Identification of Atg8 Isoform in Encysting Acanthamoeba

  • Autophagy-related protein 8 (Atg8) is an essential component of autophagy formation and encystment of cystforming parasites, and some protozoa, such as, Acanthamoeba, Entamoeba, and Dictyostelium, have been reported to possess a type of Atg8. In this study, an isoform of Atg8 was identified and characterized in Acanthamoeba castellanii (AcAtg8b). AcAtg8b protein was found to encode 132 amino acids and to be longer than AcAtg8 protein, which encoded 117 amino acids. Real-time PCR analysis showed high expression levels of AcAtg8b and AcAtg8 during encystation. Fluorescence microscopy demonstrated that AcAtg8b is involved in the formation of the autophagosomal membrane. Chemically synthesized siRNA against AcAtg8b reduced the encystation efficiency of Acanthamoeba, confirming that AcAtg8b, like AcAtg8, is an essential component of cyst formation in Acanthamoeba. Our findings suggest that Acanthamoeba has doubled the number of Atg8 gene copies to ensure the successful encystation for survival when 1 copy is lost. These 2 types of Atg8 identified in Acanthamoeba provide important information regarding autophagy formation, encystation mechanism, and survival of primitive, cyst-forming protozoan parasites.

Cutaneous Leishmaniasis of the Eyelids: A Case Series with Molecular Identification and Literature Review

  • Cutaneous leishmaniasis (CL) is a protozoan disease which is endemic in Iran. It is transmitted by the Phlebotomus sand fly. The eyelid is rarely involved possibly because the movement of the lids impedes the sand fly from biting the skin in this region. Here, we report 6 rare cases of eyelid CL. The patients were diagnosed by skin scraping, culture, and PCR from the lesions. Skin scraping examination showed Leishmania spp. amastigotes in the cytoplasm of macrophages. Culture examination was positive for Leishmania spp. PCR was positive for Leishmania major and Leishmania tropica. The lesions were disguised as basal cell carcinoma, chalazion, hordeolum, and impetigo. The patients were treated with intramuscular meglumine antimoniate (20 mg/kg/day) for at least 3 weeks. They showed a dramatic response, and the lesions almost completely disappeared. We emphasized the importance of clinical and diagnostic features of lesions, characterized the phylogenetic relationship of isolated parasites, and reviewed the literature on ocular leishmaniasis.

  • Malaria is an infectious disease affecting humans, which is transmitted by the bite of Anopheles mosquitoes harboring sporozoites of parasitic protozoans belonging to the genus Plasmodium. Despite past achievements to control the protozoan disease, malaria still remains a significant health threat up to now. In this study, we cloned and characterized the full-unit Plasmodium yoelii genes encoding merozoite surface protein 1 (MSP1), circumsporozoite protein (CSP), and Duffy-binding protein (DBP), each of which can be applied for investigations to obtain potent protective vaccines in the rodent malaria model, due to their specific expression patterns during the parasite life cycle. Recombinant fragments corresponding to the middle and C-terminal regions of PyMSP1 and PyCSP, respectively, displayed strong reactivity against P. yoelii-infected mice sera. Specific native antigens invoking strong humoral immune response during the primary and secondary infections of P. yoelii were also abundantly detected in experimental ICR mice. The low or negligible parasitemia observed in the secondary infected mice was likely to result from the neutralizing action of the protective antibodies. Identification of these antigenic proteins might provide the necessary information and means to characterize additional vaccine candidate antigens, selected solely on their ability to produce the protective antibodies.

Intestinal Parasite Survey in Seoul by Stool Examination at Hanyang University Hospital (서울지역(地城)의 양내기생충(賜內寄生蟲) 감염상태(感染狀態) 조사(調査) -한양대학병원(漢陽大學病院) 내원환자(來院患者)를 중심으로-)

  • The present study was undertaken to evaluate the present status of intestinal parasitic infection in Seoul area, Korea. During the period from June 1985 to July 1986, a total of 5,251 stool samples were collected in Department of Clinical Pathology, Hanyang University Hospital and examined by formalin-ether sedimentation technique once for helminth ova and protozoan cysts. The results were summarized as follows: 1. The overall egg positive rate of intestinal helminthes was 2.53%; and 1.43% for Clonorchis sinensis, 0.7% for Trichuris trichiura, 0.13% for Metagonimus yokogawai, 0.06% for hookworm, and 0.02% for Ascaris lumbricoides, Diphyllobothrium latum and Hymenolepis nana, respectively. 2. The overall cyst positive rate of intestinal protozoa was 1.07%. Cyst positive rate was 0.06% for Entamoeba histolytica and 0.13% for Giardia lamblia, respectively. 3. Higher prevalence of intestinal parasitic infection was observed in male and in $21{\sim}50$ year-old groups. 4. The highest prevalence of Clonorchis sinensis infection was observed in 40th age group (3.4%), and male (2.3%) was more infected than female (0.7%) in general.

Atg3-Mediated Lipidation of Atg8 Is Involved in Encystation of Acanthamoeba

  • Autophagy is a catabolic process involved in the degradation of a cell's own components for cell growth, development, homeostasis, and the recycling of cellular products. Autophagosome is an essential component in the protozoan parasite during differentiation and encystation. The present study identified and characterized autophagy-related protein (Atg) 3, a member of Atg8 conjugation system, in Acanthamoeba castellanii (AcAtg3). AcAtg3 encoding a 304 amino acid protein showed high similarity with the catalytic cysteine site of other E2 like enzymes of ubiquitin system. Predicted 3D structure of AcAtg3 revealed a hammer-like shape, which is the characteristic structure of E2-like enzymes. The expression level of AcAtg3 did not increase during encystation. However, the formation of mature cysts was significantly reduced in Atg3-siRNA transfected cells in which the production of Atg8-phosphatidylethanolamine conjugate was inhibited. Fluorescent microscopic analysis revealed that dispersed AcAtg3-EGFP fusion protein gathered around autophagosomal membranes during encystation. These results provide important information for understanding autophagic machinery through the lipidation reaction mediated by Atg3 in Acanthamoeba.

Involvement of Src Family Tyrosine Kinase in Apoptosis of Human Neutrophils Induced by Protozoan Parasite Entamoeba histolytica

  • Tyrosine kinases are one of the most important regulators for intracellular signal transduction related to inflammatory responses. However, there are no reports describing the effects of tyrosine kinases on neutrophil apoptosis induced by Entamoeba histolytica, In this study, isolated human neutrophils from peripheral blood were incubated with live trophozoites in the presence or absence of tyrosine kinase inhibitors. Entamoeba-induced receptor shedding of CD16 and PS externalization in neutrophils were inhibited by pre-incubation of neutrophils with the broad-spectrum tyrosine kinase inhibitor genistein or the Src family kinase inhibitor PP2. Entamoeba-induced ROS production was also inhibited by genistein or PP2, Moreover, genistein and PP2 blocked the phosphorylation of ERK and p38 MAPK in neutrophils induced by E. histolytica. These results suggest that Src tyrosine kinases may participate in the signaling event for ROS-dependent activation of MAPKs during neutrophil apoptosis induced by E. histolytica.

Toxoplasmosis in a Pet Peach-Faced Lovebird (Agapornis roseicollis)

  • Toxoplasma gondii atypical type II genotype was diagnosed in a pet peach-faced lovebird (Agapornis roseicollis) based on histopathology, immunohistochemistry, and multilocus DNA typing. The bird presented with severe neurological signs, and hematology was suggestive of chronic granulomatous disease. Gross post-mortem examination revealed cerebral hemorrhage, splenomegaly, hepatitis, and thickening of the right ventricular free wall. Histologic sections of the most significant lesions in the brain revealed intralesional protozoan organisms associated with malacia, spongiform changes, and a mild histiocytic response, indicative of diffuse, non-suppurative encephalitis. Immunohistochemistry confirmed the causative organisms to be T. gondii. DNA isolated from the brain was used to confirm the presence of T. gondii DNA. Multilocus genotyping based on SAG1, altSAG2, SAG3, BTUB, GRA6, c22-8, c29-2, L358, PK1, and Apico markers demonstrated the presence of ToxoDB PCR-RFLP genotype #3 and B1 gene as atypical T. gondii type II. The atypical type II strain has been previously documented in Australian wildlife, indicating an environmental transmission route.

Internal parasites of cats in Iri and its vicinity (이리지방 고양이의 장내기생충 감염상황)

  • As a series of basic study for the prevention of zoonotic parasites, fecal samples and internal organs of cats in Iri and its vicinity were examined for helminth and their ova or protozoan oocysts from November 1994 to March 1995. The results obtained were summarized as follows; Out of the 133 samples, 101(75.9%) were positive and 135(101.5%) were cumulative positive for parasites. The following were identified in the samples and positive numbers of each species detected were ; Toxocura cari, 51;Isospora spp., 27;Spirometra erinacei, 26;Tasnka taentaeformis, 24;Diphyllobothrium latum, 4;and Capillaria spp., 3, respectively. In analyzing the state of infection, 64.9%(61 heads) of all the cats inspected had a single infection, while the rest(excluding the 32 uninfected cats) had mixed infections : 27.6%(26 heads), 6.4%(6 heads) and 1.1%(1 heads) had double, triple, quadruple Infections, respectively. From these results, it can be concluded that since the helminth, zoonotic parasites, in Korean autochthonal cats cause not only various disorders, but also can be very harmful to human beings, they are considered to be of great significance to public health.

Preliminary survey of swine internal parasites at the sawdust fermentation floor system (발효톱밥돈사에 대한 돼지 내부기생충 조사)

  • Preliminary survey on the prevalence of swine internal parasites was carried out at 91 pens of 4 piggeries being operated by the sawdust fermentation floor system in Kyunggi province. To investigate the effects of viability of eggs., larva and cysts, temperatures at the underlayer of the sawdust floor and rooms of pens were taken three times a day for 8 days. In a pig farm which has been operated by the sawdust fermentation floor system for G years, eggs and larva of Ascaris suum, Trichuris suis, Oesophagostomum sp., Hyostrongylus sp. and Strongyloides ransomi were detected from 42(70.0%) among 60 pens. Cysts and oocysts of coccidia, Balantidium coli, Entamoeba sp. were also detected from 50(83.3%) among 60 pens. In three pig farms which have been operated by the sawdust fermentation floor system for one year, eggs and larva of nematoda were not detected at all, and the contamination rates of protozoan cysts and oocysts was relatively low situation. The change of the temperature in the pig room was variable according to outside condition, whereas that in underfloor was consistently stable between $28^{\circ}C$ to $30^{\circ}C$ which was strictly suitable for the development of eggs, larva and cysts.

Development of indirect fluorescent antibody test and the prevalence of the antibody titer for Neospora caninum of domestic animal in Korea (Neospora caninum 간접형광항체진단법 개발 및 국내 가축에서의 항체가 조사)

  • The present study was undertaken to develop the kit for indirect immunofluorescence antibody test(IFAT) and to investigate the prevalence of N caninum in cattle and dogs in Korea. The neo-antigen kit of IFAT developed in our laboratory was proved diagnostic efficacy by compared with the kit of veterinary medical research and development(VMRD). A survey of N caninum infections among cow and dogs in elevn areas of southern part of country was performed using a IFAT. The infection rate of 190 nationwide cattle was 8.4%(16/190) but was 75%(45/60) at P area in Chunnam province where the cattle showed the abortion repeatedly. Any of the dogs was not N caninum -positive in the Kangwon and Kyonggi areas.
