Neurochemical Characterization of the TRPV1-Positive Nociceptive Primary Afferents Innervating Skeletal Muscles in the Rats

  • Objective: Transient receptor potential vanilloid subfamily type 1 (TRPV1), a most specific marker of the nociceptive primary afferent, is expressed in peptidergic and non-peptidergic primary afferents innervating skin and viscera. However, its expression in sensory fibers to skeletal muscle is not well known. In this study, we studied the neurochemical characteristics of TRPV1-positive primary afferents to skeletal muscles. Methods: Sprague-Dawley rats were injected with total $20{\mu}l$ of 1% fast blue (FB) into the gastrocnemius and erector spinae muscle and animals were perfused 4 days after injection. FB-positive cells were traced in the L4-L5 (for gastrocnemius muscle) and L2-L4 (for erector spinae muscle) dorsal root ganglia. The neurochemical characteristics of the muscle afferents were studied with multiple immunofluorescence with TRPV1, calcitonin gene-related peptide (CGRP) and $P2X_3$. To identify spinal neurons responding to noxious stimulus to the skeletal muscle, 10% acetic acids were injected into the gastrocnemius and erector spinae muscles and expression of phospho extracellular signal-regulated kinase (pERK) in spinal cords were identified with immunohistochemical method. Results: TRPVl was expressed in about 49% of muscle afferents traced from gastrocnemius and 40% of erector spinae. Sixty-five to 60% of TRPV1-positive muscles afferents also expressed CGRP. In contrast, expression of $P2X_3$ immnoreaction in TRPV1-positive muscle afferents were about 20%. TRPV1-positive primary afferents were contacted with spinal neurons expressing pERK after injection of acetic acid into the muscles. Conclusion: It is consequently suggested that nociception from skeletal muscles are mediated by TRPV1-positive primary afferents and majority of them are also peptidergic.

Effect of Ascorbic Acid on the Gravitropic Response of Primary Roots in Maize (옥수수 일차뿌리에서 Ascorbic acid가 굴중성 반응에 미치는 효과)

  • Ascorbic acid (AA) is a multifunctional metabolite in plants that is essential for plant development and growth. We examined the effect of AA, an antioxidant, on the gravitropic response of primary roots in maize. The application of $10^{-3}$ M AA to the elongation zone did not affect the gravitropic response and slightly inhibited the root growth. However, treatment with both $10^{-5}$ M and $10^{-3}$ M AA at the root tip increased the gravitropic response and inhibited root growth. Differences in indole-3- acetic acid (IAA) activity between the upper and lower hemispheres of the root resulted in differential elongation along the horizontal root. Roots are extremely sensitive to IAA, and increasing the amount of IAA in the lower hemisphere of the root inhibited elongation. Therefore, we examined the effect of IAA in the presence of AA. The inhibitory effect of AA on the gravitropic response was greater in combination with IAA. To understand the role of AA in the regulation of root growth and the gravitropic response, we measured ethylene production in the presence of AA in the primary roots of maize. AA stimulated ethylene production via the activation of the 1-aminocyclopropane-1-carboxylic acid (ACC) oxidase gene, which regulates the conversion of ACC to ethylene. These results suggest that AA alters the gravitropic response of maize roots through modification of the action of ethylene.

THE SECOND MESIOBUCCAL CANAL OF UPPER PRIMARY MOLAR : CASE REPORT (제 2근심협측근관이 존재하는 상악유구치의 치험례)

  • In a primary teeth, dental caries is rapidly advanced the pulp disease, because the primary teeth have the thinner and the weaker enamel layer and the wider pulp chamber than relatively the permanent teeth. And the pulps of primary teeth are exposed during caries removal or even they are exposed by unexpected movement of the children or by trauma. For successive pulp treatment in primary teeth, it is necessary to understand completely about multiple canal morphology, variation of root canal anatomy and specific problem related to root formation and resorption of primary teeth. In upper primary molar, canal configuration of mesial root has the most variation same as upper molar. If not canal treatment is completely, most of all endodontic treatment should be failed. In a clinical case report, upper primary molars existed persistent pain or bleeding during treatment were founded the second MB canal and were performed the endodontic treatment of theirs. As a result, the upper primary molars have no symptom and good prognosis. In the examination of extracted upper primary molar, we found that 8 of 35 teeth(22.8%) in the upper primary first molars and 22 of 33 teeth(66.6%) in the upper primary second molar had the second mesiobuccal canal. It has revealed the high prevalence of two canals in mesiobuccal roots of upper primary molars. The frequency of occurrence of the second mesiobuccal canal must be taken into consideration when endodontic treatment is planned and as a possible cause of otherwise un explained failure.

  • Thirty seven human maxillary third mooars were injected with china ink, decalcified, cleared, and used in vitro study to determine the number of root canals the frequency and location of lateral canals, canals per root, and frequency of apical deltas. The results were as follows: 1. Less than half of the examined teeth showed 3 roots and 3 canals. 2. The mesiobuccal root was ound to contain a single primary canal in 65.4% of the teeth examined and two canals in 34.6% of the teeth studed. 3. In mesiobuccal roots with two canals, the separated apical foramen appeared 30.8% of the specimen concerned, but no common apical foramen appeared. 4. Of the 106 canals studied, 46.2% of the canals were found to contain lateral canals and these ramifications were mainly located in the apical third of the root.

Relationships between Lodging and Root Characteristics of Soy bean Plants (대두의 도복과 근부특성과의 관계)

  • The field experiment was carried out in order to find out the factors affecting lodging of Korean soybean varieties. Among the variety groups which were grown under the same cultural conditions, 25 Korean local varieties were selected by 5 lodging degree, and investigated their root characteristics in relation to lodging. The results obtained were as follows; There were no significant relationships between root dry weight, number of primary roots, number of adventitious roots and lodging. There also were no clear relationships between shoot/root weight ratio and lodging. However, it was recognized that the shoot weight/number of thick roots was highly related to soybean lodging. The susceptible varieties to lodging showed less shoot/thick root ratio.

  • One hundred human maxillary first molars were injected with china ink, decalcified, cleared, and used in vitro study to determine the number of root canals, the frequency and location of lateral canals, canals per root, and frequency of apical deltas. The results were as follows; 1. The mesiobuccal root was found to contain a single primary canal in 53% of the teeth studied and two canals in 47 % of the teeth studied. 2. In mesiobuccal roots with two canals, the seperated apical foramen appeared 32% of the specimens and the common apical foramen 15% of the specimens. 3. Of the 347 canals studied, 26.9% of the canals were found to contain lateral canals and these ramifications were mainly located in the apical third of the root.

  • Comparative biological activities of 70fr methanol extracts from the main roots and rhizomes of both red and white ginsengs were investigated using several in vitro experimental models. The main root of red ginseng and the rhizome of white ginseng strongly inhibited lipld peroxidation of hepatic microsomes induced by the non-enzymatic $Fe^{+}$ / Ascorbate system. The main root and rhizome of red ginseng markedly inhibited the release of G07, GPT and LDH by $CCl_4$-induced cytotoxicity in primary cultured rat hepatocytes as compared with those of white ginseng. And also, the main root of red ginseng showed a slight differentiating activity on HL-60 cancer cell line. The results suggest that the rhizome of ginseng have potential as a source of medicinal crude drug with possible pharmacolobica1 applications .

In mesiobuccal roots with two canals, the seperated apical foramen appeared 24% of the specimens and the common apical foramen 14% of the specimens. 3. Of the 169 canals studied, 23.1% of the canals were found to contain lateral canals and these ramifications were mainly located in the apical third of the root.

  • Morphology of primary root divergency and curvature determines the pattern of root resorption and periapical lesion, and affects successful root canal treatment. With the purpose of analysing the morphology of primary mandibular second molar roots and canals, the frequency, angle, radius, and start of curvature of the canal were measured. Fifty clinical radiographs were taken from $3{\sim}6$ years old children, followed by digitizing after scanning and analyzing by Scion image Beta 4.02TM(Scion Co. USA). The angle of curvature was determined by Schneider's method and the radius of curvature was determined by Schaefer's method. The results were as follows: 1. The angle of curvatures were $17.3^{\circ}{\pm}5.0$ (mesial), and $27.9^{\circ}{\pm}6.0$ (distal). Distal curvature was significantly larger than mesial.(p<0.05) 2. The radius of curvatures were $8.7mm{\pm}2.5$ (mesial), and $5.8\;mm{\pm}1.5$ (distal). Mesial curvature was significantly larger than distal.(p<0.05) 3. The start of curvatures were $4.1mm{\pm}0.6$ (mesial), and $4.2mm{\pm}0.6$ (distal). There were no difference between two groups. (P<0.05)

The effect of root canal preparation on the surface roughness of WaveOne and WaveOne Gold files: atomic force microscopy study

  • Objectives: To examine the surface topography of intact WaveOne (WO; Dentsply Sirona Endodontics) and WaveOne Gold (WOG; Dentsply Sirona Endodontics) nickel-titanium rotary files and to evaluate the presence of alterations to the surface topography after root canal preparations of severely curved root canals in molar teeth. Materials and Methods: Forty-eight severely curved canals of extracted molar teeth were divided into 2 groups (n = 24/each group). In group 1, the canals were prepared using WO and in group 2, the canals were prepared using WOG files. After the preparation of 3 root canals, instruments were subjected to atomic force microscopy analysis. Average roughness and root mean square values were chosen to investigate the surface features of endodontic files. The data was analyzed using one-way analysis of variance and post hoc Tamhane's tests at 5% significant level. Results: The surface roughness values of WO and WOG files significantly changed after use in root canals (p < 0.05). The used WOG files exhibited higher surface roughness change when compared with the used WO files (p < 0.05). Conclusions: Using WO and WOG Primary files in 3 root canals affected the surface topography of the files. After being used in root canals, the WOG files showed a higher level of surface porosity value than the WO files.
