Effects of Cosmetics and Their Preservatives on the Growth and Composition of Human Skin Microbiota (피부 미생물총의 생장과 구성에 대한 화장품과 그 방부제의 영향)

  • We investigated the growth-inhibitory activities of cosmetics and their preservatives against pathogens and resident skin bacteria. Of the tested cosmetics, preservatives such as parabens, 1,2-hexanediol, phenoxyethanol-contained toner, emulsion, cream and baby cream exhibited potent antibacterial effects against Staphylococcus aureus, Escherichia coli and Pseudomonas aeruginosa. Parabens, 1,2-hexanediol and phenoxyethanol inhibited the growth of pathogens, as well as skin-resident bacteria such as Staphilococcus epidermidis, Shigella flexneri, Enterobacter aerogenes and so on. The application of a basic cream containing phenoxyethanol to human skin was shown to disturb the skin microbiota: at the phylum level, Proteobacteria increased and at species level, 4P004125_s increased and Propionibacterium humerusii decreased. Based on these findings, parabens, 1,2-hexanediol and phenoxyethanol have antimicrobial activity and cosmetics containing phenoxyethanol may disturb skin microbiota.

  • To develop a natural food preservatives from medicinal herbs (62 species), antimicrobial test was performed with hot water and ethanol extracts against Escherchia coli, Listeria monocytogens and Candida albicans by paper disk method. And its result as a first screening test, 7 kinds of medicinal herbs were selected, and more precise analysis was conducted at last 3 kinds of medicinal herbs was finally selected. Therminalia chebula Retz and Rhus javanica has a broad antimicrobial spectrums and has a low MIC values. In growth inhibition test, they showed a very strong antimicrobial activity aganist Listeria monocytogens. If there has a elaborate approaches in its characteristics and properties, some good result must followed in making a powerful natural antimicrobial preservatives.

Investigation on the Safety of Hydroquinone and Preservatives among Whitening Functional Cosmetics Containing Albutin in Korea (국내 유통 알부틴 함유 미백 기능성화장품 중 히드로퀴논 및 살균보존제 안전성 조사)

  • Arbutin, which is used as a whitening ingredient, can produce hydroquinone, known as causing skin disease and carcinogen. Preservatives are essential to prevent microbial contamination during long-term storage and use of cosmetics, but safety issues such as toxicity and skin irritation are being raised. This study was conducted to determine hydroquinone and 21 preservatives levels in 40 arbutin-containing whitening functional cosmetics sold on-line and off-line. Result showed that 9 products contained hydroquinone. The concentrations in 7 products were ranged from 0.3 to 0.9 ppm, which were within the maximum allowed amount established by the Ministry of Food and Drug Safety. However, 2 products were 8.4 and 50.5 ppm and exceeded the allowed amount. Preservatives were detected 20 products. Detected items and ranges were phenoxy ethanol 0.1 ~ 0.7% (N = 15), Methyl paraben 0.19 ~ 0.21% (N = 2), Chlorphenesin 0.13% (N = 1), chlorhexidine 0.006% (N = 1), Propyl paraben 0.06% (N = 1), which were within maximum allowed amount established by the Ministry of Food and Drug Safety. Also, in cases of functional cosmetics the phrase "functional cosmetics" should be expressed on the primary or secondary package of cosmetics by cosmetics act. However, 1 product did not state the phrase as functional cosmetics. This study suggest that preservatives were safely managed. However, hydroquinone in hydroquinone-detected products could be produced by the decomposition of arbutin. Thus, further studies on the decomposition of arbutin are required to improve the quality control of the cosmetics.

Simultaneous determination of preservatives in sausages using liquid chromatography with electrospray ionization tandem mass spectrometry (LC-MS/MS를 이용한 소시지 중 보존료 동시분석)

  • A simultaneous determination method was developed for nine preservatives (benzoic acid, sorbic acid, dehydroacetic acid, methyl-, ethyl-, isopropyl-, propyl-, isobutyl- and butyl-parabens) in sausage by liquid chromatography with electrospray ionization tandem mass spectrometry (LC/ESI-MS/MS). Each parameter was established by multiple reaction monitoring in negative mode. Separation was achieved on a phenyl-hexyl ($2.5{\mu}m$, $2.1{\times}150mm$, Waters) with A-20 mM ammonium acetate containing 0.1% acetic acid in water, B-Acetonitrile as mobile phase with gradient mode at a flow rate of 0.3 mL/min. The developed method was validated for specificity, linearity, accuracy and precision in sausages samples. Linearity was over 0.998 with calibration curve of the mixed standards. The mean recoveries from sausages fortified at the level of 2.0~10.0 mg/L were in range of 98.60~109.16% with RSDs lower than 8.93%. The limits of detection (LOD) and the limits of quantification (LOQ) were in the range between 0.0003~0.085 mg/L and 0.01~0.257 mg/L, respectively. Intra-day precision and inter-day precision were 0.45~6.16% and 2.81~13.33%, respectively. Using presently developed determination method, 33 field sausage samples from Gwangju city in Korea were screened over nine preservatives. As a result, no preservatives were detected in all samples.

  • This study was conducted to investigate the preservative effects of low molecular weight chitosan with and without other preservatives on kimchi at $10^{\circ}C$ during 12 days of fermentation. The pH and titratable acidity of kimchi with preservatives (PSV) were higher and lower, respectively, than those of control kimchi (CON). The decrease in reducing sugar content was more remarkable in CON than in PSV as the fermentation proceeded. Contents of lactic acid and acetic acid were lower in PSV, especially in kimchi with chitosan dissolved in acetic acid (CH-B) and in the one containing both chitosan and Na-benzoate (CHS) than those in CON. The numbers of total viable cells, Leuconostoc sp. and Lactobacillus plantarum were higher in CON than those in PSV. Sensory firmness and green flavor were significantly lower in CON than in CH-B and CHS. Sourness and staled flavor of CH-B and CHS tended to be lower than those of other kimchi samples.

The effect of nonionic surfactants on the antimicrobial activity of preservatives in cosmetic products (비이온 계면 활성제가 화장품의 방부력에 미치는 영향)

  • In order to investigate the effect of nonionic surfactants on the antimicrobial activity of preservatives in the presence and absence of p.0.E(20) Sorbitan fatty acid ester commonly used in cosmetics and pharmaceutical systems, these experiments were carried out by determining Minimum Inhibitory Concentration(MIC) values and MIC values of adaptation against test organisms. And also the inactivation of the preservative against each microorganism in formula added with various concentrations of P.0.E(20) Sorbitan monostearate were measured by use of a preservative death time curve The results obtained were as fort low : 1) Nonionic surfactant inactivated Methylparaben to varying extents, but not Imidazolidinyl urea. 2) A combined preservative system was inactivated to a little extent (range of 0.16-0.20% Conc.), no adaptation was observed for the 5. aureus ATCC 6538. Imidazolidinyl urea complex combined with Methylparaben had a broad antibacterial spectrum against the Gram(.) and the Gram(-) bacteria It was found that preservatives had a synergistic effect by use of mixed form of preservatives, 3) In formula preserved with 0.2% Methylparaben containing 0.5, 1.0 and 2.0% P.0.E(20) Sorbitan monostearate, E. coli ATCC 10s36 and P. aeruginosa NCTC 10490 died quickly within in 2hr 4) However, from Fig.5, S. aereus ATCC 6538 died more slowly within increasing surfactant concentration and the D-values(Decimal reduction time) were 5.2, 8 and 14 hr. for samples containing 0.5, 1 0 and 2.0% P 0. E(20) Sorbitan monostearate, respectively. 5) In the case of Methylparaben, no adaptation for the E. coli ATCC 10536 6) All of the nonionic surfactant, p.0. E(20) Sorbitan fatty acid ester used in the experiments decreased the effectiveness of Methylparaben, but not of Imidazolidinyl urea.

Penetration of ACQ Treatment and its Effect of Degradation on Wood Tissues (Structure) (ACQ 약제의 목재 내 침투 및 부후 특성에 미치는 목재 조직의 영향)

  • To obtain the basic information on wood preservative process using copper, the penetration ACQ-2 preservative agent into Douglas fir wood and its degradational characteristics were investigated. Douglas fir is known to be difficult for preservative agent injection. To find the hindrance factor of the preservatives movement, the ray height, ray density, and length of tracheid were measured and observed by a stereoscopic microscope and scanning electron microscope. The results were compared in the well-penetrated parts of preservatives and insufficient parts. There were no significant differences in height, length, and density between both parts, except for the pits of the wood elements observed in SEM micrograph. More declosed pits were found in the well-penetrated parts of preservatives. Decay characteristics of wood specimen treated with various ACQ concentrations by brown-rot fungi was observed by an optical microscope. The decayed properties of tracheid, rays, and resin canals was found in the non-treated wood specimens and it was also found that some extent of preservatives concentration is necessary to protect the decay.

  • Benzoic acid, methyl paraben, and butyl paraben are preservatives that have been used in pharmaceutical, cosmetic, and food products. However, as their toxicities for human have been reported, many nations and organizations including Korea have established a regulation limit for thier usage of these preservatives in food products. The present study developed the isotope dilution liquid chromatography mass spectrometry method for accurate determination of three target preseratives in soysauce. In this study, the isotope dilution liquid chromatography mass spectrometry method was developed for accurate determination of three target preservatives in soy sauce. LC separation was optimized considering the pKa of benzoic acid which is lower than those of methyl and butyl parabens. A C18 column was used with 5 mM ammonium acetate and methanol as mobile phases. Mass spectrometry was operated in negative mode and selected reaction monitoring mode (SRM). Soy sauce sample was cleaned-up with C18 SPE cartridge for removing matrix inferences and color material. Optimized conditions and the method were validated with soy sauce reference materials for the analysis of food preservatives from Health Science Authority in Singapore. The measured values of benzoic acid, methyl and butyl paraben agreed well with reference values within their uncertainties.

Simultaneous determination of 9 preservatives in processed foods using high-performance liquid chromatography with photo diode array detector (HPLC-PDA를 이용한 가공식품 중 보존료 9종 동시분석)

  • This study was performed to develop an analytical method using Carrez reagents as the precipitant to effectively and easily remove proteins and lipids while pretreating samples for the simultaneous determination of preservatives, including dehydroacetic acid (DHA), sorbic acid (SA), benzoic acid (BA), methyl ρ-hydroxybenzoate (MP), ethyl ρ-hydroxybenzoate (EP), propyl ρ-hydroxybenzoate (PP), isopropyl ρ-hydroxybenzoate (IPP), butyl ρ-hydroxybenzoate (BP), and isobutyl ρ-hydroxybenzoate (IBP). The effective selectivity was determined by HPLC separation analysis for nine preservatives in the test solution, after removing interfering materials such as lipids and proteins. The method developed in this study showed excellent linearity at 0.999 or higher. The limit of detection (LOD) ranged from 0.09 to ~0.12 mg/L and the limit of quantitation (LOQ) was ~0.280.37 mg/L. The results of the recovery test on processed foods, including pickles, cheeses, processed meat products, beverages, sauces, and emulsified foods showed DHA, SA, BA, MP, EP, IPP, PP, IBP, and BP at 90.9~107.7 %, 85.4~113.7 %, 90.7~111.6 %, 84.5~111.2 %, 81.3~110.9 %, 82.5~102.2 %, 81.1~110.0 %, 80.9~109.0 %, and 82.4~110.3 %, respectively. The probability of the simultaneous analytical method developed in this study as a quantitative method was confirmed for various processed foods.

A Study on UV Filters, Preservatives and Heavy Metals Contained in the Children's and Inorganic Sunscreens Distributed in Korea (국내 유통 어린이 및 무기 자외선차단제에서 자외선 차단성분, 보존제 및 중금속 실태 조사)

  • In this study, 27 inorganic sunscreens and 23 sunscreens for children were collected to investigate the use and incorporation of 17 types of sunscreen agents, 13 types of preservatives, and 5 types of heavy metals. As a result, sunscreen agents were detected in the order of titanium dioxide (41 cases), zinc oxide (29 cases), bis-ethylhexyloxyphenyltriazine (10 cases), ethylhexyl salicylate (8 cases), and ethylhexylmethoxynamate (8 cases), and preservatives were detected in order of phenoxyethanol (6 cases), benzoic acid (1 case), and dihydroacetic acid(1 case). All of the identified sunscreen agents were suitable for labeling, but phenoxyethanol, a preservative component other than labeling, was detected at a concentration of 0.1%. Heavy metals such as lead, cadmium, arsenic, antimony, and nickel were not detected or were detected at various concentrations of less than 3.6 ㎍/g. All the detected UV filters, preservatives, and heavy metals were less than the allowed maximum amount stipulated by the Regulations on Cosmetic Safety Standards in Korea.
