A report of 10 unrecorded bacterial species of Korea, belonging to the phylum Firmicutes

  • To investigate the indigenous prokaryotic species diversity in Korea, various environmental samples from diverse ecosystems were examined taxonomically. The isolated bacterial strains were identified based on 16S rRNA gene sequences, and those exhibiting at least 98.7% sequence similarity with known bacterial species but never reported in Korea were selected as unrecorded species. As an outcome of this study, 10 unrecorded bacterial species belonging to the phylum Firmicutes were discovered from various sources such as soil, tidal flat, fresh water, sea water, kimchi and gut of Fulvia mutica. The unrecorded species were assigned to 7 different genera of 5 families, namely Bacillus and Ornithinibacillus of Bacillaceae, Exiguobacterium of Exiguobacteriaceae, Brevibacillus and Paenibacillus of Paenibacillaceae, Staphylococcus of Staphylococcaceae, and Lactococcus of Streptococcaceae. The selected isolates were subjected to further taxonomic characterization including the analysis of Gram reaction, cellular and colonial morphology, biochemical activities, and phylogenetic trees. The descriptive information on the 10 unrecorded species are provided.

  • In 2018, as a subset study to discover indigenous prokaryotic species in Korea, a total of 18 unreported bacterial strains were discovered. From the high 16S rRNA gene sequence similarity (>98.8%) and formation of a robust phylogenetic clade, it was determined that each strain belonged an independent and predefined bacterial species. There were no official report that these 18 species were previously described in Korea; therefore, one strain of Williamsia, one strain of Rhodococcus, three strains of Microbacterium, three strains of Agromyces, one strain of Arthrobacter, one strain of Paeniglutamicibacter, one strain of Pseudarthrobacter, one strain of Nocardioides, one strain of Fibrella, one strain of Hymenobacter, one strain of Deinococcus, two strains of Fictibacillus, and one strain of Paenibacillus are described as unreported bacterial species in Korea. Gram reaction, basic biochemical characteristics, and colony and cell morphologies are described in the species description section.

  • The phylum Actinobacteria includes many groups of aerobic, Gram-stain-positive, rod, or filamentous shaped bacteria. Actinobacteria are known for multicellular differentiation in some groups, and also for production of various secondary metabolites such as antibiotics. During a series of extensive surveys of indigenous prokaryotic species diversity in Korea, bacterial strains belonging to Actinobacteria were isolated from various sources of terrestrial environments. A total of 21 bacterial strains, belonging to 10 genera in 8 families, were isolated as unrecorded species in Korea. Among them, 11 were assigned to the family Streptomycetaceae, two species assigned to each of the families Microbacteriaceae, Mycobacteriaceae and Nocardioidaceae, and one species assigned to each of the families Euzebyaceae, Corynebacteriaceae, Micrococcaceae and Intrasporangiaceae. At the genus level, Streptomyces (10 species) was the most abundant, followed by Microbacterium and Mycolicibacterium(2 species each), and one species in each of the genera Corynebacterium, Euzebya, Arthrobacter, Terracoccus, Kribbella, Nocardioides and Yinghuangia.   • As a subset study to discover indigenous prokaryotic species in Korea, we isolated 20 bacterial strains and assigned them to the phyla Actinobacteria, Bacteroidota, Firmicutes, and Proteobacteria. From the high 16S rRNA gene sequence similarity (≥98.7%) and formation of a robust phylogenetic clades, we determined that each strain belonged to independent, predefined bacterial species. There are no official reports of these 20 species in Korea; therefore, 7 strains of the Actinobacteria, 2 strain of the Bacteroidota, 3 strains of the Firmicutes, and 8 strains of the Firmicutes are described in Korea for the first time. Gram reaction, colony and cell morphology, basic biochemical characteristics, and isolation sources are also described in the species description section.

Twelve unrecorded UV-resistant bacterial species isolated in 2020

  • In 2020, a total of 12 bacterial strains were isolated from soil after a comprehensive investigation of indigenous prokaryotic species in Korea. It was determined that each strain belonged to independent and predefined bacterial species, with high 16S rRNA gene sequence similarity (>98.7%) and formation of a robust phylogenetic clade with the closest species. This study identified four families in the phylum Actinobacteria, two families in the phylum Proteobacteria, one family in the phylum Bacteroidetes one family in the phylum Firmicutes; and four species in the family Nocardiaceae, two species in the family Nocardioidaceae, one species in the family Cellulomonadaceae, one species in the family Hymenobacter, one species in the family Methylobacteriaceae, one species in the family Microbacteriaceae, one species in the family Bacillaceae and one species in the family Sphingomonadaceae. There is no official report of these 12 species in Korea, so they are described as unreported bacterial species in Korea in this study. Gram reaction, basic biochemical characteristics, colony, and cell morphology are included in the species description section.

A report of 12 unrecorded bacterial species isolated from Suncheon Bay in Korea

  • Suncheon Bay Ecological Park, possessing abundant fisheries and biological diversity, was registered as a Ramsar wetland in Korea. Approximately 300 bacterial strains were isolated from the Suncheon Bay in a comprehensive study of indigenous prokaryotic species conducted during 2019-2020 in South Korea. A total of 12 bacterial strains were identified using 16S rRNA gene sequencing, demonstrating >98.7% sequence similarity with validly published species. These species were determined to be unrecorded bacterial species in Korea. A total of six strains were isolated from brackish water and Phragmites communis Trin (reed) species. These unrecorded species were phylogenetically diverse and belonged to three classes, six orders, and ten genera. Regarding the genus and class levels, the previously unrecorded species belonged to Jiella, Martelella, Rhizobium, Paracoccus, Rhodovulum, and Altererythrobacter of the class Alphaproteobacteria; Mycolicibacterium, Demequina, and Microbacterium of the class Actinobacteria; Confluentibacter of the class Flavobacteria. The twelve species were further characterized by gram staining, colony and cell morphology, biochemical properties, and phylogenetic position.

  • During recent screening to discover indigenous prokaryotic species in South Korea, a total of 31 bacterial strains assigned to the class Gammaproteobacteria were isolated from a variety of environmental samples including soil, tidal flat, freshwater, seawater, and plant roots. From the high 16S rRNA gene sequence similarity (>98.7%) and formation of a robust phylogenetic clade with the closest species, it was determined that each strain belonged to each independent and predefined bacterial species. There is no official report that these 31 species have been described in South Korea; therefore 5 species of 3 genera in the order Alteromonadales, 11 species of 3 genera in the order Pseudomonadales, 8 species of 6 genera in the order Enterobacteriales, 2 species of 1 genera in the order Vibrionales, 1 species of 1 genera in the order Oceanospirillales, 3 species of 3 genera in the order Xanthomonadales, and 1 species in the order Spongiibacter_o within the Gammaproteobacteia are reported for proteobacterial species found in South Korea.   • During the extensive survey of the prokaryotic species diversity in Korea, bacterial strains belonging to Betaproteobacteria and Epsilonproteobacteria were isolated from various sources including freshwater, sediment, soil and fish. A total of 23 isolates were obtained, among which 22 strains were assigned to the class Betaproteobacteria and one strain to the class Epsilonproteobacteria. The 22 betaproteobacterial strains were further assigned to Comamonadaceae (11 strains), Burkholderiaceae (6 strains), Oxalobacteraceae (2 strains), Neisseriaceae (1 strain) and unclassified family groups (2 strains). For the strains of Burkholderiaceae, 3 strains were identified as 3 species of Burkholderia, and 2 strains were as 2 species of Cupriavidus. For the strains of Comamonadaceae, 4 strains were identified as 2 species of the genus Hydrogenophaga, 2 strains as 2 species of Acidovorax, 2 strains as 2 species of Limnohabitans, and each of the remaining strains as single species of Comamonas, Curvibacter and Rhodoferax, respectively. For the strains of Oxalobacteraceae, 1 strain was identified as a species of Undibacterium, and the other strain as a species of Herbaspirillum. The strain belonging to Neisseriaceae was identified as a species of Iodobacter. The remaining strains of Betaproteobacteria were identified as species of Sphaerotilus and Methylibium respectively (family unassigned). The epsilonproteobacterial strain was identified as a species of Arcobacter of the family Camplyobacteraceae. The detailed description of each unrecorded species is provided.
