Determination of Oxidation Stability of Perilla Oil by the Rancimat Method (랜시매트법에 의한 들기름의 산화안정성 측정)

  • Comparison of induction times among the Rancimat Method, the Oven Test and the Weight Method were investigated, and the oxidation stability of perilla oil was determinated by the Rancimat Method. The induction times determinated by three methods using perilla oil show a good correlation. And the temperature dependence of the induction times of perilla oil was as follows. $t_p=e^{-0.0815T+9.183}$. The antioxidant effect of Tocopherols on the oxidation of perilla oil was not observed, and the antioxidant effect of the organic acids was increased in the order of Ascorbic acid>Ascorbyl stearate>Malic acid>Tartaric acid>Citric acid. The synergistic effect of Ascorbic acid (500 ppm)and Tocopherols was not observed below 200ppm of Tocopherols, but became larger with increasing Tocopherols level over 200ppm.

Identification of Antioxidative Components from Ethanol Extract of Rhus verniciflua STOKES (옻나무 에탄올 추출물로부터 항산화 활성 물질의 구조동정)

  • The free phenolic acid fraction of the chloroform extract from 75% ethanol extract of Rhus verniciflua STOKES (RCF) showed stronger antioxidative activity than BHT, BHA and ${\delta}-tocopherol$ at the same concentration. RCF components were isolated and identified by silica gel column chromatography, thin layer chromatography, mass spectrometer and $^1H-NMR\;and\;^{13}C-NMR$. The antioxidative activity was confirmed by electron donating activity, Rancimat method and thiobarbituric acid test in liposome system. RCF-11 could be further separated into three fractions. The antioxidative active compounds were purified and identified as gallic acid, butin and butein. The RCF-13 was purified and identified as sulfuretin.

Study on Antioxidative Activity of Plant Extracts in Fish Oil (식물 추출물이 어유의 산화 안정성에 미치는 영향)

  • Extract from forty four species of natural plants in Korea were obtained. Each extinct (1,000 ppm) was added into fish oil and their antioxidative activities were screened by rancimat, From the results, seven species including Brassica juncea, Solanum tuberosum, Capsella bursa, Ulums davidiana, Broussonetia kazinoki, Oenanthe javanica, and Rhus chinensis were selected due to their relatively prolonged induction periods. When the concentration (100 - 1,000 ppm) of the selected extract was considered, the most prolonged induction period was observed from Solanum tuberosum throughout the studied concentrations, suggesting that its extinct showed the strongest antioxidative activity against fish oil. In addition, peroxide value (POV) and thiobarbituric acid reactive substance (TBARS) value were considered at $60^{\circ}C$ for 5 days. Among the selected extract, extract from Solanum tuberosum showed the lout POV while the extract from Brassica juncea var showed the least TBARS values after 5 days. Also, the free radical scavenging effect against DPPH radical were studied with the selected extract, showing that the extract from Rhus chinensis was found to be the most effective.

Antioxidative Effectiveness of Methanol Extract in Galla Rhois (오배자(Rhus japonica Linne) Methanol 추출물의 황산화효과)

  • Free-, soluble- and insoluble phenolic acids were extracted from defatted Galla Rhois. The extracts were then dissolved in equal amounts of an soybean oil, and POV (peroxide value) of the resulting substrates, portion of the soybean oil (control) and 0.02% BHT were measured by AOM (active oxygen method) test at 97.8$^{\circ}C$ for 40 hours through Rancimat method. Induction period of control, BHT, free phenolic acids, soluble phenolic acids and insoluble phenolic acids by the Rancimat method were 4.8, 10.5, 23.9 and 30.5hr. The phenolic acids separated and tentati-vely identified by gas chromatography were catechol, gallic acid, vanillin, protocatechuic acid, syri-ngic acid, ferulic acid.

Screening of the Physiological Activity of Solvent Extracts of Paulownia coreana Bark and Antioxidative Effect of the Extracts on an Edible Oil

  • Paulownia coreana is a medicinal, edible and industrial plant with the largest leaf, and is native to Korea. We evaluated the reducing power activities, 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical scavenging activities (RSAs), nitrite scavenging activities (NSAs) and 2,2-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) cation RSAs by solvent extraction of P. coreana bark by using $50^{\circ}C$ hot water and $25^{\circ}C$ methanol. The antioxidative effect of P. coreana bark extract on corn seed oil was evaluated using the Rancimat test. The extraction yields on dry weight basis with 15 folds of hot water and methanol were 23.88% and 5.30%, respectively; further the flavonoid content in the hot water extract was over 2.5 times more than that in the methanol extract. The DPPH RSA of the methanol extract was substantially higher than that of the water extract, whereas the NSA of the water extract was higher than that of the methanol extract at pH 1.2. The ABTS RSAs of the two extracts were almost the same as that of ascorbic acid and 2,6-ditertiarybutyl hydroxytoluene. The two extracts of P. coreana bark in this study were found to slightly improve the oxidation stability of corn seed oil.

Isolation of Antioxidative Components from the Bark of Rhus verniciflua STOKES Screened from Some Chinese Medicinal Plants (한약재로부터 선발된 옻나무 수피 추출물로부터 항산화 활성물질의 분리)

  • To develop new natural antioxidants, antioxidative activity of ethanol (75%) extracts from 50 edible or medicinal plants were examined on lard and palm oil by Rancimat method ($120^{\circ}C$, 20 L/hr). The extracts from Rhus verviciflua STOKES showed comparatively strong antioxidative activity on test. Of the solvents used for extraction, chloroform extract exhibited the strongest antioxidant activity. AI (antioxidant index: induction period of oil containing extract/induction period of control oil) of chloroform extract was higher than that of commercial antioxidant, such as BHT, BHA and ${\delta}-tocopherol$. Free phenolic acid fraction (200 ppm) of the chloroform extract from 75% EtOH extract of Rhus verniciflua STOKES (RCF) showed stronger activity than that of BHT, BHA, and ${\delta}-tocopherol$ at the same concentration. RCF-11 and RCF-13 fractions separated by silicagel column chromatography from the RCF showed stronger activity than other fractions by the Rancimat method.

Extraction Characteristics and Antioxidant Activity of Ethanol Extract of Rhus javanica Bark (붉나무 껍질 에탄올 추출물의 추출특성과 항산화 활성에 관한 연구)

  • This study was conducted to investigate the several physicochemical extraction characteristics and the antioxidant activities of ethanol extract from Rhus javanica bark. The contents of soluble solid and sugar in extract was measured to 73.5 mg/100g dry basis and $17.8^{\circ}Brix$, respectively. The contents of total aromatics, total flavonoids and total phenolic compounds was measured to 0.508 in absorbance, 49.88 mg/100g and 296.6 mg/100g, respectively. The reducing power of extract was about 27.5 % of ascorbic acid with the same soluble solid contents of the extract. But the ferric reducing antioxidant power and the DPPH radical scavenging ability of extract were measured to equivalent to those of ascorbic acid. The metal ion chelating ability of the extract was 81.58 % whereas that of ascorbic acid was 74.73 %. The nitrite scavenging ability of the extract was measured to 51.76 % at pH 2. And the antioxidative effect of the extract on soybean oil was observed with Rancimat test.

Antioxidative Effects of Doenjang Fermented Using Bacillus subtilis DJI (Bacillus subtilis DJI을 이용하여 제조한 된장의 항산화효과)

  • We investigated the antioxidative effects of solvent extracts of doenjang fermented using Bacillus subtilis DJI (DJI doenjang) in vitro. The solvents used for extraction were ethanol, n-hexane, and water. The antioxidative activities of DJI doenjang solvent extracts were measured by estimation of peroxide value, the presence of linoleic acid level, and nitrite scavenging activity, the Rancimat test, and 1,1-diphenyl-2-picryl hydrazyl (DPPH) free-radical generation, in comparison with the commercial antioxidant butylated hydroxytoluene (BHT). The peroxide value of an ethanol extract was lower than those obtained using n-hexane and water extracts. Furthermore, the peroxide value of the ethanol extract was similar to that obtained after BHT treatment. The nitrite scavenging activity was 23.36% after addition of 600 ppm DJI doenjang ethanol extract, and the DPPH free-radical scavenging activity was 19.06% under same condition, which shows that DJI doenjang ethanol extract exhibited lower antioxidative capacities than did BHT. In the Rancimat test, the ethanol extract (11.20 min induction time), n-hexane extract (7.58 min induction time), and water extract (8.26 min induction time) after treatment with 600 ppm DJI doenjang extracts demonstrated longer induction periods than did BHT (6.94 min). These results indicate that DJI doenjang has potential anti-oxidative activity.

Antioxidative Effects of Volatile Oil and Oleoresin Extracted from Rosemary, Sage, Clove and Nutmeg (Rosemary, Sage, Clove 및 Nutmeg의 휘발성 및 비휘발성 성분의 항산화성)

  • This study was designed to investigate the antioxidative effects of volatile oil and oleoresin of rosemary mary (Rosmarinus officinalis Linne), sage (Salvia officinalis Linne), clove (Syzygium aromaticum Merrill) and nutmeg (Myristica fragrans Houttuyn) using the Rancimat and POV test. The antioxidative effects of steam volatile oils of rosemary, sage clove. and nutmeg were not statistically significant, at the levels of 0.01, 0.05, 0.1% added to lard. The antioxidative effects of oleoresin of above 4 spices were increased with the oleoresin concentration. The comparison of the results between Rancimat and POV test of above 4 spices was almost same.

The Antioxidant Activities of Acetone Extracts of Chestnut Inner Shell, Pine Needle and Hop (율피, 솔잎, 호프의 아세톤 추출물의 항산화작용)

  • The extracts 70% acetone from chestnut inner shell, pine needle and hop flower were added to soybean oil, lard, soybean oil-water emulsion and lard-water emulsion in order to test the antioxidant activity compared with ${\alpha}-tocopherol$, respectively. Induction period was determined by Rancimat and the antioxidant activities were recorded in the order of pine needle>chestnut inner shell>hop>tocopherol in the soybean oil, and in the order of tocopherol>chestnut inner shell>pine needle>hop in the lard treatment. These three extracts showed similar high antioxidant activity by peroxide value test in soybean oil, soybean oil-water emulsion, lard and lard water emulsion, respectively. The chestnut inner shell extract would be the highest antioxidant agent among the extracts tested in this study. Ellagic acid of chestnut inner shell and flavanol of pine needle would increase the antioxidant activity according to analysis result of polyphenol compound, respectively.
