 PDA-HPLC에 의한 조제분유 중 비타민 A, E의 동시분석)

  • Rapid and simple method was developed for simultaneous determination of vitamins A and E contents in infant formula. Vitamins A and E were extracted by PDA-HPLC with reversed phase column using organic solvent, and their contents in Certified Reference Material (CRM) and infant formula were determined and compared with results of Food Standards Codex and AOAC method for evaluation of developed method, Vitamins A and E contents in CRM determined by developed method were within certified range of standard values. Developed method has great advantages of simple and rapid sample preparation and simultaneous determination of vitamins A and E by PDA-HPLC using reversed phase column.

Microwave-assisted extraction of paclitaxel from plant cell cultures (Microwave를 이용한 식물세포배양으로부터 paclitaxel 추출)

  • A simple and efficient microwave-assisted extraction procedure was developed and optimized for the extraction of paclitaxel from the plant cell cultures of Taxus chinensis. The biomass, immersed in a methanol-water mixture, was irradiated with microwaves in a closed-vessel system. The microwave-assisted extraction was compared with the existing conventional solvent extraction in terms of yield, extraction time, and solvent consumption. The use of microwave energy allows rapid recovery of paclitaxel from biomass and dramatically reduces extraction time and solvent usage compared to conventional solvent extraction. The paclitaxel was completely extracted from biomass by microwave-assisted extraction for 3 min at $50^{\circ}C$, for 6 min at $30^{\circ}C$ and $40^{\circ}C$, respectively.

Process Development for Production of Antioxidants from Lipid Extracted Microalgae Using Ultrasonic-assisted Extraction (탈지미세조류로부터 초음파추출을 이용한 항산화 물질 생산 공정 최적화)

  • Ultrasound-assisted extraction (UAE) has attracted growing interest, as it is an effective method for the rapid extraction of bioactive compounds from plants with a high extraction efficiency comparable to the conventional extraction. In this study, UAE was used for the extraction of polyphenols from lipid extracted microalgae (Tetraselmis KCTC 12236BP) and the effects of five extraction variables on the total phenolic compounds (TPC) were studied. For the optimization of extraction parameters, particle size, solid-to-liquid (L/S) ratio, ethanol concentration, extraction temperature and extraction time have been examined as independent variables. All variables exhibited the significant effects on the extraction of TPC and extraction temperature showed the most significant effect among five variables. The optimal extraction conditions were the extraction using mixed particle, S/L ratio of 10%, ethanol concentration of 60%, extraction temperature of $100^{\circ}C$ and extraction time of 30 min, which gave the 8.7 mg GAE/g DW for TPC. Compared with conventional hot-water extraction, TPC extraction under UAE was increased by up to 1.8 fold with same extraction condition. This study showed that UAE under low temperature and short extraction time was proven to be an effective extraction process for TPC production from LEA compared to conventional hot-water extraction process.
