Activation Changes of Hafnia alvei Aspartase by Acetic Anhydride

  • The Hafnia alvei aspartase activity with acetic anhydride treatment gradually increased and reached 7.5-fold that of the native one. The activity of the acetylated aspartase was a little higher than that of the native enzyme, indicating that the cooperativity between a substrate and enzyme is increased. The optimum temperature of the native asparatse was $45^{\circ}C$, and that of the acetylated enzyme shifted to $40^{\circ}C.$ The pH vs. the activity profile of the acetylated asparatse was also different from that of the native enzyme. The initial velocity pattern of the acetylated aspartase intersects to the left of the ordinate, indicating the sequential kinetic mechanism other than a rapid equilibrium ordered one. The reciprocal plots for aspartate of the native aspartase were curved, but those of the acetylated aspartase were linear, indicating the Michaelis-Menten kinetics. The helical content of the acetylated aspartase was rather decreased to $9{\textperthousand}$ than that $(63{\textperthousand})$ of the native one.

  • Gear is an essential component of an automotive. Crowning is used for tooth modification of a gear. The basic concept of gear tooth crowning is to reduce the stress concentration in edge of contact area and appropriate profile modifications can help gears to resist scoring, pitting, and tooth breakage. In this study, a method to determinate spur gear tooth crowning amount to make smooth surface stress and subsurface stress distribution is proposed. This method is based on the contact analysis.

A Blind Watermarking Algorithm using CABAC for H.264/AVC Main Profile (H.264/AVC Main Profile을 위한 CABAC-기반의 블라인드 워터마킹 알고리즘)

  • This paper proposed a watermark embedding/extracting method using CABAC(Context-based Adaptive Binary Arithmetic Coding) which is the entropy encoder for the main profile of MPEG-4 Part 10 H.264/AVC. This algorithm selects the blocks and the coefficients in a block on the bases of the contexts extracted from the relationship to the adjacent blocks and coefficients. A watermark bit is embedded without any modification of coefficient or with replacing the LSB(Least Significant Bit) of the coefficient with a watermark bit by considering both the absolute value of the selected coefficient and the watermark bit. Therefore, it makes it hard for an attacker to find out the watermarked locations. By selecting a few coefficients near the DC coefficient according to the contexts, this algorithm satisfies the robustness requirement. From the results from experiments with various kinds and various strengths of attacks the maximum error ratio of the extracted watermark was 5.02% in maximum, which makes certain that the proposed algorithm has very high level of robustness. Because it embeds the watermark during the context modeling and binarization process of CABAC, the additional amount of calculation for locating and selecting the coefficients to embed watermark is very small. Consequently, it is highly expected that it is very useful in the application area that the video must be compressed right after acquisition.

  • In recent years, world is faced with a transportation energy dilemma, and the transportation is almost dependent on a single fuel - petroleum. However, Hybrid Electric Vehicle (HEV) technology holds more advantages to reduce the demand for petroleum in the transportation by efficiency improvements of petroleum consumption. Therefore, there is a trend that lower gear noise levels are demanded in HEV for drivers to avoid annoyance and fatigue during operation. And meshing transmission error (T.E.) is the excitation that leads to the tonal noise known as gear whine, and radiated gear whine is also the dominant source of noise in the whole gearbox. In this paper, the analysis of gear tooth profile and lead modification is firstly presented, and then, the different transmission error of no mesh misalignment and mesh misalignment under one loaded torque for the 1st gear pair of HEV gearbox was investigated and compared. At last, the appropriate tooth modification was used to minimize and compare the transmission error of the gear pair with mesh misalignment under the loaded torque.

Study of Dynamic Characteristics of 2.5-MW Wind Turbine Gearbox (2.5MW 풍력발전기 기어박스 동특성 연구)

  • In this study, a gearbox and blade were modeled in the MASTA program, and the housing and carrier components were modeled using a finite element method. Using substructure synthesis, all the components were combined and used to establish a vibration model of a 2.5-MW wind turbine gearbox. In addition, the safety displacement factor was evaluated using an AGMA data sheet about bearing's outer race for the input shaft and output shaft. As a result, the bearing's outer race for the input shaft, and the radial and axial responses were satisfied by the $1^{st}$ and $2^{nd}$ planetary gears and the $3^{nd}$ helical gear transmission error(TE), respectively. However, the output shaft support bearing's outer race responses were not satisfied with the radial response by the $2^{nd}$ TE and axial response by the $3^{rd}$ TE. To reduce the vibration, tooth modification was needed. After profile tooth modification, at the outer race of the output shaft support bearing, the radial response was reduced by approximately $20{\mu}m$, and the axial response was reduced by approximately $6{\mu}m$.

  • The aging process is multifactorial and results from the combined effects of inherited(genetic) and acquired factors including life style, food habits, physical activity, and diseases. That give rise to the various approaches in aging. We are trying to study biological changes with aging, In detail we are focused on gene and protein function accompanied by normal or abnormal aging process, especially our efforts are aimed at revealing the functional relationship of proteins in aging as a final product of gene. We expect that proteomic approach to the study of protein function involved in aging should give us variety of integrated data to understand biological changes of long lived lives, We have applied expression proteomics to rat liver bred in dietary restriction or in at libitum to elucidate the effects of food habit on aging. Expression proteomics shows us protein profile in a selected tissue or cells as a whole and gives us the information about protein expression level, posttranslational modification and degenerative modification of expressed proteins. Comparative analysis of young and old rat liver by two dimensional gels shows that gene expression of several proteins was down regulated in old rats and some protein expression level is increased with aging. Dietary restriction slows down these changes of gene expression and in some proteins there's no difference in protein expression level at same ages in comparison with rats bred in at libitum. About forty protein was identified by peptide mass fingerprint with MALDI-TOF and rest of the protein of interest is in the course of identification, Also we are trying to make mitochondrial and cytosolic proteom reference map. These suborganelle proteom map will gives us the information about low abundance proteins and cellular localization of proteins. Proteomics is a growing methodology to study biological system. High throughput qualitative and qualitative aspect of this approach will gives us large amount of integrated information and speed up our understanding about biological system

  • This research aims to suggest web-based 3D face modeling system for hairline modification surgery. As public interests in beauty regarding face escalate with era of wide persoanl mobile smart iCT devices, need for medical information system is urgent and increasing demand. This research attempted to build 3D facing modeling library deploying conventional technology and proprietary software available. Implications from the our experiment found that problems and requirement for developing new web based standard. We suggest new system from our experiment and literature review regarding relevant technologies. Main features of our suggested systems is based on studies regarding hair loss treatment such as medical science, beauty studies and information technology. This system processes input images of 2D frontal and profile pictures of face into 3D face modeling with mesh-data. The mesh data is compatible with web standard technology including SVG and Canvas Tag supported natively by HTML5.

A Study on the Leading Phase Operation of Single Phase PWM Converter Train (단상PWM컨버터 차량의 진상운전에 관한 연구)

  • This paper presents a new operation method for the single phase PWM(Pulse Width Modulation) converter train. Recently, the trains adopting the PWM converter have become the majority in the electric locomotives since there are distinct advantages over the predecessors, which can be operated at near unity power factor. However, a slight modification of the control scheme makes this kind of vehicles run in the region of leading power factor. Although this feature seems to be of no significant use by itself, the leading phase operation can improve the voltage profile and the line loss of the feeding systems is decreased by compensating the reactive power loss along the line when it considered together with the feeding systems. This method is even more economical and efficient comparing with the installation of SVC that is mainly used for this purpose since the train can become a movable compensator. With the conditions and some essential formula for the leading phase operation, a new power factor control algorithm has been proposed to implement this scheme. The results of simulation through SIMULINK model show that the proposed method is suitable enough for practical use.

  • NAC(Network Access Control) has been developed as a solution for the security of end-point user, to be a target computer of worm attack which does not use security patch of OS and install Anti-Virus, which spreads the viruses in the Intra-net. Currently the NAC products in market have a sufficient technology of pre-connect, but insufficient one of post-connect which detects the threats after the connect through regular authentication. Therefore NAC users have been suffered from Zero-day attacks and malware infection. In this paper, to solve the problems in the post-connect step we generate the normal behavior profiles using the traffic information of each host, host information through agent, information of open port and network configuration modification through network scanner addition to authentication of host and inspection of policy violation used before. Based on these we propose the system to detect the hosts infected malware.

Expression Profiling of Lipopolysaccharide Target Genes in RAW264.7 Cells by Oligonucleotide Microarray Analyses

  • In inflammatory responses, induction of cytokines and other immune regulator genes in macrophages by pathogen-associated signal such as lipopolysaccharide (LPS) plays a crucial role. In this study, the gene expression profile changes by LPS treatment in the macrophage/monocyte lineage cell line RAW264.7 was investigated. A 60-mer oligonucleotide microarray of which probes target 32381 mouse genes was used. A reverse transcription-in vitro translation labeling protocol and a chemileuminescence detection system were employed. The mRNA expression levels in RAW264.7 cells treated for 6 h with LPS and the control vehicle were compared. 747 genes were up-regulated and 523 genes were down-regulated by more than 2 folds. 320 genes showing more than 4-fold change by LPS treatment were further classified for the biological process, molecular function, and signaling pathway. The biological process categories that showed high number of increased genes include the immunity and defense, the nucleic acid metabolism, the protein metabolism and modification, and the signal transduction process. The chemokine-cytokine signaling, interleukin signaling, Toll receptor signaling, and apoptosis signaling pathways involved high number of genes differentially expressed in response to LPS. These expression profile data provide more comprehensive information on LPS-target genes in RAW264.7 cells, which will be useful in comparing gene expression changes induced by extracts and compounds from anti-inflammatory medicinal herbs.
