Effects of Cobalt-60 Gamma Ray Irradiation on Blood Cells and Enzyme Activity of Albino Rats (코발트-60 감마선(線) 조사(照射)가 흰쥐의 혈액성분(血液成分) 및 수종(數種) 효소(酵素)의 활성(活性)에 미치는 영향(影響))

  • This paper was aimed to study the effects of Cobalt-60 gamma ray irradiation on Albino rat blood cells and the activity of enzymes were measured using blood cell auto analyzer (Cell-Dyn 900) and enzyme autoanalyzer (Gilford 3500) respectively. Cobalt-60 gamma rays those are grouped into 200R, 400R, 600R, 800R, 1,000R, 1,200R, 1,300R, 1,400R, 1,500R and 1,600R to the rats of male and female and measured the numeral varieties of blood corpuscles of the rats and the active varieties of enzyme of acid, alkaline phosphatase (ACP, ALP), lactic acid dehydrogenase (LDH), glutamic oxaloacetic transaminase (GOT) and glutamic pyruvic transaminase. (GPT) those came out after 1, 3, 5 or 7 days. The results are summarized as follows: 1. In both case of male and female, the numeral varieties of the erythrocytes, in the 200R group of irradiation showed the minimum value in the 3 days group and a tendency of increase in the 5 and 7 days groups, while the numeral varieties of the leucocytes in every groups of irradiation showed a tendency of a rapid decrease after a day. The numeral varieties of the blood platelet in both case of male and female didn't show generally any great change in a day and 3 days groups, but showed a rapid decreasing appearance after the 5 days. 2. In both case of male and female, the activities of ACP were on the decrease gradually after 3 days of the irradiation and the activities of ALP were on the decrease after 5 days. Similarly in both case of mate and female, the activities of LDH showed the decrease after 3 and 5 days, and the phenomenous of GOT showed an appearance of increase in a day, but decreased after 3 days. As for the GPT activities couldn't find any great change in the male rats in comparison with the control groups, but in the female rats a tendency of the increase in 600R, 800R, 1,000R groups after 3 days. 3. In the case of the irradiation of high quantity of ray more than 1,200R, all rats died after 4 days, and by the irradiation of 1,600R all rats died after 3 days, and it showed the sensitive response of a living body to the irradiation of high quantity.

Changes in quality characteristics of sliced garlic with different freezing conditions during storage (냉동조건에 따른 편마늘의 냉동저장 중 품질변화)

  • This study was performed to evaluate the effect of different freezing conditions and storage periods on physicochemical and microbial characteristics of garlic. Garlics were washed, dried, sliced to 0.3 cm then packed in LDPE+LLDPE film bags. They were treated with still-air freezing at $-20^{\circ}C$ (SAF20), $-40^{\circ}C$ (SAF40) and immersed-liquid freezing at $-40^{\circ}C$ (ILF40). Frozen garlics were stored under frozen storage conditions for 7 months at $-20^{\circ}C$ and quality characteristics were measured monthly during the frozen storage. Freezing rate of garlic was the fastest in ILF 40 (10 min), SAF40 (40 min) and SAF20 (1,600 min) sequentially. In ILF40, drip loss, cutting force, total aerobic bacteria count and pH were the lowest, whereas pyruvic acid and allicin content were the highest (p<0.05) during frozen storage, these results were the most similar characteristics with the fresh garlic. During frozen storage, drip loss, color difference and total organic acid content were significantly fluctuated in SAF20 (p<0.05), while they were not changed in ILF40. Overall, total aerobic bacteria count and pH decreased, cutting force, pyruvic acid and allicin content remained unchanged in all groups. In conclusion, the optimal freezing conditions for garlic with the least quality changes was considered to be ILF40 (immersed liquid freezing), keeping quality characteristics up to 7 months by freezing storage.

Preparation of Korean Traditional Alcoholic Beverage (Yakju) by a Protoplast Fusion Yeast Strain Utilizing Starch and its Quality Characteristics (전분분해 효모융합체를 이용한 전통 발효주의 제조와 품질특성)

  • The objectives of this study were to evaluate the physico-chemical and sensory characteristics of a Korean traditional alcoholic beverage (yakju) prepared using different nuruk (Korean-style koji) concentrations and yeasts such as the fusant FA776 and Saccharomyces cerevisiae KOY-1, respectively. The fusant FA776, which has alcohol-fermenting and starch-utilizing properties, was formed by Saccharomyces cerevisiae KOY-1 and Saccharomyces diastaticus KCTC1804. The fermentation trial was conducted in a 5 L lab-scale jar at $25^{\circ}C$. The maximum alcohol production of the K-100 and F-50 reached levels of 135.0 mg/mL and 119.4 mg/mL, respectively. The pH values were in a range of 4.3-4.5. Total acidity was in a range of 0.47-0.60%. Organic acids and amino acids were analyzed in order to evaluate variations in its composition and content via HPLC analysis. Organic acids including lactic acid, citric acid, malic acid, and pyruvic acid, and 16 kinds of amino acids, including aspartic acid, were detected in all treatments. K-100 showed the highest amino acid contents, whereas F-50 exhibited the lowest amino acid contents. Volatile flavor components such as phenylethyl alcohol, isoamyl alcohol, 2-methylthiophane, isobutyl alcohol, and ethyl succinate were detected as a major component in all treatments, as determined via gas chromatography. The results of our sensory evaluation demonstrated that Yakju fermented by the FA776 fusant yielded more favorable results than S. cerevisiae KOY-1.

Protective Efficacy of Alpha-lipoic Acid against AflatoxinB1-induced Oxidative Damage in the Liver

  • Alpha-lipoic acid (${\alpha}$-LA) is not only involved in energy metabolism, but is also a powerful antioxidant that can protect against hepatic oxidative stress induced by some drugs, toxins, or under various physiological and pathophysiological conditions. Here, we investigated the effect of ${\alpha}$-LA against liver oxidative damage in broilers exposed to aflatoxin $B_1$ ($AFB_1$). Birds were randomly divided into four groups and assigned different diets: basal diet, 300 mg/kg ${\alpha}$-LA supplementation in basal diet, diet containing 74 ${\mu}g/kg$ $AFB_1$, and 300 mg/kg ${\alpha}$-LA supplementation in diet containing 74 ${\mu}g/kg$ $AFB_1$, for 3 weeks. The results revealed that the addition of 300 mg/kg ${\alpha}$-LA protected against the liver function damage of broilers induced by chronic low dose of $AFB_1$ as estimated by a significant (p<0.05) change in levels of plasma total protein, albumin, alkaline phosphatase and the activities of liver glutamic-oxalacetic transaminase and glutamic-pyruvic transaminase. The histopathological analysis also showed that liver tissues were injured in the $AFB_1$ diet, but this effect was alleviated by the addition of 300 mg/kg ${\alpha}$-LA. Additionally, $AFB_1$ induced a profound elevation of oxidative stress in birds, as indicated by an increase in malondialdehyde level, a decrease in glutathione peroxidase activity and a depletion of the glutathione content in the liver. All of these negative effects were inhibited by treatment with ${\alpha}$-LA. Our results suggest that the inhibition of $AFB_1$-induced excess production of lipid peroxides and the maintenance of intracellular antioxidant status may play important roles in the protective effects of ${\alpha}$-LA against $AFB_1$-induced oxidative damage in the liver.

On the Changes in Organic Acids of Strawberry in Air with Different $CO_2$ Concentration (공기중에 혼합된 탄산가스 농도에 따른 저장딸기의 유기산 변화에 관하여)

  • Changes in pH, titratable acidity, some organic acids and brix of the strawberry stored at the different levels of $CO_2$, in air were measured. The changing trends in pH and the acidity of the strawberries were not clear among the storage treatments, and pH of the strawberries in all the treatments was increased(r=0.9615) by contrast to decrease in the acidity(r=-0.8483). Ascorbic acid content of the strawberries at control lot was higher than the contents of the CA stored for the initial 2 weeks after storage. This trend was reversed after that period and the strawberries in air with 30% $CO_2$, showed the highest retention rate, 28% of the initial content after 5 weeks. Among the analyzed organic acids, malic acid was gradually decreased in all the treatments during storage and the decreasing rates of the strawberries in air with 30% $CO_2$, was somewhat stupider than those in the others. Pyruvic acid was reduced in the control lot during storage but drastically increased in the strawberries stored at the CA storage conditions between the 2nd and the 3rd week after storage, and then it was decreased.

Effects of Scutellariae Radix Extract on Lipid Metabolism, Oxidation and Production of Pro-Inflammatory Cytokines in Rats Fed Highly Oxidized Fat (황금추출물이 과산화지질을 급여한 흰쥐의 지질대사, 산화반응 및 전염증성 Cytokine의 생산에 미치는 영향)

  • Objectives: This study investigated the effects of Scutellariae Radix extract (SRE) on lipids metabolism, oxidation and the production of pro-inflammatory cytokines in rats fed highly oxidized fat. Methods: To induce obesity, male Sprague‐Dawley rats were fed a highly oxidized fat diet for 10 weeks. SRE at 100 mg/kg were administered orally to obesity-induced rats for 6 weeks, and their lipid metabolism, oxidation and production of pro-inflammatory cytokines were examined. Results: The concentrations of free fatty acid, triglyceride, total cholesterol, and low density lipoprotein-cholesterol in plasma decreased in SRE-treated groups, although the difference was not significant between control and SRE-treated groups, while that of high density lipoprotein-cholesterol significantly increased in SRE group. The concentrations of total cholesterol and triglyceride in the liver were tended to decrease in SRE-treated group. The concentrations of thiobarbituric acid in plasma and liver were lower in SRE group than in control group. The levels of glutamic oxaloacetic transaminase and glutamic pyruvic transaminase in plasma were decreased in SRE group. Activities of glutathione peroxidase, superoxide dismutase, and catalase in liver were tended to increase in the SRE group. The plasma concentrations of interleukin $(IL)-1{\beta}$, tumor necrosis factor $(TNF)-{\alpha}$ and IL-6 were lower in SRE group than in control group, while that of IL-10 was higher. The liver concentrations of $IL-1{\beta}$, $TNF-{\alpha}$, and IL-6 were tended to decrease while that of IL-10 tended to increase in SRE group. Conclusions: Finally SRE could be used in the production of nutraceuticals for lowering lipids and exerting anti-oxidation and anti-inflammatory effects in obesity rats fed highly oxidized rat.

Physicochemical Properties of Breeding Lines of Colored Barleys (유색보리 육성계통의 이화학적 특성)

  • The below is the results of physicochemical analysis of 40 breeding lines of colored barley (CB) whose systems are different Water content of CB No. 24 showed the lowest value of 7.4% while CB No. 9 showed the highest value of 10.8%. Crude protein of CB $9.7{\sim}12.9%$ range was found. Crude fat content of CB No. 6 showed the highest value of 4.35% while CB No. 34 showed the lowest of 1.35%. Crude ash content of CB No. 31 showed the lowest value of 1.20%. Ca content of CB No. 10 showed the highest value of 717.50 mg% while general barley showed the lowest value of 442.82 mg%. Mg content of CB No. 10 showed the highest value of 1320.00 mg%. Cu content of CB No. 20 showed the lowest value of 2.20 mg% while CB No. 33 showed the highest value of 6.25 mg%. K content of CB No. 20 showed the lowest value of 723.24 mg% while CB No. 1 showed the highest value of 1002.50 mg%. Mn content of CB No. 28 showed the lowest value of 31.72 mg% while general barley showed the highest value of 94.56 mg%. ${\beta}-Glucan$ content of CB No. 25 showed the lowest value of 5.20 mg% while CB No. 28 showed the highest value of 4.46 mg%.

Phosphate Solubilization and Gene Expression of Phosphate-Solubilizing Bacterium Burkholderia multivorans WS-FJ9 under Different Levels of Soluble Phosphate

  • Phosphate-solubilizing bacteria (PSB) have the ability to dissolve insoluble phosphate and enhance soil fertility. However, the growth and mineral phosphate solubilization of PSB could be affected by exogenous soluble phosphate and the mechanism has not been fully understood. In the present study, the growth and mineral phosphate-solubilizing characteristics of PSB strain Burkholderia multivorans WS-FJ9 were investigated at six levels of exogenous soluble phosphate (0, 0.5, 1, 5, 10, and 20 mM). The WS-FJ9 strain showed better growth at high levels of soluble phosphate. The phosphate-solubilizing activity of WS-FJ9 was reduced as the soluble phosphate concentration increased, as well as the production of pyruvic acid. Transcriptome profiling of WS-FJ9 at three levels of exogenous soluble phosphate (0, 5, and 20 mM) identified 446 differentially expressed genes, among which 44 genes were continuously up-regulated when soluble phosphate concentration was increased and 81 genes were continuously down-regulated. Some genes related to cell growth were continuously up-regulated, which would account for the better growth of WS-FJ9 at high levels of soluble phosphate. Genes involved in glucose metabolism, including glycerate kinase, 2-oxoglutarate dehydrogenase, and sugar ABC-type transporter, were continuously down-regulated, which indicates that metabolic channeling of glucose towards the phosphorylative pathway was negatively regulated by soluble phosphate. These findings represent an important first step in understanding the molecular mechanisms of soluble phosphate effects on the growth and mineral phosphate solubilization of PSB.

  • The purpose of this study was carried out to determine the possibility of in vitro maturation of tiger oocytes. Immature oocytes were recovered from a pairs of ovaries. A total of 78 oocytes were collected, of which forty threes were identified as compact cumulus cells and uniform cytoplasm. 43 COCs were in vitro matured at 39℃, 5% CO₂ in air atmosphere for 48 h in a IVM medium (TCM-199 supplement with 10% FBS, 0.6 mM cysteine, 0.2 mM pyruvic acid and 10 IU/㎖ HMG). Toxicity of doxycycline, a broad-spectrum semi-synthetic antibiotic, in Pacific whiteleg shrimp (Litopenaeus vannamei) (흰다리새우(Litopenaeus vannamei)에서 광범위 반합성 항생제 doxycycline의 독성연구)

  • Doxycycline is a semi-synthetic broad-spectrum antibiotic, and it has been used to get rid of bacteria in animals and humans. The use of antibiotics has greatly contributed to the aquaculture production although its misuse sometimes presents public health problems. This study was performed to investigate the toxic effects of doxycycline on whiteleg shrimp (Litopenaeus vannamei) administered for possible infection treatments. The shrimp were allocated into four groups and doxycycline was fed three times a day for 7 days at 0, 20, 50 and 100 mg/kg to each group. After 24 hr following the 7-day treatment, hemolymph and hepatopancreas were used for blood and biochemical analysis: Total hemocyte counts, Total protein, Total cholesterol, Gluscose, Glutamic pyruvic transaminase, Glutamic oxaloacetic transaminase, Glutathione peroxidase, Superoxide dismutase, Glutathione-s-transferases, Total antioxidant capacity colorimetric and Acid phosphatase. In addition, histopathological examination was performed on the hepatopancreas and muscle. It was observed that body weight gain was significantly retarded in 100 mg/kg doxycycline group. Doxycycline was found to induce biochemical or functional disorders at 100 mg/kg as observed many of the blood and biochemical parameters were significantly reduced. In conclusion, it was judged that there will be no major toxicity problems with doxycycline when used for shrimp aquaculture at regular doses.
