Establishment of γ-irradiation-induced Hematopoietic Tissue Damage Model in ICR Mice (ICR 마우스에서 감마선 조사로 유도된 조혈조직 손상 모델 확립)

  • Ionizing radiation causes the massive generation of reactive oxygen species, resulting in cellular and tissue damage. The present study was performed to evaluate ${\gamma}$-irradiation induced cellular damage in ICR mice. The mice were divided into four groups with ten mice in each group. Group 1 served as an unexposed control group. Groups 2, 3, and 4 were exposed to 3, 5, and 7 Gy of ${\gamma}$-radiation, respectively. Five mice per group were sacrificed 1 and 7 days after ${\gamma}$-radiation. Exposure to ${\gamma}$-irradiation resulted in hematopoietic damage in a dose-dependent manner when compared with the unexposed control group, which featured a significantly decreased spleen index. However, the exposed mice showed no significant differences in their serum AST, ALT and in the histopathological change of their liver. These results suggest that ${\gamma}$-irradiation is a good tool to prepare a hematopoietic damage model. This animal model can be employed to study the hematopoietic efficacy of biologically active compounds.

  • Radiation damage due to neutrons has been calculated in Ni-based alloys in Wolsong CANDU reactor environments. Two damage parameters are considered: displacement damage, and transmutation gas production. We used the SPECTER and SRIM computer codes in quantifying radiation damage. In addition, damage caused by Ni two-step reactions was considered. Estimations were made for the annulus spacers in a CANDU reactor that are located axially along a fuel channel and made of Inconel X-750. The calculation results indicate that the transmutation gas production from the Ni two-step reactions is predominant as the effective full power year increases. The displacement damage due to recoil atoms produced from Ni two-step reactions accounts for over 30% out of the total displacement damage.

Protection of Radiation-Induced DNA Damage by Functional Cosmeceutical Poly-Gamma-Glutamate

  • This study compared the radioprotective effects of high-molecular-weight poly-gamma-glutamate (${\gamma}-PGA$, average molecular mass 3,000 kDa) and a reduced form of glutathione (GSH, a known radioprotector) on calf thymus DNA damage. The radiation-induced DNA damage was measured on the basis of the decreased fluorescence intensity after binding the DNA with ethidium bromide. All the experiments used $^{60}Co$ gamma radiation at 1,252 Gy, representing 50% DNA damage. When increasing the concentration of ${\gamma}-PGA$ from 0.33 to $1.65{\mu}M$, the DNA protection from radiation-induced damage also increased, with a maximum of 87% protection. Meanwhile, the maximal DNA protection when increasing the concentration of GSH was only 70%. Therefore, ${\gamma}-PGA$ exhibited significant radioprotective effects against gamma irradiation.

  • As utilization of radiation in medicine, industry and biochemical research increases, the protection against radiation damage has become an important issue. Natural products such as herbal medicines are beginning to receive attention as modifiers on the radiation response. In the present study, the protective effect of a herb, Menthae Herba, against radiation-induced DNA damage was evaluated using alkaline single-cell gel electrophoresis (SCGE; comet assay) in the mouse peripheral blood Iymphocytes and the micronucleus formation test in the Chinese hamster ovary (CHO) cells. The tail moment, which was a marker of DNA damage in the SCGE, and the frequency of micronuclei was decreased in groups treated with Mentae Herba extract before exposure to 200 cGy of gamma-ray. We also confirmed its activities to scavenge DPPH and hydroxyl radicals. These experiments demonstrated that Menthae Herba was effective at reducing the radiation-induced damage of DNA and scavenging free radicals. It is plausible that scavenging of free radicals by Menthae Herba may have played an important role in providing the protection against the radiation-induced damage to the DNA. These results indicated that Menthae Herba might be a useful radioprotector and that radical scavenging appears to be one of the mechanisms of radiation protection.

  • For a long life photovoltaic cell the degradation of the device characteristics with 1 MeV electron radiation must be known so as to be able to predict the life of the cell. Hence, a study was made of radiation damage effects on the bulk properties of the silicon crystal. From the results of the data, it is concluded that there appeared to be a steads rotate damage level reached in f type material.

  • The dependence of cell survival on exposure dose and the duration of the liquid-holding recovery (LHR) was obtained for diploid yeast cells irradiated with ionizing radiation of different linear energy transfer (LET) and recovering from radiation damage without and with various concentrations of cisplatin - the most widely used anticancer drug. The ability of yeast cells to recover from radiation damage was less effective after cell exposure to high-LET radiation, when cells were irradiated without drug. The increase in cisplatin concentration resulted in the disappearance of this difference whereas the fraction of irreversible damage was permanently enlarged independently of radiation quality. The probability of cell recovery was shown to be constant for various conditions of irradiation and recovery. A new mechanism of cisplatin action was suggested according with which the inhibition of cell recovery after exposure to ionizing radiations was completely explained by the production of irreversible damage.

  • The bacterium Deinococcus radiodurans is one of the most resistant organisms to the effects of ionizing radiation and other DNA-damaging agents. In this study, distributions of 10 ddr (DNA damage response) genes were investigated in 8 species of Deinococcus by polymerase chain reaction (PCR). We have compared the sequences of ddr genes of D. radiodurans, D. geothermalis and D. deserti, and selected primers which are suitable for the detection of ddr in different species of Deinococcus. A sequence homology search and PCR assay showed that ddrO, which encodes a global regulator of the radiation-desiccation response, was most well conserved in the Deinococcus lineage.

  • Single cell gel electrophoresis (SCGE) was used to the experiment with the variation on the amount of smoking and low dose radiation exposure to find how much the Lymphocyte DNA was damaged, and especially for whom smoke a lot(about 20 or more than 20 cigarettes a day) it was found to be highly damaged. While, the damage of 'not more than 20 cigarettes a day' was found to be not so much significant as like for whom smoke about or more than 20 cigarettes a day And, according to the different amount of the radiation exposure, the Lymphocyte DNA was found to be considerably damaged for 0-13m Sv (P<0.01), it was not able to prove the relationship between the DNA damage and the radiation exposure.

  • When a light is projected upon a material, part of its radiation energy is absorbed and the rest is reflected or transmitted according to the nature of the material. The molecules of the substance absorbing a light obtains the radiation energy to the wavelength of the light to make photochemical degradation by ultraviolet ray or thermal reactions like physical damage by infrared ray. The degree of damage by radiation energy varies to the substances of materials, the spectral power distribution of the light source and the duration of irradiation. Because the damage brings about a devaluation of material and once damaged, it is irretrievable, it is necessary to minimize the damage and conserve the native quality of a material by a protective lighting system. A measuring system was set up to measure the temperature rise of each sample by infrared radiation from light sources. And the temperature rise and temperature distribution by various infrared lamps were measured with varying time.
