갈근류 생약의 puerarin 함량분석 및 품질평가 (Quality Evaluation and Quantitative Analysis of Puerarin in Various Puerariae Radix)

  • This study was carried out to obtain the basic information that can be used to index arrowroots (Pueraria species) in fourteen regions of China and Korea. The puerarin content in various arrowroots produced in the different areas were quantitatively analysed by HPLC. The average of puerarin content of the 'Galgeun' (Pueraria lobata) was higher than that of the 'Bungalgeun'(Pueraria thomsonii) produced in China and Korea. As a result, the order of the puerarin content was 1) Korean puerariae radix (P. lobata, 0.609%), 2) Chinese puerariae radix (P. lobata, 0.546%), 3) Chinese puerariae radix (P. thomsonii, 0.066%). This result showed that the content of puerarin in the 'Galgeun'(P. lobata) are about 10 times higher than those of the 'Bungalgeun'(P. thomsonii).

Effects of Gal geun (Puerariae Radix) on lowering lipid and antioxidant

  • Effects of Gal geun (Puerariae Radix) EtOH ext. on lipid lowering and antioxidant were investigated in hyperlipidemic rat. Concentration of FFA and triglyceride in plasma showed a tendency to decrease in Gal geun ext. groups. Concentration of plasma total cholesterol and LDL-cholesterol decreased in Gal geun ext. groups. However the concentration of HDL-cholesterol showed no significantly different in all treatment groups. Concentration of liver total cholesterol and triglyceride showed a tendence to decrease in Gal geun ext. groups. Concentration of plasma and liver TBARS showed a low values in Gal geun ext. groups. The values of GSH-Px and SOD activity showed no significantly different among all the treatment groups. However the values of SOD and CAT activity showed a high value in the Gal geun ext. groups.

Scientific Analysis of the Formulation Theory of Chungpesagan-tang; In Vitro Cytotoxicity of Chungpesagan-tang

  • To analyse scientifically the fundamental formulation theory and drug interaction of Chungpegagan-tang, the extraction level of puerarin and daidzin, the transforming activity of puerarin and daidzin to daidzein by human intestinal bacteria and in vitro cytotoxicity against tumor cell lines of Chungpesagan-tang were investigated. When Puerariae Radix was extracted with Chungpesagan-tang composing herbal medicines, the puerarin extraction level from these polyprescriptions was decreased by the extraction with Raphani Semen or Cimicifugae Rhizoma, but the other herbal medicines increased it. The activity transforming puerarin and daidzin to daidzein by human intestinal bacteria was increased by Raphani Semen, Cimicifugae Rhizoma and Angelicae Tenuissimae Radix, but decreased by Scutellariae Radix and Rhei Rhizoma. Puerariae Radix did not showed in vitro cytotoxicity against tumor cell lines. However, by its anaerobic incubation with human intestinal bacteria, it showed a potent cytotoxicity. When the main components, puerarin and daidzin, of Puerariae Radix were incubated with human intestinal bacteria, the main metabolites were daidzein and calycosin. These metabolites had the most potent cytotoxicity, compared to those of puerarin and daidzin. Raphani Semen, Rhei Rhizoma and Chungpesagan-tang had also the potent cytotoxicity against tumor cell lines by the anaerobic incubation with human intestinal bacteria.

갈근과 옻 혼합추출물의 성분변화 및 항알러지 효과 (Constituent Alterations of Pueraiae Radix and Lacquer Tree Mixture Extract and their Anti-allergic Effects)

  • 옻나무는 옻오름 같은 알러지 반응으로 인해 식품으로의 활용이 제한되므로 이러한 알러지 반응을 완화시키기 위하여 갈근 추출물과 혼합하여 알러지 억제효과를 평가하였으며, 알러지를 유발시키는 성분의 변화를 측정하여 화학적 구조변화 및 생물학적 알러지 반응 억제 양상을 관찰하였다. 이에 갈근 추출물에 대한 옻나무 추출물의 알러지 반응 완화효과를 RBL-2H3 세포에서 ${\beta}$-hexosaminidase의 양을 측정하여 평가하였으며, 갈근 추출물에서 매우 우수한 억제효능을 나타내었다. 또한, 갈근 추출물을 혼합한 결과 양성대조군 ketotifen보다 다소 우수한 효능이 나타났다. 옻나무 추출물의 알러지 유발물질 확인하고, 갈근과 혼합 추출 하였을 때의 알러지 유발물질 함량변화를 평가하기 위하여 GC/MS 분석을 실시하였다. 그 결과 4종의 알러지 유발물질을 확인할 수 있었으며, 갈근 추출물과 혼합시 알러지 유발물질은 그 함량이 적어지거나 소멸되었다. 따라서, 갈근과 혼합하여 추출하였을 때 함량이 낮아지거나 소멸되는 알러지 유발물질 4종에 대하여 탈과립 억제 효능을 측정하였다. 그 결과, 옻의 알러지 유발물질 4종으로부터 알러지를 유도한 RBL-2H3 세포에서 탈과립화 억제효능 양상이 유사하였으며, 이를 통해 갈근 추출물은 항알러지 효과를 나타냄을 알 수 있었다. 이러한 효능을 통하여 옻과 갈근을 혼합하여 추출한 복합추출물의 경우 옻의 문제점인 알러지 반응을 효과적으로 억제할 수 있을 것으로 사료된다.

생약추출물이 Aspergillus parasiticus Aflatoxin $B_1$ 생성에 미치는 영향 (Effects of Some Herbal Extracts on Aflatoxin $B_1$ Production from Aspergillus parasiticus)

  • 본 연구에서 8종의 생약 추출물이 A. parasiticus의 배양시 aflatoxin $B_1$ 생성에 미치는 효과를 조사하였다. 배지의 pH는 배양 3일 후에 모든 생약 추출물이 pH 4 이하를 나타냈으며 구기자, 오매, 계피, 두충은 배양 6일에 다시 pH 4 이상으로 상승하였고 이중 대추가 배양 기간 중 가장 낮은 pH를 나타냈다. 균체 생성량은 모든 실험군이 대조군보다 높았으며 갈근, 두충, 오미자, 대추, 오매,구기자, 목과의 순으로 나타났다. 이중 갈근이 최대 생성량을 나타냈으며 목과가 가장 낮은 생성량을 나타냈다. Aflatoxin $B_1$은 갈근과 대추 추출물을 제외한 모든 실험군에서 생성이 억제되었다. 특히 계피, 오매, 두충 구기자, 오미자 추출물 에서 aflatoxin $B_1$ 생성이 현저히 저하되었으며 계피가 가장 큰 억제 효과를 나타냈다. 균체량이 많이 생성되면 aflatoxin $B_1$ 생성이 적어지고 균체량이 적게 생성되면 aflatoxin $B_1$의 생성이 많아졌다. Aspergillus parasiticus에 의해 aflatoxin $B_1$을 가장 적게 생성하는 계피와 가장 많이 생성하는 갈근 추출물의 총단백질 생성량은 계피는 3일째 (34.5%), 갈근 ext ract는 4일째 (36.4%) 총단백질의 함량이 최대가 되어 균체 내의 총단백질의 함량은 계피와 갈근 추출물을 첨가한 시험군이 게조군의 총단백질 함량 (32.7%)보다 약간 많았으며 aflatoxin $B_1$ 생성 및 축적이 최대가 되는 시기는 총단백질량이 최대가 되는 시기보다 대체로 1일 정도 늦게 나타났다.

High Performance Liquid Chromatographic Analysis of Isoflavones in Medicinal Herbs

  • Phytoestrogens have been used as a food supplement to prevent osteoporosis. The isoflavones in the phytoestrogens are daidzein, genistein and formononetin which are present in various herbs. This study examined the quantity of isoflavones in medicinal herbs, which can be used as a phytoestrogen supplement; soybean. These isoflavones were quantified using high performance liquid chromatography (HPLC) with a UV/VIS detector. The concentration of daidzein in Puerariae Radix was $10,436.16{\pm}2,143.83\;mg/kg$ of the dried herb, which was much higher than that extracted from soybeans, $341.47{\pm}18.96\;mg/kg$. The amount of genistein in Sophorae flavescentis Radix ($336.09{\pm}50.89mg/kg$) was approximately 11 times higher than that extracted from soybean ($30.03{\pm}7.17mg/kg$). The level of formononetin in Dalbergiae odoriferae Lignum, $2,189.14{\pm}136.46mg/kg$, was the highest among the herbs tested. The total isoflavone content of Puerariae Radix was approximately 30 times higher than that extracted from soybean. Therefore, plants from the family Leguminosae, particularly Puerariae Radix, can be a good source of phytoestrogens.

갈근 Butanol Fraction의 약리 작용에 관한 연구 (Pharmacological Studies on Butanol Fraction of Puerariae Radix)

  • Effective BuOH fraction was extracted from Puerariae radix (Pueraria thunbergiana), and several pharmacological activities were examined. From several pharmacological examinations, it was found that BuOH fraction has an antipyretic activity against typhoid vaccine pyrexia in rabbit, a papaverine-like antispasmodic activity on isolated ileum and jejunum of rat, an analgesic effects on mice treated by 0.7% HAc, and an anti-inflammatory activity of carrageenin edema in rat. It was found that BuOH fraction has detoxication effects on acute and subacute toxic rat induced by alcohol in consequence of biochemical tests of serum. Therefore it was considered that BuOH fraction has preventive and therapeutic effects in alcohol intoxication.

Scientific Analysis of Formulation Theory of Chungpesagan-tang; The purgative Action of Chungpesagan-tang

  • To analyze scientifically the fundamental formulation theory and drug interaction on the purgative action of Chungpesagan-tang, which is one of the traditional Korean medical prescriptions that has been most frequently used for stroke, water extracts of herbal medicines with Rhei Rhizoma were administered into mice and the purgative action was measured. Water extracts of Raphani Semen or Cimicifugae Rhizoma with Rhei Rhizoma had more potent purgative activity than Rhei Rhizoma alone, although Raphani Semen alone displayed no purgative action. However, Platycodi Radix and Puerariae Radix inhibited the purgative action of Rhei Rhizoma. When Rhei Rhizoma with Raphani Semen or Cimicifugae Radix was extracted with water, sennoside A content in the water extract was increased. However, Puerariae Radix and Platycodi Radix decreased the content of sennoside A in the extracted water. The transforming rate of sennoside A of Rhei Rhizoma was inhibited by Puerariae Radix and Platycodi Radix. The transforming rate of sennoside A of Chungpesagan-tang to rheinan-throne by human intestinal microflora and its purgative activity were similar to those of Rhei Rhizoma. Finally we thought that the purgative action of Chungpesagan-tang could be controlled by the addition and subtraction of Chungpesagan-tang-composing herbal medicines.

갈근 중의 Polyphenol Oxidase에 관한 연구 (Studies on Polyphenol Oxidase from Puerariae Radix)

  • Polyphenol oxidase(PPO) was purified from an extract of Puerariae Radix by ammonium sulfate fractionation followed by Sephadex G-150 column chromatography, which resulted in a 56-fold increase in specific activity. The enzyme was optimum of pH 6.5. The optimum temperature of enzymic reaction was about $40^{\circ}$. The enzyme was thermostable with a half-life equal to 32 min at $70^{\circ}$. Km values of the PPO for catechol and pyrogallol from Lineweaver Burk plots were $1.3{\times}10^{-2}M$, $1.16{\times}10^{-2}M$, respectively. The substrate specificity of the Puerariae Radix PPO showed high affinity toward pyrogallol. Reducing reagents such as cysteine, potassium metabisulfite, ascorbic acid, 2-mercaptoethanol completely inhibited the PPO activity at $10^{-2}M$ level. Linewear-Burk analysis of inhibition data revealed that the inhibition by cysteine, 2-mercaptoethanol, 4-nitrocatechol, potassium cyanide was competitive with Ki values of $4.3{\times10^{-2}M,\;0.73{\times}10^{-6}M,\;6.9{\times}10^{-6}M,\;6.4{\times}10^{-7}M$, respectively. The browning reaction by PPO was observed to decrease temporarily with the addition of sodium diethyl dithiocarbamate, a well known copper chelating agent. Among the divalent cations, $Cu^{2+}$ ion was strong activator on PPO and $Mn^{2+},\;Co^{2+}$ ions was effect on PPO activity. $Zn^{2+},\;Mg^{2+}$ ions was inhibitor on PPO.
