Studies on the Effects of Piperidine Derivatives on Blood Pressure and Smooth Muscles Contractions

  • Ten substituted phenacyl derivatives of 4-hydroxypiperidine were synthesized and studied for their effects on the mean arterial blood pressure (MABP) in normotensive anaesthetized rats and smooth muscles contractions of isolated rabbit jejunum. Two derivatives caused fall in blood pressure at the dose of 10-20 mg/kg and one rise in blood pressure at the dose of 20 mg/kg. Two compounds exhibited biphasic response (hypotensive followed by hypertensive) and one gave triphasic response at 10 mg/kg dose. Rest of four derivatives were found devoid of any effect on mean arterial blood pressure up to the dose of 30 mg/kg. All the derivatives except two caused relaxant effect on the spontaneous contraction of rabbit jejunum at the dose range of 0.1 -2 mg/kg.

만형자(蔓荊子)의 가토혈압(家兎血壓)에 미치는 영향 (Effect of Viticis rotundifoliae Fructus Water Extract on the Blood Pressure in the Rabbit)

  • In this study the effect of water extract of Viticis rotundifoliae Fructus(VRE) on the blood pressure of the rabbit was investigated. The results of the experiment are as follows. 1. VRE produced a fall of blood pressure, exhibiting dose-action curve in the rabbit. 2. The depressor effect of VRE was inhibited by atropine and chlorisondamine, potentiated by physostigmine, and then potentiated slightly by bethanidine. 3. Effect of VRE was not influenced by propranolol and cyproheptadin. The above results indicate that hypotension of VRE is produced by stimulation of parasympathetic nervous system induced centrally.

Haloperidol 이 심혈관계에 미치는 영향 (Experimental Studies on the Cardiovascular Effects of Haloperidol in Cat and Rabbit)

  • Haloperidol, a butyrophenone, was synthetized by Janssen and introduced for the treatment of psychosis. Although structurally different from the phenothiazines, the butyrophenones share many of their pharmacological properties, such as inhibition of conditioned avoidance response, blocking effect of amphetamine reaction, producing catalepsy, antishock effect and protection against the lethal effects of catecholalmines. Chlorpromazine can lower the arterial blood pressure through its adrenergic blocking activity, its direct effect in relaxing vascular smooth muscle, its direct effect in depressing the myocardium and its action in a complex manner on the central nervous system. In the case of haloperidol, however, was not clarified the mechanism of lowering the blood pressure. The present paper describes the effects of haloperidol on cardiovascular system to investigate the mechanisms of its actions on the arterial blood pressure. The results are followings; 1. In anesthetized cats, intravenous administration of haloperidol and chlorpromazine in the dose of 0.1mg/kg produced a slight decrease in the blood pressure, which followed by complete recovery within $30{\sim}60$ minutes. In the dose of 3mg/kg, however, both produced an abrupt and marked decrease of the blood pressure, which followed by delayed recovery. 2. Haloperidol in the dose ranges of 0.1mg to 3.0mg/kg tended to produce the heart rate slowing in the cats, while chlorpromazine has no effect on the rate. 3. Following administration of haloperidol or chlorpromazine, epinephrine reversal in the arterial blood pressure was observed in the cat, however the responses of norepinephrine and acetylcholine were little affected. 4. In the isolated rabbit atrium the contractility was depressed by haloperidol in the doses over 0.5mg per 100ml, but the rate was not affected. In contrast, the epinephrine-induced contractility was not depressed after haloperidol treatment. However, the increased rate of atrium by epinephrine was partially blocked after haloperidol. 5. In the isolated rabbit aortic strip, epinephrine-induced contraction was blocked by haloperidol. With the above results, it may be concluded that the hypotensive effect of haloperidol was largely due to ${\alpha}$-adrenergic blocking properties and the direct effect in depressing the myocardium as well as its action on central nervous system.

  • It is well known that pharmacological actions of bradykinin are smooth muscle dilatation, increase in capillary permeability, accumulation and migration of leucocytes and inducement of pain. The most significant action of bradykinin is the dilatation of blood vessels. The responses of bradykinin (0.5ug/kg, i. v. injection) on blood pressure were observed before and after single i. v. administration of guanethidine (2mg/kg) in the rabbits. The result of experiment was as follows: In the rabbit pretreated with guanethidine, the depressor response of bradykinin was much pronounced in comparison with that of normal rabbit.

  • Fructus Tribuli has been used in Korea for many centuries as a therapeutic agent for headache and dizziness due to hyperactivity of the liver-yang. The effect of Fructus Tribuli on the blood pressure is not known. The purpose of this study was to determine the effect of Fructus Tribuli on blood pressure and artery rats and rabbits. 1. Fructus Tribuli exerted a dose-dependent relaxation of isolated rabbit common carotid artery. 2. Blood pressure was significantly decreased by Fructus Tribuli (10.0mg/kg) in rats.

  • According to the original documents, Sagungsan is considered as an effective drug for controlling the hypertensive epistaxis induced by tension of autonomic nerve and it's hyperfunction. The present experiment was designed to understand the effect of Sagungsan extract on the hemostatic action, intracranial pressure, blood pressure and cardiovascular system in experimental animals. And thus the bleeding time, prothrombin time, capillary dilation, blood pressure, Intracranial pressure, and enzymatic analysis of the ATPase activities were studied. The result obtained here were as followings: 1. Sagungsan water extract reduced the bleeding time in mouse, and prolonged the prothrombin time in rabbits. 2. The drug extract increased the tail volume by capillary dilation in rats. 3. The drug extract inhibited the increase of intracranial pressure and arterial blood pressure in rabbits. 4. At the early time, the increase of arterial blood pressure by the drug extract significantly inhibited by pretreated atropin and regitine in rabbits. 5. The drug extract relaxed the smooth muscle by stimulating the Mg2+-Ca2+-ATPase activities of gastric sarcoplasmic reticulum isolated from rabbit stomach. 6. The drug extract stimulated the heart contraction by inhibiting the $Mg^{2+}-Ca^{2+}-ATPase$ activities of cardiac sarcoplasmic reticulum isolated from rabbit heart. The inhibitory mechanism was reversible and noncompatitive. 7. The drug extract increased the hepatic blood volume by stimulating the hepatic total ATPase activities and hepatic metabolism. 8. The drug extract acted as a tranquilizer by inhibiting the neural Na+-K+-ATPase activity. According to the results, Sagungsan water extract dilated the capillaries, stimulated the heart beat, and thus increased the blood flow with decreasing the intracranial pressure and blood pressure. These effects stanches the epistaxis collectively.

강활(羌活)이 가토(家兎) 혈관평골근(家兎 血管平滑筋)의 긴장성(緊張性) 조절(調節)에 미치는 영향(影響) (The Effects of Notopterygii Rhizoma on the Carotid Arterial Tension in Rabbit)

  • The purpose of this study was to analyze the Rhizoma on the blood pressure, heart rate and to define the mechanism of Notopterygii Rhizoma-induced relaxation in rabbit common carotid arterial contracted by agonists. Method : In order to explore the effect of Notopterygii Rhizoma on the blood pressure and heart rate, Notopterygii Rhizoma extract was injected in vein of rabbit ear. In order to investigate the effect of Notopterygii Rhizoma on norepinephrine(NE)-induced contracted rabbit carotid arterial strips, transverse strips with intact or damaged endothelium were used for the experiment using organ bath. To analyze the mechanism of Notopterygii Rhizoma-induced relaxation, Notopterygii Rhizoma extract infused into NE-induced contracted strips induced by agonists after treatment of methylene blue, propranolol, ouabain and it infused into serotonin, potassium chloride-induced contracted strips. Result : The blood pressure was significantly decreased by Notopterygii Rhizoma, but heart rate was insignificantly. In addition, Notopterygii Rhizoma significantly relaxed the norepinephrine, serotonin, potassium-induced contracted strips with intact endothelium or damaged endothelium. The relaxing effect of Notopterygii Rhizoma In NE-induced contracted strips with damaged endothelium by pretreatment of methylene blue, propranolol was not changed, but Ouabain was significantly decreased. Conclusion : These results were shown that Notopterygii Rhizoma affected the NE -induced contracted smooth muscle without the participation of endothelium, and demonstrated that the mechanism of NotoDtervgii Rhizoma-induced relaxation was the obstruction of receptor-operated Ca2+ channel.

고혈압(高血壓) 치료제(治療劑)의 개발(開發)에 관(關)한 연구(硏究)(I) -등심초(燈心草)의 혈압강하(血壓降下) 작용(作用) 및 기전(機轉)- (A Study on the Development of Hypotensive Agent (I) -Hypotensive Action and Mechanism of Junci Herba in the Rabbit-)

  • The blood pressure response to Junci Herba water and methanol extracts in rabbit and $LD_{50}$ to Junci Herba water extract in mouse were investigated in order to develop a hypotensive agent from natural resources. $LD_{50}$ of Junci Herba water extract (WE) was 600 mg/kg in mouse, when WE was injected intraperitonealy. Junci Herba water and methanol extract, when injected into the vein of rabbit, produced a fall of blood pressure. Hypotensive effect of WE was suppressed by atropine and potentiated by phentolamine, while not affected by avil, propranolol, and phenoxybenzamine. Intravenous injection of chlorisondamine weakened the hypotensive effect of WE, and WE produced hypertensive effect in this rabbit. Intravenous injection of bretylium did not affect the hypotensive effect of WE, but WE produced hypertensive effect in this rabbit. In the rabbit treated with chlorisondamine, hypertensive effect of WE was suppressed by methysergide or bretylium, but not affected by atropine or phenoxybenzamine.

  • Isoimperatorin and Oxypeucedanin isolated from root of Angelica Koreana Max. (Umbelliferae) that is used as "Kang-Whal" a botanical drug, in Korea, Show following general pharmacologic activities. The treatment with Oxypeucedanin on the exercised duodenum of the rabbit increased the motility of it while isoimperatorin shows little effects. But on the exercised heart of the frogs, both components depressed its contractility. Whereas Oxypeucedanin depressed both blood pressure of the carotied artery and respiration of the rabbit, Isoimperatorin tends to increase blood pressure, respiration and the tonus of the muscle of the earth worm. Oxypeucedanin did not affect the tonus of the muscles of the earth worm, but depressed the uterus contraction of the rabbit, which was, on the contrary, increased by Isoimperatorin.

Ginnol이 가토호흡(家兎呼吸)에 미치는 영향(影響) (Respiratory effect of Ginnol on rabbits)

  • Ginnol was obtained from Gingko Biloba L. by purification of its extract. Author observed the hyperventillation effect on pentobarbital anesthetized rabbit with Ginnol but there was no significant change in blood pressure. Morphine sulfate, even in large doses, did not alter the hyperventillation effect of Ginnol on rabbit. Blood chemistry of Ginnol treated rabbit showed slight increase in blood pH but within a physiological normal range, and increased total $CO_2$ content. While slight decreased $pCO_2$, increased ${HCO_3}^-$ and decreased $H_2CO_3$ and with decreased blood $Cl^-$ meant an uncompensatory alkalotic change but methemoglobin was not detected.
