Effect of Colored Potato Flakes Against Acetaminophen-induced Liver Damage in Rats

  • We examined the hepatoprotective effects of colored potato flakes on acetaminophen (AAP)-induced liver damage in rats. F344/DuCrj (8 week-old) rats were fed a cholesterol-free diet with 54.9486 g of ${\alpha}$-corn starch/100g diet and were orally treated with 25% colored flakes of Kitamurasaki (KM: light purple), Northern Ruby (NR: red), and Shadow Queen (SQ: medium purple) potatoes co-administered with AAP (0.5 g/100 g diet) for 4 weeks. The hepatic thiobarbituric acid-reactive substances (TBARS) values in the KM, NR, and SQ groups were significantly lower (p<0.05) than those in the control groups with and without AAP. Furthermore, the hepatic catalase, Mn-superoxide dismutase (SOD), and Cu/Zn-SOD mRNA levels in the KM, NR, and SQ groups were higher than those in the control groups with and without AAP. The present findings suggest that colored potato flakes are useful as a prophylactic agent against oxidative liver damage.

  • This study was carried out to evaluate antioxidant and anticancer activity of crudes extracts from colored corn (Zea mays. L.) among particle size with different pulverizing methods (pin mill and ultra fine pulverizer). In cytotoxicity test using extracts from the flours grounded by pin mill and ultra fine pulverizer respectively, it showed that A-549 was the highest anticancer activity among in vitro using 4 cancer cell line types (Hep-G2, A-549, HCT-116 and MCF-7). Assessing the antioxidant activities of crude extracted from different pulverizing methods was measured by trolox equivalent antioxidant capacity (TEAC) and ferric reducing antioxidant power (FRAP). Higher scanvening activity against free radical was observed in the crude extracted from small particle size flours of colored corn than in those of the big particle size.

  • Our review begins with the maize hybrid for grain, called 'Seakso 1,' which was developed in 2008 by the Gangwon Agricultural Research and Extension Services in Korea, and subsequently registered in 2011. In this study, we aimed to investigate the lipid metabolic enzyme activity and inhibitory effect on the adipocyte differentiation, in 3T3-L1 cells of the identified Seakso 1 corn husk and cob extracts (EHCS). We investigated the pancreatic lipase inhibitory effect and anti-adipogenic effect of EHCS.The lipid accumulation and adipocyte differentiation were measured by the procedure of Oil Red O staining, Real-time PCR and the Western blot analysis. The pancreatic lipase inhibitory activity of EHCS was measured at higher levels than those of the positive control (orlistat) at 100, 500, and $1,000{\mu}g/mL$. In particular, EHCS was noted as being significantly inhibited and including a measured adipocyte differentiation and lipid accumulation, when treated during the adipocyte differentiation process in 3T3-L1 cells. Based on the Oil Red O staining, EHCS inhibited lipid accumulation at 19.19%, 33.30% at $1000{\mu}g/mL$, $2000{\mu}g/mL$, respectively. The real-time PCR and Western blot analysis showed that EHCS significantly decreased in the mRNA expression and protein level of obesity-related factors, such as peroxisome-proliferatorsactivated-receptor-${\gamma}$ ($PPAR{\gamma}$) and CCAAT enhancer-binding-proteins ${\alpha}$ ($C/EBP{\alpha}$). This study potentially suggests that the Saekso 1 corn husk and cob extracts may improve lipid metabolism and reduce lipid accumulation.

  • Colonial morphology of the various yeasts often encountered in sputum or other clinical specimens was investigated on the corn meal-potato-yeast extract agar medium (GJCPY) containing orange-yellow pigments extracted from Gardenia jasminoides fruits in hopes of differential identification on primary cultures. The results obtained are as follows. 1) Cryptococcus neoformans which is a medically important yeast and whose colony showed brown to purple brown on GJCPY medium was distinguishable not only from buff colored Cr. laurentii after one week incubation but also from Candida spp. 2) Colony color of Candida albicans, a most common species in sputum specimens and of Ca. parapsilosis, a rare isolate, remained unchanged even after 15 days incubation. 3) Ca. tropicalis, second common isolate from sputums and Ca. krusei, a rare isolate, formed a characteristic rough and wrinkled colonies that permit to differentiate them from others. 4) Rare isolates, Ca. guilliermondii and Ca. lusitaniae, turned to prussian blue within three days of incubation. 5) Torulopsis sp. and Saccharomyces cerevisiae showed glossy grayish blue or light blue after one week incubation. The findings clearly showed that Ga. jasminoides pigments medium was useful to the morphological differentiation of medically important yeasts that were often encountered in sputum or other clinical specimens.

Reproduction of Kwanmo worn during the Hoeryeyeon(會禮宴) Performances in the reign of King Sejong - Focusing on Jinhyunkwan, Pybyun, Buyongkwan - (세종조 회례연시 관모 재현 - 진현관, 피변, 부용관을 중심으로 -)

  • This paper is on the reproduction of kwanmo, which is worn by the dancers in Taepyeongjiak at the National Classical Music Institute, focusing on Jinhyunkwan, Pybyun, Buyongkwan. Taepyeongjiak is a performance of the court banquet called Hoeryeyeon held during the 15th year of Sejong'reign(1433). The Hoeryeyeon performances were held on January 1st and December 22nd, to strengthen the good relationship between the King and his officials. The reproduction of kwanmo worn by the dancers was reproduced for the modern performances on the basis of the literature and relic research. The results of the study are as follows. Jinhyunkwan is the kwanmo that is associated with the literature and it comes from an old Chipokwan, is confirm that liang(梁) of Chipokwan's characteristic vanished. Pybyun is kwanmo which is associated with the military, comes from Jeolpung. The incision line was formed to make the corn shape, and this became the liang(梁). Buyongkwan is corolla adorned with lotus, and is highlighted with colored string of beads. Unlike other corollas, its gilt lotus was adorned with purple-yellow braids.

Manufacture of Sikhe(a Traditional Korean Baverage) Using Corn Silk Extracts (옥수수 수염 추출액을 이용한 식혜 제조)

  • We prepared sikhe (CSE/BR-SH) using corn silk extract and black rice. The pH decreased during saccharification, from pH $5.88{\pm}0.03$ to pH $5.67{\pm}0.02$ after 6 h. However, the brix and reducing sugar contents increased during saccharification of CSE/BR-SH, with the highest levels ($7.6{\pm}0.05$ brix and $4.012{\pm}0.05$ g/l, respectively) being attained at 6 h. Amylase activity increased to 116.12% of control values 1 h after saccharification of CSE/BR-SH, and decreased thereafter. CSE/BR-SH was light purple in color. Soluble phenolic concentration increased markedly from an initial 8.43 g/l to 23.09 g/l at the end of saccharification (6 h), as did DPPH radical-scavenging activity (from an initial 17.3% to 70.98%), Increases were noted in all of ABTS radical-scavenging activity (from 40.25% to 75.32%), reducing power (from 0.241 to 0.682), and ferric reducing antioxidant power (FRAP) (from 0.288 to 1.071).

Ruminal Degradability of Tropical Feeds and Their Potential Use in Ruminant Diets

  • The objective of this study was to determine the degradability of cassava chip (CC), cassava waste (CW), yellow sweet potato (YP), white sweet potato (WP), purple sweet potato (PP), corn meal (CM), and rice bran (RB) using in situ technique. Two ruminally fistulated steers with an average weight of $303{\pm}10kg$ were used to determine in situ degradabilities of DM and OM. Seven feed sources were weighted in nylon bags ($38{\mu}m$ pore size) and incubated ruminally for 1, 2, 4, 6, 8, 12, 24, and 48 h. The results showed that asymptote (a+b) and effective degradability (ED) of DM of energy sources ranked from the highest to the lowest; CC, YP, WP, PP, RB, CW, and CM (99.3, 92.5; 97.6, 87.9; 97.5, 87.9; 97.2, 87.8; 87.5, 63.6; 78.6, 63.0 and 81.7; 59.3, respectively) and for OM asymptote (a+b) and effective degradability (ED) were similar to those of degradation of DM (99.4, 93.4; 98.8, 89.8; 98.5, 89.4; 98.4, 88.1; 92.4, 65.8; 85.1, 66.9 and 83.6, 63.3, respectively). It was concluded that disappearance characteristic of CC was the highest and it may potentially facilitate the achievement of optimal ruminal availability of energy: protein especially with NPN for microbial protein synthesis.

Effect of Saekso 2 Corn Kernels and Cobs Extracts on Antioxidant Activity in Rats Fed High Fat-cholesterol Diet (옥수수 색소 2호 품종의 알곡과 속대 추출물을 첨가한 고지방-고콜레스테롤 식이가 흰쥐의 항산화 활성에 미치는 영향)

  • The objective of this study was to determine the effect of sakso 2 corn kernels and cobs extracts on antioxidant activity in rats fed a high fat-cholesterol diet (HFC) for 2 weeks. 48 male Sprague-Dawley (4-weeks-old) were randomly divided into 6 groups: normal diet (N), HFC (C), HFC and 0.05% kernel extracts of Saekso 2 (T1), HFC and 0.25% kernel extracts of Saekso 2 (T2), HFC and 0.05% cob extracts of Saekso 2 (T3), HFC and 0.25% cob extracts of Saekso 2 (T4). The weight gain in all treatment groups were significantly lower and the food efficiency ratio (FER) in all treatment groups except T3 were lower than C group. Liver index (liver weight/100 g body weight) in N group and T2 were significantly lower than C group. The level of total cholesterol in plasma of N group and T2 were significantly lower than C group and HDL-cholesterol in plasma of N group and T2 were significantly lower than C group. Malondialdehyde (MDA) concentration of thiobarbituric acid reactive substances in N group, T3 and T4 were significantly lower than C group. Activity of catalase (CAT) in all treatment groups were lower than C group. These result suggest that saekso 2 corn kernels and cobs extracts may reduce oxidative damage through the activation of antioxidative defense systems in rats fed high fat-cholesterol diets.

Assessment of Nutritional Components, Antioxidant Contents and Physiological Activity of Purple Corn Husk and Cob Extracts (자색옥수수 포엽과 속대 혼합 추출물의 영양성분, 항산화 활성 물질 함량분석 및 생리활성 평가)

  • The objective of this study was to investigate the worth of extract husk and cobs of the Seakso 1 (EHCS) for the functional foods. We aimed to investigate the proximate composition, fatty acids, amino acids, antioxidant active substance contents, antioxidant activity, inhibitory activity of the ${\alpha}-amylase$ and ${\alpha}-glucosidase$. The proximate composition of the EHCS have represented 6.90% moisture, 7.31% crude ash, 0.52% crude fat and 7.07% crude protein. Among the 17 kinds of amino acids that were analyzed in thd EHCS, the glutamic acid was the highest, with 736.08 mg / 100 g. The fatty acids detected in the EHCS were palmitic acid oleic acid and linoleic acid. The proportion of the unsaturated fatty acids was 83.33%. We determined the contents of the antioxidant active substance by the total polyphenol and flavonoid. The total polyphenol and flavonoid contents were 99.87 mg/g and 25.02 mg/g, respectively. The antioxidative activity of the EHCS were determined using a DPPH and ABTS assay. In the antioxidative activity determination, the DPPH and ABTS radical scavenging activities were 95.62% ($1,000{\mu}g/mL$) and 92.00% ($10,000{\mu}g/mL$), respectively. The inhibitory activity of ${\alpha}-amylase$ and ${\alpha}-glucosidase$ (10 mg/mL) were 95.86% and 76.92%, respectively. These results suggest that the EHCS could be potentially used as a resource for the bioactive materials for health functional foods.
