당첨가가 B형 간염 바이러스 백신의 안정성에 미치는 영향 (Effects of Addition of Sugars on the Stability of Hepatitis B Virus Vaccine)

  • Most of the current licenced hepatitis B vaccines are being produced by recombinant DNA technology in large fermentation cultures of Saccharomyces cerevisiae of yeast cells which carry the gene coded for hepatitis B virus surface antigen. These vaccines are proved very effective clinically and the immunogenicity of vaccines could be maintained for a long time under refrigeration. To develope the stabilizer that could increase the stability of hepatitis B virus vaccine which could be stored for a long period at room temperature or higher conditions, glucose, lactose and sucrose solutions in phosphate buffered saline were added into hepatitis B vaccine respectively to make 2.5%, 5%, 7.5% and 10% final concentration in vaccines. These sugar-vaccine mixtures were stored at room temperature for one month, two months and three months respectively and then inoculated into ICR mice intramuscularly. On the fourteenth day after inoculation, mice were bled and sera were tested for the evaluation of efficacies of vaccines. The results showed that 5% glucose, 7.5% lactose and sucrose increased the stability of vaccines in some degree and this method could be applied for the production of other viral vaccines and bacterial vaccines.

In Vitro Stability of β-galactosidase Microcapsules

  • The present study was carried out to examine the efficiency of microcapsules and a stability of lactase in vitro in the simulated gastric and intestinal conditions. As a coating materials, medium-chain triacylglycerol (MCT) and polyglycerol monostearate (PGMS) were used. The highest efficiency of microencapsulation was found in the ratio of 15:1 as coating to core material with both MCT (91.5%) and PGMS (75.4%). In a subsequent experiment, lactose content was measured to study a microcapsule stability. Lysis of microcapsules made by MCT in simulated gastric fluid was proportionally increased such as 3% in pH 5 and 11% in pH 2 for 20 min incubation. In the case of PGMS microcapsulation, 11-13% of lactose was hydrolyzed at 20 min in all pHs and also very little amount (less than 3%) of lactose was hydrolyzed after 20 min in all pHs. The highest percentages of lactose hydrolysis in MCT and PGMS microcapsules were 68.8 and 60.8% in pHs 7 and 8 during 60 min, respectively. Based on our data, the lactase microcapsules seemed to be stable when they stay in the stomach, and hydrolyzed rapidly in small intestine where the bile acid was excreted.

색소생성 균주 Kocuria sp. K70의 특징과 색소생성 최적 조건 및 물리적 안정성 (Characterization of Pigment-Producing Kocuria sp. K70 and the Optimal Conditions for Pigment Production and Physical Stability)

  • Marine bacterium producing pigment was isolated from the solar saltern of Mijo-myeon, Namhae, Korea. Based on phenotypic characteristics and 16S rRNA sequence analysis, the strain was identified as Kocuria sp., which produced a yellow pigment. The pigment showed UV absorption maximum at 469nm. The bacterial strain grew well on Marine broth 2216 culture medium. Productivity of the pigment reached the maximum value after 44 hours at $30^{\circ}C$, 2% NaCl and pH 6.0. The pigment was produced best when supplied by 1% lactose as a carbon source and 1% beef extract as a nitrogen source. The result of the color stability study showed that pigment extracted from the strain by ethanol was stable at $-20-25^{\circ}C$ and also showed higher stability over 70% for 14 days in light conditions at $25^{\circ}C$. The pigment extract was also stable for all metal ions tested, except for $FeCl_2$.

대두 지방산을 활용한 화장품 기초제 개발 (Studies on Application of Soybean Fatty Acid for Development of Cosmetic Formulation)

  • Soybean fatty acid, the largest byproduct in the production of soybean oil, was formulated for hand cream, oil in water emulsion base, to improve the suppleness and elasticity of skin. The stability of emulsion observed by a macroscopic method was used as a characteristic index for deciding an optimum formula of hand creams. The optimum formula of the most stable hand cream was obtained from polynomial regression equation, contour graphs and partial derivative graphs. The values of soybean fatty acid and stearyl alcohol in the obtained optimum formula were 9.75 and 14.75 w/w%, respectively, and sodium lauryl sulfate was not needed. Experimental value for the stability of hand cream prepared according to the optimum formula was 76,14 days, and the prediction value by computation method was 73.25 days. From the results of accelerated tests by elevated temperature, the stability of hand cream by optimum formula was 1.7 year at room temperature $(25^{\circ}C)$. The hand cream containing soybean fatty acid was found to be free of primary irritant substance to the skin by Draize technique.

GLOBAL STABILITY OF HIV INFECTION MODELS WITH INTRACELLULAR DELAYS

  • In this paper, we study the global stability of two mathematical models for human immunodeficiency virus (HIV) infection with intra-cellular delays. The first model is a 5-dimensional nonlinear delay ODEs that describes the interaction of the HIV with two classes of target cells, $CD4^+$ T cells and macrophages taking into account the saturation infection rate. The second model generalizes the first one by assuming that the infection rate is given by Beddington-DeAngelis functional response. Two time delays are used to describe the time periods between viral entry the two classes of target cells and the production of new virus particles. Lyapunov functionals are constructed and LaSalle-type theorem for delay differential equation is used to establish the global asymptotic stability of the uninfected and infected steady states of the HIV infection models. We have proven that if the basic reproduction number $R_0$ is less than unity, then the uninfected steady state is globally asymptotically stable, and if the infected steady state exists, then it is globally asymptotically stable for all time delays.

Power Devolution and Economic Stability: Evidence from Pakistan

  • The current study analyzed the impacts of fiscal decentralization (FD) on the economic stability of Pakistan. This study used time series data from 1981 to 2017. The collected data was first passed through the unit root analysis. ARDL estimation techniques were employed to scrutinize the data where long-run associations were tested through Wald F-statistics. The long-run estimates were extracted by applying Ordinary Least Square, and error correction mechanisms were employed to find the speed of adjustment for disequilibria between the long and the short run. Wald F-statistics confirmed the existence of long-run cointegration. Long-run elasticities suggested that fiscal decentralization because of limited institutional capabilities of provincial governments failed in bringing stability in the economy of Pakistan. Similarly, transparency issues and misspecification of projects hinder the outcome of investment to stabilize the economy. High service payments on debt cut the amount that can be used for skills improvements and destabilize the economy. High Population growth puts pressure on infrastructure and reduces production capacity, ultimately destabilizing the economy by increasing unemployment and inflation. Based on these findings, the government is suggested to improve the institutional capacity of lower governments for the desired outcome of power devolution.

알칼리 수전해를 위한 상용 음이온교환막의 고온 특성 (High Temperature Characteristics of Commercially Available Anion Exchange Membrane for Alkaline Water Electrolysis)

  • In order to evaluate the possibility as a separator in alkaline water electrolysis, the high temperature characteristics were evaluated by measuring the membrane resistance and durability of 5 types of commercial anion exchange membranes in 7 M KOH solution and at 80℃. The membrane resistance of AEM membrane measured in 7 M KOH solution and at 80℃ had a lower value of about 8-24 times compared to the other membranes. The durability of AEM membrane tested with the soaking time in 7 M KOH solution and at 80℃ showed a very good stability and that of FAAM40 and FAAM75-PK showed secondly a good stability. The thermal stability with the soaking time in 7 M KOH solution and at 80℃ of FAAM40 and FAAM75-PK membrane analyzed by thermo-gravimetric analysis showed a good stability compared to the other membranes.

The Influences of Addition of Sugar with or without L. buchneri on Fermentation and Aerobic Stability of Whole Crop Maize Silage Ensiled in Air-stress Silos

  • The whole plant of crop maize was chopped and ensiled in double-layered polyethylene bags to determine the influence of residual sugar on the fermentation of lactic acid and aerobic stability by L. buchneri in whole crop maize silage made in airstress condition. There were a total of six treatments used in this experiment as follow: added 25 g de-mineralised water per kg chopped maize serving as control (con), 37.5 g glucose solution containing 12.5 g glucose ($g_1$), 75 g glucose solution containing 25 g glucose ($g_2$), 25 g, L,.buchneri suspension intended for $10^6$ cfu $g^{-1}$ (L.b.), $g_1$+L.b. and $g_2$+L.b. All silos were opened at day 91 after ensiling for measuring the pH values, microbiological enumeration, fermentative products and aerobic stability. The dry matter loss increased significantly (p<0.01) due to inclusion of sugar or L. buchneri. The lower lactic acid concentrations were observed (p<0.01) in silages inoculated with L. buchneri only or in combination with sugar addition than the correspondent uninoculated silages. Compared with control silage, ethanol production was about 3 or 6-fold higher due to addition 12.5 or 25 g glucose per kg chopped maize at ensiling. The silages added with sugar contained less acetic acid concentration (p<0.01) than control, but silages inoculated with L. buchneri showed the contrary effects (p<0.01) at different sugar levels. No butyric acid was found in uninoculated silages, silages inoculated with L. buchneri. producted more propionic acid, 1-propanol and butyric acid. Lactic acid bacteria counts increased markedly (p<0.01) due to inoculation with L. buchneri, whereas it was reduced (p<0.01) by added sugar. No significant difference was observed in count of yeast, but inoculation with L. buchneri shows a decreasing trend. Mould count in all silages was less than 2 (log cfu $g^{-1}$). The added sugar had negative effects on aerobic stability of maize silage made under air-stress conditions, whereas inoculation with L. buchneri improves (p<0.01) the aerobic stability.

Effect of Applying Molasses and Propionic Acid on Fermentation Quality and Aerobic Stability of Total Mixed Ration Silage Prepared with Whole-plant Corn in Tibet

  • The objective of this study was to evaluate the effects of molasses and propionic acid on the fermentation quality and aerobic stability of total mixed ration (TMR) silages prepared with whole-plant corn in Tibet. TMR (354 g/kg DM) was ensiled with four different treatments: no additive (control), molasses (M), propionic acid (P), and molasses+propionic acid (PM), in laboratory silos (250 mL) and fermented for 45 d. Silos were opened and silages were subjected to an aerobic stability test for 12 days, in which chemical and microbiological parameters of TMR silages were measured to determined the aerobic deterioration. After 45 d of ensiling, the four TMR silages were of good quality with low pH value and ammonia/total N (AN), and high lactic acid (LA) content and V-scores. M silage showed the highest (p<0.05) LA content and higher dry matter (DM) recovery than the control and P silages. P silage had lower (p<0.05) LA content than the control silage. During aerobic exposure, lactic acid contents decreased gradually in the control and M silages, while that of P and PM silages increased, and the peak values were observed after 9 d. M silage had similar yeast counts with the control silage (> $10^5$ cfu/g FM), however, it appeared to be more stable as indicated by a delayed pH value increase. P and PM silages showed fewer yeasts (< $10^5$ cfu/g FM) (p<0.05) and were more stable than the control and M silages during aerobic exposure. It was concluded that M application increased LA content and improved aerobic stability of TMR silage prepared with whole-plant corn in Tibet. P application inhibited lactic acid production during ensiling, and apparently preserved available sugars which stimulated large increases in lactic acid during aerobic exposure stage, which resulted in greater aerobic stability of TMR silage.

The Effect of Bacterial Inoculants and a Chemical Preservative on the Fermentation and Aerobic Stability of Whole-crop Cereal Silages

  • Three microorganisms and one chemical preservative were tested for their effects on the fermentation and aerobic stability of whole-crop wheat, sorghum and maize silages. Wheat at the early dough stage, sorghum at the late milk stage and maize at the one-third milk line stage were harvested and ensiled in 1.5-l anaerobic jars untreated or after the following treatments: control (no additives); Lactobacillus plantarum (LP) at $1.0{\times}10^6$ colony-forming units (CFU)/g of fresh forage; L. buchneri (LB) at $1.0{\times}10^6$ CFU/g; Propionibacterium acidipropionici (PA) at $1.0{\times}10^6$ CFU/g; and a formic acid-based preservative (FAP) at 3 ml/kg of fresh forage weight. Three jars per treatment were sampled on d 90 after ensiling, for chemical and microbiological analysis. At the end of the ensiling period, 90 d, the silages were subjected to an aerobic stability test lasting 5 d. In this test, $CO_2$ produced during aerobic exposure was measured along with chemical and microbiological parameters which serve as spoilage indicators. The silages inoculated with LP had higher concentration of lactic acid compared with the controls and the other treated silages (p<0.05). The controls and LP-inoculated silages spoiled upon aerobic exposure faster than LB, PA and FAP-treated silages. The controls and LP-inoculated silages spoiled upon aerobic exposure faster than LB, PA and FAP-treated silages due to more $CO_2$ production (p<0.05) in these two groups and development of yeasts unlike the other groups. In the experiment, the silages treated with LB, PA and FAP were stable under aerobic conditions. However, the numbers of yeasts was higher in the LP-inoculated wheat, sorghum and maize silages compared with the LB, PA and FAP-treated silages. The LB, PA and FAP improved the aerobic stability of the silages by causing more extensive heterolactic fermentation that resulted in the silages with high levels of acetic and propionic acid. The use of LB, PA and FAP as silage additives can improve the aerobic stability of whole-crop wheat, sorghum and maize silages by inhibition of yeast activity.
