압력구배가 변하는 표면 위의 Bypass 천이 유동의 예측 (Prediction of Bypass Transition Flow on Surface with Changing Pressure Gradient)

  • A modified $textsc{k}$-$\varepsilon$model is proposed for calculation of transitional boundary-layer flows with changing pressure gradient. In order to develop the model for this problem, the flow is divided into three regions; pre-transition region, transition region and fully turbulent region. The effect of pressure gradient is taken into account in stream-wise intermittency factor, which bridges the eddy-viscosity models in the pre-transition region and the fully turbulent region. From intermittency data in various flows, Narashima's intermittency function, F(${\gamma}$), has been found to be proportional to $\chi$$^{n}$ according to the extent of pressure gradient. Three empirical correlations of intermittency factor being analyzed, the best one was chosen to calculate three benchmark cases of bypass transition flows with different free-stream turbulence intensity under arbitrary pressure gradient. It was found that the variations of skin friction and shape factor as well as the profiles of mean velocity in the transition region were very satisfactorily predicted.

  • 한국형 B형 간염바이러스(HBV) DNA의 표면항원 유전자를 포유동물 세포에서 발현시켜 항원의 검출과 유전자의 분자유전학적인 연구를 하기 위하여 pre-surface antigen 지역과 표면항원 유전자 그리고 poly(A) addition site가 포함된 DNA 조각을 simian virus 40(SV 40)의 DNA 복제 원점과 promoter가 포함된 유전자 운반체에 재조함 시켰다. 우선, HBV DNA가 들어있는 pHBV 107을 Bam HI으로 부분절단한뒤 self-ligation시켜 두 HBV DNA가 같은 방향으로 들어간 pHBVD 107을 만들었다. 이 plasmid를 Bgl II로 절단하였을때 pre-surface지역과 표면항원 유전자 그리고 poly(A) addition site가 함께 포함된 2.7 kb의 insert DNA 조각을 얻었다. 유전자 운반체로는 포유동물세포에서 복제할 수 있도록 하기 위하여 SV40의 DNA 복제 원점부위와 72 bp repeats(enhancer)가 포함된 pSVOE를 만든다음 이 vector의 Pvu II 절단자리에 Bam HI linker를 붙여 insert DNA가 vector의 SV40 late promoter지역 가까이에 들어갈 수 있도록 변형시킨 pSVOB를 만들었다. 이상과 같이 만들어진 pre-surface 지역-표면항원유전자-poly(A)-addition site가 포함된 2.7 kb DNA 절편을 pSVOB promoter 뒤의 Bam HI site에 삽입하여 재조합된 plasmid pSVBS를 얻었다. 예비실험으로 pSVBS를 T-antigen이 생산되는 COS cell에 이주시켰더니 $HB_sAg$가 발현됨을 보았다.

  • In this study, we measured upper right, lower right, upper left region to find the optimal region which represent liver function. We found that the upper right region is optimal because of high accuracy and low standard deviation. For normal subjects all the calculated HEFs were greater than 100% and all those of abnormal ones were less than 80%. Thus HEF can be assumed to discriminate abnormal livers from normal ones. It was found that the patients with surgical operation would survive if both the pre-and the post-operative HEFs are greater than 50%. Therefore HEF method can be a good estimator for surgeon to determine the surgical operation and to evaluate remaining hepatic function after surgery as well as it is reliable and simple.

Neutron Cross Section Evaluation on Mo-95, Tc-99, Ru-101 and Rh-1()3 in the Fast Energy Region

  • The neutron induced nuclear data for Mo-95, Tc-99, Ru-101 and Rh-103 was calculated and evaluated in the fast energy region. The energy dependent optical model potential parameters were extracted based on the recent experimental data and applied up to 20 MeV. The s-wave strength function was calculated from the parameters. Spherical optical model, statistical model in equilibrium energy, multistep direct and multistep compound model in pre-equilibrium energy and direct capture model were used in the calculation. The theoretically calculated cross sections were compared with the experimental data and the evaluated files The model- calculated total and capture cross sections were in good agreement with the reference experimental data. The direct capture contribution improved the capture cross sections in pre- equilibrium region. The evaluated cross section results were compiled to ENDF-6 format and will improve the ENDF/B-Vl.

  • Equatorial Plasma Bubble (herafter, EPB) is a common feature in low-latitude F-region during the night time. Since EPB causes significant impacts on the satellite communication and navigation systems, its accurate forecast is highly demanded by the GNSS users. Thus, further understanding of these features and their configuration is a challenging issue in space weather studies. The day-to-day variability of the plasma bubble activity was investigated by analyzing the TIMED/GUVI, ROCSAT-1, DMSP, and CHAMP satellite data. The pre-reversal enhancement (PRE) is known as the most important single parameter for the onset of plasma bubbles but we do not know yet to what extent the day-to-day variability of the bubble activity can be attributed to the PRE. We obtained the magnitude of the PRE from ROCSAT-1 and the occurrence of bubbles in relation to the PRE was investigated by using the coincident observations of EPBs from TIMED/GUVI, DMSP, and CHAMP. By conducting one-to-one comparison of the PRE characteristics with the EPB occurrence we examined the role of the PRE in the onset of EPBs.

오스테나이트계 25Cr-20Ni 스테인리스강의 고온 예변형에 의한 크리프 거동 (Creep Behavior of High Temperature Prestrain in Austenitic 25Cr-20Ni Stainless Steels)

  • In the present study, we examined the influence of prestrain on creep strength of Class M alloy(STS310S) and Class A(STS310J1TB) alloys containing precipitates. Prestrain was given by prior creep at a higher stress than the following creep stresses. Creep behaviour before and after stress change and creep rate of pre-strained specimens were compared with that of virgin specimens. Pre-straining produced the strain region where the strain rate was lower than that of a virgin specimen both for STS310J1TB and STS310S steels. The reason for this phenomenon was ascribable to the viscous motion of dislocations, the interaction between dislocations and precipitates in a STS310J1TB steel, and the interaction of dislocations with sub-boundaries in a STS310S steen which has the higher dislocation density and smaller subgrain size resulted from pre-straining at higher stress.

  • One of the essential functions of virus surface proteins is the recognition of specific receptors on target cell membranes, and cellular receptors play an important role in viral pathogenesis. But the earliest steps of hepatitis B virus (HBV) infection, such as hepatocyte receptor interaction with the virus, are poorly understood. Previous work has suggested an important role of the preS1 region of HBV envelope protein in mediating viral binding to hepatocytes. Although hepatitis B virus (HBV) infection appears to be initiated by specific binding of virions to cell membrane structures via one or potentially several viral surface proteins, data showing the identification or isolation of the HBV receptor (s) are not yet available. The receptor-like proteins on the plasma membrane surface of HepG2 cells that bind to PreS1 were separated and identified using affinity chromatography, and the amino-terminal amino acid sequences of the receptor-like proteins were determined.

Affinity Maturation of an Anti-Hepatitis B Virus PreS1 Humanized Antibody by Phage Display

  • In a previous study we generated an anti-Hepatitis B Virus (HBV) preS1 humanized antibody (HzKR127) that showed in vivo HBV-neutralizing activity in chimpanzees. However, the antigen-binding affinity of the humanized antibody may not be sufficient for clinical use and thus affinity maturation is required for better therapeutic efficacy. In this study, phage display technique was employed to increase the affinity of HzKR127. All six amino acid residues (Glu95-Tyr96-Asp97-Glu98-Ala99-Tyr100) in the heavy (H) chain complementary-determining region 3 (HCDR3) of HzKR127 were randomized and phage-displayed single chain Fv (scFv) library was constructed. After three rounds of panning, 12 different clones exhibiting higher antigen-binding activity than the wild type ScFv were selected and their antigen-binding specificity for the preS1 confirmed. Subsequently, five ScFv clones were converted to whole IgG and subjected to affinity determination. The results showed that two clones (B3 and A19) exhibited an approximately 6 fold higher affinities than that of HzKR127. The affinity-matured humanized antibodies may be useful in anti-HBV immunotherapy.

How do diverse precipitation datasets perform in daily precipitation estimations over Africa?

  • Characterizing the performance of precipitation (hereafter PRE) products in estimating the uncertainties in daily PRE in the era of global warming is of great value to the ecosystem's sustainability and human survival. This study intercompares the performance of different PRE products (gauge-based, satellite and reanalysis) sourced from the Frequent Rainfall Observations on GridS (FROGS) database over diverse climate zones in Africa and identifies regions where they depict minimal uncertainties in order to build optimal maps as a guide for different climate users. This is achieved by utilizing various techniques, including the triple collection (TC) approach, to assess the capabilities and limitations of different PRE products over nine climatic zones over the continent. For daily scale analysis, the uncertainties in light PRE (0.1 5mm/day) are prevalent over most regions in Africa during the study duration (2001-2016). Estimating the occurrence of extreme PRE events based on daily PRE 90th percentile suggests that extreme PRE is mainly detected over central Africa (CAF) region and some coastal regions of west Africa (WAF) where the majority of uncorrected satellite products show good agreement. The detection of PRE days and non-PRE days based on categorical statistics suggests that a perfect POD/FAR score is unattainable irrespective of the product type. Daily PRE uncertainties determined based on quantitative metrics show that consistent, satisfactory performance is demonstrated by the IMERG products (uncorrected), ARCv2, CHIRPSv2, 3B42v7.0 and PERSIANN_CDRv1r1 (corrected), and GPCC, CPC_v1.0, and REGEN_ALL (gauge) during the study period. The optimal maps that show the classification of products in regions where they depict reliable performance can be recommended for various usage for different stakeholders.
