Let (R, m) be a 2-dimensional regular local ring with algebraically closed residue field R/m. Let K be the quotient field of R and $\upsilon$ be a prime divisor of R, i.e., a valuation of K which is birationally dominating R and residually transcendental over R. Zariski showed that there are finitely many simple $\upsilon$-ideals $m\;=\;P_0\;{\supset}\;P_1\;{\supset}\;{\cdots}\;{\supset}\;P_t\;=\;P$ and all the other $\upsilon$-ideals are uniquely factored into a product of those simple ones [17]. Lipman further showed that the predecessor of the smallest simple $\upsilon$-ideal P is either simple or the product of two simple $\upsilon$-ideals. The simple integrally closed ideal P is said to be free for the former and satellite for the later. In this paper we describe the sequence of simple $\upsilon$-ideals when P is satellite of order 3 in terms of the invariant $b_{\upsilon}\;=\;|\upsilon(x)\;-\;\upsilon(y)|$, where $\upsilon$ is the prime divisor associated to P and m = (x, y). Denote $b_{\upsilon}$ by b and let b = 3k + 1 for k = 0, 1, 2. Let $n_i$ be the number of nonmaximal simple $\upsilon$-ideals of order i for i = 1, 2, 3. We show that the numbers $n_{\upsilon}$ = ($n_1$, $n_2$, $n_3$) = (${\lceil}\frac{b+1}{3}{\rceil}$, 1, 1) and that the rank of P is ${\lceil}\frac{b+7}{3}{\rceil}$ = k + 3. We then describe all the $\upsilon$-ideals from m to P as products of those simple $\upsilon$-ideals. In particular, we find the conductor ideal and the $\upsilon$-predecessor of the given ideal P in cases of b = 1, 2 and for b = 3k + 1, 3k + 2, 3k for $k\;{\geq}\;1$. We also find the value semigroup $\upsilon(R)$ of a satellite simple valuation ideal P of order 3 in terms of $b_{\upsilon}$.
This study was conducted to investigate the genotypes and frequencies of Melanocortin 1 Receptor(MC1R) genes in pigs which plays a central role in regulation of eumelanin (black/brown) and phaeomelanin(red/yellow) pigment synthesis within the mammalian melanocytes. Four different breeds of pigs(20 Landrace, 20 Yorkshire, 20 Duroc, and 93 Jeju native black pigs) were used and PCR-RFLP analysis of MC1R gene was also carried out. Two regions of MC1R genes (428bp and 405bp) were amplified using two specific primers (MERL1-EPIG2, EPIG1-EPIG3), respectively and MC1R allele were determined using 2 restriction enzymes (BspHⅠ, AccⅡ). The results of this experiment indicated that MC1R allelic type in Landrace, Large Yorkshire and Duroc were MC1R *2 (Ep), MC1R *2 (Ep), MC1R *4 (e), respectively. However, various allelic types of MC1R genes were detected in Jeju native black pigs. MC1R allelic type of Jeju black pigs was MC1R*2 type as in Meishan and Large black breeds or MC1R*3 type as in Hampshire and Berkshire breeds and the gene frequencies of ED1 and ED2 were 0.554 and 0.446 in average.
A number of 7-[(3-methylthio or methylthiomethyl)pyrrolidiny]qui nolone-3-carboxylic acids were synthesized by condensation of 7-fluoro substituted quinolone-3-carboxylic acid with 3-methylthiopyrrolidine or 3-methylthiomet hylpyrrolidne. 3-Methylthiopyrrolidine or 3-methylthio-methylpyrrolidine which was prepared from N-benzyl-3-hydroxy pyrrolidine or 3-hydroxymethylpyrrolidine. The in vitro antimicrobial activity of them were tested against twenty species of Gram-positive or Gram-negative microorganisms. It showed remarkable antibacterial activity, particularly against Gram-positive microorganisms. Among those 1-cyclopropyl-6,8-difluoro-7-(3-methylthiomethy-lpyrrolidinyl)-1,4-d ihydro-4-oxo-3-quinolinecarboxylic acid(7d) and 1-cyclopropyl-6- fluoro-8-chloro-7-(3-methylthiomethyl pyrrolidinyl)-1,4-dihydro-4-oxo-3-quinolinecarboxylic acid (7f) showed the most potent in vitro antibacterial activity.
Mutations in the DNMT3A and IDH genes represent the most common genetic alteration after FLT3/NPM1 in acute myeloid leukemia (AML). We here analyzed the frequency and distribution pattern of DNMT3A and IDH mutations and their associations with other molecular markers in normal karyotype AML patients. Fortyfive patients were screened for mutations in DNMT3A (R882), IDH1 (R132) and IDH2 (R140 and R172) genes by direct sequencing. Of the 45 patients screened, DNMT3A and IDH mutations were observed in 6 (13.3%) and 7 (15.4%), respectively. Patients with isolated DNMT3A mutations were seen in 4 cases (9%), isolated IDH mutations in 5 (11.1%), while interestingly, two cases showed both DNMT3A and IDH mutations (4.3%). Nucleotide sequencing of DNMT3A revealed missense mutations (R882H and R882C), while that of IDH revealed R172K, R140Q, R132H and R132S. Both DNMT3A and IDH mutations were observed only in adults, with a higher frequency in males. DNMT3A and IDH mutations were significantly associated with NPM1, while trends towards higher coexistence with FLT3 mutations were observed. This is the first study to evaluate DNMT3A/IDH mutations in Indian patients. Significant associations among the various molecular markers was observed, that highlights cooperation between them and possible roles in improved risk stratification.
The aminolysis of diphenyl thiophosphinic chloride (2) with substituted anilines in acetonitrile at 55.0 oC is investigated kinetically. Kinetic results yield large Hammett ρX (ρnuc = ?3.97) and Bronsted βX (βnuc = 1.40) values. A concerted mechanism involving a partial frontside nucleophilic attack through a hydrogen-bonded, four-center type transition state is proposed on the basis of the primary normal kinetic isotope effects (kH/kD = 1.0-1.1) with deuterated aniline (XC6H4ND2) nucleophiles. The natural bond order charges on P and the degrees of distortion of 42 compounds: chlorophosphates [(R1O)(R2O)P(=O)Cl], chlorothiophosphates [(R1O)(R2O)P(=S)Cl], phosphonochloridates [(R1O)R2P(=O)Cl], phosphonochlorothioates [(R1O)R2P(=S)Cl], chlorophosphinates [R1R2P(=O)Cl], and chlorothiophosphinates [R1R2P(=S)Cl] are calculated at the B3LYP/ 6-311+G(d,p) level in the gas phase.
In order to upgrade native sheep, Rambouillet (R) rams were mated to Kaghani (K) ewes to generate F1 ($R{\times}K$) crossbred ewes. Crossbred ewes were backcrossed to Rambouillet rams to produce B1 ($R{\times}F1$), B2 ($R{\times}B1$) and B3 ($R{\times}B2$) genotypes. Weaning weight of 2605 lambs and wool weight of 2378 mature ewe records, representing R, K, F1, B1, B2 and B3 genotypes, were analyzed to compare genetic variation among genotypes produced during upgrading process and identify genotypes of the highest performance. Performance of Rambouillets was also evaluated under semi-temperate climate. Data were adjusted for yearly variation considering Rambouillet as a control. Genotypes influenced lambs weaning weight (p<.01). B1 lambs were heaviest (18.4 kg) followed in order by B2, F1, B3, R and K lambs (18.3, 17.9, 16.9, 16.8 and 13.2 kg, respectively). The highest wool production was 2.5 kg from R ewes followed by B2 (2.3), B3 (2.3), F1 (2.0) and K (1.2) ewes (p < .01). Ewe mating weight, reproduction, growth and wool production of Rambouillets deteriorated significantly after the first decade of their importation. Compared with the first phase (1959-1971), ewe mating weight, litter size, birth weight, lamb weaning weight and wool production declined by 20, 23, 32 and 36%, respectively, in the second phase (1972-1988).
The nucleophilic substitution reactions of (2R,4R,5S)-(+)-2-chloro-3,4-dimethyl-5-phenyl-1,3,2-oxazaphospholidine 2-sulfide (3) with substituted anilines ($XC_6H_4NH_2$) and deuterated anilines ($XC_6H_4ND_2$) are investigated kinetically in acetonitrile at $5.0^{\circ}C$. The anilinolysis rate of 3 involving a cyclic five-membered ring is considerably fast because of small negative value of the entropy of activation (${\Delta}S^\neq=-2cal\;mol^{-1}\;K^{-1}$) over considerably unfavorable enthalpy of activation (${\Delta}H^\neq=18.0\;kcal\;mol^{-1}$). Great enthalpy and small negative entropy of activation are ascribed to sterically congested transition state (TS) and bulk solvent structure breaking in the TS. A concerted $S_N2$ mechanism with a backside nucleophilic attack is proposed on the basis of the secondary inverse deuterium kinetic isotope effects, $k_H/k_D$ < 1.
The black rockfish (Sebastes schlegeli) is an important commercial species in Korean fisheries. We used acoustic telemetry using coded ultrasonic transmitters to track the home ranges and homing routes of the black rockfish that inhabit in the Tongyeong marine ranching area. Twenty-four fish were released at five points. The distance from the capture point (C1) to the released point was 0 (R7), 0.2 (R1), 0.9 (R2), 1.3 (R3) and 1.9 (R4) km. Two of the three fish released at R1 (0.2 km) returned home in under 48hrs. Another was found 0.8km from C1. Two of the five fish released at R2 returned home, but it took over 250 days. None of the ten fish released at R3 and R4 returned home. Three of the ten fish moved 1.1-2.6 km from C1 and were found near a cage or a natural reef. Six fish were released at R7. The younger the fish, the wider was its movement range around the artificial reefs. Fish over 3 years old were likely to move less far.
PICK1, a PDZ domain-containing protein, is known to increase the reuptake activities of dopamine transporters by increasing their expressions on the cell surface. Here, we report a direct and functional interaction between PICK1 and dopamine $D_3$ receptors ($D_3R$), which act as autoreceptors to negatively regulate dopaminergic neurons. PICK1 colocalized with both dopamine $D_2$ receptor ($D_2R$) and $D_3R$ in clusters but exerted different functional influences on them. The cell surface expression, agonist affinity, endocytosis, and signaling of $D_2R$ were unaffected by the coexpression of PICK1. On the other hand, the surface expression and tolerance of $D_3R$ were inhibited by the coexpression of PICK1. These findings show that PICK1 exerts multiple effects on $D_3R$ functions.
Objective: Numerous studies have indicated that the apoptosis and proliferation of granulosa cells (GCs) are closely related to the normal growth and development of follicles and ovaries. Previous evidence has suggested that miR-126-3p might get involved in the apoptosis and proliferation of GCs, and phosphatidylinositol 3-kinase regulatory subunit 2 (PIK3R2) gene has been predicted as one target of miR-126-3p. However, the molecular regulation of miR-126-3p on PIK3R2 and the effects of PIK3R2 on porcine GCs apoptosis and proliferation remain virtually unexplored. Methods: In this study, using porcine GCs as a cellular model, luciferase report assay, mutation and deletion were applied to verify the targeting relationship between miR-126-3p and PIK3R2. Annexin-V/PI staining and 5-ethynyl-2'-deoxyuridine assay were applied to explore the effect of PIK3R2 on GCs apoptosis and proliferation, respectively. Real-time quantitative polymerase chain reaction and Western Blot were applied to explore the regulation of miR-126-3p on PIK3R2 expression. Results: We found that miR-126-3p targeted at PIK3R2 and inhibited its mRNA and protein expression. Knockdown of PIK3R2 significantly inhibited the apoptosis and promoted the proliferation of porcine GCs, and significantly down-regulated the mRNA expression of several key genes of PI3K pathway such as insulin-like growth factor 1 receptor (IGF1R), insulin receptor (INSR), pyruvate dehydrogenase kinase 1 (PDK1), and serine/threonine kinase 1 (AKT1). Conclusion: MiR-126-3p might target and inhibit the mRNA and protein expressions of PIK3R2, thereby inhibiting GC apoptosis and promoting GC proliferation by down-regulating several key genes of the PI3K pathway, IGF1R, INSR, PDK1, and AKT1. These findings would provide great insight into further exploring the molecular regulation of miR-126-3p and PIK3R2 on the functions of GCs during the folliculogenesis in female mammals.
