Development of herbicide-tolerant Korean rapeseed (Brassica napus L.) cultivars (한국 고유의 품종을 이용한 제초제 저항성 유채 개발)

  • An interest in the production of seed-oil based fuel and raw materials, which comes from renewable plant sources, has been intrigued by the phenomenon of global warming and shortage of fossil fuels. Rapeseed (Brassica napus) is the most important oilseed crop, which produces seeds with 40% oil. It is desirable to develop genetically modified rapeseed producing oils, which can be easily converted to biodiesel. As an initial step for development of genetically modified rapeseed for the production of biofuels or bio-based materials, Korean rapeseed cultivars, Naehan, Youngsan, Tammi and Halla, were analyzed. Four Korean rapeseed cultivars produce 32 to 40% oil of seed dry weight, which is rich in oleic acid (more than 60 mole%). The cotyledonary petioles of rapeseed cultivar, Halla, were transformed using Agrobacterium tumefaciens strain GV3101, carrying the uidA gene encoding $\beta$-glucuronidase (GUS) as a reporter gene and the phosphinothricin acetyltransferase (PAT) gene as a selectable marker. The stable integration of PAT gene in the genome of transgenic rapeseeds was confirmed by PCR analysis. Expression of uidA gene in various rapeseed organs was determined by fluorometric assay and histochemical staining. Transformation efficiency of a Korean rapeseed Halla cultivar was 10.4%. Genetic inheritance of transgenes was confirmed in $T_2$ generation.

Overexpression of Ice Recrystallization Inhibition Protein (HvIRIP) from Barley Enhances Cold Tolerance in Transgenic rapeseed plants (HvIRIP 과발현 유채 형질전환체의 내한성 증진)

  • Rapeseed (Brassica napus) is now the second largest oilseed crop after soybean. Cold temperature tolerance is an important agronomic trait in winter rapeseed that determines the plant's ability to control below freezing temperatures. To improve cold tolerance of rapeseed plants, an expression vector containing an Barley Ice recrystallization inhibition protein (HvIRIP) cDNA driven by a cauliflower mosaic virus 35S promoter was transferred into rapeseed plants. Transgenic expression of HvIRIP was proved by southern- and northern-blot analyses. The level of freezing tolerance of transgenic $T_3$ plants was found to be significantly greater than that of wild-type rapeseed plants by freezing assay. Proline accumulation during cold stress was also highly induced in the transgenic rapeseed plants. The transgenic plants exhibited considerable tolerance against oxidative damage induced by cold stress. Our results indicated that heterologous HvIRIP expression in transgenic rapeseed plants may induce several oxidative-stress responsive genes to protect from cold stress.

Development of Rapeseed Precleaner and Cleaner for Biodiesel Production (바이오디젤 생산을 위한 유채종자 조정선기 및 정선기 개발)

  • Mechanization such as machine harvesting, precleaning, drying and cleaning for rapeseed harvested with high moisture content should be accomplished for biodiesel production. In addition, machine drying and cleaning is inevitable in the mechanization of work, just because rice should be transplanted right after harvesting rapeseed in Korea. Particularly, early harvested rapeseed with the combine have high moisture content and undesirable materials such as stalks and stones which make drying-process difficult and lower the efficiency of drying. Therefore, this study was conducted to develop precleaner and cleaner which could remove foreign substances from harvested rapeseeds. The precleaner consists of throw-in hopper, conveyor, feeding hopper, two precleaning sieves and discharging sections. Precleaning capacity was 1,505 kg/hr in shaking frequency of 370 cpm (cycles per minute) and tilt angles of between $5^{\circ}$ and $7^{\circ}$. The efficiency of precleaning was between 90.9% and 91.5%. The cleaner consists of feeding, shaking, blowing, cleaning and discharging sections. Cleaning performance was 435.4kg/hr in the number of rocking motions of 475 cpm and tilt angle of $10^{\circ}$. The ratios of cleaning, foreign substances and loss were 96.5%, 3.5% and 0.2%, respectively.

Effect of Rapeseed [Recommendable cultivars(Mokpo Dangy 19)] oil an Growth and Lipid Metabolism in Rats (개량유채 목포 단교 19호유가 흰쥐의 성장 및 지방대사에 미치는 영향)

  • This study was performed to investigate nutritional effect and the utilization possibility of rapeseed oil which could replace the imported edible oils and oil crops. The proximate compositions of a recommendable cultivar(Mokpo Dangyo 19) and a native kind(Asahi) and the characteristics of these rapeseed oils were analyzed. The animal experiment of these rapeseed oils was carried out during 8 weeks of growing periods after weanling. Forty male weanling Sprague-Dawley rats were randomly assigned to 3 diet groups of Dangyo 19 oil, Asahi oil and commercial Soybean oil. Mokpo Dangyo 19 variety contained more lipids than Asahi variety by 8% and there was no difference in physiochemical characteristics between Dangyo 19 oil and Asahi oil except that erucic acid was contained little in Dangyo 19 oil but 50% in Asahi oil. Body weight gain, FER(Feed efficiency ratio) and PER(Protein efficiency ratio) of rats fed Dangyo 19 oil were higher than those of rats fed Asahi oil and organ weights such as liver, kidney and epididymal fat pads weights of rats fed Dangyo 19 oil were significantly higher(P<0.05) thanthose of rats fed Asahi oil. the apparent digistibility of total diet and total lipid were higher in rats fed the diets containing Dangyo 19 oil than Asahi oil. The content of total lipid in heart in heart were not significantly different with dietary oil kinds. The content of total lipid in liver increased with age in all rapeseed oil groups.

A Study on Skin Care Effects of Rapeseed Meal Extract (유채박 추출물의 피부미용 효과에 관한 연구)

  • This research was to investigate the inhibitory effects of tyrosinase, elastase and collagenase using rapeseed meal extract for the functional cosmetics. Gamma-tocopherol and alpha-tocopherol contents were 304.9 and 212.2 mg/kg, respectively. In the case of delta-tocopherol and plastochromanol-8, they were 12.1 and 35.7 mg/kg, respectively. The total phenol content of methanol extract was the highest (49.6 mg/g) which was about 4.96 fold higher than that of water extract. The maximum nitrite scavenging activities of methanol and acetone extract at pH 1.2 were 85.2 and 80.1%, respectively, at 8.0 mg/mL. When the extract concentration of rapeseed meal increased upto 2.0 mg/mL, cell viabilities did not appear to have any significant cytotoxic effect, irrespective of extracts. Tyrosinase and elastase inhibition activities increased from 25.2 to 42.5% and 25.3 to 48.0%, respectively, as methanol extract concentration increased from 0.5 to 1.0 mg/mL. The collagenase inhibition activities of methanol and acetone extracts at 1.0 mg/mL were 67.2 and 68.0%, respectively. These results showed that the methanol and acetone extract of rapeseed meal can be used as a new source of functional cosmetic agent.

Thin-layer Drying Characteristics of Rapeseed

  • Purpose: The aims of this study were to define the drying characteristics of rapeseed and to determine the optimum thin-layer drying model for rapeseed by considering the effects of drying temperature and relative humidity. Methods: The thin-layer drying experiments were conducted at different combinations of drying air temperature levels of 40, 50, and $60^{\circ}C$ and relative humidity levels of 30, 45, and 60%, on both of which drying rate depends. The drying rate increased with increasing air temperature as well as decreasing relative humidity. The 13 models were fitted to the experimental data. Results: From the results of the regression analysis for empirical constants of the Page model, the values of $R^2$ were the highest (ranging from 0.9924 to 0.9966) and the values of RMSE were the lowest (ranging from 0.0169 to 0.0296). Conclusions: For all drying conditions considered, the Page model was determined to be the most suitable model for describing the thin-layer drying of rapeseed (P-value < 0.01). The moisture diffusion coefficients were calculated using the moisture diffusion equation for a spherical shape, based on Fick's second law.

Effects of Rapeseed Oil Diet on Serum and Liver Lipid Levels in Rats (유채유 함유사료가 쥐의 혈청 및 간의 지질조성에 미치는 영향)

  • The fatty acid composition of a rapeseed oil being on the market was analyzed and the effect on gain of the body weight and lipid levels in serum and liver tissue of male rats of Sprague-Dawely strain fed the diet containing the rapeseed oil were studied. The fatty acid components of marketed rapeseed oil was oleic acid 29.4%, erucic acid 26.52%, linoleic acid 20.39% and linolenic acid 8.68%. The contents of total lipid in serum W3S Significantly higher in RSO20 group than Contr01 group(P< 0.01) . But that in the liver tissue did 001 show significant differences. The contents of triglyceride in serum was control group 84.14mg/dll, RSO15 group 100.33mg 141 and RSO20 group 122.00mg 141 and showed significant difference between each group, but that in the livertissue did not show significant differences. The contents of phospholipid in serum did not show significant differences. But that in the liver tissue showed significant difference between the control group 8.42mg /g and Rs02o group 7.34mg /g(p<0.001). The contents of total-cholesterol and free-cholesterol in serum and liver tissue of the RSO20 group showed the highest levels compared with control group, but there did not show significant differences. The contents of ester-cholesterol in serum showed significant.

Effects of Sowing Date on Grain Yielding and Related Traits in Rapeseed

  • The objectives of this experiment was to evaluate the effect of seeding and transplanting date on yield and quality of rapeseed experiment were conducted in the research field of Chungcheongnam Do(Taejun) Agricultural Research and Extension Services with Yudal, one of the rapeseed cultivar of different growth characteristics among the six treatments were examined and recorded. Results showed that yield components such as plant height, length of ear and pod, number of branches and pods and percentage of pod setting and seed set were highest at the plot with direct seeding in Sep. 10. In this treatment yield of seed, 1,000 grains weight and weight of $1{\ell}$ were highest. On the basis of vegetative and yield parameter of direct seeding in Sep. 10 was the best sowing date of rapeseed crop in Chungcheongnam Do region of Korea. Experiments were conducted in the research field of Chungcheongnam Do(Taejun) Agricultural Research and Extension Services with Yudal variety, which showed different growth characteristics. Yield components such as plant height, length of ear and pod, number of branches and pods and percentage of pod setting and seed set were highest at the plot with direct seeding in Sep. 10. Yield of seed, 1,000 grains weight and weight of $1{\ell}$ were highest at the direct seeding in Sep. 10. Judging from the results reported above, seemed to be direct seeding in Sep. 10.

Extraction of Liberated Reducing Sugars from Rapeseed Cake via Acid and Alkali Treatments (산 및 알칼리 처리에 의한 유채박의 유리당 추출)

  • Rapeseed cake, which is the organic waste remaining after rapeseed oil production, is readily available and considered an ecologically-friendly resource with very low cost and high dietary fiber content. This research was carried out for two reasons. First, it was done to analyze the liberated reducing sugar content of rapeseed cake. Second, it was done to investigate the effects on the sugar yield of the various concentrations of acidic and alkaline catalysts used for the hydrolysis of rapeseed cake and the concentrations of rapeseed cake in each catalyst. Several amounts of ground rapeseed cake, 0.5 g, 1 g, and 2 g, were put into 100 mL of catalysts such as sulfuric acid (0.5~2%), hydrochloric acid (0.5~2%), and sodium hydroxide (0.5~2%). Then they were hydrolyzed for 5 min at 121$^{\circ}C$. After hydrolysis, HPLC equipped with an RI detector was used to analyze liberated reducing sugars such as sucrose, glucose, galactose, fructose, and arabinose separated from rapeseed cake. The degradation rate of rapeseed cake was the highest in hydrochloric acid. As the catalyst concentrations used for hydrolysis of rapeseed cake increased, the degradation rate of rapeseed cake also significantly increased. Total reducing sugar content was the highest in hydrochloric acid, and it increased with the increase of catalyst concentrations. However, as the amount of rapeseed cake increased, the total reducing sugar content decreased, exceptionally sucrose in the case of sodium hydroxide.

  • Biological conversion of biomass into fuels and chemicals requires hydrolysis of the polysaccharide into monomeric sugars. In this study, dilute sulfuric acid used as a catalyst for the pretreatment of rapeseed straw. Hydrolysis can be performed enzymatically, and with dilute or concentrate mineral acids. Dilute-acid hydrolysis of rapeseed straw was optimized through the utilization of combined severity. Evaluation criteria for optimization of the pretreatment conditions were based on high xylose recovery and low inhibitor contents in the hydrolyzates. In addition, this paper reports the compositional analysis of hydrolyzate liquors and solid residues, xylose and glucose mass balance closures, and digestibility results of the acid pretreated rapeseed straw.
