Effect of Pretreatments and Holding Solutions on Vase Life and Quality of Cut 'Saphir' Rose (전처리와 보존용액이 절화장미 'Saphir'의 수명과 품질에 미치는 영향)

  • This experiment was carried out to investigate the effect of pretreatments and holding solutions on quality and vase life of cut 'Saphir' rose (Rosa hybrida L.). Pulsing with aluminum sulfate or NaOCl prolonged vase life held in HQS+sucrose+ethionine. Solution absorption held in distilled water were more increased than held in HQS+sucrose+ethionine. Holding solution with 8-hydroxyquinoline sulfate (HQS)+sucrose+ethionine increased fresh weight compared with distilled water except pulsing with STS+sucrose. Pulsing with aluminium sulfate delayed bent-neck held in HQS+sucrose+ethionine. Flower diameter and dry weight of petal held in HQS+sucrose+ethionine were more increased than held in distilled water except pulsing with STS+sucrose. There were no significance in Hunter color value 'L' and value 'a', but value 'b' was decreased held in distilled water.

Shear bond strength of a new self-adhering flowable composite resin for lithium disilicate-reinforced CAD/CAM ceramic material

  • PURPOSE. The purpose of this study was to evaluate and compare the effects of different surface pretreatment techniques on the surface roughness and shear bond strength of a new self-adhering flowable composite resin for use with lithium disilicate-reinforced CAD/CAM ceramic material. MATERIALS AND METHODS. A total of one hundred thirty lithium disilicate CAD/CAM ceramic plates with dimensions of $6mm{\times}4mm$ and 3 mm thick were prepared. Specimens were then assigned into five groups (n=26) as follows: untreated control, coating with $30{\mu}m$ silica oxide particles ($Cojet^{TM}$ Sand), 9.6% hydrofluoric acid etching, Er:YAG laser irradiation, and grinding with a high-speed fine diamond bur. A self-adhering flowable composite resin (Vertise Flow) was applied onto the pre-treated ceramic plates using the Ultradent shear bond Teflon mold system. Surface roughness was measured by atomic force microscopy. Shear bond strength test were performed using a universal testing machine at a crosshead speed of 1 mm/min. Surface roughness data were analyzed by one-way ANOVA and the Tukey HSD tests. Shear bond strength test values were analyzed by Kruskal-Wallis and Mann-Whitney U tests at ${\alpha}=.05$. RESULTS. Hydrofluoric acid etching and grinding with high-speed fine diamond bur produced significantly higher surface roughness than the other pretreatment groups (P<.05). Hydrofluoric acid etching and silica coating yielded the highest shear bond strength values (P<.001). CONCLUSION. Self-adhering flowable composite resin used as repair composite resin exhibited very low bond strength irrespective of the surface pretreatments used.

Effects of Seed Pretreatments on Emergence, Growth and Yield of Bupleurum falcatum L. (시호 입모향상을 위한 파종전 노천매장 효과)

  • The objectives of this study were to evaluate the effects of some seed pretreatments on seed germination, emergence, growth and yield of Bupleurum falcatum L. Seed treatments included stratification in open-field during winter, washing with running water, low temperature storage, soaking into $H_2SO_4$ solution, and scarification. Germination increased 15% by stratification and 9% by washing treatment compared to non-treatment, control. It was suggested that washing treatment leached inhibiting substance, while stratification treatment leached inhibiting substance and promoted growth of immatured embryo. In the field mulched with rice straw, stratification and washing treatments were effective to shorten days to emergence by 31 and 21 days and to increase emergence rate by 7 and 5 %, respectively, compared with control. By these two treatments, root growth was improved, the number of harvested plants was increased considerably and dry root yield per ha were increased by 29 and 22 %, respectively, compared with control.

  • The cultural characteristics of fruit bodies formation in the two selected strains(brown and yellow strain) of winter mushroom collected in Korea were investigated. The effects of various environmental conditions and physicochemical pretreatments were evaluated by the character of fruit bodies. There was no differences between two strains in non-selectivity of media, optimum temperature($11^{\circ}C{\sim}15^{\circ}C$), relative humidity(90%), and illumination(60 lux). In the application of alternated temperature (the best condition : $5^{\circ}C{\cdot}10^{\circ}C$ and $5^{\circ}C{\cdot}15^{\circ}C$), ultraviolet ray(30~50 seconds), electricity(5 seconds), thiamine hydrochloride(0.05~0.1%), urea, and IAA as a pretreatments, there was no differences between two strains. The brown strain had larger pileus and longer stipes than these of the yellow strain, while the yellow strain yielded more weight and number of the fruit body formation than the brown strain.

  • This research was carried out to promote the seed germination of Styrax japonica, by several pretreatments and to examine its seedling growth and seed component variations according to time and pretreatment. The results are as follows ; 1. Styrax japonica seeds are usually germinated after more than one year cold stratification. However, when the seeds were stratificated for 5 months after dipped in the solution of 70% $H_2SO_4$ for 30 minutes and also treated with $GA_3$(3000ppm), they showed 55% germination. This demonstrated that the seeds could be germinated in one year when pretreated. 2. The growth of one-year-old seedlings after pretreatment was the same as that of two-year-old seedlings when compared 160 days after seeding. 3. Before seeding, one-year-old, pretreated seeds of Styrax japonica showed increased moisture content but decreased crude fat, carbohydrate and exude ash, which were similar to the components of two-year-old seeds.

Influences of Pretreatment Procedures, and Refractive and Absorptive Indices in Grain Size Analysis of Sandy Samples by Laser Diffraction Grain Size Analyzer (레이저 회절 입도분석기를 이용한 사질 시료의 입도분석에 있어서 전처리 및 굴절율과 흡수율의 영향)

  • This study aims to compare the inf luences of pretreatment procedures, and refractive and absorptive indices in grain size analysis of sandy samples collected from Sagot Beach, Baengnyeong Island by laser diffraction grain size analyzer, and propose the proper procedure and method in grain size analysis of sandy samples. The analyzed samples do not indicate large differences by the three pretreatment procedures applied in this study. However, the organic matters should be removed by hydrogen peroxide, because the samples without hydrogen peroxide pretreatments show differences from the samples with hydrogen peroxide pretreatments. The results with a refractive index of 1.3 and absorptive index less than 0.01 also indicate differences from those with other indices. Compared to the differences in fine samples, these differences are not significant and thus, it can be concluded that the results in grain size analysis of sandy samples are not greatly influenced by the refractive and absorptive indices. However, other indices out of the ranges should be applied.

A Study for the Utilization of Wild Herbaceous Species -Effects of Gibberelic Acid Treatment on Seed Germination- (야생초류의 자원화를 위한 연구 -지베렐린($GA_3$) 처리가 몇 야생초류의 종자발아에 미치는 영향-)

  • To examine the effects of gibberelin treatment on seed germination of six wild herbaceous species -Dicentra spectabilis, Aster pekinensis, Agastache rugosa, Elsoholtzia ciliata, Boehmeria tricuspis and Pulsatillia cerrma var. chaerophyiloides - in Kangwon-do, pretreated seeds of five species were sowed on plastic pots with mixed soil(sand:peat moss:perlite: vermiculite, 1:1:1:1, v/v) in 23,April 1995. Seeds were soaked in 25,50 ppm gibberelin solution and ground water during 24 hours. Pretreated seeds of Pulsatiilia cerrma var. chaerophylloides were sowed on nursery soil in 1, June 1995. Seed pretreatments in 31, May 1995 were same method. Seed germination rate was measured and compared among species and treatments. Seed germination rate of Dicentra spectabilis, Agastache rugosa and Elsoholtzia ciliata was differed higly significant among pretreatments ,but those of that of Aster pekinensis, Boehmeria tricuspis and Pulsatillia cerrma var chaerophylloides did not significant at 5% level. Gibberelin pretreatment raised seed germination rate of Dicentra spectabilis and Agastaohe rugosa and proper concentration of GA$_3$ was 25ppm and seed germination rate of Elsoholtzia ciliata was highest at GA$_3$ 50ppm plot. The utilization of wild herbaceous species were discussed.

Determination of oxymatrine in Sophora Radix by High Performance Liquid Chromatography (고성능 액체 크로마토그래피를 이용한 고삼 중의 oxymatrine 분석에 관한 연구)

  • A high performance liquid chromatographic method was designed for the quantitative analysis of oxymatrine in Sophora Radix. The separation of oxymatrine was performed by reversed-phase chromatography with a $C_{18}$ column and a buffered aqueous solution containing acetonitrile, and monitored by UV absorption at 215 nm. Extraction of oxymatrine in Sophora Radix was carried out using various solvents and extraction methods. The optimum extraction efficiency for the crushed Sophora Radix was achieved by reflux at $80^{\circ}C$ in 50% ethanol for five hours. Most extraction methods used to complicate pretreatments. In this study, sublimation was employed for a extraction method without going through complicate pretreatments. Sublimation was carried out under high vacuum ($1{\times}10^{-3}$ torr) and at high temperature ($200^{\circ}C$). Extraction efficiency using Sublimation was found to be inferior to other extraction methods.

Assessment of different pretreatments to breakage dormancy and improve the seed germination in Elaeocarpus serratus L. - an underutilized multipurpose fruit tree from South India

  • The seeds of Elaeocarpus serratus, a tropical underutilized fruit tree are characterized by hard seed coat and consequent poor water uptake and low germination. To improve the regeneration through seeds, various parameters such as viability of seeds, water uptake, and effect of seed mass on germination and pretreatments were performed using a completely randomized design (CRD). Tetrazolium (TZ) test was conducted using fresh, mature seeds revealed $50{\pm}2.56%$ mean viability. Seeds of different weight classes showed similar pattern of water uptake and the saturation level was achieved at 60 hrs of soaking. Seeds belong to weight class 2.6-3.5g were germinated ($12.5{\pm}1.26%$) with $175{\pm}1.75days$ (d) of mean time taken for germination (MTG). Germination capacity of seeds varied significantly among different populations and Varkala population gave $12.5{\pm}1.1%$ germination with $174.6{\pm}2.5d$ MTG. Among various seed treatments, mechanical scarification was superior in germination and significant reduction in MTG ($p{\leq}0.05$). The mechanical scarification by complete removal of seed coat resulted in $49.2{\pm}1.52%$ germination within a short period of time ($9.52{\pm}0.89d$ MTG). However, the complete removal of seed coat without damaging to embryo is a difficult task. An alternate treatment (Mechanical scarification II) by making cracks on nut faces vertically followed by soaking in distilled water for 24 hrs gave $48.4{\pm}1.73%$ germination with significantly reduced MTG ($12.14{\pm}0.56d$) over unsoaked, untreated control ($6.5{\pm}1.84%$ germination and $197.18{\pm}1.79d$ MTG; $p{\leq}0.05$). This treatment (Mechanical scarification II) is therefore recommended for E. serratus seeds as it can adopt easily and can achieve 7 fold increases in germination over control. The recorded germination through mechanical scarification is in tune with realized viability percentage of the seeds.

Effect of Biological and Liquid Hot Water Pretreatments on Ethanol Yield from Mengkuang (Pandanus artocarpus Griff)

  • This study aimed to increase the sugar and ethanol yield from the mengkuang plant biomass through biological and liquid hot water (LHW) pretreatment and their combination. The results showed that biological pretreatments with 5% inoculum of the fungus Trametes versicolor resulted in the highest alpha cellulose content incubated for 30 days, and 10% inoculum resulted in the lowest lignin content. LHW pretreatment decreased the hemicellulose content of pulps from 10.17% to 9.99%. T. versicolor altered the structure of the mengkuang pulp by degrading the lignin and lignocellulose matrix. The resulting delignification and cellulose degradation facilitate the hydrolysis of cellulose into sugars. The alpha cellulose content after biological-LHW pretreatment was higher (78.99%) compared to LHW-biological pretreatment (76.85%). Scanning electron microscopy analysis showed that biological-LHW combinated treatment degrades the cell wall structures. The ethanol yield for biological-LHW pretreated sample was observed 43.86% (13.11 g/L ethanol by weight of the substrate, which is much higher than that of LHW-biological pretreatment (34.02%; 9.097 g/L). The highest reducing sugar content about 45.10% was observed with a resulting ethanol content of 15.5 g/L at LHW pretreatment temperature of $180^{\circ}C$ for 30 min.
