Structural Characteristics of Immunostimulating Polysaccharides from Lentinus edodes

  • There is a significant amount of experimental evidence suggesting that polysaccharides from mushrooms enhance the host immune system by activating various mechanisms in immune cells, including macrophages. In this study, polysaccharides from Lentinus edodes were found to stimulate the functional activation of macrophages to secrete inflammatory mediators and cytokines and increase the phagocytotic uptake. The chemical properties of the stimulatory polysaccharides, CPFN-G-I, CPBN-G, and CPBA-G, were determined based on their monosaccharide composition, which mainly consisted of glucose and mannose. According to FT-IR and GC/MS, the structure of CPFN-G-I, purified from the fruiting body of L. edodes, was found to consist of a $\beta$-1,6-branched-$\beta$-1,4-glucan, whereas CPBN-G and CPBA-G, purified from the liquid culture broth, were found to be composed of a heteromannan. The configuration of the p-linkage and triple helical conformation of each polysaccharide were confirmed using a Fungi-Fluor kit and Congo red, respectively.

  • To develop potent anitumor polysac-charides from higher hungi of Korea the alcohol precipitated polysaccharides, which were obtained from hot water extracts of shake-cultured mycelia of eight species of basidiomycetes, were subjected to antitumor screening tests, using sarcoma 180 as a tumor cell. When administered i. p, at the dose level of 20mg/kg/day for ten days into the mice which were implanted with $1{times}10^{8}$ cells of sarcoma 180 twenty-four hour before the start of polysaccharide injection, the polysaccharides of Plutcus cervinus ISA-Pc-1004, Pleurotus pulmonarius, Hydnum repandum ISA-Hr-1006, and Laccaria laccata ISA-LI-1008 respectively showed inhibition ratios of 62.0%, 51.7%, 64.8%, and 57.8%. They were selected for further study, while those of Pancillus serotina ISA-Ps-1012, Laccaria amethystina ISA-La 1001, Pholiota adiposa ISA-Pa-1010, and Lepista spISA-Ls-1007 respectively showed inhibition ratios of -9.3%, 25.3%, 27.3% and 33.0%.

Effects of Mushroom Protein -bound Polysaccharides on Blood Glucose Levels and Energy Metabolism in Streptozotocin-Induced Diabetic Rats (버섯 단백다당체의 당뇨 유발 흰쥐의 혈당수준과 에너지원 조성에 미치는 영향)

  • The hypoglycemic effects of 2 mushrooms, Pleurotus ostreatus and Lentinus edodes, on streptozotocin(STZ) induced diabetic rats were investigated in this study . Diabets mellitus was induced in male Sprague-Dawley rats by the injection of STZinto the tail vein at a dose of 45mg/kg. Sprague-Dawley male rats(200-250g) were assigned to one control and three STZ-diabetic groups. Diabetic groups were assigned to STZ-control, pleurotus ostreatus and Lenitinus edodes groups. All groups were fed a AIN 76 diet. The two experimental groups were fed with each protein-bound polysaccharide(150mg/kg BW) for 14 days and with carboxymethyl cellulose for STZ-control group. The body weight gain was monitored and the blood levels of glucose and cholesterol were measured . Levels of protein triglyceride, and free fatty acid in plasma were analysed. Serum aminotransferase activity as also measured. The body weight gain was lower in the all diabetic groups than in the of normal group. The weight of spleen was reduced by adminstration of the Lentinus edodes protein-bound polysaccharides. The result suggest that orally administered Lentinus edodes protein-bound polysaccharides exhibited hypoglycemic effect in STZ -induced diabetic rats and that these protein-bound polysaccharides may be useful for the management of diabetes mellitus.

Antioxidant Properties of Different Polysaccharides Extracted with Water and Sodium Hydroxide from Rice Bran

  • The antioxidant properties of water extracted polysaccharides (PW), 1%(w/v) NaOH extracted polysaccharides (PN1), and 5%(w/v) NaOH extracted polysaccharides (PN5) were assessed in this paper. PW showed good capability of scavenging $H_2O_2$, anti-lipid peroxidation, reduction power, and scavenging 1,1-diphenyl-2-picrylhydrazyl (DPPH). The maximum values were all observed at the dose of 1 mg/mL sample. As far as the antioxidant activities of PN1 were concerned, the capability of scavenging superoxide radical, chelating metal, and total antioxidation showed higher than those of PN5. When polysaccharide concentration was 1 mg/mL, the 3 index described above were attached 71.8%, 51.6%, and $446.3{\mu}M$ Trolox equivalent, respectively. With respect to PN5, higher capabilities of scavenging hydroxyl free radicals were obtained while the peak of 84.8% was observed at the concentration of 1.0 mg/mL. Moreover, the concentration-dependent influences were characterized in all cases.

Purification, Chemical Composition, and in vitro Antioxidant Activity of Two Protein-bound Polysaccharides from Rapeseed Meal

  • Crude polysaccharides from rapeseed meal (PRM) were extracted with 0.3% NaOH aqueous solution, followed by further purifications and 2 fractions, namely PRM1 and PRM2, were separated with a DEAE-cellulose DE-52 column. Their primary compositions were analysed and antioxidant activity was determined, including scavenging activity toward superoxide anion radicals, hydroxyl radicals, and nitric oxide radicals, reducing power, and inhibitory effects against the microsomal lipid peroxidation, compared to that of L-ascorbic acid. The results indicated that PRM1 and PRM2 exhibited not only good reducing power and inhibitory effects on the microsomal lipid peroxidation, but also strong scavenging activity toward superoxide anion radicals, nitric oxide radicals, and hydroxyl radicals. In addition, positive correlations were also observed between the superoxide anion radical scavenging activity and the protein contents of the polysaccharides, and the reducing power and the sulfate contents. These findings thus clearly suggest the polysaccharides possess direct and potent antioxidant activity.

Preparation of Heparinoids from Acidic Plant Polysaccharides (식물성 산성당으로부터 헤파리노이드의 제조)

  • Anticoagulant activities were tested for the fifteen kinds of medicinal plants by measuring activated partial thromboplastin time (aPTT). Of them five kinds or species (Artemisia princeps, Sanguisorba officinalis, Artemisia apiacea, Eclipa alba, Schizonepeta tenuifolia) were selected and fractionated for the preparation of acidic polysaccharides. They were extracted with water by refluxing and the extracts were precipitated with ethanol. The precipitates were separated based on charge using a DEAE-Sephadex. The low salt and high salt fractions were sulfated with anhydrous pyridine and chlorosulfonic acid complex. In vitro anticoagulant activities of sulfated polysaccharides were tested by measuring aPTT, prothrombin time (PT), and factor Xa clotting time using normal human plasma. No relationship was found between the amount of uronic acids and anticoagulant activities, but the sulfated ones show the increase of activities. In vivo anticoagulant properties of the sulfated polysaccharide from Artemisia apiacea were also tested by the intraveneous administration of three different doses (3,5 and 10 mg/kg) to rats. APTT and PT were increased significantly and the action of factor Xa and thrombin mediated through antithrombin III were inhibited slightly.

Polysaccharide Extraction and Comparison of Free Radical Scavenging Activities from Tremella fuciformis and Auricularia auricula Fruit Body (흰목이와 흑목이 버섯의 다당추출 및 유리라디칼 소거활성 비교)

  • The polysaccharides from fruit body of Auricularia auricula and Tremella fuciformis were extracted using hot water, and partially purified through ethanol precipitation and dialysis. Free radical scavenging activities of the crude and purified polysaccharides were examined and compared each other. Free radical scavenging activities of the partially purified polysaccharides were higher than those of crude polysaccharides. DPPH free radical, ABTS radical and SOD-like activities of partially purified polysaccharide at 1 mg/mL of concentration from A. auricula were 61.7, 9.6 and 38.9%, respectively, while those of T. fuciformis were 9.6, 5.7 and 15.3%, respectively. Results of site and non-site specific hydroxyl radical scavenging activities indicated that the partially purified polysaccharide fractions from A. auricula and T. fuciformis exhibited the hydroxyl radical scavenging effect by hydrogen donating ability and iron ion chelating ability. Also, reducing powers of A. auricula and T. fuciformis were 77.1 and 14.7% of BHT (0.1%) as standard, respectively. It was suggested that antioxidant activities of A. auricula were about 1.4~6.4 times higher than those of T. fuciformis due to different levels of polyphenol content.

Improvement of Inflammation, Diabetes, and Obesity by Forest Product-Derived Polysaccharides through the Human Intestinal Microbiota

  • The intestinal microbiota plays a crucial role in determining human health, rendering it a major focus of scientific investigation. Rather than eliminating all microbes, promoting the proliferation of beneficial microorganisms within the gut has been recognized as a more effective approach to improving health. Unfavorable conditions potentially alter gut microbial populations, including a reduction in microbial diversity. However, intentionally enhancing the abundance of beneficial gut microbes can restore a state of optimal health. Polysaccharides are widely acknowledged for their potential to improve the gut microbiota. This review emphasizes the findings of recent studies examining the effects of forest product-derived polysaccharides on enhancing the gut microbiota and alleviating inflammation, diabetes symptoms, and obesity. The findings of several studies reviewed in this paper strongly suggest that forest products serve as an excellent dietary source for improving the gut microbiota and potentially offer valuable dietary interventions for chronic health problems, such as inflammation, diabetes, and obesity.

A Review of the Relationship between the Theory of 'Pizhuweiwei(脾主爲衛)' and Intestinal Flora ('비주위위(脾主爲衛)'와 장내(腸內) 세균총(細菌叢)의 상관성(相關性)에 대한 고찰(考察))

  • Objectives : This paper focuses on recent research related to the relationship between 'Pizhuweiwei(脾主爲衛)' and intestinal flora, and suggests Korean Medical treatment methods that can treat and prevent diseases related to Spleen Deficiency(脾虛) such as ulcerative colitis, atrophic gastritis, diabetes, and obesity that is prevalent today. Methods : This study summarizes recent research results based on various literature on the relationship between the spleen and intestinal flora. Results : Polysaccharides contained in Jianpi herbs(健脾藥) have the effect of increasing beneficial bacteria and maintaining the diversity of intestinal microorganisms to improve intestinal function, managing intestinal metabolites to improve the body's immune function, and regulating the intestinal immune defense system. Therefore, based on the theory of 'Pizhuweiwei(脾主爲衛)', if the symptoms are treated through the spleen with Spleen-strengthening herbal medicinals, it could provide a substantial starting point for improving immunity. Conclusions : Polysaccharides contained in Jianpi herbs(健脾藥) could be considered as potential probiotics based on research findings which show that polysaccharides can regulate the intestinal flora and strengthen weak spleen, playing an important role.

Effects of Mono- and Polysaccharides on In Vitro Fertility of Boar Spermatozoa

  • This study was conducted to examine the effect of several saccharides on the induction of capacitation and acrosome reaction (AR) and to examine the effects of mono and polysaccharides on the penetration activity of boar spermatozoa. Spermatozoa were inseminated in medium with fucose, galactose and mannose as monosaccharide, and fucoicIan. galactan and marman as polysaccharide. The penetration rates were significantly (p<0.05) lower in medium with galactose (40.6%), mannose (38.1%), fucose (41.6%) and fucoidan (36.6%) compared with control (56.7%). The rates of AR were increased (40.7 to 59.8%) by the preincubation periods prolonged from 0 to 4 hr (p<0.05). Similar tendencies were observed in AR when spermatozoa were treated with monosaccharides, but not significantly differ among the groups treated with different time of preincubation with some exception of galactose. When spermatozoa were treated with polysaccharides, the rates of AR were significantly (p<0.05) increased by preincubation time prolonged from 0 to 4 hr with an exception of fucoidan. In conclusion, the present study suggests that penetration rate of spermatozoa is higher in presence of polysaccharides than monosaccharides. Also, it may resume that the comparing to control, the all saccharides (L-fucose, D-galactose, D-mannose, fucoidan. galactan and mannan)-treated groups slightly increase the AR pattern as preincubation time prolonged.
