한국산 배 (Pyrus pyrifolia Nakai)로부터 polyphenol 화합물의 구조결정 (Chemical Structure of Polyphenol Isolated from Korean Pear (Pyrus pyrifolia Nakai))

  • 한국산 배를 60% acetone으로 추출하여 Sephadex LH-20 gel column chromatography, MCI-CHP 20 gel column chromatography, Bondapack $C_{18}$ gel column chromatography을 이용하여 TLC와 HPLC로 순도를 검증한 후 4종의 polyphenol 화합물을 분리, 정제하였다. Compound A와 B는 Sepadex LH-20 gel column chromatography에서 증류수상에서 용출되었고 compound C는 40% methanol상에서 용출됨을 보아 compound B와 compound C는 흡착성이 강한 polyphenol 화합물이라 추정되었다. 분리, 정제한 4종의 compound를 NMR, FAM-mass 및 FT-IR를 이용하여 화학구조를 결정한 결과 compound A는 (+)-catechin, compound B는 (+)-gallocatechin, compound C는 (-)-epigallocatechin이고 compound D는 procyanidin B-3-3-o-gallate로 밝혀졌다.

한국산 인삼의 polyphenol 화합물의 생리활성 효과 (Identification of Biologically Active Compounds from Panax ginseng C. A. Meyer)

  • 한국산 인삼 5년근을 60% acetone으로 추출하여 Sepadex LH-20 gel column chromatography, MCI-CHP 20 gel column chromatography, Bondapak $C_{18}$, column chromatography을 이용하여 TLC와 HPLC로 순도를 검증한 후 3 종의 polyphenol 화합물을 분리하였다. Compound I과 II는 Sepadex LH-20 gel column chromatography에서 증류수상에서 용출되었고 compound Ⅲ는 40% methanol 상에서 용출됨을 보아 compound II와 compound III는 흡착성이 강한 polyphenol 화합물이라 추정되었다. 혈압강하에 관여하는 효소인 angiotensin converting enzyme(ACE)의 저해효과는 compound II가 157 ppm에서 31.86%로 3 가지 분획물 중 가장 우수하였으며 compound I과 III는 높은 활성을 보이지 않았다. 통풍예방에 관여하는 xanthine oxidase 저해효과는 compound I, II에서 666 ppm에서 100%저해효과를 나타내었으나 compound III는 저해효과가 낮았다. Melanin을 형성하는 tyrosinase 저해효과를 실함한 결과 compound III는 400 ppm에서 28.6%의 저해효과를 나타내었고 Compound I과 II의 저해효과는 낮았다.

Cacao bean으로부터 분리된 polyphenol 성분의 화학구조분석과 ACE 저해효과 (The chemical structure of polyphenols isolated from cacao bean and their inhibitory effect on ACE)

  • Ghana산 cacao bean으로부터 acetone으로 추출하고 Sephadex LH-20, MCI-gel CHP-20, Bondapak $C_{18}$ 및 Fuji gel ODS $G_3$, chromatography 등을 이용하여 7종의 polyphenol 화합물(compound 1 - compound 7)을 분리 정제하였고, 이들 화합물의 화학구조를 $^1H-NMR$, $^{13}C-NMR$, IR 및 MS를 사용하여 검색하였고 아울러 angiotensin converting enzyme(ACE) 저해효과를 조사 검토하였다. 분리정제한 7종의 polyphenol 화합물의 구조를 분석한 결과, compound 1: [(+)-catechin], compound 2: [(-)-epicatechin], compound 3: [procyanidin B-1, (-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(+)-catechin], compound 4 : [procyanidin B-2, (-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin], compound 5: [procyanidin B-7, (-)-epicatechin-$(4{\beta}{\rightarrow}6)$-(+)-catechin], compound 6 : (procyanidin B-2,3,3'-O-digallate) 및 compound 7: [cinnamtannin A-2, (-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin-$(4{\beta}{\rightarrow}8)$-(-)-epicatechin]임을 동정하였다. ACE의 저해효과는 procyanidin B-2,3,3'-O-digallate (compound 6)가 $100\;{\um}M$에서 94.6%로 매우 우수하였으며, (+)-catechin (compound 1), (-)-epicate -chin (compound 2), procyanidin B류 (compound 3, 4, 5) 및 cinnamtannin A-2 (compound 7)도 각각 67.9%, 61.9%, 88.6%, 82.5%, 72.2% 및 82.3%의 비교적 우수한 저해효과가 있었다. 아울러 결합방식에서는 $4{\beta}{\rightarrow}6$보다 $4{\beta}{\rightarrow}8$의 결합방식이, 그리고 procyanidin 류에서는 gallate를 갖는 물질이 이를 함유하지 많은 물질보다 더욱 높은 ACE의 저해효과를 나타내었으며, 또한 hydroxyl 기가 많을수록 효소 저해효과도 증가하는 것으로 나타났다. 이와 같은 결과는 chocolate의 주원료인 cacao bean의 polyphenol 성분 또한 녹차 등에서 볼 수 있는 생리활성효과에 손색없는 것으로 판단되고, 이러한 기능성에 기초하여 chocolate, 음료 등의 식품이나 의약품의 기능성 소재로서의 산업적 응용 가능성이 높은 것으로 사료된다.

  • 배로부터 polyphenol 화합물군을 분리 및 정제하여5주간 고지방식이를 한 후, 혈장 및 간장에서의 지질대사에 미치는 영향을 분석한 결과 혈장에서는 배의 polyphenol 화합물군 PR-II에서 총지질, 총콜레스테롤이 유의성 있게 감소하였다 또한 간장에서는 총지질, 중성지질이 대조군에 비해서 유의성 있는 결과가 나타났다 5주 동안 고지방 식이를 한 후 분변을 채취하여 총지질, 총콜레스테롤 및 중성지질을 측정한 결과 배에서 추출한 polyphenol화합물 군이 고지방식이만을 섭취시킨 대조군에 비해 다소 감소되는 경향을 보였다. 배의 polyphenol 화합물군을 섭취시킨 후 총단백질과 알부민 함량을 측정한 결과 대조군과 실험군에서 유의성 있는 차이는 나타나지 않았다.

흑마늘 첨가 김치 양념소의 항산화 활성 (Antioxidant Activity of Kimchi Seasoning with Black Garlic)

  • To enhance the physiological activities and reduce the off-flavor of garlic in the Kimchi, we manufactured the Kimchi seasoning replaced with 25~75% black garlic instead of raw garlic and measured the contents of antioxidants and antioxidant activities. The polyphenol compound content was $270.91{\mu}g/mL$ in the control and $278.18{\sim}305.75{\mu}g/mL$ in the Kimchi seasoning with black garlic. The polyphenol compound content increased by increasing the amount of black garlic in the Kimchi seasoning. The polyphenol compound contents gradually decreased during storage period. The flavonoid compound content was 26.18 mg% in the control and 26.80~38.72 mg% in the Kimchi seasoning replaced with 25~75% black garlic. The flavonoid compound contents increased by increasing the amount of black garlic. The flavonoid contents were different from those of polyphenol compounds during storage periods. In the case of DPPH electron donating abilities, DPPH electron donating ability of the control was 84.82%, which showed a lower antioxidative activity than the Kimchi seasoning replaced with black garlic. The electron donating abilities of the Kimchi seasoning replaced with black garlic were increased as the amount of black garlic was increased. In the lipid peroxidation test, the control showed no lipid peroxidation during the storage period. The Kimchi seasoning replaced with black garlic also inhibited lipid peroxidation during storage as in the control. However, it showed lower activity than the control. Reducing power was higher in the Kimchi seasoning replaced with black garlic than in the control. The higher the ratio of black garlic, the higher the reducing power.

고초균에서 폴리페놀로 유도된 DNA 손상에 대한 폴리페놀산화효소의 억제효과 (The Inhibitory Effect of Polyphenol Oxidase on Polyphenol-Induced DNA Damage of Bacillus subtilis)

  • Antimutagenic activity of the enzymatic browning reaction products (EBRPs) was investigated by using the spore rec-assay with Bacillus subtilis strains H17 $(rec^+)\;and\;M45 (rec^-)$. The EBRPs tested were prepared from the reactions of five different kinds of polyphenols with polyphenol oxidase isolated from the leaves Perilla frutescens. In the spore rec-assay, most of the polyphenolic compounds tested showed positive, whereas only their tested compound showed negative respectively. In addition of polyphenol oxidase inhibitors such as cysteine, glutathione and ascorbic acid to the reaction mixtures consisted with the polyphenol oxidase and polyphenols, the mutagenic effects were increased in the spore recassay. These results show that the activity of polyphenol oxidase may play an important role in the reduction of mutagenicity of polyphenols.

TLC에 의한 한국산 인삼과 서양상 페놀성 성분의 비교 (Comparison of Phenolic Components between Korean and American ginsengs by Thin-Layer Chromatography)

  • The distribution of phenolic components of Korean ginseng (Panax ginseng KG) and American ginseng (Panax quinquefolium, AG) were compared by thin-layer chromatography (TLC). Silica gel TLC gave 3~4 spots, while $NH_2$ HPTLC 5~6 spots, which were colored by both $FeCl_3$/$K_3Fe(CN)_6$ and Folin-Ciocalteu. The distribution of phenolic components was quite different between KG and AG. Especially, a polyphenol (m.w. 578), which had been isolated from KG by the author, was not found in AG. This result suggests that the polyphenol could be used as an index compound for the differentiation of KG from AG.

소리쟁이 뿌리 열수 추출물의 폴리페놀 함량 및 추출물의 항산화 효과 (Polyphenol Contents of Rumex crispus Root Extract with Hot Water and its Antioxidative Effect)

  • This study was conducted to investigate the extraction characteristics including total polyphenol compound content (TPC) and the antioxidative effect of Rumex crispus root extract on commercial corn oil. Extraction yield was increased with extraction temperature but decreased with extraction ratio. No significant differences in aromatics content were found among the extracts prepared with various extraction conditions; extraction ratio, extraction temperature, extraction pH and composition of extractant. Total flavonoid content of the extract was increased with extraction temperature and extraction pH, and highest value of it was found when extractant composition of ethanol in water was 75%. Total polyphenol compound content (TPC) of the extract with 10 fold extraction ratio showed the highest value, but no significant difference in TPC was found with extraction temperature. Reducing power and DPPH (${\alpha}$,${\alpha}$-diphenyl-${\beta}$-picrylhydrazyl) radical scavenging ability (RSA) of the extract in spectrophotometric absorbance were decreased with extraction ratio but increased with extraction temperature and showed 63.1%~98.4% and 67.6%~86.7% of those of ascorbic acid in reducing power and DPPH RSA, respectively. The antioxidative effects of the extract on corn oil were 84.8~93.0% of that of commercial butylated hydroxytoluene, the antioxidant index value was highest when the ethanol composition to water in extractant was equal ratio.

Anti-Allergic and Anti-Asthmatic Activity of Helioscopinin-A, a Polyphenol Compound, Isolated from Euphorbia helioscopia

  • During the course of searching for anti-allergic substances from unexplored plant sources, an inhibitor of leukotriene $D_4$-induced tracheal contraction was isolated from Euphoribia helioscopia. This isolated polyphenol compound, known as helioscopinin-A, showed a certain inhibitory activity on capillary permeability in passive cutaneous anaphylaxis responses of rats and also on antigen-induced bronchial constriction in an experimental asthma model of guinea pigs. The compound at a high concentration weakly inhibited histamine release from isolated mast cells of rats. It is suggested that this compound is an anti-allergic and anti-asthmatic which exerts its activity through antagonism on leukotrene $D_4$-induced responses. A partial inhibition of allergic mediator relase may also bee involved.

오배자와 적포도과피 폴리페놀 분획물의 항균성 및 Glucosyltransferase 저해효과 (Effect of Inhibition on Glucosultransferase and Antimicrobial Activity of Polyphenol Fraction of Gallnut and Red Grape Husk)

  • The antimicrobial and glucosyltransferase(GTase) inhibition activity were searched for 30 species of various folk drugs and by products of food industry. Among them, two species, gallnut and red grape husk water extracts, were selected for the powerful antimicrobial and GTase inhibition activity. The polyphenol fractions of gallnut and red grape husk were showed very greater antimicrobial activity on both Gram(+) and (-), B. subtilis and E. coli. The minimum antimicrobial activity of gallnut polyphenol fraction were 1.0mg for B. subtilis and 3,0mg for E. coli. Red grape husk was 2.0mg for B. subtilis and 3.0mg for E coli. The polyphenol fractions of gall nut and red grape husk were showed powerful GTase inhibition activity. The concentrations of these fractions for 80% inhibition of GTase activity were 1.08$\times$10$\^$-3/mg/㎖ and 1.08$\times$10$\^$-2/mg/㎖, respectively. The most abundant compound in these fraction seems to be polyphenol derivatives. From these results, we think that the gallnut and the red grape husk polyphenol fraction had more antimicrobial and anti-plaque activities than artificial synthetic preservatives as an economic point of view.
