  • Axenic tissue culture of a marine red algae Porphyra yezoensis foma narawaensis was established for the vegetative propagation of tissues as a seed stock and for the development of a low salinity-tolerant cell line. Callus tissues have been induced from the vegetative area of blade away from the hold fast when grown on PES-agar medium. The brownish red fragile callus was maintained under fluorescent light of ca. 2000 lux with 12 : 12 hr L : D at $16^{\circ}C$. Amounts of carbohydrate and protein was determined against the weight of callus. Optimum temperature of the callus growth was $14^{\circ}C\~18^{\circ}C$. Optimun concentration of sodium chloride was $2.0\%$ for the callus growth in PES-agar medium.

Cryopreservation of Sporothalli of the Genus Porphyra (Bangiales, Rhodophyta) from Korea

  • Cryopreservation of sporothalli of the red alga Porphyra (P. seriata, P. yezoensis, P. tenera, P. pseudolinearis and P. dentata) was performed by the two-step cooling method in liquid nitrogen. The algal samples were suspended in various cryoprotective solutions, and slowly cooled to -40$^{\circ}C$ in 4 hours using a programmed freezer. After the first slow cooling the suspensions with cryoprotectants were immediately immersed in liquid nitrogen. The suspension from the programmed freezer was thawed quickly by agitation of the vial in a water bath at 40°C. When ice in the suspension of cryogenic vial was mostly melted, the vial was transferred to an ice bath for complete melting of the residual ice. The cryoprotectants in the vial were washed off by gradual dilution with seawater. The viability of the cell was assessed with neutral red staining. The viability of Porphyra samples ranged 54.6-70.9% when the mixed suspension of 10% dimethylsulfoxide and 0.5 M sorbitol in 50% seawater used as a cryoprotectant.

Anti-proliferating Effects and Gene Expression Profiles through Antioxidant Activity of Porphyra yezoensis Fractions on Human HepG2 Cell Lines (인간 간암세포주 HepG2에서 김 분획물의 항산화 활성을 통한 증식 억제 및 유전자 발현 양상)

  • In this study, the total polyphenol contents, antioxidant activities and anti-proliferation effects of HepG2 cell lines in organic slovent fractions obtained from the main methanolic extract of P. yezoensis were analyzed. The polyphenol content of the $CHCl_3$ fraction was $10.3{\mu}g/mg$, slightly less than $13.08{\mu}g/mg$ of the water fraction, but $ED_{50}$ estimated by measuring DPPH free radical scavenging activity exhibited the highest $16.96{\mu}g/ml$ in the $CHCl_3$ fraction. The proliferation effects of $CHCl_3$ and EtOAc fraction toward HepG2 cells inhibited in a dose-dependent manner, showed 90% inhibition when treated for 24 hr at $900{\mu}g/ml$ of $CHCl_3$ fraction. Meanwhile gene expression patterns in HepG2 cells treated $50{\mu}g/ml$ of $CHCl_3$ fraction were identified with microarray analysis. Concerning the efficacy of P. yezoensis, gene ontology analysis explored the genes associated with response to molecule of bacterial origin, vitamin D metabolic process, and response to nutrient. Thus IL6R, CYP1A1 were selected as significant genes based on expression patterns of HepG2 cells, and pathway analysis indicates that ARNT might be considered as a upstream regulator. Also, expression analysis of IL6R and CYP1A1, activity of upstream regulator ARNT in HepG2 cells was confirmed based on Western blotting analysis at the protein level after being treated with 50 and $100{\mu}g/ml$ of $CHCl_3$ fraction.

  • This study examined the effects of dietary glycoprotein extracted from Porphyra yezoensi on growth performance and resistance against the pathogenic bacteria Edwardsiella tarda in olive flounder. A porphyra-originated glycoprotein (P) was extracted using sequential processes of water and ethanol treatment. P extracts were added to a fish-meal-based diet at concentrations of 0.0, 0.5, and 1.0% (designated as Con, $P_{0.5}$, and $P_{1.0}$, respectively). Fish were fed one of the three experimental diets for 10 weeks. All fish groups exhibited over 96.7% survival during the experimental period. Results indicated that the fish fed diets containing P showed an increase in growth performance, including enhanced weight gain, specific growth rate, and feed efficiency. An increase in insulin-like growth factor (IGF-1) was observed in the fish fed the $P_{1.0}$ diet, as compared to those fed Con. At the end of the 10-week feeding trial, all fish were infected with E. tarda, and accumulated mortality was monitored for 8 days. Fish fed the Con diet exhibited increasing mortality from day 3 to the end of the challenge test, whereas the mortality of P-fed fish ceased at day 5. We suggest that supplementation with P-originated glycoprotein in aquafeed may increase growth performance and resistance against pathogenic bacteria in olive flounder juveniles.

Proximate Composition and Mineral Content of Laver Porphyra yezoensis from the Korean Coast (한국 연안산 방사무늬김(Porphyra yezoensis)의 일반성분 및 미네랄 함량)

  • To measure the proximate composition and mineral content of laver Porphyra yezoensis, we collected 30 raw and 30 dried laver from the major production area of the south coast of Korea (Busan, Goheung, Haenam, Wando). The approximate composition of a 100 g edible portion of raw laver was $89.9{\pm}1.4$ g moisture, $3.7{\pm}1.0$ g protein, $0.5{\pm}0.2$ g lipids, $2.1{\pm}1.8$ g carbohydrate and $3.9{\pm}0.6$ g ash. The approximate composition of a 100 g portion of dried laver was $8.9{\pm}1.6$ g moisture, $31.5{\pm}6.5$ g protein, $1.9{\pm}0.3$ g lipids, $48.4{\pm}6.5$ g carbohydrate and $9.3{\pm}1.1$ g ash. No clear regional variation in laver composition was observed. The mineral content of laver was expressed as dry weight. The mean macro mineral content per 100 g portion of raw laver was (in descending order): K ($1,979{\pm}863.0$ mg), Na ($1,063.2{\pm}498.8$ mg), P ($658.7{\pm}101.8$ mg), Mg ($432.3{\pm}83.5$ mg) and Ca ($394.2{\pm}136.5$ mg). In comparison, the mean micro mineral content of raw laver was (in descending order): Fe ($243.72{\pm}154.75\;{\mu}g/g$), Zn ($72.76{\pm}30.61\;{\mu}g/g$), Mn ($41.53{\pm}15.33\;{\mu}g/g$), Cu ($4.16{\pm}1.66\;{\mu}g/g$) and Ni ($0.43{\pm}0.70\;{\mu}g/g$) No clear regional variation in the mineral content of laver was observed; however, raw laver contained a higher mineral content than dried laver.

Effect of Porphyran isolated from Laver, Porphyra yezoensis, on Liver Lipid Peroxidation in Hyperlipidemic Rats and on Immunological Functions in Mice (김 다당류 porphyran의 급이가 흰쥐의 혈청과 간의 효소활성 및 마우스의 면역에 미치는 영향)

  • This study was carried out to investigate the effect of porphyran on enzyme activity in rats and immunity in mice. Animals were divided into 5 groups, and were given porphyran diet for 4 weeks. Porphyran was extracted from Porphyra yezoensis: Diet groups were normal diet, control diet fed high fat, cholesterol and sodium cholate, control and 1% porphyran diet (1% PD), control and 5% porphyran diet (5% PD), control and 10% of porphyran diet (10% PD). Also Balb/c female mouse were injected i.p. with porphyran extract every other day for 20 days at levels of 1%, 2% and 5%. Alanine aminotransferase (ALT), aspartate aminotransferase (AST), alkaline phosphatase (ALP) and lactate dehydrogenase (LDH) activities were lower in the porphyran diet group than those in control group. Superoxide dismutase and catalase activities in liver homogenates were reduced in porphyran diet group compared to those of control group. Also, the level of liver thiobarbituric acid reactive substance (TBARS) was lower in porphyran group than that of control group. Porphyran increased IL-1 production in a dose-dependent manner, however, interleukine-2 production was reduced as the amount of porphyran increases. These results showed that supplementation of porphyran lowered antioxidant enzyme activities and has possibility of modulating immunological function.

Comparative Analysis of the Amino Acid Composition and Phylogenetic Diversity of Five Seaweed Species (해조류 5종의 아미노산 조성 및 계통 다양성 비교 분석)

  • Seaweeds represent a widely harnessed marine resource that are valued for their abundant supply of essential nutrients, particularly proteins and amino acids. In Korea, where over 500 species of seaweed thrive and more than 50 are utilized for culinary purposes, seaweed has become a staple in regular diets. In this study, we focused on five of the most commonly consumed seaweed species in Korea: Capsosiphon fulvescens, Hizikia fusiforme, Porphyra yezoensis, Saccharina japonica, and Undaria pinnatifida. We closely examined the amino acid compositions of these five species. High-performance liquid chromatography showed that aspartic acid, glutamic acid, alanine, and leucine were the most abundant amino acids in the seaweeds. Principal component analysis revealed that the five seaweed species could be classified into three clusters according to their amino acid composition, partially corroborating findings from the phylogenetic analysis. Among various amino acids, glutamic acid, aspartic acid, and alanine were the primary amino acids driving differentiation. Notably, U. pinnatifida and C. fulvescens, which demonstrated close phylogenetic proximity, exhibited remarkably similar amino acid profiles. Conversely, although P. yezoensis and S. japonica shared a phylogenetic relationship, they displayed distinctly different amino acid compositions. H. fusiforme emerged as a distinct group in both analyses.

Effects of Salinity on Chlorophyll Fluorescence from Porphyra Thalli and Comparison of Species with Different Intertidal Distribution

  • Characteristic changes in chlorophyll fluorescence from thalli of red alga, Porphyra, under high salt stress and during subsequent recovery were investigated, and the differences in the sensitivity to the stress among four species of Porphyra with different intertidal distributions were compared. By the treatment of NaCl with $9\%$ or higher concentrations, photochemical efficiency (Fv/Fm) decreased in a biphasic pattern: a rapid decrease was observed within 1­3 h and followed by a slow decline. The decrease of Fv/Fm was mainly due to the increase of Fo without significant increases of Fm. When the thalli treated with $15\%$ NaCl for 6 h were returned to natural sea water for recovery, the increase of Fv/Fm also showed a biphasic pattern: a rapid increase of Fv/Fm was observed within 2 h and followed by a slow increase. Differences in the sensitivity to salt stress among the four species could be found during recovery after the treatment of severe salt stress. After the treatment of $20\%$ NaCl for 6 h, Fv/Fm decreased below 0.3 in all of the four Porphyra species, and the species living in upper parts of the intertidal zone (P. suborbiculata and P. pseudolinearis) could recover better compared with the species in lower parts of the intertidal zone (P. seriata and P. yezoensis), during recovery for 24 h. The species collected from the coast of the South Sea seemed to be more tolerant than those in the East Sea.
