Identification of Bacteriocin-Producing Lactobacillus paraplantarum First Isolated from Kimchi

  • A bacteriocin-producing lactic acid bacterium with inhibitory activity against the growth of Lactobacillus plantarum was isolated from kimchi, a traditional Korean fermented vegetable. For the identification of the isolate, its 16S rDNA was sequenced. As a result, the sequence showed $99\%$ homology with those from Lactobacillus paraplantarum, Lb. plantarum, and Lactobacillus pentosus. For further identification of the isolate, the sequence of its 16S/23S rDNA spacer region was determined, and the sequence matched perfectly with that of Lb. paraplantarum. SDS­PAGE fingerprinting of whole-cell proteins of the isolate was almost identical with that of Lb. paraplantarum. The isolation and identification of Lb. paraplantarum suggest that Lb. paraplantarum is one of the lactic acid bacteria involved in kimchi fermentation.

Genetic Diversity and Pathogenicity of Cylindrocarpon destructans Isolates Obtained from Korean Panax ginseng

  • We analyzed the genetic diversity of Cylindrocarpon destructans isolates obtained from Korean ginseng (i.e., Panax ginseng) roots by performing virulence tests and nuclear ribosomal gene internal transcribed spacer (ITS) and mitochondrial small subunit (mt SSU) rDNA sequence analysis. The phylogenetic relationship analysis performed using ITS DNA sequences and isolates from other hosts helped confirm that all the Korean C. destructans isolates belonged to Nectria/Neonectria radicicola complex. The results of in vivo and ex vivo virulence tests showed that the C. destructans isolates could be divided into two groups according to their distinctive difference in virulence and the genetic diversity. The highly virulent Korean isolates in pathogenicity group II (PG II), together with foreign isolates from P. ginseng and P. quinquefolius, formed a single group. The weakly virulent isolates in pathogenicity group I, together with the foreign isolates from other host plants, formed another group and exhibited a greater genetic diversity than the isolates of PG II, as confirmed by the mt SSU rDNA sequence analysis. In addition, as the weakly virulent Korean isolates were genetically very similar to the foreign isolates from other hosts, they were likely to originate from hosts other than the ginseng plants.

사과푸른곰팡이병에 관여하는 Penicillium의 계통분석, 형태 및 병원성 (Phylogenetic Analysis, Morphology and Pathogenicity of Penicillium spp. associated with Blue Mold of Apple in Korea)

  • Blue mold is the most important postharvest disease of apples in Korea. Apple fruits with blue mold symptoms were collected from storages in different locations in Korea and were investigated for their association with Penicillium species. A total of sixty five isolates of Penicillium were sampled from the collected apples. Based on DNA sequence analysis of ${\beta}$-tublin gene and ITS and lsu rDNA (ID region) and morphological characteristics, they were identified as P. crustosum, P. expansum, P. italicum, P. solitum and P. sp.. P. sp. which is closely related to P. hirsutum is a new species, not reported before. P. expansum (35%) was predominant species followed by P. crustosum. The phylogenetic tree inferred from combined ${\beta}$-tublin and ID region sequence showed good correlation with species that are defined by morphological characteristics. In pathogenicity test, apples were wound-inoculated with conidial suspension and incubated at $20-22^{\circ}C$. The most severe and destructive species was P. expansum. The species caused a decayed area 42-50mm in diameter after 8-10days. Decayed area caused by P. crustosum and P. sp. was 26-32mm and 20-26mm, respectively. This is the first record of P. crustosum, P. italicum and P. sp. from apple in Korea.

벼 뿌리 내생 항균성 Serratia marcescens의 분리 및 동정 (Isolation and Identification of Rice Root Endophytic Antagonistic Serratia marcescens)

  • 벼에서 문제시되는 도열병과 잎집무의마름병을 생물학적으로 방제하기 위해 병원균과 생태학적 지위가 비슷한 벼 뿌리에서 내생하는 S. marcescens 23 균주를 분리하였다. 선발 균주들을 공시하여 R. solani와 P. grisea에 대한 길항능력을 검정하여 R. solani와 P. grisea에 각각 83.9%. 88.3%의 높은 억제율을 보인 RSm220 균주를 선발하였다. 선발된 RSm220은 생리ㆍ생화학적 특성 검정결과 S. marcescens type strain과 높은 상관성을 나타내었고. 16S rDNA sequencing에 의한 계통 분석에 의해 S. marcescens의 16S rDNA sequence에 98.2% 유사성을 나타내어 S. marcescens로 동정되었다 내생성 S. marcescens RSm220은 벼 도열병과 잎집무늬마름병에 대한 생물학적 방제제로의 사용이 가능할 것으로 사료된다.

제주도에 서식하는 목본 식물의 잎에서 분리한 미기록 내생균 (Identification of Unrecorded Endophytic Fungi Isolated from Leaves of Woody Plants in Jejudo, Korea)

  • 본 연구는 제주도 사려니숲과 동백동산에서 편백나무(Chamaecyparis obtusa), 삼나무(Cryptomeria japonica), 비자나무(Torreya nucifera), 꽝꽝나무(Ilex crenata), 동백나무(Camellia japonica) 등 5종의 목본 식물의 잎에서 내생균을 분리하고, 형태 및 분자생물학적 특징 통해 동정하였다. 이를 위해 primer인 ITS1F와 ITS4를 이용하여 internal transcribed spacer (ITS) rDNA영역과 primer LR0R과 LR16을 이용하여 large subunit rDNA 영역을 그리고 primer Bt2a과 Bt2b를 이용하여 ${\beta}$-tubulin 영역의 염기서열을 동시에 분석하였다. 그 결과 Mycosphaerella aleuritidis, Neofusicoccum eucalyptorum, Neofusicoccum parvum, Phyllosticta citrichinensis, Phyllosticta cryptomeriae, Phomopsis cotoneastri, Sphaerulina rhododendricola, Guignardia mangiferae, Lophodermium jiangnanense, Lophodermium minus 등 10종의 미기록 내생균을 동정하였고, 이를 보고하고자 한다.

수확 후 저장 배에서 분리한 Penicillium속 균의 유전적 다양성 (Genetic Diversity of Penicillium isolates Isolated from Pears with Postharvest Decay in Storage)

  • 평택일원의 6개의 배 저장고로부터 300개의 부패된 배를 수집 하여 병반부위로부터 곰팡이를이분리 하여 조사한 결과 Penicillium 균이 이병배중에서 84%가 분리되었다. 84 Penicillium 균주를 대상으로 URP2R primer로 균주 간의 DNA 다양성을 조사한결과 18 type의 Penicillium 균주를 검출 할 수 있었으며 URP1F, URP2F, URP2R, URP4R의 PCR 다형성밴드를 이용한 유전적 유연관계분석으로 4 group으로 나눌 수 있었다. 18 Penicillium균주의 형태적특징을 조사한결과 3가지의 특징적인 배양적 특징을 갖는 것으로 확인 되었으며 포자형성과 포자형태에서도 3가지 type이 관찰 되었는데 SP-15균주 type, KP-1-1 균주 type, SP-17 type으로 PCR 다형성결과와 유사한 경향을 나타내었다. SP-15형은 형태적, rDNA의 ITS 염기서열 분석에서 P. expansum로 동정 되었으며 분리된 Penicillium 균주 중 70%를 차지하는 우점종인 것으로 나타났으므로 향 후 URP-PCR profile은 종간, 종내 계통 분류에 유용하게 이용 할 수 있다.

  • To understand the functional roles of the neuron-specific $\beta$-subunit of large-conductance calcium-activated potassium ($BK_{Ca}$) channel, we isolate the full-length complementary DNA of $\beta$4-subunit from rat brain library and investigated its effects on the function of $\alpha$-subunit (Slo). The deduced amino acid sequence of rat $\beta$4 (r$\beta$4), 210 amino acids in length, was closely related to those of $\beta$4 subunits in other mammalian species but showed only a limited sequence homology to the other $\beta$-subunits, $\beta$1 to $\beta$3.(omitted)d)

  • We isolated two parvalbumin cDNAS by expressed sequence tag analysis (1,577 ESTs in total) from the self-fertilizing fish Rivulus marmoratus (Cyprinodontiformes, Rivulidae). Two isoforms of parvalbumin genes showed high similarity to those of carp at 88% and 91% amino acid residues identity, respectively, and showed 79.8% similarity between two parvalbumin isoforms. Of 1,577 ESTs from R. marmroatus sequenced, parvalbumin 1 gene was most abundant. This gene was strongly expressed in the order of muscle, eye, and brain, while it was expressed slightly in other tissues. In this paper, we discussed on the R. marmoratus parvalbumin genes on its sequence and basic characteristics.

Two New Species of Cryptococcus sp. and Candida sp. from Wild Flowers in Korea

  • Among 80 types of yeast isolated from wild flowers in Daejeon, Korea, two species that have not yet been identified by phylogenetic analysis of the internal transcribed spacer-2 (ITS2) genes and 26S rDNA sequences were identified as Candida sp. 44-C-1 and Cryptococcus sp. 9-D-1. Neither of the newly identified species formed ascospores, while Candida sp. 44-C-1 formed pseudomycelium and Cryptococcus sp. 9-D-1 did not.

TFSCAN 검색 프로그램 TFSCAN의 개발

  • TFD is a transcription factor database which consists of short functional DNA sequences called as signals and their references. SIGNAL SCAN, developed by Dan S. Prestridge, is used to determine what signals of TFD may exist in a DNA sequence. This program searches TFD database by using a simple algorithm for character string comparison. We developed TFSCAN that aims at searching for signals in an input DNA sequence more efficently than SIGNAL SCAN. Our algorithms consist of two parts, one constructs an automata by scanning sequences of rFD, the other searches for signals through this automata. Searching for signal-related references is radically improved in time by using an indexing method. Usage of TFSCAN is very simple and its output is obvious. We developed and installed a TFSCAN input form and a CGI program in GINet Web server, to use TFSCAN. The algorithm applying automata showed drastical results in improvement of computing time. This approach may apply to recognizing several biological patterns. We have been developing our algorithm to optimize the automata and to search more sensitively for signals.
