Exo-Polysaccharide Production from Liquid Culture of Lentinus edodes (Lentinus edodes 액체배양을 통한 세포외 다당체 생산)

  • The optimum liquid culture conditions were investigated for cell growth and polysaccharide production from liquid culture of Lentinus edodes. In flask culture, the optimal medium compositions for the polysaccharide production contained glucose 60 g/L, yeast extract 10 g/L, $KH_2PO_4$ 2.0 g/L, and $MgSO_4{\cdot}7H_2O$ 1.0 g/L. The maximum mycelial growth and polysaccharide production were 11.01 g/L and 1.64 g/L, respectively. In bioreactor, through the variation of aeration in order to increase mycelial growth and polysaccharide production, the maximum mycelial growth and polysaccharide production were 55.9 g/L at 8th day and 7.34 g/L at 7th day of cultivation with 1.5 vvm, respectively.

  • Cytoprotective ability of polysaccharides isolated from different edible mushrooms was investigated on the 7-ketocholesterol-induced damaged cell line. Polysaccharide extracts from six different edible mushrooms-Flammulina velutipes, Peurotus ostreatus, Lentinus edodes, Agrocybe aegerita, Agaricus blazei, and Cordyceps militaris- were prepared by hot water extraction and alcohol precipitation. Cytoprotective ability was evaluated by measuring the viable cells of the normal embryonic liver cell line (BNL CL. 2) in the presence of 7-ketocholesterol. At $80\;{\mu}g/mL$ of 7-ketocholesterol, cytotoxicity was very high with a loss of 98% of viable cells after 20 h of incubation. With the addition of $200\;{\mu}g/mL$ of each polysaccharide isolate to the cell line containing $80\;{\mu}g/mL$ of 7-ketocholesterol, polysaccharide isolates from both Flammulina velutipes and Peurotus ostreatus could significantly inhibit the 7-ketochoelsterol-induced cytotoxicity in the cells. But other polysaccharide isolates were not effective in inhibiting cell damage caused by the oxLDL-induced cytotoxicity.

  • The polysaccharide has been known to have an antitumor activity, which were extracted from the fruiting bodies, mycelia, and culture broths of Agaricus blazei. For the cell growth and the polysaccharide production, the optimal medium contained 8% glucose, 2% yeast extract, 0.1% $MgSO_4{\cdot}7H_2O$, 0.2% $KH_2PO_4$, 0.2% $CaCl_2{\cdot}2H_2O$ and 0.2% $MnSO_4{\cdot}5H_2O$. When 0.2% of glutamic acid was added at 4day, the cell concentration was 13.5 g/L and the polysaccharide production was 9.9 g/L, respectively.

  • Identification of escherchia coli K1 polysaccharide antigen isolated from urine specimens of urinary tract infections in children were performed from of 1992 to 1993 in Kyoto, Japan. The serotypes of E. coli were categorized that O1:H7, O2:H6, O2:H7, O16:H6, O18:H7, O18:H ̄, and O135:H44 among 14 strains isolated from urine specimens of urinary tract infections in children by the serological test. And, one strain (O18:H ̄, isolation rate: 7.1%) of E. coli K1 polysaccharide antigen among 14 strains were isolated from urine specimens of urinary tract infections in children by the bacteriophage test.

  • It is well known that the numbers and functions of immune-associated cells are increased by saponins and polysaccharides in ginseng. In this study, the mixture of polysaccharide and saponin (MPS) from Panax ginseng is applied to LPS- activated RAW 264.7 cells. The production of NO and the gene expression of IL-6 and TNF-$\alpha$ are decreased in LPSactivated RAW 264.7 cells and the expression of arginase II and PD-1L genes is decreased in LPS-untreated macrophages. Therefore, the mixture of saponin and polysaccharide from Panax ginseng could be used in order to regulate immune responses.

  • The environmental and physiological factors affecting the production of exopolysaccharide (Methylan) and Poly-${\beta}$-hydroxybutyrate(PHB) by Methylobacterium organophilum were investigated. The maximum PHB content was obtained at $38^{\circ}C$ whereas maximum polysaccharide concentration was $3.54g/\ell$ at $30^{\circ}C$. Optimum pH was pH 7-8 for PHB production and pH 6-7 for polysaccharide production, respectively. Under the condition of $Mo^{2+}, Mg^{2+} or Mn^{2+}$ limitation with nitrogenlimitation, the PHB accumulation was increased, whereas the polysaccharide production was decreased as compared with that of solenitrogenlimitation. Under the condition of sole K+ limitation, cell growth was significantly inhibited and no polysaccharide was produced. However, the PHB content was as high as 60% of dry cell weight. Effect of C/N ratios (methanol/ammonium) in the feeding solution was examined for the simultaneous production of polysaccharide and PHB. The higher ratio of C/N showed the lower cell growth, higher content of PHB in cells, and higher yield of polysaccharide.

  • A bacterial isolate FRI-33 which produces hydrophillic polysaccharide was identified and its cultural condition was investigated. FRI-33 was identified as Enterobacter agglomerans. The optimum cultural conditions for polysaccharide production were 3$0^{\circ}C$, pH 5.7, using medium composed of glucose 25 g/$\ell$, peptone 2.0 g/$\ell$, yeast extract 0.5 g/$\ell$, KH$_2$PO$_4$ 1.0g/$\ell$, MgSO$_4$.7H$_2$ O 1.0g/$\ell$, CaCO$_3$ 2.5g/$\ell$. The polysaccharide production after 72 hours was 8.41 g/$\ell$. The polysaccharide was composed of galactose (1.0 mole), xylose (1.5 mole), gluconodeltalactone (1.9 mole) and ribose (0.03 mole). The apparent viscosity of 1% polysaccharide solution was 504 mPa.s at 60 rpm and intrinsic viscosity was 45.80 d$\ell$/ g.

Effects of Culture Conditions on Mycelial Growth and Polysaccharide Production of Tricholoma matsutake in Bioreactor

  • This experiment was carried out to obtain the optimal liquid culture conditions for the mycelial growth and the polysaccharide production of Tricholoma matsutake. For the mycelial growth and polysaccharide production, the synthetic medium was optimized with containing glucose 40 g/L, yeast extract 30 g/L, $KH_2PO_4$ 1.5 g/L and $MgSO_4.7H_2O$ 1 g/L. The effects of agitation and aeration were investigated for the cell growth and the polysaccharide production in batch culture. The biomass and polysaccharide concentrations were 21.87 g/L at 150 rpm and 8.86 g/L at 300 rpm, respectively. And the biomass concentration and the polysaccharide production were 20.85 g/L at 0.5 vvm and 8.83 g/L at 1.5 vvm, respectively.

The Functional Effects of Anti-microbial Activity and Anti-inflammatory of Seaweed polysaccharide Extracts (해조 다당류 추출물의 항균성 및 항염증 기능성평가)

  • This study aimed to evaluate seaweed polysaccharide extracts as a cosmetic material. To assess anti-microbial efficacy, Staphylococcus aureus (S. aureus) was treated with seaweed polysaccharide extracts and zones of inhibition were measured. In addition, the anti-inflammatory effect was confirmed in RAW 264.7cells, and seaweed polysaccharide extracts was applied to the dorsal skin of Sprague-Dawley (SD) rats to evaluate single-dose toxicity. As a results, seaweed polysaccharide extracts did not exhibit cytotoxicity at concentrations up to $1,000{\mu}g/mL$ in skin fibroblasts. Furthermore, when S. aureus was treated with 1% seaweed polysaccharide extracts, clear zones of $1.52{\pm}0.34cm$ formed, confirming sufficient anti-microbial activity. When RAW 264.7 cells were treated with seaweed polysaccharide extracts extract, nitric oxide (NO) production decreased in a concentration-dependent manner and the production of inflammation-related cytokines, such as interleukin 1 beta ($IL1{\beta}$), tumor necrosis factor alpha ($TNF{\alpha}$), and prostaglandin E2(PGE2), decreased. When seaweed polysaccharide extracts extract was applied at various concentrations to rats, symptoms did not change for more than 14 d, and there was no change in body or organ weights. In conclusion we found that seaweed polysaccharide extracts is not cytotoxic and has anti-microbial and anti-inflammatory effects. Therefore, it is suitable for use as a cosmetic material.

Immune Enhancing Effects of Intracellular and Extracellular Polysaccharides Extracted from Mycelial Cultivate of Agaricus blazei Murill (신령버섯(Agaricus blazei Murill) 균사체내외 다당체의 면역활성효과)

  • This study was performed to compare in vitro immune enhancing effects of polysaccharides extracted from cultivated mycelia of Agaricus blazei Murill. Carbohydrate contents of semi-purified polysaccharides were 85.6% and 95.3%, while ${\beta}$-glucan conents were 67.9% and 88.1% for intracellular and extracellular polysaccharide, respectively. Samples were adjusted to the same in their carbohydrate contents before efficacy tests. Both intracellular and extracellular polysaccharide increased nitric oxide (NO) synthesis of macrophage RAW 264.7 in dose dependent manner, and the maximum increase rate was 53.9 and 53.1% in intracellular and extraceltular polysaccharide, respectively. The polysaccharides also increased synthesis of cytokines such as interleukin (IL)-$1{\beta}$, IL-6 and tumor necrosis factor (TNF)-${\alpha}$ in RAW 264.7. For all the 3 cytokines, the increase rate of synthesis was much higher in extracellular polysaccharide compared to intracellular polysaccharide, especially at low concentration. Both polysaccarides increased the proliferation of splenocytes in vitro, intracellular polysaccharide showed increase in dose dependent manner while extraceltular polysaccharide showed increase untill medium concentration ($250\;{\mu}g/ml$). They did not show direct cytotoxicity against cancer cells such as B16F0 melanoma. As results, it was regarded that the both intracellular and extracellular polysaccharide from A. blazei showed immune enhancing effects in vitro, but the activity is higher in extracellular polysaccharide compared to intracellular polysaccharide.
