The Screening and Characterization of Promoters Inducible by Superoxide Radical in Escherichia coli (대장균에서 Superoxide 라디칼에 의하여 유도되는 프로모터의 탐색 및 특성 분석)

  • We screened promoters inducible by superoxide radical from Escherichia coli. For this. we constructed random promoter library from E. coli MG 1655 using a promoter-probing plasmid. pJAC4. Six hundred and sixty clones in this library were classified based on their promoter strength by ampicillin gradient plate assay. Three hundred and eighty three clones with relatively weak to medium promoter strength were selected and then screened for their inducibility by superoxide radical on ampicillin gradient plate containing paraquat. Three clones (clones 5. 15 and 34) were detected to be induced by paraquat treatment and the level of induction were between 1.4 and 4 folds. Comparison of nucleotide sequences of the cloned promoter fragment with registered sequences in GENBANK and EMBL databases suggests that the cloned DNA fragments have not been yet characterized in E. coli. Transcription start sites in these clones were determined by rrimer extension and S I nuclease protection analysis. S 1 analysis of clones 5 and IS indicated that the mRNA levels were increased by paraquat treatment. Especially. clone 5 \vas found to have two transcription start sites. the upstream start site of which was selectively used by paraquat treatment. Searching for promoter clements. we found that only the downstream promoter of clone 5 has -10 and - 35 promoter elements recognized by RNA polymerase ($E\sigma^{70}$) and the others have no conserved promoter elements. This suggests that these superoxideinducible promoters may require transcription initiation protein(s) other than $E\sigma^{70}$.

  • To assess the relationship between anti-oxidant and immunomodulatory activities of Diospyros kaki Thunb. fruit, we harvested persimmons on the first Friday of every month from July to October, and conducted a DPPH assay, a T-bet promoter assay, and an ELISA for IL-4 determination. Anti-oxidant activity increased as fruit weight rose. When the fruit was extracted with five different solvents, and the extracts examined for radical scavenging activity, such activity exhibited a pattern similar to that seen when anti-oxidant activity was assessed. T-bet promoter activity decreased on ripening, whereas IL-4 expression increased, as tested in a DNFB-induced animal model. Collectively, the results suggest that inflammation-inhibitory activity, valuable in treatment of some immune diseases, rises as persimmons ripen.

  • Advanced (Chemical) oxidation processes (AOP) differ from most conventional ones in that hydroxyl radical(OH.) is considered to be the primary oxidant. Hydroxyl radicalcan react non-selectively with a great number of organic and inorganic chemicals. The typical rate constants of true hydroxyl radical reactions are in the range of between 109 to 1012 sec-1. Many processes are possible to generate hydroxyl radical. These include physical and chemical methods and their combinations. Physical means involves the use of high energy radiation such as gamma ray, electron beam, and acoustic wave. Under an applied high energy radiation, water molecules can be decomposed to yield hydroxyl radicals or aqueous electrons. Chemical means include the use of conventional oxidants such as hydrogen peroxide and ozone, two of the most efficient oxidants in the presence of promoter or catalyst. Hydrogen peroxide in the presence of a catalyst such as divalent iron ions can readily produce hydroxyl radicals. Ozone in the presence of specific chemical species such as OH- or hydrogen peroxide, can also generate hydroxyl radicals. Finally the combination of chemical and physical means can also yield hydroxyl radicals. Hydrogen peroxide in the presence of acoustic wave or ultra violet beam can generate hydroxyl radicals. The principles for hydroxyl radical generation will be discussed. Recent case studied of AOP for water treatment and other environmental of applications will be presented. These include the treatment of contaminated soils using electro-Fenton, lechate treatment with conventional Ponton, treatment of coal for sulfur removal using sonochemical and the treatment of groundwater with enhanced sonochemical processes.

Effect of LG Herbal Medicine Complex (LG-HMC) on Diesel Particulate Matter (DPM) Induced Skin Inflammation and Xenobiotic Response Activity In Vitro (LG-HMC의 미세먼지 유발 염증의 완화와 생체이물대사완화 효과)

  • Diesel particulate matter (DPM) induces inflammatory cytokines in HaCaT and stimulates XRE promoter activity through AhR binding. Thus, it increases CYP gene family expression. In this study, we have elucidated inhibitory effect of LG Herbal Medicine Complex (LG-HMC) on DPM-induced XRE promoter activity and inflammatory cytokine expression. First of all, the XRE promoter activity was overexpressed by DPM treatment and the LG-HMC abrogated the XRE promoter activity. Morus alba bark extract, Scutellaria baicalensis root extract and constituents of LG-HMC, were found to be main effecters against DPM induced XRE promoter activation. We also found that DPM treatment elevated inflammatory cytokines in HaCaT and the treatment of LG-HMC, Morus alba bark extract and Scutellaria baicalensis root extract down-regulated the DPM induced inflammatory cytokine expression. Additionally, Morus alba bark extract and Scutellaria baicalensis root extract were found to act as free radical scavengers. In conclusion, we confirmed skin protective effect of LG-HMC and uncovered two components, Morus alba bark extract and Scutellaria baicalensis root extract, that play major role in protecting epidermal kertinocytes from damage.

Effects of Mole Crickets (Gryllotalpa orientalis) Extracts on Anti-oxidant and Anti-inflammatory Activities. (땅강아지(Gryllotalpa orientalis) 추출물의 항산화 및 항염증 활성)

  • Tremendous natural product extracts were used as a herb medicine remedy or therapy for centuries. Because these extracts have various biological activities, we examined the effects of Gryllotalpa orientalis extract for anti-oxidant and anti-inflammation activity by 1,1-diphenyl-2-picrylhydrazyl (DPPH), ferric reducing ability of plasma (FRAP), hydroxy radical scarvenging and cyclooxygenase-2 promoter assays. Gryllotalpa orientalis extracts were prepared from solvents such as distilled water (DW), dimethyl sulfoxide (DMSO), ethanol and methanol. The results showed that Gryllotalpa orientalis extracts have potent DPPH (methanol extract), FRAP (DW extract) and hydroxy radical scavenging (DW and methanol extracts) activity than any other extracts used. A significant inhibition of cyclooxygenase-2 (Cox-2) promoter activity was detected in the presence of DMSO extract or ethanol extract. Collectively, the present results suggested that Gryllotalpa orientalis extract could be used for anti-oxidant and/or anti-inflammation agent for human or agricultural purposes.

Effects of Gene Expression of Photobacterium leiognathi CuZn Superoside Dismutase (PSOD) by lacZ Promotor Control under Oxidative Stress

  • The effect of PSOD expression on lacZ-sodP fusion (pYK4) was explored in Escherichia coli sodA sodB mutants (QC774) under oxidative stress. In this system, although .betha.-galactosidase activity was not fully induced by isopropyl-1-thio-.betha.-galactosidase (IPTG) and was inhibited by glucose, functional PSOD was under lacZ promotor control and was induced by IPTC, lactose, PQ and copper isons, finally, the results show that higher PSOD expression leel was consistently importnat in defending against superoxide radicals.

The estimation of Hydroxyl radical generation rate in Ozonation (오존산화공정에서 수산화라디칼(OH.)의 생성속도 측정)

  • During ozonation process, the hydroxyl radical generation rates were measured under different experimental conditions (ozone feed rate, nitrobenzene concentration, hydroxyl radical scavenger, pH, HO$_2$O$_2$/O$_3$ etc.) Nitrobenzene could be decomposed by hydroxyl radical rather than ozone only and nitrobenzene decomposition rate was expressed with functions of ozone and nitrobenzene concentration. The rate was decreased as the hydroxyl radical scavenger concentration was increased, and all results were followed pseudo first-order reaction. Using a competitive method, hydroxyl radical generation rate was measured with probe compound and scavenger. It was proportional to ozone concentration, and 0.24mo1 of hydroxyl radical was produced with 1mol of ozone. Under different pH conditions, hydroxyl radical generation rates were measured (pH 10.2 (0.91Ms$^{-1}$ ) > pH 7.3 (0.72Ms$^{-1}$ ) > pH 5.6 (0.67Ms$^{-1}$ ) > pH 3.4 (0.63Ms$^{-1}$ )) showing higher generation rate at high pH values. Addition of hydrogen peroxide promoted the generation rate of hydroxyl radical. Considering the results of pH experiments and addition of hydrogen peroxide experiments, the hydroxyl radical generation rate was 1.6 times higher in hydrogen peroxide solution than in high pH solution, indicating addition of hydrogen peroxide is better promoter to produce the hydroxyl radical in ozonation. These results could be applied to AOPs to remediate the contaminated wastewater and groundwater.

Adhesive Properties of Epoxy Composite According to the Surface Treatment of Cu Substrate and Adhesion Promoter Content (구리기판의 표면처리 및 접착증진제 함량에 따른 에폭시 컴포지트의 접착특성)

  • In this study, we synthesized poly(itaconic acid-co-acrylamide) (IAcAAM) used as a novel polymer adhesion promoter to improve the adhesion strength of surface-treated Cu lead frames and epoxy composites. IAcAAM comprising itaconic acid, acrylamide was prepared through radical aqueous polymerization. The chemical structure and properties of IAcAAM was analyzed by FT-IR, 1H-NMR, GPC, and DSC. The surface of the copper lead frame was treated with high temperature, alkali, and UV ozone to reduce the water contact angle and increase the surface energy. The adhesive strength of Cu lead frame and epoxy composite increased with the decrease of contact angle. The adhesive strength of Cu lead frame/epoxy composite increased with the addition of IAcAAM in epoxy composite. As silica content increased, the adhesive strength of Cu lead frame and epoxy composite tended to slightly decrease.

  • Chemical degradation of aqueous humic acid by ozonation was studied with respect to the direct reactions of ozone and the indirect reactions due to its preliminary decomposition to secondary oxidant, OH radical. This was characterized by analyzing TOC, $UV_{254}$ and ozone consumption measured in different experimental conditions in which ozone reacted in the presence of various concentrations of $H_2O_2$ and $HCO_3{^-}$ concentrations ranging from 20 to 100 mg/L. and different pH (5-9). The results suggest that the TOC removal is mainly dependent on indirect reactions of OH radical whereas $UV_{254}$ reduction is mainly dependent on direct reactions of ozone with humic acid molecules. It has been also found that ozone consumption was most likely to be affected by pH and alkalinity in the solution.

  • Objectives : Reactive oxygen species (ROS) have been associated with pathogenic processes including carcinogenesis through direct effect on DNA directly and by acting as a tumor promoter. Therefore, it has been regarded that ROS may be a major target for cancer prevention. The root of Paeonia lactiflora pall (PL), a traditional Chinese herb, has been a component of effective prescriptions for treatment of liver disease. Also, there are some reports about the antioxidant activities of the extracts from PL. However, little has been known about the effects of PL against oxidative damage. This work aimed to elucidate the anti-oxidant effects of Paeonia lactiflora pall (PL) in the non-cellular system and cellular system. Methods : Antioxidant activities of PL were evaluated by hydroxyl radical scavenging assay and $Fe^{2+}$ chelating assay. Anti-oxidative effect of PL was evaluated by ${\varphi}X$-174 RF I plasmid DNA cleavage assay in non-cellular system. In addition, DNA migration assay, expression level of phospho-H2AX, MTT assay and lipid peroxidation assay were performed for evaluate the anti-oxidative effect of PL in cellular system. Results : PL had a dose-dependent hydroxyl radical scavenging and $Fe^{2+}$ chelating capacity. In addition, PL inhibited oxidative DNA and cell damage induced by hydroxyl radical in non-cellular system and cellular system. Conclusion : Taken together, P. lactiflora pall may be possible for the application to a potential drug for treating the oxidative diseases such as cancer.
