One of the major problems with sweetpotato (Ipomoea batats Lam.) is the tough thread tissue that occurs in the storage root, which has a negative impact on the sales of sweet potato because it impairs the texture during cooking and the processing quality. The fiber contents in storage roots of sweetpotato is affected by cultivation conditions and environment. To investigate the effect of fiber generation at different levels of irrigation, the sweetpoatoto "Hogammi" was transplanted in greenhouse. Sweetpotato was grown in styrofoam beds(W1605*D330*H300mm) to block moisture flowing from the outside. The irrigation was carried out as 3 levels (5,10, and 20 mm through drip irrigation facilities) at 20-day intervals. Five plants were harvested per plot at 90, 100 and 120 days after transplanting (DAT). The size of the storage root was large in the order of irrigation conditions 10mm>20mm>5mm treatment. And the longer cultivation period, the larger size of the storage root was observed. As a result of the analysis of the fiber content, it showed a tendency to decrease as the cultivation period increased (90days→120days). In addition, the fiber contents of sweetpotato harvested at 90, 100 and 120 DAT in the level of 5 mm irrigation plot were 351, 324 and 207 mg/100g, respectively, which were higher than those of other irrigation level plots. During all cultivation periods, the 10mm treatment group showed the lowest fiber content of 280, 228 and 127 mg/100g. At 20 mm irrigation level, the fiber content was less than that of 5mm irrigation level, but showed a tendency to increase compared to that of 10 mm irrigation level. These results suggested that drought stress or excessive-irrigation increases the fiber content of sweetpotato, which reduces their commercial value.

A comprehensive survey of mungbean-growing areas was conducted to observe leaf spot disease caused by Alternaria alternata. Alternaria leaf spot symptoms were observed on the leaves. Diversity of 50 genotypes of mungbean was assessed against A. alternata and data on pathological traits was subjected to cluster analysis. The results showed that genotypes of mungbean were grouped into four clusters based on resistance parameters under the influence of disease. The principal component biplot demonstrated that all the disease-related parameters (% disease incidence, % disease intensity, lesion area, and % of infection) were strongly correlated with each other. Alt a 1 gene that is precisely found in Alternaria species and is responsible for virulence and pathogenicity. Alt a 1 gene was amplified using gene specific primers. The isolated pathogen produced similar symptoms when inoculated on mungbean and tobacco. The sequence analysis of the internal transcribed spacer (ITS) region, a 600 bp fragment amplified using specific primers, ITS1 and ITS2 showed 100% identity with A. alternata. Potato virus X (PVX) -based silencing vector expressing Alt a 1 gene was constructed to control this pathogen through RNA interference in tobacco. Out of 50 inoculated plants, 9 showed delayed onset of disease. Furthermore, to confirm our findings at molecular level semi-quantitative reverse transcriptase polymerase chain reaction was used. Both phenotypic and molecular investigation indicated that RNAi induced through the VIGS vector was efficacious in resisting the pathogen in the model host, Tobacco (Nicotiana tabacum). To the best of our knowledge, this study has been reported for the first time.
Host plants of the Henosepilachna vigintioctomaculata are known as Solanaceae plants such as potato, tomato, eggplant and Lycium chinense. Henosepilachna vigintioctomaculata (Motschulsky) cause continuous damage from early June to mid-August in leaves of L. chinense under environmental-friendly management in Cheongyang, Chungnam. The developmental period of egg and larva of H. vigintioctomaculata were investigated. H. vigintioctomaculata were reared on L. chinense at several temperatures, for example, 15, 20, 25 and $30^{\circ}C$ in the laboratory. In addition, the control effect of 5 eco-friendly agricultural materials were examined. As results, hatching rate is the highest to 91% at $25^{\circ}C$. Egg periods were 10.8, 7.7, 5.1 and 3.7 days at each tested temperature, respectively. And the total developmental periods from egg to adult were 42.7, 26.3, 18.4 and 19.4 days at each tested temperature, respectively. Otherwise, their eclosion rates from pupa to adult were showed 58%, 77%, 85%, 65% at each tested temperature, respectively. As a result of the control effect, both adults and larvae of H. vigintioctomaculata were shown a high mortality to mixture with extract of Sophora flavescens, Chenopodium ambrosioides and Melia azedarach. Additional, larvae is shown high mortality to extract of Azadirachta indica and Cymbopogon nardus. Thus H. vigintioctomaculata might be controled by using plant extract of eco-friendly agricultural materials in due season.
Four species of Pythium and two species of Sclerotium previously not recorded in Korea during 1976 and Leptosphaeria salvinii which previosly reported but reidentified. Pythium aristosporm Vantery, Pythium sp. and Pythium myriotylum Drechsler were isolated from diseased rice seedlings and from green withered rice plants and Pythium irreglare Buisman was isolated from paddy soil. Three species of Pythium except P. irregulare grew well at $40^{\circ}C$ on Potato dextrose agar and were confirmed as highly pathogenic but P. irregulare showed low pathogenicity on Yushin variety of rice. Sclerotium hydrophilum and Sclerotium oryzae-sativae appeared to be weakly pathogenic, but Leptosphaeria salvinii was confirmed as a highly pathogenic. Ordinally the two species of Sclerotium grew and produced many sclerotia on dead sheath and stems of rice. There are still some problems to clarify and reconsider in regard to the pathogenicity of the sclerotial fungi because their populations were so very high in paddy fields, but their role might be wound parasite.
Survey of sesame crop in the cultivators' field in the district of Daejeon, Yuseong, Sintanjin, Nonsan and Keumsan revealed that Alternaria and Corynespora blights of sesame were wide spread and caused severe damage to sesame plants. Symptoms of a new disease of sesame in Korea caused by Alternaria sesami were spreading, dark-brown to black, water-soaked lesions which often could be traced the entire length of the stem. In severe infections, several lesions coalesced together involving a major portion of the blade and the infected leaves dried and usually dropped off. Symptoms of Corynespora blight, caused by Corynespora cassiicola, were irregular shaped, concentrically-zoned, light brown to reddish brown lesions which later coalesced and caused defoliation. Stem lesions were long, reddish brown streaks that often coalesced, blighting the plants. The optimum temperatures for mycelial growth of A. sesami and C. cassiicola were about $27^{\circ}C$ and sporulations of these 2 fungi were stimulated under alternating light and darkness. Mycelial growth and sporulation of A. sesami and C. cassiicola were the greatest on sesame oatmeal agar (SOA) and potato dextrose agar (PDA), respectively.
In April 2014, Fusarium wilt was found on kohlrabi seedlings in Iksan, Korea. Symptoms included wilting of foliage, drying and withering of older leaves, and stunting of the plants. The infected plants eventually died during growth. Colonies on potato dextrose agar were pinkish white, and felted with cottony and aerial mycelium. Macroconidia were falcate to almost straight, thin walled and usually 3-septate. Microconidia were usually formed abundantly in false-heads on short monophialides on the hyphae and were hyaline, smooth, oval to ellipsoidal, aseptate or medianly 1-septate, very occasionally 2-septate, slightly constricted at the septa, $4-11{\times}2.5-5{\mu}m$. On the basis of the morphological characteristics and phylogenetic analyses of molecular markers (internal transcribed spacer rDNA and translation elongation factor $1{\alpha}$), the fungus was identified as Fusarium oxysporum. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch's postulates. To our knowledge, this is the first report on the occurrence of Fusarium oxysporum on kohlrabi in Korea.
This study was conducted to dentify the optimum plantlet type of 'Solara' potatoes (Solanum tuberosum L.) for growth in an aeroponics system. Plantlets of 'Solara' were transplanted on March 16, 2015 in a greenhouse, and growth and yield characteristics were investigated at 70 and 78 days after transplanting, respectively. Stem length was shorter in plantlet of 15-day-old stem cuttings and acclimatization of culture, and the stem length of plantlets of stem cuttings tended to increase with increasing stem cutting age. The fresh weight of plants was the highest in the plantlets of 40-day-old stem cuttings and the lowest in non-rooted stem cuttings and acclimatization of culture. The highest number of first stolons was obtained in 35-day-old stem cuttings. The number of second stolons was the highest in plantlets of 35-day-old stem cuttings, acclimatization of culture, and 30- day-old stem cuttings. The total number of tubers was higher in plantlets of 35-day-old stem cuttings and acclimatization of culture, and the number of tubers above 3 g was the highest in plantlets of 35-day-old stem cuttings. The weight of tubers above 3 g was the heaviest in plantlets of 35-day-old stem cuttings(1,947 g per 10 plants), followed by plantlets of 30-day-old stem cuttings. These results indicate that plantlets of 30 to 35-day-old stem cuttings could be the best for production of 'Solara' potato tubers in an aeroponics system.

This study was conducted to evaluate five different strains of rhizobacterial isolates viz. PA1, PA2, PA4, PA5 and PA12 for biological control against Colletotrichum acutatum, C. coccodes, C. gloeosporioides, C. dematium, Botrytis cinerea, Rhizoctonia solani, Sclerotinia minor and Fusarium sp. In vitro inhibition assay was performed on three different growth mediums, potato dextrose agar (PDA), tryptic soy agar (TSA), and PDA-TSA (1:1 v/v) for the selection of potential antagonistic isolates. According to the result, isolate PA2 showed the highest inhibitory effect with 65.5% against C. coccodes on PDA and with 96.5% against S. minor on TSA. However, the same isolate showed the highest inhibition with 58.5% against C. acutatum on PDA-TSA. In addition, an in vivo experiment was performed to evaluate these bacterial isolates for biological control against fungal pathogens. Plants treated with bacteria were analyzed with phytopathogens and plants inoculated with phytopathogens were treated with isolates to determine the biological control effect against fungi. According to the result, all five isolates tested showed inhibitory effects against phytopathogens at various levels. Mode of action of these rhizobacterial isolates was evaluated with siderophore production, protease assay, chitinase assay and phosphate solubilizing assay. Bacterial isolates were identified by 16S rDNA sequencing, which showed that isolates PA1 and PA2 belong to Bacillus subtilis, whereas, PA4, PA5, and PA12 were identified as Bacilus altitudinis, Paenibacillus polymyxa and Bacillus amyloliquefaciens, respectively. Results of the current study suggest that rhizobacterial isolates can be used for the plant growth promoting rhizobacteria (PGPR) effect as well as for biological control of various phytopathogens.
From 2014 to 2016, Fusarium wilt disease was found on fassionfruit in Iksan and Jeju, Korea. Symptoms included wilting of foliage, drying and withering of leaves, and stunting of the plants. The infected plants eventually died during growth. Colonies on potato dextrose agar were pinkish white, and felted with cottony and aerial mycelia with 35 mm after one week. Macroconidia were falcate to almost straight, thin-walled and usually 2-3 septate. Microconidia were usually formed on monophialides of the hyphae and were hyaline, smooth, oval to ellipsoidal, aseptate or medianly 1-septate, very occasionally 2-septate, slightly constricted at the septa, $3-12{\times}2.5-6{\mu}m$. On the basis of the morphological characteristics and phylogenetic analyses of two molecular markers, internal transcribed spacer rDNA and translation elongation factor $1{\alpha}$, the fungus was identified as Fusarium oxysporum. Pathogenicity of a representative isolate was proved by artificial inoculation, fulfilling Koch's postulates. To our knowledge, this is the first report on the occurrence of F. oxysporum on fassionfruit in Korea.
Anthracnose symptoms were observed on the community area in Sancheong-gun, Gyeongnam Province in Korea. The symptoms occurred on leaf and stem, and then plants eventually were died. Colony color was whitish gray to dark gray on potato dextrose agar. Conidia were single celled . colorless, cylindrical and was 10.2~20.4$\times$3.6$\times$6.0$\mu$m in size. Appressoria were dark brown, ovate to obovate and 6.2~10.5$\times$4.3$\times$8.6$\mu$m in size. Perithecia were brown to black in color and shaped as globose to obpyriform and 72.6~284.7$\mu$m in size. Asci were clavate to cylindrical in shape and 63.6~98.8$\times$8.3~12.5$\mu$m in size. Ascospores were cylindrical. fusiform, slightly curved at the center and 9.6~21.8$\times$4.0~6.0$\mu$m in size. Optimum temperature fur growth was 3$0^{\circ}C$. On the basis of mycological characteristics and pathogenecity test on host plants, the fungus was identified as Glomerella cingulata . This is the first report on the Anthracnose of achyranthes caused by Glomerella cingulata in Korea.
