Enzymatic Characteristics of Polyphenol Oxidase from Apple (Ralls Janet) (사과(Ralls Janet) Polyphenol Oxidase의 효소학적(酵素學的) 성질(性質))

  • In order to obtain elementary data of enzymatic browning of apples and apple products and to examine effectively inhibitory method of browning, we extracted polyphenol oxidase (EC 1.10.3.1) from apple (Ralls Janet) and investigated its general properties. The optimum conditions for the enzyme reaction were pH 6.0 and temperature of $30^{\circ}C$. The enzyme was very stable at pH 4.0, and at the range of pH 5.0-9.0 its activity was above 80% compared with pH 4.0. The enzyme was very stable by heating at $40^{\circ}C$ for 1 hour, and almost 50% of enzyme activity was lost by heating at $60^{\circ}C$ for 30 minutes. The polyphenol oxidase activity was enhanced by the addition of $Cu^{2+}$ and $Mn^{2+}$, respectively, meanwhile $Na^+$, $Hg^{2+}$ and $Co^{2+}$ inhibited the enzyme activity. The enzyme activity was greatly decreased in the presence of inhibitors such as cysteine, sodium metabisulfite and ascorbic acid. The polyphenol oxidase greatly catalyzed the oxidation of o-diphenols such as chlorogenic acid and catechol, which suggests that main substrate of polyphenol oxidase is o-diphenol compounds.

Induction of Extracellular Polyphenol Oxidase from Two White-rot Fungi (목재(木材) 부후균(腐朽菌)의 목질소(木質素) 분해효소(分解酵素) 유도(誘導)에 관하여)

  • Among the representative phenolic compounds in relation to lignin derivatives and protein synthesis inhibitors, the most effective inducer for the extracellular polyphenol oxidase (PO) of Lentinus edodes JA01 was gallic acid and ferulic acid for Pleurotus ostreatus. Optimum concentration of these inducers was 2.0 mM and 1.0 mM, respectively. Addition of gallic acid after two days culture had the best effect on production of PO enzyme of L. edodes JA01 and for P. ostreatus, and addition of ferulic acid after three days culture had the best effect. Also, in case of L. edodes JA01, polyphenol oxidase activity was parallel to growth curve, whereas the maximum enzyme activity of P. ostreatus was shown at exponential growth phase and declined thereafter.

Total Polyphenol, Flavonoid Contents and Antioxidant Activities of Dolsan Leaf Mustard Pickle during Storage (저장 기간 중 돌산갓 피클의 폴리페놀, 플라보노이드 및 항산화 활성에 대한 변화)

  • Dolsan leaf mustard has various biological functions, including those in the immune system and cancer prevention. It contains high amounts of nutritional and medicinal compounds, which are important for maintaining optimum health. The objective of this study was to determine the total phenolics contents and antioxidant activity in Dolsan leaf mustard pickle (DLMP) during storage. DLMP methanol extracts had the highest total polyphenol and flavonoid contents of $35.56{\pm}0.01GAEmg/g$ and $4.54{\pm}0.03QEmg/g$, respectively. The DPPH and ABTS radical scavenging activities in the DLMP methanol extracts showed the highest activities of 79.4 and 85.36%, respectively. The ferric reducing antioxidant power (FRAP) assay showed 2.24-3.82 mM $FeSO_4$ eq. (p<0.05) in DLMP extracts. Overall, storage at day 14 showed the highest antioxidant activity.

Antioxdant Activity of Hibiscus cannabinus L. Leaves in Different Growth Time (생육시기에 따른 양마 잎의 항산화 활성)

  • The results on the useful functional compounds of kenaf (Hibiscus cannabinus L.) leaves cultivated in reclaimed lands and the biological activity effects of extracts were as follows. On 98 days after sowing (DAS) Tainung-2 showed the highest total chlorophyll content (1.68 mg/g), and on 141 DAS Dowling showed higher chlorophyll content (1.50 mg/g) than the other two did. In all cultivars the total chlorophyll content was higher on 141 DAS than on 98 DAS. Total polyphenol and total flavonoid contents were the highest in Tainung-2 (30.50 mg/g and 57.03 mg/g, respectively), and total polyphenol and total flavonoid contents (30.50 mg/g and 57.03 mg/g, respectively) were the highest in 30% ethanol extraction. Ascorbic acid contents were higher on 141 DAS than on 98 DAS in three cultivars. SOD activities of kenaf leaf extract were generally over 90%. DPPH radical scavenging activity of Tainung-2 was higher than others.

Bioactivity of Trifoliate Orange (Poncirus trifoliate) Seed Extracts

  • Trifoliate orange seed extracts (TSEs) were made using either distilled water (TW), ethanol (TE), or n-hexane (TH), to measure total polyphenol contents, DPPH and ABTS radical scavenging activities, and anti-complementary activity. The results showed that the total polyphenol content showed higher value at TE (235.24 ${\mu}g/mL$, p<0.05) than those of TW (132.65 ${\mu}g/mL$) and TH (165.44 ${\mu}g/mL$) at 10 mg/mL and TE exerted the highest DPPH radical scavenging activity (61.77%, p<0.05), which occurred in the following order: TE TW (56.87%)>TH (39.78%). The results of ABTS radical scavenging activity showed that TW (34.26%) and TE (31.81%) showed similar activities, which were higher than TH (12.74%, p<0.05). Anti-complementary activity of TE (61% at 500 ${\mu}g/mL$) showed a higher activity when compared with the positive control (60% at 1,000 ${\mu}g/mL$) polysaccharide-K (PSK), a known immuno-active polysaccharide from Coriolus versicolor. Consequently, among TSEs, TE is a byproduct from trifoliate orange and could be an important source of dietary polyphenolic antioxidant compounds and immunopotentiating activity, including complement activation.

Total polyphenol and ferulic acid analysis of a new variety of corn, Bandiburichodang, according to steaming time and roasting temperature

  • Bandiburichodang (BDC) is a new variety of Zea mays L. Total polyphenol content (TPC) assay and quantitative analysis of ferulic acid (FA) were performed to determine the steaming, roasting conditions of BDC kernels that lead to the highest content. TPC levels increased after roasting under all conditions. TPC levels in samples steamed at 115 ℃ for 25 min were 3.157 mg/g before roasted, and increased to 3.825 and 4.739 mg/g after roasting at 160 and 200 ℃, respectively. Whether BDC kernels were roasted was relevant with TPC content. BDC kernels were extracted to perform quantitative analysis of FA. Roasting temperature affected FA content: the higher the temperature, the lower the content. BDC kernels that were steamed at 115 ℃ for 25 min had 0.178 mg/g of FA content before roasting, and levels decreased to 0.132 and 0.115 mg/g after roasting. Under different roasting conditions, FA content decreased 15 to 50%. We hypothesize that this phenomenon is due to a breakdown of phenolic compounds or cell wall disruption.

  • Polyphenol oxidase in apple (Golden Delicious) was extracted, partially purified and its properties were found as follows; Polyphenol oxidase showed optimum pH for activity at 6.5 and optimum temperature at $30^{\circ}C$ and high affinity to o-diphenol compounds. Cysteine, ascorbic acid and sodium metabisulfite appeared to be most effective inhibitors. EDTA showed a slight inhibition. During the enzyme was kept in test tube at $4^{\circ}C\;and\;20^{\circ}C$ for a week, polyphenol oxidase activity decreased sharply during the first four days at $20^{\circ}C$, then decreased slowly as the storage was prolonged. At $4^{\circ}C$, the polyphenol oxidase activity appeared to be relatively stable during the first two days before activity began to decline sharply. Polyacrylamide disc gel electrophoresis indicated four bands with polyphenol oxidase activity. Three bands and one band of the active bands were observed after heating for 1hr at $60^{\circ}C\;and\;70^{\circ}C$ respectively. The enzyme activity was observed 40% after treatment at $60^{\circ}C$ and 5% after treatment at $70^{\circ}C$. Therefore, no difference in the thermal stability was observed between active bands and the enzyme activity.

  • To search for antioxidant compounds from plant resources, ethanol extracts of 10 plants species were investigated using DPPH method and SOD activity by NBT method. The highest activity was shown in the ethanol extracts of Schizandra Chinesis Baillon. And than, antioxidant effects and total polyphenol concentration were investigated in solvent fraction of ethanol extracts from Schizandra Chinesis Baillon. The DPPH inhibitory activity and SOD activity of ethyl acetate fraction (DPPH 51.6%, SOD 66.2%) and butanol fraction(DPPH 60.7%, SOD 67.4%) showed strong biological activities.

Antioxidant Activities of Agrimonia pilosa Ledeb: In Vitro Comparative Activities of Its Different Fractions

  • The antioxidant potentials of the methanol extract and its various fractions from Agrimonia pilosa Ledeb were measured as 1,1-diphenyl-2-picrylhydrazyl (DPPH), and 2,2'-azinobis-(3-ethylbenzothiazoline-6-sulphonic acid) (ABTS) radical scavenging abilities as well as the reducing power decreasing NO in vitro. The methanol extract of Agrimonia pilosa Ledeb showed significantly strong scavenging effects on free radicals of DPPH, ABTS, and NO. Hexane fraction (HF), ethyl acetate fraction (EF) and n-butyl alcohol fraction (BF) were prepared by solvent fractionation. By comparison of polyphenol contents among BF and HF, EF with high polyphenol contents showed the highest DPPH, ABTS, and NO scavenging effects with the value of 50.2%, 80.4%, and 65.7%, respectively, at the highest tested dose. HPLC analysis revealed the presence of various phenolic compounds in EF of Agrimonia pilosa Ledeb. These results suggest that EF could be considered as natural antioxidant sources and dietary nutritional supplements to prevent oxidation-related diseases.
