Ruminal pH pattern, fermentation characteristics and related bacteria in response to dietary live yeast (Saccharomyces cerevisiae) supplementation in beef cattle

  • Objective: In this study we aimed to evaluate the effect of dietary live yeast supplementation on ruminal pH pattern, fermentation characteristics and associated bacteria in beef cattle. Methods: This work comprised of in vitro and in vivo experiments. In vitro fermentation was conducted by incubating 0%, 0.05%, 0.075%, 0.1%, 0.125%, and 0.15% active dried yeast (Saccharomyces cerevisiae, ADY) with total mixed ration substrate to determine its dose effect. According to in vitro results, 0.1% ADY inclusion level was assigned in in vivo study for continuously monitoring ruminal fermentation characteristics and microbes. Six ruminally cannulated steers were randomly assigned to 2 treatments (Control and ADY supplementation) as two-period crossover design (30-day). Blood samples were harvested before-feeding and rumen fluid was sampled at 0, 3, 6, 9, and 12 h post-feeding on 30 d. Results: After 24 h in vitro fermentation, pH and gas production were increased at 0.1% ADY where ammonia nitrogen and microbial crude protein also displayed lowest and peak values, respectively. Acetate, butyrate and total volatile fatty acids concentrations heightened with increasing ADY doses and plateaued at high levels, while acetate to propionate ratio was decreased accordingly. In in vivo study, ruminal pH was increased with ADY supplementation that also elevated acetate and propionate. Conversely, ADY reduced lactate level by dampening Streptococcus bovis and inducing greater Selenomonas ruminantium and Megasphaera elsdenii populations involved in lactate utilization. The serum urea nitrogen decreased, whereas glucose, albumin and total protein concentrations were increased with ADY supplementation. Conclusion: The results demonstrated dietary ADY improved ruminal fermentation dose-dependently. The ruminal lactate reduction through modification of lactate metabolic bacteria could be an important reason for rumen pH stabilization induced by ADY. ADY supplementation offered a complementary probiotics strategy in improving gluconeogenesis and nitrogen metabolism of beef cattle, potentially resulted from optimized rumen pH and fermentation.

Optimization of an Industrial Medium and Culture Conditions for Probiotic Weissella cibaria JW15 Biomass Using the Plackett-Burman Design and Response Surface Methodology

  • The objective of this study was to optimize industrial-grade media for improving the biomass production of Weissella cibaria JW15 (JW15) using a statistical approach. Eleven variables comprising three carbon sources (glucose, fructose, and sucrose), three nitrogen sources (protease peptone, yeast extract, and soy peptone), and five mineral sources (K2HPO4, potassium citrate, ⳑ-cysteine phosphate, MgSO4, and MnSO4) were screened by using the Plackett-Burman design. Consequently, glucose, sucrose, and soy peptone were used as significant variables in response surface methodology (RSM). The composition of the optimal medium (OM) was 22.35 g/l glucose, 15.57 g/l sucrose, and 10.05 g/l soy peptone, 2.0 g/l K2HPO4, 5.0 g/l sodium acetate, 0.1 g/l MgSO4·7H2O, 0.05 g/l MnSO4·H2O, and 1.0 g/l Tween 80. The OM significantly improved the biomass production of JW15 over an established commercial medium (MRS). After fermenting OM, the dry cell weight of JW15 was 4.89 g/l, which was comparable to the predicted value (4.77 g/l), and 1.67 times higher than that of the MRS medium (3.02 g/l). Correspondingly, JW15 showed a rapid and increased production of lactic and acetic acid in the OM. To perform a scale-up validation, batch fermentation was executed in a 5-l bioreactor at 37℃ with or without a pH control at 6.0 ± 0.1. The biomass production of JW15 significantly improved (1.98 times higher) under the pH control, and the cost of OM was reduced by two-thirds compared to that in the MRS medium. In conclusion, OM may be utilized for mass producing JW15 for industrial use.

고지방식이 마우스의 간에서 Lactobacillus acidophilus NS1에 의한 글리코겐 함량 조절 효과 (Effect of Lactobacillus acidophilus NS1 on the Hepatic Glycogen Contents in High-Fat Diet-Fed Mice)

  • 본 연구는 고지방식이 마우스에서 체중 감소, 혈당 감소 및 인슐린 저항성 개선 효과를 가지는 LNS1 균주의 간 내 글리코겐 함량에 미치는 영향을 조사하여 고지방식이에 의한 비정상적인 글리코겐 대사 개선을 위한 활용 가능성을 검토하고자 실시하였다. LNS1을 12주간 경구 투여한 고지방식이 마우스의 간에서 포도당 운반체 단백질인 GLUT2와 글리코겐 합성의 주요 효소인 GCK, GYS2의 유전자 발현 변화를 확인한 결과, LNS1의 경구 투여는 고지방식이 마우스에 비해 GLUT2와 GYS2의 유전자 발현을 각각 약 2배, 1.8배 증가시켰으며, GCK의 발현에는 영향을 주지 않는 것으로 확인되었다. 또한, GCK의 regulatory unit으로 작용하여 GCK의 활성을 억제하는 GCKR와 글리코겐 분해 과정의 주요 효소인 G6PC의 발현은 LNS1 투여에 의해 HFD마우스에 비해 각각 약 53%, 32% 감소함을 보였다. 간 조직에서의 결과와 마찬가지로 HepG2 세포에 LNS1-CM의 처리는 GLUT2와 GYS2의 유전자 발현을 약 1.9배, 2배 증가시켰으며, GCK의 발현 변화에는 영향을 주지 않는 것으로 확인되었다. GCKR과 G6PC의 유전자 발현 또한 LNS1-CM 처리에 의해 각각 77%, 47% 감소함을 보였다. 또한, 간 조직 내 글리코겐 함량은 고지방식이와 LNS1 투여를 병행한 마우스에서 고지방식이 마우스에 비해 약 1.5배 증가한 것으로 조사되었다. 위의 결과들을 종합해 볼 때, LNS1은 GLUT2, GYS2, GCKR와 G6PC의 발현 조절을 통해 간 조직내 글리코겐 함량을 증가시켜 고지방식이에 의한 글리코겐 대사 이상을 개선시키는 효과를 가지는 것으로 사료된다.

유산균에 의한 양파 착즙액의 발효효과와 이화학적 특성 (Fermentation of onion extract by lactic acid bacteria enhances its physicochemical properties)

  • 본 연구는 우수한 생리활성을 나타내는 것으로 알려진 양파 껍질을 식품으로 활용하고 유산균 발효를 통해 기능성 증진을 도모하고자 과육과 함께 양파 껍질을 착즙하고, 착즙액 발효에 적합한 유산균을 선정하고자 발효에 따른 품질특성 및 항산화 활성을 검토하였다. 양파 착즙액의 유산균 발효에 따라 pH와 당도는 감소하였으며, 적정 산도는 증가하였고 유산균 수는 발효 48시간까지 증가한 후 감소하였다. 총 유리당 함량은 모든 발효물에서 감소하였으며, 유리당 조성 변화의 경우 Bb와 Lb 접종 양파 착즙액 발효물에서는 fructose, glucose 및 sucrose가 감소하였으며, 이외의 균주 접종 양파 착즙액 발효물에서는 fructose와 sucrose는 감소한 반면, glucose는 증가하였다. 총 유기산 함량은 발효에 의해 증가하였으며, 유기산 중 lactic acid가 가장 많이 증가하였다. 발효에 따른 quercetin 함량은 Ll, Ls 및 Pp 접종 양파 착즙액 발효물은 감소한 반면, Bb, Ef, Lb 및 St 접종 양파 착즙액 발효물은 증가하였다. 총 폴리페놀 함량은 Bb, Ll, Lb 및 Ls 접종 양파 착즙액 발효물은 증가하였으나, Ef, St 및 Pp 접종 양파 착즙액 발효물은 감소하였고, 총 플라보노이드 함량은 St 접종 양파 착즙액 발효물을 제외한 모든 발효물에서 증가하였다. DPPH 및 ABTS 라디칼 소거능은 유산균 발효에 의해 증가하였다. 본 연구 결과 양파 착즙액의 유산균 발효는 생리활성과 기능성을 증진 시키는 것으로 나타났으며, 특히 B. breve KCTC 3441 균주 발효를 통해 quercetin, polyphenol, flavonoids 함량이 각각 50%, 57%, 39% 그리고 DPPH와 ABTS 라디칼 소거능은 74%와 29% 증가되었다. 따라서 양파착즙액의 발효 균주는 B. breve KCTC 3441이 적합한 것으로 판단된다.

16S rDNA 염기서열 분석을 통한 제주연안 소라(Turbo cornutus) 장내세균 다양성 조사 (Analysis of Intestinal Microbial Communities of Topshell (Turbo cornutus) fromCoast of Jeju Island, Korea by 16S rDNA Sequence Analysis)

  • 본 논문은 제주연안에서 채집한 소라(Turbo cornutus)의 장내세균을 분리하고 군집의 다양성을 조사하였다. 표준배지를 사용하여 순수 정체 배양 결과 MA agar에서 가장 많은 군집을 나타났다. 일반 배양 CFU값은 평균 1.8×105, 혐기 배양 CFU값은 평균 0.4×10으로 보다 적게 나타났다. 기존 표준균주와 16S rDNA sequence 유사도 비교 분석 결과 크게 4문 12과 26속 67종으로 분류되었다. 표준균주와의 상동성 범위는 93-100%로 나타났다. 소라장내세균 군집은 크게 Proteobacteria 39% (α-proteobacteria; Phyllobacteriaceae (1), Rhodobacteraceae (8) / γ-proteobacteria; Alteromonadaecae (1), Shewanellaceae (4), Vibrionaceae(12))로 가장 우점하였고, Firmicutes 34% (Bacillaceae (21), Paenibacillaceae (2)), Actinobacteria 21% (Cellulomonadaceae (1), Mycobacteriaceae (6), Nocardiaceae (4), Streptomycetacea (3)), Bacteroidetes 6% (Flavobacteriaceae (4))로 각각 나타났다. Bacillus sp., Vibrio sp.이 가장 우점 하였으며, 그 외 대부분의 분리 균주는 해양 유래 관련 균주로 나타났다. 이전 보고된 제주 연안에 서식하는 해양동물 장내세균군과 비슷한 양상을 보였다. 분리된 일부 균주가 단당류(Cellulose), 다당류(alginate, agar)분해능을 갖고 있는 것으로 나타났는데, 대부분이 해조류 유래 세균으로 소라의 섭이가 장내세균군과 관련됨을 알 수 있었다. 동시에 probiotics 기능을 갖고 있는 일부 균주도 분리되었다.

Saudi Experts Consensus on Diagnosis and Management of Pediatric Functional Constipation

  • Although functional gastrointestinal disorders (FGIDs) are very common in pediatric patients, there is a scarcity of published epidemiologic data, characteristics, and management patterns from Saudi Arabia, which is the 2nd largest Arabic country in terms of area and the 6th largest Arabic country in terms of population, with 10% of its population aged <5 years. Functional constipation (FC) is an FGID that has shown a rising prevalence among Saudi infants and children in the last few years, which urges us to update our clinical practices. Nine pediatric consultants attended two advisory board meetings to discuss and address current challenges, provide solutions, and reach a Saudi national consensus for the management of pediatric constipation. The pediatric consultants agreed that pediatricians should pay attention to any alarming signs (red flags) found during history taking or physical examinations. They also agreed that the Rome IV criteria are the gold standard for the diagnosis of pediatric FC. Different therapeutic options are available for pediatric patients with FC. Dietary treatment is recommended for infants with constipation for up to six months of age. When non-pharmacological interventions fail to improve FC symptoms, pharmacological treatment with laxatives is indicated. First, the treatment is aimed at disimpaction to remove fecal masses. This is achieved by administering a high dose of oral polyethylene glycol (PEG) or lactulose for a few days. Subsequently, maintenance therapy with PEG should be initiated to prevent the re-accumulation of feces. In addition to PEG, several other options may be used, such as Mg-rich formulas or stimulant laxatives. However, rectal enemas and suppositories are usually reserved for cases that require acute pain relief. In contrast, infant formulas that contain prebiotics or probiotics have not been shown to be effective in infant constipation, while the use of partially hydrolyzed formula is inconclusive. These clinical practice recommendations are intended to be adopted by pediatricians and primary care physicians across Saudi Arabia.

Enhanced γ-aminobutyric acid and sialic acid in fermented deer antler velvet and immune promoting effects

  • Deer antler velvet is widely used in traditional medicine for its anti-aging, antioxidant, and immunity-enhancing effects. However, few studies have reported on the discovery of probiotic strains for deer antler fermentation to increase functional ingredient absorption. This study evaluated the ability of probiotic lactic acid bacteria to enhance the concentrations of bioactive molecules (e.g., sialic acid and gamma-aminobutyric acid [GABA]) in extracts of deer antler velvet. Seventeen strains of Lactobacillus spp. that were isolated from kimchi and infant feces, including L. sakei, L. rhamnosus, L. brevis, and L. plantarum, and those that improved the life span of Caenorhabditis elegans were selected for evaluation. Of the 17 strains, 2 (L. rhamnosus LFR20-004 and L. sakei LFR20-007) were selected based on data showing that these strains increased both the sialic acid and GABA contents of deer antler extract after fermentation for 2 d and significantly improved the life span of C. elegans. Co-fermentation with both strains further increased the concentrations of sialic acid, GABA, and metabolites such as short-chain fatty acids and amino acids. We evaluated the biological effects of the fermented antler velvet (FAV) on the antibacterial immune response in C. elegans by assessing worm survival after pathogen infection. The survival of the C. elegans conditioned with FAV for 24h was significantly higher compared with that of the control worm group fed only normal feed (non-pathogenic E. coli OP50) exposed to E. coli O157:H7, Salmonella typhi, and Listeria monocytogenes. To evaluate the protective effects of FAV on immune response, cyclophosphamide (Cy), an immune-suppressing agent was treated to in vitro and in vivo. We found that FAV significantly restored viability of mice splenocytes and immune promoting-related cytokines (interleukin [IL]-6, IL-10, inducible nitric oxide synthase [iNOS], interferon [IFN]-γ, and tumor necrosis factor [TNF]-α) were activated compared to non-fermented deer antlers. This finding indicated the protective effect of FAV against Cy-induced cell death and immunosuppressed mice. Taken together, our study suggests that immune-promoting antler velvet can be produced through fermentation using L. rhamnosus LFR20-004 and L. sakei LFR20-007.

가자미식해에서 분리한 Lactobacillus spp.의 항헬리코박터 활성 평가 (Anti-Helicobacter pylori Activity of Lactobacillus spp. Isolated from Gajami Sikhae)

  • 본 연구에서는 전통 가자미식해에서 분리한 LAB에 의한 H. pylori 억제효과를 살펴보았다. 가자미식해에서 분리한 40종의 LAB는 L. plantarum, L. brevis, Lc. mesenteroides, and W. paramesenteroides 등으로 확인되었다. 40종의 LAB중 liquid inhibition assay에 의해 H. pylori 억제효과가 있는 것으로 판단되는 10 종류의 젖산균을 선별하였다. 선별된 젖산균 배양액을 1%, 5%, and 10% 농도로 적용하였을 때 H. pylori 52, 51, e-53와 309에 대해서 생육 억제효과가 나타났다. LAB 배양 상등액의 pH를 7.0으로 조절하여 적용시켰을 LAB 10 종류 모두 5%와 10%의 농도에서 H. pylori 52와 LAB 7 종류의 10% 농도에서 H. pylori e-53에 대해 억제효과를 나타내었다. 그것은 또한 urease 활성을 억제시키는 영향도 나타내었다. 결과적으로 가자미식해에서 분리한 L. plantarum spp., L. brevis spp., Lc. mesenteroides spp., W. paramesenteroides spp. 등에 의해 H. pylori가 제균되거나 억제되는 효과를 나타내는 것을 확인하였다.

Mulberry Low-Fat Ice Cream Supplemented with Synbiotic: Formulation, Phytochemical Composition, Nutritional Characteristics, and Sensory Properties

  • For this study, we designed and produced mulberry low-fat ice cream supplemented with synbiotics (MLF-ISS). The sensory characteristics and physical, chemical, and microbiological qualities of MLF-ISS were then determined. Mulberry juice inoculated with or without probiotic (Lactobacillus plantarum TISTR 926 and Saccharomyces boulardii CNCM I-745) was also tested at 37℃ for 24 h to determine probiotic growth rate, pH, total anthocyanin content (TAC), total phenolic content (TPC), and antioxidant activity (AA). Only the TAC of mulberry juice inoculated with S. boulardii CNCM I-745 increased considerably (p < 0.05) among these parameters. MLF-ISS was produced with varied mulberry fruit concentrations (0, 10, 20, 30, or 40%) (w/w). The MLF-ISS prepared with 30% mulberry fruit (w/w) (30-MLF-ISS) had a higher score in appearance, color, and sweetness (p < 0.05) when sensory qualities were measured using the 9-point hedonic scale method. In the CIE lab system (L*, a*, b*), the color values of 30-MLF-ISS were 27.80 ± 0.26, 12.99 ± 0.59, and 1.43 ± 0.05, respectively. The 30-MLF-ISS was also subjected to a proximate analysis. The melting rate of 30-MLF-ISS was 0.29 ± 0.03 g/min and the time it took for the first drop to fall was 37.00 ± 7.00 min. TAC, TPC, and AA of 30-MLF-ISS were observed to alter significantly (p < 0.05) during varied intervals of storage at - 18℃ (0, 30, and 60 days). The viability of probiotics in 30-MLF-ISS slightly decreased after storage at -18℃ for 8 weeks, but remained about 6 log CFU/g. During storage at -18℃ for 0 and 120 days, no pathogenic bacteria were detected in 30-MLF-ISS. These findings show that 30-MLF-ISS has nutritional and functional value, is free of foodborne pathogenic bacteria, is safe for consumers' health, and is suitable for application in the ice cream and related food industries.

비병원성 야생효모 Candida oleophila WP5-19-1로부터 장내유용세균 유해 효소 β-glucuronidase 저해제의 생산 및 특성 (Production and Characterization of the Beneficial β-glucuronidase Inhibitor from non pathogenic wild yeast, Candida oleophila WP5-19-1, and its effects on gut microbes )

  • 비병원성 야생효모로부터 대장에서의 독성물질 활성화에 관여하여 대장암을 유발시키는 효소 중의 하나인 β-glucuronidase를 저해하는 물질을 생산하기 위하여 대전광역시 월평공원으로부터 분리 동정한 야생효모들 중 비병원성 야생효모들을 선별하여 배양 상등액과 무세포 추출물을 각각 제조한 후 β-glucuronidase 저해활성을 측정하였다. 133균주의 비병원성 야생효모들 중 C. oleophila WP5-19-1의 무세포추출물이 49.0%의 가장 높은 저해활성을 보여 우수 균주로 최종 선발하였다. β-Glucuronidase 저해물질 최적 생산 조건은 C. oleophila WP5-19-1를 초기 pH를 6.0으로 조정한 dextrose 5% 함유한 potato dextrose 배지에 접종하여 30℃에서 24시간 배양하였을 때였고 이때 IC50값; 8.4 mg으로 가장 높은 저해활성을 보였다. C. oleophila WP5-19-1의 β-glucuronidase 저해물질을 한외여과와 Sephadex G-50으로 gel 여과를 실시하여 부분 정제하였고 이들은 30-60℃, pH 6.0-9.0에서 80% 이상의 잔존 저해활성을 보여 안정하였다. 또한 철, 구리, 아연과 같은 중금속 이온들은 저해활성을 활성화시켰다.
