Magnolol Inhibits LPS-induced NF-${\kappa}B$/Rel Activation by Blocking p38 Kinase in Murine Macrophages

  • This study demonstrates the ability of magnolol, a hydroxylated biphenyl compound isolated from Magnolia officinalis, to inhibit LPS-induced expression of iNOS gene and activation of NF-${\kappa}B$/Rel in RAW 264.7 cells. Immunohisto-chemical staining of iNOS and Western blot analysis showed magnolol to inhibit iNOS gene expression. Reporter gene assay and electrophoretic mobility shift assay showed that magnolol inhibited NF-${\kappa}B$/Rel transcriptional activation and DNA binding, respectively. Since p38 is important in the regulation of iNOS gene expression, we investigated the possibility that magnolol to target p38 for its anti-inflammatory effects. A molecular modeling study proposed a binding position for magnolol that targets the ATP binding site of p38 kinase (3GC7). Direct interaction of magnolol and p38 was further confirmed by pull down assay using magnolol conjugated to Sepharose 4B beads. The specific p38 inhibitor SB203580 abrogated the LPS-induced NF-${\kappa}B$/Rel activation, whereas the selective MEK-1 inhibitor PD98059 did not affect the NF-${\kappa}B$/Rel. Collectively, the results of the series of experiments indicate that magnolol inhibits iNOS gene expression by blocking NF-${\kappa}B$/Rel and p38 kinase signaling.

The Electrical Characteristics of SRAM Cell with Stacked Single Crystal Silicon TFT Cell (단결정 실리콘 TFT Cell의 적용에 따른 SRAM 셀의 전기적 특성)

  • There have been great demands for higher density SRAM in all area of SRAM applications, such as mobile, network, cache, and embedded applications. Therefore, aggressive shrinkage of 6T Full CMOS SRAM had been continued as the technology advances, However, conventional 6T Full CMOS SRAM has a basic limitation in the cell size because it needs 6 transistors on a silicon substrate compared to 1 transistor in a DRAM cell. The typical cell area of 6T Full CMOS SRAM is $70{\sim}90F^{2}$, which is too large compared to $8{\sim}9F^{2}$ of DRAM cell. With 80nm design rule using 193nm ArF lithography, the maximum density is 72M bits at the most. Therefore, pseudo SRAM or 1T SRAM, whose memory cell is the same as DRAM cell, is being adopted for the solution of the high density SRAM applications more than 64M bits. However, the refresh time limits not only the maximum operation temperature but also nearly all critical electrical characteristics of the products such as stand_by current and random access time. In order to overcome both the size penalty of the conventional 6T Full CMOS SRAM cell and the poor characteristics of the TFT load cell, we have developed $S^{3}$ cell. The Load pMOS and the Pass nMOS on ILD have nearly single crystal silicon channel according to the TEM and electron diffraction pattern analysis. In this study, we present $S^{3}$ SRAM cell technology with 100nm design rule in further detail, including the process integration and the basic characteristics of stacked single crystal silicon TFT.

A WblA-Binding Protein, SpiA, Involved in Streptomyces Oxidative Stress Response

  • The Streptomyces coelicolor wblA gene is known to play a negative role in both antibiotic biosynthesis and the expression of genes responding to oxidative stress. Recently, WhcA, a WblA ortholog protein, was confirmed to interact with dioxygenase-encoding SpiA ($\underline{s}$tress $\underline{p}$rotein $\underline{i}$nteracting with Whc$\underline{A}$) in Corynebacterium glutamicum. We describe here the identification of a SpiA ortholog SCO2553 protein ($SpiA_{sc}$) that interacts with WblA in S. coelicolor. Using heterologous expression in E. coli and in vitro pull-down assays, we show that WblA specifically binds $SpiA_{sc}$, and is influenced by oxidants such as diamide. These data indicate that the interaction between WblA and $SpiA_{sc}$ is not only specific but also modulated by the redox status of the cell. Moreover, a $spiA_{sc}$-disruption mutant exhibited a less sensitive response to the oxidative stress induced by diamide present in solid plate culture. Real-time RT-PCR analysis also showed that transcription levels of oxidative stress response genes (sodF, sodF2, and trxB) were higher in the $spiA_{sc}$-deletion mutant than in wild-type S. coelicolor. These results show that $SpiA_{sc}$ negatively regulates WblA during oxidative stress responses in S. coelicolor.

Inverse Telecine by Using Frequency Analysis (주파수 분석을 통한 인버스 텔레시네 기법)

  • When a cinema being composed of 24 frames/sec is converted to NTSC video or TV program, several fields we repeated to have 60 fields/sec, which is called the telecine or 3:2 pull-down. Hence, when encoding the telecine NTSC video into MPEG format, if we convert it into the original 24 frames/sec Progressive cinema, then we can save 20% of bits compared to the case of encoding all the 60 fields. In this Paper, we propose an algorithm for performing the inverse telecine by using the properties of the frames. Specifically, the algorithm exploits the fact that there Is much Inconsistency between the even and odd fields in the case of telecine frame, which results in high magnitude at the Nyquist frequency In the vertical direction. The experiment shows that the proposed algorithm performs very well regardless of the quality of video with a very few computations, whereas the conventional motion based method requires much computational complexity and its performance is degraded when the video is of low (eg. VHS) quality.

An Experimental Study on the Characteristics of a Composite Structure of Lattice Girder and Shotcrete (격자지보와 숏크리트 복합구조체의 특성 실험 연구)

  • Lattice girder is a new steel support developed in Europe for the replacement of an existing H-shaped steel set, which is installed after tunnel excavation. Lattice girder has the following several advantages : 1. Lattice girder minimizes the amount of shotcrete shadow which happens to occur behind a steel support. 2. A triangular shape of lattice girder makes shotcrete placed efficiently. 3. Lattice girder provides a good bond strength for shotcrete, which makes the composite structure of lattice girder and shotcrete behave monolithic, and therefore, the rock load can be supported effectively by the lattice girder system, This paper presents the results from a model wall test, a strength test for shotcrete shot on the model wall and a strength test for the bond between lattice girder and shotcrete. These tests proved that lattice-girder system is superior to H-shaped steel-set system concerning the shotcrete rebound rate, the developed shotcrete strength and the adhesion characteristics.

Mechanical Properties of AlN/hBN Ceramic Composites (AlN/hBN 복합재료의 기계적 성질)

  • AlN-BN ceramic composites were fabricated and their mechanical properties were investigated. The relative density of hot-pressed composites decreased with increasing BN content, but over $99\%$ could be obtained with 30 $vol\%$ BN in AlN. YAG was formed in the composites and monolithic AlN as a second phase by the reaction between $Y_2O_3$, added as sintering aid, and $Al_2O_3$. As expected, Vickers hardness and Young's modulus decreased with increasing BN content. The three-point flexural strength also showed similar behavior decreasing from 500 MPa of monolith down to 250 MPa by the addition 30 $vol\%$ BN. However, interestingly, the standard deviation of the strength values decreased significantly as BN was added to AlN. As a result, the Weibull modulus of the AlN-30 $vol\% BN composite was 21.3, which was extremely high. Fractography and crack path studies revealed that BN platelets induced grain pull-out and crack bridging in a bigger scale during crack propagation. Consequently, fracture toughness increased as more BN was added, reaching 4.5 $MPa\sqrt{m}$ at 40 $vol\%$ BN.

Development of Spatial Information System for Regional Ground Stability Assessment near Dam area (댐 주변지역 광역적 지반 안정성 평가를 위한 공간 정보시스템 개발)

  • Ground failure such as landslide, rock fall land subsidence by heavy rainfall have damaged to people and property. Especially, the damage to important facility such as dam, bridge, tunnel and industrial complex may be possible. Therefore the ground failure must be assessed and counter plan must be prepared. So, the object of this study is to develop the spatial information system for regional ground stability assessment. For this, the topographic, geologic, soil, forest, land use, rainfall frequency map, and satellite image near 40 dams were collected and constructed to the spatial information system. The spatial information system was developed using Avenue in ArcView 3.2 environment and consists of pull down menus and icons. For application of the spatial information system, regional ground stability was assessed in Andong dam. The assessment was ground failure susceptibility and possibility. The spatial information can be used for regional ground stability assessment, prevention and mitigation of hazard, and management of ground as basic data.

Heat Shock Protein $90{\beta}$ Inhibits Phospholipase $C{\gamma}-1$ Activity in vitro

  • Phospholipase $C-{\gamma}1\;(PLC-{\gamma}1)$ is an important signaling molecule for cell proliferation and differentiation. $PLC-{\gamma}1$ contains two pleckstrin homology (PH) domains, which are responsible for protein-protein interaction and protein-lipid interaction. $PLC-{\gamma}1$ also has two Src homology (SH)2 domains and a SH3 domain, which are responsible for protein- protein interaction. To identity proteins that specifically binds to PH domain of $PLC-{\gamma}1$, we prepared and incubated the glutathione S-transferase(GST)-fused PH domains of $PLC-{\gamma}1$ with COS7 cell lysate. We found that 90 kDa protein specifically binds to PH domain of $PLC-{\gamma}1$. By matrix-assisted laser desorption ionization time of flight-mass spectrometry, the 90 kDa protein revealed to be heat shock protein (Hsp) $90{\beta}$. Hsp $90{\beta}$ is a molecular chaperone that stabilizes and facilitates the folding of proteins that are involved in cell signaling, including receptors for steroids hormones and a variety of protein kinases. To know whether Hsp $90{\beta}$ affects on $PLC-{\gamma}1$ activity, we performed $PIP_2$ hydrolyzing activity of $PLC-{\gamma}1$ in the presence of purified Hsp $90{\beta}$ in vitro. Our results show that the Hsp $90{\beta}$ dose-dependently inhibits the enzymatic activity of $PLC-{\gamma}1$ and further suggest that Hsp $90{\beta}$ regulates cell growth and differentiation via regulation of $PLC-{\gamma}1$ activity.

Nebulin C-terminus Interacts with NCBP51, a New Isoform of RING Finger Protein 125 (RNF125)

  • Nebulin, a giant modular protein from muscle, is thought to act as molecular ruler in sarcomere assembly. In skeletal muscle, the C-terminal ${\sim}50 kDa$ region of nebulin extends into the Z-line lattice. The most recent studies implicated highlighting its extensive isoform diversity and exciting reports revealed its expression in cardiac and non-muscle tissues containing brain. Also these novel findings are indicating that nebulin is actually a multifunctional filament system, perhaps playing roles in signal transduction, contractile regulation, and myofibril force generation, as well as other not yet defined functions. However the binding protein of nebulin and function in brain is still unknown. A novel binding partner of nebulin C-terminal region was identified by screening a human brain cDNA library using yeast two-hybrid system. Nebulin C-terminus binding protein 51 (NCBP51) was contained a RING-finger domain and identified a new isoform of RING finger protein 125 (RNF125). The interaction was confirmed using the GST pull-down assay. NCBP51 belongs to a family of the RING finger proteins and its function remains to be identified in brain. The role of nebulin and NCBP51 will be studied by loss-of-function using siRNA technique in brain.
