Effect of Sodium Acetate on the Production of Sisomicin by Micromonospora inyoensis (Micromonospora inyoensis에 의한 Sisomicin 생산에서 Sodium Acetate의 영향)

  • This study was carried out to enhance the production of sisomicin by Micromonospora inyoensis ATCC 276000. The effect of various sugars and organic acids as the supplements to a basal mineral medium was tested for the sisomicin production by fully grown mycelium. Among the substances tested, acetate and citrate greatly increased the production of sisomicin at the sixth day of culture. Especially, in the basal mineral medium containing 0.1M sodium acetate, sisomicin production was 6.7 times more than that in the same medium with glucose. When 0.1M sodium acetate was added in the fermentation medium initially, the cell growth was inhibited by sodium acetate, although specific productivity was higher than that in the same medium without sodium acetate. On the contrary, when sodium acetate was added to the culture after three days, the sisomicin production and specific productivity were 1.6 times higher than those in the same medium without sodium acetate. The results suggested that sodium acetate was a stimulating substance of sisomicin production by M. inyoensis.

Influence of pretreatment medium, fresh medium addition, and culture plate size on the production of embryos in isolated microspore culture of hot pepper (Capsicum annuum L.) (고추의 소포자 배양 시 전처리 배지, 새 배지의 첨가, 및 배양 용기의 크기가 배의 생산에 미치는 영향)

  • The influences of pretreatment medium, the addition of fresh medium, and the size of culture plate on the production of embryos were investigated in isolated microspore culture of hot pepper (Capsicum annuum L.). Among the media used for heat shock pretreatment ($32{\pm}1{^{\circ}C}$), high frequency embryo production was obtained when the sucrosestarvation medium A was used. On the other hand, neither 0.37 M mannitol solution nor NLNS medium supplemented with sucrose was not efficient for embryo production. The addition of culture medium to pretreatment media considerably decreased the embryo production even though embryo development proceeded further. The embryo production was not improved by the addition of fresh medium after 2 or 3 weeks from starting culture. Increase in the size of the culture plate from $3.5{\times}1.0$ cm to $6.0{\times}1.5$ cm improved embryo quality. These results will provide valuable information for developing an efficient microspore culture system of hot pepper for high frequency embryo production.

  • Scopolia parviflora Nakai, a rare and endangered species, is the sole plant producing tropane alkaloids (TA) among the Korean native species. In order to enhance TA productivity the SP72 root line was selected by screening 100 of root line, and the optimal culture media for root growth and TA production were investigated with the SP72 roots. Based on the several media, SH and 2B5 medium were determined as growth medium and White and NN medium as production medium. Among the four combinations of two-stage culture, 2BN (2B5 as growth medium plus NN as production medium) showed more enhanced root growth and TA production as compared with production media of White and NN medium and growth media of SH and 2B5 medium, respectively. However, bubble column bioreactor (BCB) cultures applying two-stage culture did not reveal the effective results despite of the each successful operation of two-stage culture in conical flasks and BCB cultures.

  • In Korea Small-medium Companies Constitutes 96.5% of all of the mining and manufacturing industries, about 35.7% of the total production of value added business, an average of 35% of the actual export of industrial goods and 47.6% of employment since present at the end of 1979. Now then Small-medium Industry Companies organization style constitutes 78.6% of a form of private company and 81.2% of the total on an small scale under 50 persons inregular employee. Specially, the trouble of business management of the mining and manufacturing Companies in Korea are constituted average 28.3% of production management ill the worst trouble of business management from 1973 to 1979 and small-medium industry Companies are the same of about more than 30% of production management in the trouble of business management. In this cases, the status of production management is effected by small-medium industry companies are the style of non-system and non-organization, but then 13parts in production management operations have the many trouble problems. In this circumstances a change for the better of production management operations in small medium industry companies are suggested the application effects and achievement points of production management, it is suggested a kind of variety and a small quantity, production management system of small-medium industry Company. And then Industrial Engineering technics is applied systematic route and the company grow more than better its physical condition for the improve productivity applied system approach for production management. There for the chapter 4-5 of this paper treats of a rational production management system approach, it make a emphatic major point of high level of economic growth of small medium Industry enterprise.

An Optimization of Flavonoid Production from the Suspension Culture of Scutellaria baicalensis Georgi Cells

  • Flavonoid production by suspended cells of Scutellaria baicalensis Georgi was studied and the medium was optimized for cell growth and baicalin production. In SH medium the flavonoid production was not closely associated with the cell growth. A modified SH medium, FPM, was therefore designed for enhanced baicalin production. In FPM, both cell growth and baicalin production were increased by 1.5 times and 1.67 times than in the original SH medium, respectively. The increases could be attributed to the increased metabolic activities involved in the flavonoid biosynthesis as represented by enhanced activities of phenylalanine ammonia-lyase.

  • When S. fradiae was cultured in S medium, it stavted to produce neomycin in the middle of stationary phase of growth. Antibitoic production is regulated not only by glucose but also by metabolites formed from glucose. A chemically defined minimal salt broth was developen for the study of metabolites activating produition of antibiotic in a neomycin producer. When growth and production or antibiotic in minimal salt broth was examined with a full grown or a vefctativc mycelium, the medium was found not to be good for the growth, but to be good enough for the production of antibiotic with a full grown mycelium. When many carbotlydrates, organic acids, or alcohol were supplmented with instead of glucose in the medium suspcndcn with a full grown mycelium, the amount of antibiotic produced in the medium containing fumaratc was 5 times more than that in the medium with glucose. Further study indicated that the medium is not good also for the growth but good for the production of antibiotic. The antibiotic produced in this medium was identified to be neomycin. The activation of the production of neomycin by fumarate was further confirmed in a complex medium. Fuinarate is suspected to initiate and to activate the biosynthesis of neomycin at the gene level.

  • Serum is a potential source of bacterial, mycoplasmal and viral contamination, and it has a possibility of the introduction of serum proteins, prion and pyrogens into the final vaccine product. For porcine Rotavirus vaccine production, it is necessary to develop serum free medium which do not cause those problems. A new serum free medium was developed for porcine Rotavirus vaccine based on DMEM, and the performance of developed serum free medium was evaluated in terms of Vero cell growth and Rotavirus vaccine production. The cell density, gown in serum free medium developed, was similar with that in serum supplemented medium. Also, it was higher than that in other commercially available serum free medium. The productivity of Rotavirus vaccine using serum free medium developed and optimum production strategies will be also discussed.

Media Optimization for L-tryptophan Production by Genetically Engineered Klebsiella pneumoniae (유전자 조작한 Klebsiella pneumoniae로부터 L-트립토판 생산을 위한 발효배지 조건)

  • The optimum medium composition for the production of L-tryptophan with Klebsiella pnuemoniae pheA tyrA trpE trpR/pSC 101-trp$^{+}$ and the effect of precusors in the optimum medium were studied. The specific growth rate in the optimum medium was almost the same as that in the basal medium, the former showing 1.01 and the latter 1.07 hr $$^{-1}$, but the produced tryptophan was increased 45% in the optimum medium. The maximum amount of produced tryptophan was 159 mg/l within 14 hours. Tryptophan production was ceased by casamino acid addition over 4 g/l in medium, but cell maSS increased with its addition. Indole and anthranilate as precusors had toxic effect on growth and tryptophan production at experimented concentration range (over 20 mg/l), but L-serine had good effect on tryptophan production, resulting in 175 mg/l tryptophan within 14 hours.

  • The effects of inorganic salts and feather concentrations on pretense production by Bacillus megaterium F7-1 were investigated. Pretense production was dependent on the presence of phosphates in the medium. Supplementation of medium with calcium ion slightly increased protease production. The highest protease production was obtained at $1.4\%$ feather. The optimal medium contained $2.0\%$ glucose, $0.8\%$ skim milk, $0.06\%\;K_{2}HPO_{4}\%,\;0.04\%\;KH_{2}PO{4},\;0.06\%\;NaCl,\;0.03\%\;MgCl_{2}\cdot6H_{2}O,\;0.002\%\;CaCl_{2}\cdot2H_{2}O,\;and\;1.4\%$ whole feather. By using this optimized medium, increased production of the protease was achieved compared with the cases of using basal medium.

A Study on the Pullulan Production Using Whey (Whey를 이용한 Pullulan의 발효생산에 관한 연구)

  • In this study, Pullulan Production from whey medium by Aureohsidium pullulans was investigated. When the Pullulan production from whey medium was carried outs final cell concentration was similar to sucrose basal medium but Pullulan concentration was less than 5g/$\ell$. For pullulan production from whey medium, adaptation culture technique was tried on lactose and galactose base medium When the adaptation culture technique was not applied, the maximum concentration of pullulan was 3.4g/$\ell$ for lactose, 2.5g/$\ell$ for galactose. Adapted strains produced 10~12g/$\ell$ from lactose, 10~11g/$\ell$ from galactose. The pullulan production from lactose basal medium was 13.5g/$\ell$ for lactose adapted strains and 18.6g/$\ell$ for galactose adapted strains. When the adapted strains were inoculated on whey medium, maximum pullulan production was obtained at initial pH 3.0.
