Production of Korean Native Cow from Mongolian Cow following Transfer of Vitrified Blastocyst (Mongolian 수란우에 한우 동결수정란의 이식 후 산자 생산)

  • The purpose of this study was to investigate the comparison of viability of bovine blastocysts following glass micropipette (GMP) vitrification and the possibility of production of Korean Native Cow ("Hanwoo,"Bos taurus coreanae) following embryo transfer into Mongolia cows (Bos taurus mongolian). The embryos of Korean Native Cow were produced by IVMFC or superovulation methods in Korea, cryopreserved by GMP vitrification, and subsequently trans-ported to Mongolia. The recipient cows were synchronized using a CIDR plus and prostaglandin $F_2\alpha$($PGF_2\alpha$) treatment. To produce in vivo embryos, seven cows were superovulated using FSH and PGF$_2$/sub $\alpha$/ treatment. A total of 64 blastocysts ( $9.1\pm2.94$ per cow) were collected. In vitro embryos were produced using a defined culture system which cleaved in 80.1% ova (174/217), and developed to blastocyst stage embryos of 40.8% (71/174). The post-thaw survival rate of in vivo blastocysts (93.7%; 45/48) was significantly higher than that of in vitro blastocysts (82.5%; 52/63, P<0.05). Embryo transfer was carried out using 8 Mongolian recipient cows and 2 post-thaw blastocysts per recipient. Five of 8 recipients were found pregnant at Day 60 but one abortion occurred by Day 240. Two of offspring were produced from the Mongolian cows at 275 days after embryo transfer. These results indicated that a GMP vitrification method could be used as a cryopreservation technique for in vivo or in vitro bovine blastocysts and produced effectively a Korean Native Cow following embryo transfer into a Mongolian recipient cow.

Effect of Level of Feeding on the Performance of Crossbred Cows during Pre- and Post-partum Periods

  • The study was undertaken to see the effect of elevated feeding during pre-partum or pre- as well as post-partum period on the productive and reproductive performance of crossbred cows. The experiment lasted for 60 d pre-partum to 120 d post-partum. Eighteen dry pregnant crossbred cows divided into three equal groups were fed either as per NRC feeding standard (C) or 20% above NRC during 60 d pre-partum ($T_1$) or fed 20% above NRC during both 60 d pre-partum to 120 d post-partum ($T_2$) period. During prepartum period body weight gain was significantly ($p{\leq}0.05$) higher in $T_1$ and $T_2$ groups than that of control group. The animals fed at higher plane of nutrition ($T_1$ and $T_2$) took significantly lesser time for complete relaxation of pelvic muscles, act of calving and for expulsion of placenta than that of control group. Moreover, such cows delivered 2 to 3 kg heavier calves as compared to normal fed dams. During post-partum period, the average daily milk yield was significantly higher in $T_2$ group than that in $T_1$ and control groups. The peak yield was significantly higher in $T_2$ group, it took longer time to reach peak production but it was more persistent in this group as compared to $T_1$ and control groups. Average milk fat, solids-not-fat (SNF) and total solids were significantly higher in $T_1$ and $T_2$ groups as compared to control group. Body weight losses incurred during early lactation were not even compensated by end of 4th month of lactation in C and $T_1$ groups whereas the animals in $T_2$ group gained 2.0 kg. The 1st post-partum estrus and conception rate were better in high fed groups ($T_1$ and $T_2$) than that of control group. The returns over feed cost of milk production were higher in $T_2$ group followed by $T_1$ and control groups indicating the advantage of elevated feeding during pre- and post-partum periods.

  • Luteal cells produce progesterone that supports pregnancy. Steroidogenesis requires coordination of the anabolic and catabolic pathways of lipid metabolism. In the present study, the corpus luteum (CL) in early pregnancy established from luteal phase and pregnant phase was analyzed. The first study determined progesterone changes in the bovine CL at day 19 (early maternal recognition period) and day 90 in mid-pregnancy and compared them to the CL from day 12 of the estrous cycle. CL alternation was tested using two-dimensional polyacrylamide gel electrophoresis (2-DE) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF). Comparing CL from luteal phase to those from pregnant phase counterparts, significant changes in expression level were found in 23 proteins. Of these proteins 17 were not expressed in pregnant phase CL but expressed in luteal phase counterpart, whereas, the expression of the other 6 proteins was limited only in pregnant phase CL. Among these proteins, vimentin is considered to be involved in regulation of post-implantation development. In particular, vimentin may be used as marker for CL development during pregnancy because the expression level changed considerably in pregnant phase CL tissue compared with its luteal phase counterpart. Data from 2-DE suggest that protein expression was disorientated in mid pregnancy from luteal phase, but these changes was regulated with progression of pregnancy. These findings demonstrate CL development during mid-pregnancy from luteal phase and suggest that alternations of specific CL protein expression may be involved in maintenance of pregnancy.

  • Successful induction of lactation in 5 nulliparous repeat breeding cows was achieved by administering diethyl stillboestrol (0.1 mg/kg B.W.) and hydroxyprogesterone caproate (0.25 mg/kg B.W.) for 3 days followed by reserpine (4 mg/day) for 4 days, and dexamethasone (16 mg/day) for one day. The treatment caused cystic ovarian condition in all the animals. Intravaginally progesteronesoaked sponges suppressed but did not eliminate the behavioural estrus. Administration of LHRH ($300{\mu}g$) regressed the cysts in all animals in the first instance but the cysts reappeared in 2 animals. One animal became pregnant and delivered a normal calf.

  • This experiment was carried out to investigate of the early pregnancy diagnosis by finger pressure method, mucus smear method and rectal palpation method in 169 heads of pregnant dairy cow. 1. Pregnancy diagnostic rate of previous, middle and late period of pregnancy by finger pressure methods were 87.3%, 82.0% and 70% respectively. 2. Pregnancy diagnostic rate of previous, middle and late period of pregnancy by mucus smear methods were 83.1%. 94.9% and 95.0% respectively. 3. Pregnancy diagnostic rate of previous, middle and later period of pregnancy by rectal palpation methods were 83.1%, 96.1% and 100% respectively. 4. Pregnancy diagnostic rate of early pregnancy by finger pressure method, mucus smear method and rectal palpation method were 86.7%, 86.8% and 66% respectively. 5. Accordingly, finger pressure method in pregnancy diagnosis is thought to be recommendable because this early diagnostic means are simple with accurate results.

Results of Embryo Transfer with Hanwoo Embryos Produced In-Vivo or In-Vitro to Holstein Cows as Recipients (체내 또는 체외에서 생산된 한우 수정란을 젖소 수란우에 이식한 결과)

  • This study was performed to investigate the result that in-vivo or in-vitro embryos of Hanwoo cows were transferred to Holstein cows. Seventeen Hanwoo cows were used as donors for production of in-vivo embryos and fresh hanwoo in-vivo embryos were transferred to 1,150 Holsteins. And 2 embryos were transferred to 188 Holstein recipients to produce twin calves. Diagnosis on pregnancy was performed by rectal palpation at $60\sim90$ days after transfer. The pregnancy rate of Holstein recipients was 55.8% after transfer with Hanwoo in-vivo embryos and 38.2% after transfer with Hanwoo in-vitro embryos. The delivery rate of pregnant Holstein recipients was 88.4% after transfer with Hanwoo in-vivo embryos and 75.6% after transfer with Hanwoo in-vitro embryos. The rate of delivery of Holstein recipients transferred with two Hanwoo embryos was 36.2% and the rate of twin production was 25.9%. The rate of twin production by embryo transfer with in-vivo embryos was 30.4%, whereas the fate with in-vitro embryos was 15.6%. The pregnancy rate according to the grade of corpus luteum of Holstein recipients transferred with Hanwoo in-vitro embryos was 41.5 and 36.0% for A and B grade, respectively. The pregnancy rate according to the transfer in site in the uterine lumen of recipients was 40.9 and 32.7% for anterior and middle site, respectively. The pregnancy rate according to day of embryo transfer after estrus of recipients was 45.5, 38.8 and 39.7% for day 6, day 7 and day 8, respectively. There was difference of pregnancy rate according embryo transfer technician ($30.5\sim45.8%$) individual dairy farm ($21.1\sim51.0%$). These results are supposed to indicate that the rate of pregnancy after transfer with Hanwoo embryos to Holstein recipients was similar to that within the same breed, and consequently that this method would be beneficial to enhance the productivity in Hanwoo reproduction.

Effects of energy-protein supplementation frequency on performance of primiparous grazing beef cows during pre and postpartum

  • Objective: Twenty-four pregnant Nellore primiparous grazing cows were used to evaluate the effects of energy-protein supplementation and supplementation frequency during pre (105 d before calving) and postpartum (105 d after calving) on performance and metabolic characteristics. Methods: Experimental treatments consisted of a control (no supplementation), daily supplementation (1.5 kg/d of concentrate/animal) and infrequent supplementation (4.5 kg of concentrate/animal every three days). During the pre and postpartum periods, concentrations of blood metabolites and animal performance were evaluated. Ureagenesis and energy metabolism markers were evaluated at prepartum period. Results: Supplementation frequency did not alter (p>0.10) body weight (BW), average daily gain (ADG), and carcass traits during pre and postpartum. The BW (p = 0.079), adjusted BW at day of parturition (p = 0.078), and ADG (p = 0.074) were greater for supplemented cows during the prepartum. The body condition score (BCS; p = 0.251), and carcass traits (p>0.10) were not affected by supplementation during prepartum. On postpartum, supplementation did not affect animal performance and carcass traits (p>0.10). The dry mater intake was not affected (p>0.10) by supplementation and supplementation frequency throughout the experimental period. Daily supplemented animals had greater (p<0.001) glucose levels than animals supplemented every three days. Supplementation and supplementation frequency did not alter (p>0.10) the levels of blood metabolites, neither the abundance of ureagenesis nor energy metabolism markers. Conclusion: In summary, our data show that the reduction of supplementation frequency does not cause negative impacts on performance and metabolic characteristics of primiparous grazing cows during the prepartum.

Studies on the Blood Picture of Holstein Cows in Korea (Holstein 암소의 혈액상(血液像)에 관하여)

  • The literature on the hematology of dairy cattle contains very important information of physiological and clinical field, but a comprehensive survey on the blood picture of Holstein cows has not been made yet in Korea. The object of the present investigation was to make good this deficiency, and to suggest standards for the blood picture of Holstein cows kept under average farming condition in this country. Observations were made on the blood picture of 30 healthy non-pregnant Holstein cows aging from 6 to 10 years. All of them were selected at random from 15 livestock farms of Deajeon area in order to determine the hematological values from January to Feburary, 1974. The ranges and mean values of erythrocyte count, hemoglobin in blood, hematocrit value, mean corpuscular volume, mean corpuscular hemoglobin, mean corpuscular hemoglobin concentration, and total and differential leukocyte count were determined. The result obtained in this work were summerized as follows: 1. Ranges and mean values(with standard error) of erythrocyte count, hemoglobin in blood and hematocrit values were 4.90~7.82 and $5.83{\pm}0.12{\times}10^6/mm^3$, 7.8~10.3 and $8.7{\pm}0.11g/100ml$, and 26~38 and $30.2{\pm}0.53$ mI/100 ml, respectively. 2. Mean corpuscular hemoglobin, mean corpuscular volume, and mean corpuscular hemoglobin concentration showed ranges and mean values(with standard error) of 13.2~17.4 and $14.8{\pm}0.22$ pg, 46.0~71.8 and $52.3{\pm}0.31{\mu}m^3$, and 21.6~32.7 and $28.6{\pm}0.47$ g/100 ml, respectively. 3. Total leukocyte count showed a range of 6,300~13,600 and a mean value(with standard error) of $10,035{\pm}346/mm^3$. 4. Ranges and mean values(with standard error) of differential leukocyte counts of total neutrophil were 21~40 and $36.4{\pm}0.7%$, 1,638~4,080 and $3,233{\pm}111/mm^3$, of band neutrophil 0~9 and $3.9{\pm}0.5%$, 0~1,028 and $390{\pm}54/mm^3$, of segmented neutrophil 18~35 and $28.4{\pm}0.7%$, 1,386~3,672 and $2,842{\pm}103/mm^3$, of lymphocyte 46~69 and $55.3{\pm}1.1%$, 3,380~8,976 and $5,535{\pm}263/mm^3$, of monocyte 0~4 and $1.7{\pm}0.2%$, 0~324 and $116{\pm}21/mm^3$, of eosinophil 4~15 and $10.8{\pm}0.6%$, 540~1,725 and $1,082{\pm}72/mm^3$. No basophil was observed in this work.

  • Estrus cycle of cow, season and follicldar stimulating treatment, having an effect on the number of follicle, are investigated for the oocyte recovery rate in ovum pick-up(OPU). The number of follicle aspirated and oocyte collected on the different days of estrus cycle(D 4~5, D 9~10 and D 14~15) were not significantly different among the groups. The higher number of viable oocytes were produced on Jan-May(79.0%) than Jun-Aug(33.3%) by OPU in cow. The number of follicle and aspirated oocyte in cows treated FSH or PMSG combined with bovine somatotropin(bST) were 1.2~1.5 times higher than in cows treated alone follicular stimulating hormone(FSH) or pregnant mare serum gonadotropin(PMSG). In conclusion, OPU can be repeatedly practiced 2 or 3 times in an estrus cycle. In addition, the high environmental temperature is not good for ovarian function of cow and the bST co-treatment with FSH or PMSG is increasing the number of aspiratable follicle.

Embryo Production and Transfer for Dairy MOET Scheme Application (젖소 MOET Scheme의 추진을 위한 수정란 생산 및 이식)

  • The objective of this study was to apply the multiple ovulation and embryo transfer (MOET) program practically in dairy herds. Forty five superior Holstein cows ranked in 5% according to Type-Production Index(TPI) in Korea were selected as donors. The donors were superovulated with pFSH and the embryos collected from donors were frozen and preserved. The preserved embryos and frozen Holstein embryo imported from foreign country were thawed and transferred to recipients. The results obtained were as follows; 1. The total number of ova and freezable embryos collected per donor was 6.5 and 2.8, respectively. 2. The freezable embryos were obtained more(p<0.05) when the body condition score (BCS) of donors was in range of 2.50∼3.25(4.1) than in range of 3.50∼4.00(1.9), while the total number of ova was not changed. 3. The season affected on the collected number of freezable embryos(6.1 in winter, 4.5 in fall, 1.1∼1.5 in spring and summer, P<0.05), and the total number of obtained ova were more in winter than in other seasons(P<0.05). 4. Embryos were transferred to 343 recipients and 152 cows were confirmed pregnant(44.3%). 5. The higher pregnancy was obtained (P<0.05) when embryos were transferred in summer(53.3%) than in fall(36.0%), while the pregnancy rate was not affected by the origin and developmental stage of embryos, and the parity, BCS and estrus induction of recipients. From these results, the pregnancy rate was considered to be acceptable for the embryo transfer with domestic or imported Holstein embryos, however embryo production from superior Holstein donors was unsatisfactory for application of MOET scheme.
