우렁쉥이 육으로부터 Carotenoprotein의 분리.정제 및 특성 (Purification and Characterization of Carotenoprotein from the Muscle of Ascidian, Halocynthia roretzi)

  • 우렁쉥이의 육에 존재하는 carotenoprotein을 추출하여 이온교환수지 및 겔크로마토그래피를 이용하여 정제하였으며, 정제된 carotenoprotein의 분자량, pH 안정성, 열안정성, 변성제의 영향, 아미노산 조성 및 지방산 조성을 분석하였다. Carotenoprotein의 정제도는 5배, 수율은 2.9%였으며, 1 mg당 $0.15\;{\mu}g$ carotenoid가 함유되어 있었다. Carotenoprotein의 최대 흡수파장은 UV/Vis 스펙트럼에서 463 nm 및 439 nm였으며, 분자량은 64.4 kDa으로 단일 polypeptide로 이루어져 있었다. Carotenoprotein은 pH 7.0부근의 중성영역에서 안정하였고, 열안정성은$65^{\circ}C$에서 2시간 이내에 90%이상 유지하였으며, SDS 변성제에 의해 5 nm 단파장 영역으로 이동하였다. Carotenoprotein의 아미노산 조성은 Gly, hsn, Gln 및 Ser의 함량이 전체 아미노산의 35.3%로 전체적으로 산성의 성질을 보였으며, 구성지방산 조성의 함량은 monoene산이 45.2%로 가장 많았고, 다음으로 포화지방산 및 polyene산이 각각 38.1% 및 11.7%였다.

옻나무 사료의 급여수준과 기간이 비육돈의 냉장중 육질에 미치는 영향 (Effect of Feeding Levels and Periods of Dietary Rhus verniciflua Stokes at Finishing Stage on the Meat Quality of Pigs during Refrigerated Storage)

  • 비육돈에게 옻나무 사료의 첨가수준과 급여기간이 도축 후 육질에 미치는 영향을 구명하기 위해 본 실험을 실시하였다. 생시체중이 80kg과 65kg인 미경산돈(Landrace×Yorkshire×Duroc)을 도축전 5주(비육후기)와 8주(비육전기:3주; 비육후기:5주)동안 옻나무 사료를 0%(대조구), 2% 및 4%로 처리구당 5두씩 급여하였다. 도축후 24시간동안 냉각시킨 다음 발골한 등심(M. longissimus)을 3±0.2℃에서 8일 동안 저장하면서 실험을 실시하였다. 일반성분 함량에서 옻나무 사료를 4% 급여한 돈육이 대조구에 비해 조지방 함량이 감소하였다(p<0.05). 보수력은 옻 급여육이 대조구보다 높았다(p<0.05). 표면육색에서 명도(L* 값)는 저장기간 동안 옻나무 사료를 5주간 급여육이 대조구보다 높았으나(p<0.05), 적색도(a* 값)와 황색도(b* 값)는 옻나무 급여수준과 기간에 따른 영향을 받지 않았다. TBARS는 저장기간중에 옻나무 급여육이 대조구보다 낮았으며(p<0.05), 옻나무 사료를 5주간 급여육이 가장 높은 항산화 효과를 보였다. 옻나무 사료를 급여하면 돈육 지방의 불포화지방산이 증가하고 상대적으로 포화지방산이 줄어드는 것으로 나타났다. 단가 불포화지방산(MUFA)의 경우 옻나무 사료 급여량이나 기간에 상관없이 모든 옻 급여육이 대조구에 비해 유의적으로 높았고, 다가 불포화지방산(PUFA)은 8주간 옻나무 사료를 급여한 돈육에서 유의적으로 높게 나타났다(p<0.05). 반면 포화지방산의 경우 옻나무 사료를 5주간 급여한 돈육이 대조구보다 유의적으로 낮았다(p<0.05). 등지방의 융점은 옻나무 사료의 급여수준과 기간이 증가함에 따라 감소하였다(p<0.05). 따라서 옻나무 사료를 비육돈에게 급여하면 돈육의 지방 함량이 줄어드나 보수력 및 불포화지방산이 증가되며, 냉장기간중 지방산화가 억제되었다.

Growth Performance, Carcass Characteristics and Meat Quality of Boer-Cross Wether and Buck Goats Grazing Marshall Ryegrass

  • An experiment was conducted to evaluate the effects of castration on growth performance, carcass characteristics, and meat quality of goat kids. Fourteen Boer-cross buck and wether goat kids (n = 7; initial body weight (BW) $38.0{\pm}0.35\;kg$ and $34.8{\pm}0.35\;kg$, for bucks and wethers, respectively) were grazed on annual Marshall ryegrass (Lolium multiflorum Lam.) for 56 days. Body weights were recorded after 4 h withdrawal from feed and water for two consecutive days, every 2 wk. After d 56, animals were harvested and hot carcass weight (HCW), cold carcass weight (CCW), dressing percent (DP), kidney and pelvic fat (KPF), longissimus muscle (LM) area, back fat (BF), and other carcass parameters were measured. Day 0 BW was used as a covariate for analyses. However, bucks were heavier than wethers at d 15 (p = 0.09), 42 (p = 0.001) and 56 (p = 0.001). Bucks had higher ADG (146 vs. 74 g/d; p = 0.001), HCW (21.2 vs. 18.8 kg; p = 0.06) and CCW (20.3 vs. 17.9 kg; p = 0.04) when compared with wether goats. Dressing percentage (51 vs. 47%; p = 0.06), KPF (0.44 vs. 0.16%; p = 0.02) and BF (0.41 vs. 0.21 cm; p = 0.05) were higher in wethers vs bucks, respectively; however, USDA live or carcass grades were similar. Longissimus muscle tissue from wethers and bucks were similar in darkness ($L^*$) and redness ($a^*$), but wethers had more (p = 0.02) yellow tint ($b^*$). Palmitic (C16:0), stearic (C18:0) and oleic (C18:1) acids were higher (p = 0.001) in muscle tissue from wethers compared to bucks. The saturated fatty acid (SFA) and monounsaturated fatty acid (MUFA) contents of muscle tissue were lower (p = 0.001) for bucks with no difference in polyunsaturated fatty acids (PUFA). Longissimus muscle initial temperature was higher in bucks (p<0.04) and pH change post-mortem was similar for bucks and wethers. These results indicated that castration of young market goats reduced growth performance and produced carcasses with more fat and higher SFA.

Effects of Fermented Garlic Powder on Production Performance, Egg Quality, Blood Profiles and Fatty Acids Composition of Egg Yolk in Laying Hens

  • The effects of fermented garlic powder on production performance, egg quality, blood profiles and fatty acid composition of egg yolk in laying hens were studied in a 35-d experiment. Two-hundred and forty (ISA brown) layers aged 41 weeks were randomly allocated into the following four treatments: i) CON (basal diet); ii) G1 (CON+fermented garlic powder 1.0%); iii) G2 (CON+fermented garlic powder 2.0%) and iv) G3 (CON+fermented garlic powder 3.0%). There were no differences (p>0.05) among treatments in egg production, egg weight, eggshell breaking strength and eggshell thickness throughout the whole experimental period. However, yolk height was increased significantly (p<0.05) by the addition of fermented garlic powder during the 5th week while yolk color was greater (p<0.05) in G2 and G3 than in CON and G1 in the 5th week. Compared with CON, Haugh unit was increased (p<0.05) in response to fermented garlic powder treatments during the 5th week. No significant effects on total protein, albumin and IgG were observed in response to any of the treatments over the experimental period (p>0.05). There was a significant (p<0.05) reduction in plasma cholesterol concentration when the dietary level of fermented garlic powder was increased from 0.0 to 3.0%. The levels of saturated fatty acids (SFA) were significantly decreased (p<0.05) in response to G2 and G3 while monounsaturated fatty acids (MUFA) were higher (p<0.05) in G2 and G3 treatment groups than in CON and G1. Compared with other treatments, polyunsaturated fatty acids (PUFA) and PUFA:SFA ratio were higher (p<0.05) in G3. In conclusion, this study demonstrated that addition of fermented garlic powder reduced plasma cholesterol concentration and did not cause adverse effects on production performance. Moreover, addition of 3.0% garlic powder decreased SFA but increased PUFA and PUFA:SFA ratio in egg yolk.

고불포화 지방식이가 흰쥐의 Plasma High Density Lipoprotein Cholesterol 량과 혈청 및 조직 내의 지방성분에 미치는 영향 (Effects of Dietary Polyunsaturated Fat on HDL - Cholesterol, Total Cholesterol and Triglyceride in Plasma and Tissues of Adult Rats)

  • The study was designed to observe the effects of dietary intake of three different levels of polyunsaturated to saturated fatty acid (P/S) ratio on plasma HDL - cholesterol and triglyceride in plasma and tissues of adult rats. Rats were assigned into the three diet groups, composed of protein 24%, carbohydrate 46%, and fat 30% of total caloric intake. However, the P/S ratio of fat was adjusted by using beef tallow, corn oil and perilla oil to give 0.2, 1.1 and 6.0. All groups were fed ad libitum for 4 week. Plasma cholesterol level was decreased but net significantly with increasing P/S ratio of dietary fat, and showed significant negative correlation coefficient. Plasma TG level was also decreased with increasing PUFA level of dietary fat. However, the effect of P/S ratio (6.0) was not greater than that of P/S ratio (1.1) on plasma TG concentration. This implied that high level of PUFA was not required to change plasma TG level as much in the case of HDL-cholesterol. Plasma HDL-cholesterol was significantly increased only in the group of P/S ratio 6.0 which suggested that high level of dietary PUFA was required to increase HDL level. There was a decreasing trend in the levels of total cholesterol and TG per g liver with increasing P/S ratio of dietary fat. There was no effect in the levels of TG per g skeletal muscle by P/S ratio but the level of cholesterol per unit skeletal muscle was rather increased with increased P/S ratio.

어촌과 도시지역 여고생의 영양실태, 체조성 및 혈액성상에 관한 연구 (A Study on Nutrients Intakes, Body Composition and Hematological Status of High School Girls in Fishing and Urban Areas)

  • The purpose of this study was to examin height, weight and body composition nutrient intakes and serum lipids for high school girls in urban and fishing area. The subjects consisted of 78(44 fishing and 34 urban) high school girls aged in average 17.3 and 17 years. Their dietary intakes were assesed for 1 day by means of 24 hours dietary recall method. Concentrations of serum lipids, percentage of body fat and volume of total body water were measured. The mean energy intake of the subjects were 1431 ㎉ in fishing and 1659 ㎉ in urban area. The subjects in fishing area(urban) consumed 58(67) g protein, 27(36) g lipid, 459(634) mg calcium, 676(945) mg phosphorous, 11(14) mg iron, 786(574) RE retinol, 2.8(3.6) mg tocopherol, 0.9(1.0) mg thiamin, 1.2(1.1) mg riboflavin, 16.9(20.5) mg niacin and 64.1(92.8) mg ascorbic acid, respectively. Energy, calcium, iron and tocopherol intakes were lower than the Korean RDA in both girls. The mean percentage of fishing girls were 19.8% and significantly lower than 25.6% in urban. These values seem to fall in the desirable range in fishing girls, but high school girls in urban is seemed to overweight. The mean saturated fatty acid(SFA), monounsaturated fatty acid(MUFA), polyunsaturated fatty acid(PUFA) and dietary cholesterol(Chol) intakes were 7.3 g, 8.1 g, 7.2 g and 263 mg for fishing girls were 13.1 g, 12.1 g, 9.1 g and 216 mg for urban girls respectively. The fat, SFA and MUFA intakes in fishing girls were significantly lower than urban girls. The P/M/S ratio was 0.9/1.1/1 for fishing, was 0.7/0.9/1 for urban. The mean serum triglyceride(TG), total cholesterol, HDL cholesterol, LDL-cholesterol concentrations were 77.7(61.8) mg/dl, 124.5(142.3) mg/dl, 59.3(22.4) mg/dl and 49.6(107.8) mg/dl in fishing girls(urban girls), respectively. All of these values seem to fall in the desirable range but HDL-chlesterol concentrations in fishing girls were significantly higher than urban girls and LDL-cholesterol concentrations in fishing girls were significantly lower than urban girls. There were positive correlations between body fat percent and total cholesterol or LDL-cholesterol concentration : negative correlations between between body fat percent and HDL-cholesterol concentration : positive correlation between fat intake and LDL-cholesterol concentration : negative correlations between serum HDL-cholesterol concentration and fat or SFA or MUFA intakes. 진주담치 근육중의 적색 Carotenoprotein의 정제 및 특성 (Purification and Characterization of the Red Carotenoprotein from the Muscle of Blue Mussel, Mytilus edulis)

  • 우리나라에서 대량 생산되고 있는 진주담치 근육에서 Triton X-100를 사용하여 carotenoprotein을 추출, 정제하여 그 특성을 실험한 결과를 요약하면 다음과 같다. Carotenoprotein(${\lambda}_{max}=462nm$)를 $20\%$(w/v) $(NH_4)_2SO_4$, DEAE-cellulose 이온교환수지 그리고 Sephacryl S-300 겔 크로마토그래피를 통해 정제한 결과, 정제도는 5배 증가하였고 수율은 $1.85\%$였으며, carotenoprotein의 분자량은 약 372KDa(gel filtration)이었고, SDS-PAG 전기영동으로 단백질의 subunit를 확인한 결과, 60KDa($23.70\%$), 46.9KDa($9.14\%$), 26KDa($49.14\%$) 및 13KDa($18.02\%$)로 이루어져 있었다. Carotenoprotein의 안정성은 pH 4.0 이하의 강산 및 pH 10.0 이상의 강알칼리에서는 단백질이 불안정하였고, SDS 변성제를 $0.2\%$(w/v)되게 첨가하여 녹인 후 가시부 영역($400{\sim}600nm$)에서 최대파장 변화를 보면 462nm에서 456nm로 약 6nm 단파장 영역으로 이동한 것을 알 수 있었다. Carotenoprotein의 구성아미노산 조성은 threonine이 $35.90\%$로 다른 아미노산에 비해 함량이 높았으며, 필수아미노산과 소수성 아미노산의 함량은 전체아미노산 함량의 각각 $62.76\%$$25.02\%$를 차지하였고, methionine, histidine 및 proline은 존재하지 않았다. Carotenoprotein 1g당 총지질의 함량은 256mg였고, 그 구성지방산의 조성은 polyene산이 $62.4\%$로 가장 많았고 다음으로 포화산($19.58\%$)및 monoene산($18\%$) 순이었다. 특히, 16:4n-3의 조성비가 $33.90\%$로 상당히 높게 나타났고, EPA, DHA 등 w3 계의 장쇄(長鎖)고도불포화지방산의 조성비가 $22.13\%$로 높게 나타났으며, 양적으로 많은 지방산은 포화산 중에는 palmitic acid($C_{16:0}$), monoene산 중에서는 palmitoleic acid($C_{16:1}$), polyene산 중에서는 hexadecatetraenoic acid($C_{16:4}$), eicosapentaenoic acid ($C_{20:5}$), docosahexaenoic acid($C_{22:6}$) 등이 함량이 많았다. Carotenoprotein으로부터 분리한 총 caroteoid 함량은 $8.80mg\%$였고, 주 carotenoid는 mytiloxanthin($74.79\%$), 3,4,3'-trihydroxy-7',8'-didehydro-${\beta}$-carotene($18.26\%$)였다.

감마선과 Phosphine 처리가 백삼의 지방질 관련 성분에 미치는 영향 (Comparative Effects of Gamma Irradiation and Phosphine Fumigation on Lipid-Related Components of White Ginseng During Post-Treatment Period)

  • 백삼의 생물학적 품질개선이 가능한 phosphine gas와 감마선 처리(5 kGy 이하)가 저장시료의 지방질 관련 성분에 미치는 영향을 비교 검토하였다. 시료의 pH는 gas 처리에 따라 다소 염기성을 나타내었으며, 상온조건$(20^{\circ}C/70%\;RH)$ 및 가혹조건$(40^{\circ}C/90%\;RH)$에 6개월 간 포장 저장 중 전반적으로 다소 감소되었다. 가혹조건에 저장된 시료는 저장초기부터 갈변현상이 심하게 발생되었으나 상온 저장된 시료에서는 거의 갈변되지 않았고 처리구 간에도 차이가 크지 않았다. 시료의 갈변현상의 발현은 저장 4개월 경까지 추출물의 수소공여능과 비례하였다. 시료 지방질 TBA가는 감마선 조사시 다소 증가되었으나 저장기간의 경과로 점차 유사한 값을 보였으며, 카보닐가는 점차 증가되는 경향이었다. 시료의 지방산 조성은 가스나 5 kGy 감마선 처리에 의해 변화되지 않았으며, 저장 6개월 이후에는 불포화지방산조성의 감소와 포화지방산조성의 증가 현상이 부분적으로 확인되었다.

경기 일부지역 대학생의 섬유소 섭취 행동단계에 따른 영양소 섭취상태 비교 (Comparison of Nutrient Intakes Regarding Stages of Change in Dietary Fiber Increasing for College Students in Kyunggi-Do)

  • This study was conducted to compare nutrient intakes regarding stages of change in dietary fiber increasing behavior. Subjects were consisted of healthy 383 college students (2S0 females and 133 males) in Kyunggi-Do. Stages of change classified by an algorithm based on 6 items were designed each subjects into one of the 5 stages: precontemplation (PC), contemplation (CO), preparation (PR), action (AC), maintenance (MA). Nutrient intakes were assessed by 24-hr recall method. Regarding the S stages of changes, PR stage comprised the largest group $(39.4\%)$, followed by AC $(33.7\%)$, MA$(14.6\%)$, PC$(7.6\%)$, CO$(34.7\%)$. Female were more belong to either AC or MA. The higher stage of change in dietary fiber increasing behavior, the higher self-efficacy. In all male and female, there were no differences in energy, protein, monounsaturated fatty acids, polyunsaturated fatty acids and cholesterol intakes across the 5 stages. But, fiber, postassuim (K), vitamin A and vitamin C intakes of AC or MA were higer than those of PC, CO and PR $Energy\%$ from fat of $PR(25.4\~26.5\%)$ was higher than $20\%$, and those of AC and MA was lower than the other groups. Dietary P/S and ${\varepsilon}6/{\varepsilon}$ 3 ratios of AC and MA were similar to the recommended ratio. Female of PR had the most total saturated fat and palmitic acid and those of MA had the least. Male of PR had the least $\alpha-LNA\;({\varepsilon}3)$ and total ${\varepsilon}3$ fatty acids and those of MA had the most. In male and female in AC or MA, fiber and K intakes from breakfast, dinner and snack and vitamin C intakes from all meals were higher than those of the other stages. These results of our study confirm differences in stages of change in fiber intake in terms of nutritional status. To have lower $energy\%$ from fat, higher intakes of K, fiber and vitamins, desirable ratio of dietary fatty acids, it needs consistent nutritional education leading to the AC or MA of fiber increasing behavior.

Effects of Dietary Peroxidizability Index Values on Hepatic TBARS and Antioxidant Enzyme Activity in 7,12-dimethylbenz[$\alpha$]anthracene-treated Rats

  • Breast cancer may be the consequence of free radical damage, which is partially caused by the excessive intake of dietary fat and imbalances in antioxidant scavenger system;. In this experiment, we examined! the effects of dietary peroxidizability index (PI) values on hepatic thiobmbituric acid reaction substances (TBARS) and antioxidant enzyme activities in rats treated with 7,12-dimethylbenz[$\alpha$]anthracene (DMBA). Female Sprague-Dawley rats were used and 7,12-DMBA (20 mg/kg body weight) was gastrically intubated at seven weeks of age in order to induce mammary tumors (MT). The levels of dietary PI were 36, 81, 126 and 217 (LPI, MLPI, MHPI and HPI), while dietary polyunsaturated/saturated fatty acids ratio was maintained at the same level (1.0). Fat used in the experiment was mixed with soybean oil, com oil, palm oil, perilla oil, sesame oil, fish oil, and beef tallow. Experimental diets were given for the following 20 weeks. We measured tumor numbers and weights, and then assayed the hepatic TBARS levels and antioxidant enzyme activities such as superoxide dismutase (SOD), catalase, glutathione peroxidase, glutathione-S-transferase (GST) and glutathione reductase (GR). The incidence of Mr was the lowest in the MHPI group. The hepatic TBARS level was significantly raised with increasing dietary PI value. The hepatic SOD and GR activities were differed significantly by dietary PI value. The hepatic SOD activity was negatively correlated with dietary PI value and GR activity was the highest in the rats fed the MHPI diet. When the dietary P/S ratio is kept at 1.0, adequate dietary PI value (PI value of 126) may reduce the incidence and growth of Mr, but this benefit may be lost with higher dietary PI value. These results suggest that the awareness of dietary PI values may help to decrease breast cancer incidence and growth.
