Immunochemical Determination of Plant Constituents (1990년 제23회 학술발표회 논문초록)

  • Until relatively recently plant scientists have made little use of immunological techniques. Now, however, more and more researchers are discovering the powder of these techniques for the screening of immunomodulators and for the detection, quantitative determination and localization of compounds in plant materials. Especially, the recent developments in the fields of plant biotechnology and plant genetic engineering make it even more important for forkers in the plant sciences to become acquainted with the more sophisticated methods. The possible methods include onestep purification of antigens, visualization in situ by immunocytochemis try and on polyacrylam ids gel s by ni trocellulose Western blotting and quantification by various immunoassay. Among them, in this reviews, the quantitative determination methods are to be reviewed. There are several kinds of methods for the quantitative determination of plant constituents such as colorimetry, TLC, GLC, DCC, UV derivatization, densitometry and HPLC. When the complexity of plant constituents is considered. densitometry and HPLC have many advantages in sensitivity and separation ability. After a 11 some advarltages of two methods meritiorled above, all of these methods have many disadvantages and inconveniences. Previous purification for the application of all these methods make them less sensitive and more tedious. Immunoassay can solve these problems in part. But immunoassay also has some limitations. Specificity of immunoassay, contrary, can be considered to be disadvantages. Including this the advantages and disadvantages of immunoassay are to be discussed.

  • The sesaminol glucosides in 80% EtOH extract from sesame seeds were separated by high performance liquid chromatography (HPLC). A HPLC system using a Develosil ODS-5 column and gradient elution system from 30% to 80% methanol was selected for separation and quantitative determination of sesaminol triglucoside, sesaminol diglucoside, and sesaminol monoglucoside. Quantitative analyses for these sesaminol glucosides, sesaminol triglucoside, sesaminol diglucoside, and sesaminol monoglucoside were determined on the basis of standard curve of sesaminol glucosides. Sesaminol triglucoside, sesaminol diglucoside and sesaminol monoglucoside contents of the seed of one Korean sesame cultivar, Danbaekggae, were 56.4 mg/100g, 9.6 mg/100g, and 7.5 mg/100g, respectively.   • Ursolic acid was isolated from Prunellae Herba (Prunella vulgaris var. lilacina) and identified by direct comparison with an authentic sample. A method of analysis for the evaluation of ursolic acid was developed based on extraction of ground plant material, followed by quantitative determination using capillary gas chromatography of the TMS derivative. Quantitative analysis by GC after derivatisation under mild silylating conditions showed 0.31% ursolic acid in 20 samples collected throughout regions of Korea while no ursolic acid was detected in the samples of the whole plant of Thesium chinense, a substitute for Prunellae Herba in southern regions of Korean peninsula.

  • Another application of the new quantitative analytical method of prussic acid by "Microdiffusion analysis" for the determination of prussic acid in human blood was studied. The blood containing potassium cyanide was dropped in outer room of unit, and then N-sulfuric acid was added. The liberated HCN gas was absorbed into nickel sulfate solution of inner room, afterward, absorbed prussic acid was determined with EDTA by residual titration. The result was coincided with the result of Liebig Denigs' method at ordinary temperature.

  • Separation and quantitative determination of 20-hydroxyecdysone from Achyranthis Radix has been conducted by using HPLC method. 20-hydroxyecdysone in a methanol extract from the raw drug was separated on a reverse phase column using a $CH_3CN-H_2O\;(18:82)$ solvent system and the average content is $0.0931{\pm}0.0048%$. For the preparation of authentic standard, we isolated 20-hydroxyecdysone from the roots of Achyranthes fauriei by $SiO_2$ and RP-18 column chromatography.

A Convenient HPLC/ELSD Method for the Quantitative Analysis of Betaine in Lycium chinense

  • In order to facilitate the quality control of betaine from the fruits of Lycium chinense, we have developed a rapid and simple method for quantitative determination. Determination was achieved on a Discovery C18 column with an isocratic solvent system of 0.32% perfluoropentanoic acid aqueous-acetonitrile at a flow-rate of 0.5 mL/min and detected an ELSD. The method was reproducible with intra- and inter-day variations of less than 6% (R.S.D). The recoveries were in the range of 90.01~100.05%. The method turned out to be fast and simple, furthermore, to have a good selectivity and sensitivity for the quantity determination of betaine in the fruits of L.chinense.

  • The rhizomes of Salvia miltiorrhiza Bunge(Labiatae) has been used in Chinese traditional medicine for the treatment of coronary heart diseases and myocardial infarction. As a part of a research for standardization of crude oriental drugs, we have determined the content of tanshinone IIA in Salvia miltiorrhiza purchased from various regions of Korea. The HPLC method by which quantitative analysis was conducted, showed reproducible results and chromatographic isolation of tanshinone IIA was accomplished successively.

  • Qualitative and quantitative analysis for carbonyl compounds contained in the five kinds of filtrated myulchichuts have been made using HPLC. Carbonyl compounds were separated by converting them into 2,4-dinitrophenylhydrazine derivatives which, in turn extracting with hexane. The extract was dehydrated, filtered and examined on HPLC with $C_{18}column$. The quantitative determination of carbonyl species was achieved using standard calibration technique. Highly reproduceable results were obtained by this methods and the amounts of 2,4-dinitrophenylhydrazine derivatives were highly correlative to peak heights. As the results, we find that the seven kinds of carbonyl compounds are contained in each of myulchichuts examined in this study. Five of which are identified as pyruvic acid, formaldehyde, acetaldehyde, acetone and butanone. But the amounts of carbonyl compounds vary widely with the kinds of the filtrated myulchichuts.

  • Geniposide, an iridoid glucoside, was isolated from the cortex of Eucommia ulmoides by repealed silica gel chromography and identified by its spectral data. The quantitative determination of geniposide with HPLC has been developed and this method provided a tool for standardization of the commercial Eucommiae Cortex.
