고추(Capsicum annuum L.) 배양세포의 Elicitor 유도성 Phytoalexin 생성 (Elicitor-InduciblePhytoalexin from Cell Suspension Cultures of Pepper(Capsicum annuum L.))

  • Extracellular capsidiol, sesquiterpenoid phytoalexin, in the medium of pepper (Capsicum annuum L.) suspension cells was not identified from control cells, but highly accumulated in the elicitor-induced cells within 6 hours after the addition of 0.05$\mu\textrm{g}$/$m\ell$ cellulase. Capsidiol production in elicitor-induced cells was markedly suppressed by cytochrome P450 inhibitors, such as ancymidol and ketoconazole demonstrating that biosynthesis of capsidiol is catalyzed by at least on hydroxylation enzyme in the biochemical pathway. Based on protein electrophoresis, two bands, 23.0kDa and 27.5kDa, were identified as newly synthesized polypeptides in the elicitor-induced suspension cells, suggesting that pepper cells which were subjected to elicitor treatment activate specific gene(s) for capsidiol biosynthesis in cultured cells.

  • Aqueous fractions from Korean red ginseng inhibited the phosphorylations of 40 KD and 20 KD polypeptides which were induced by phorbol-12-myristate-13-acetate (100 nM) in human platelets. Much more carbohydrates were contained in the aqueous fractions than proteins. An aqueous fraction extracted with methanol, mainly, consists of glycoproteins, molecular weights of which were below 18 KD. We may infer that the aqueous fraction from Korean red ginseng do antitumorous and antiplatelet functions.

복수암 생쥐에 대한 발효녹용의 항암작용 (Antitumor Activity of Fermented Antler on Sarcoma 180 in Mice)

  • Some components and antitumor effect of antler and fermented antler with Bacillus P-92 were determined. By fermenting the antler with Bacillus P-92, the amount of free amino acids, polypeptides and other compounds were increased. The free amino acid of the fermented antler was 6-fold higher than those of the untreated antler. Sarcoma 180 cells were intraperitoneally implanted into ICR mice to determine the effect of fermented antler and untreated antler on life span prolongation. The life span of ICR mouse treated with antler and fermented antler were prolonged to 24.4% and 39.2%(p<0.05), and survival rates were 22% and 33%, respectively. In addition, the induction of neutrophil was 2.3-fold and 3.4-fold, respectively.

Improvement in the Stability of Glycinecin A through Protein Fusion of the Two Structural Components

  • Glycinecin A, a bacteriocin produced by Xanthomonas campestris pv. glycines, inhibits the growth of X. c. pv. vesicatoria. We have reported that purified glycinecin A is composed of two polypeptides, is active over a wide range of pH (6 to 9), and is stable at temperatures up to 60$\^{C}$. Glycinecin A is a heterodimer consisting of 39- and 14-kDa subunits; the two encoding genes, glyA and glyB, respectively, have been cloned (Heu et al. 2001. Appl. Environ. Microbiol. 67, 4105-4110). Co-expression of glyA and glyB in the same cell is essential for bacteriocin activity. We constructed and produced a chimeric glycinecin A connecting glyA and glyB in one open reading frame. The chimeric glycinecin A has the same bactericidal activity as the wild-type glycinecin A. However, the chimeric glycinecin A is more stable in a wider range of pH and temperature.

When a ribosome encounters a premature termination codon

  • In mammalian cells, aberrant transcripts harboring a premature termination codon (PTC) can be generated by abnormal or inefficient biogenesis of mRNAs or by somatic mutation. Truncated polypeptides synthesized from these aberrant transcripts could be toxic to normal cellular functions. However, mammalian cells have evolved sophisticated mechanisms for monitoring the quality of mRNAs. The faulty transcripts harboring PTC are subject to nonsense-mediated mRNA decay (NMD), nonsense-mediated translational repression (NMTR), nonsense-associated alternative splicing (NAS), or nonsense-mediated transcriptional gene silencing (NMTGS). In this review, we briefly outline the molecular characteristics of each pathway and suggest mRNA quality control mechanisms as a means to regulate normal gene expression.

The Effects of 20-hydroxyecdysone in Drosophila Kc Cells on the Ecdysteroidosis

  • Drosophila Kc cells are ecdysone-responsive : hormone treatment leads rapidly to increased synthesis of several ecdysone-inducible polypeptides (EIPs) and to commitment to eventual proloferative arrest. Later the treated cells undergo morphological transformation, cease to proliferate and to grow. These responses have proven useful as models for studying ecdysone action and environmental endocrine disrupting actions. In this study, we used 20-HE to check out the Kc cells properties to the ecdysone and this properties will be applied to the environmental chemicals to find out the endocrine disrupting action in ecosystem. The cell counts of cultures harvested after 3 days' growth in the presence of 20-hydroxyecdysone. In Kc cell cultures, there were statistically significant different from control cells at 20HE 10/sup 7/-10/sup 5/. The morphological effects of all the hormones were similar, differing only in the dose level at which they were initiated.

  • Protein folding is a fundamental problem in structural bioinformatics and so numerous studies have been devoted to the subject. As the most common regular secondary conformation in proteins, helix has been an important ingredient of the protein folding problem. In particular, alanine based polypeptides are widely studied to identify the helix folding process in that the aianine amino acid is known to have one of the highest helix propensities. In principle, intrinsic helix propensities can be obtained from gas-phase measurements where solvent effect is absent. Hudgins et al. studied alanine-based peptides in vacuo using high-resolution ion mobility measurement technique. It was reported that introduction of a single Iysine at the C terminus resulted in the formation of very stable, monomeric polyalanine helices. We also have investigated helix formation in vacuo with different terminal charge conditions; we have found a new type of helix motif, To the best of our knowledge, this type of helix conformation has not been characterized before and we name it as I-helix.

국내에서 분리한 encephalomyocarditis virus의 성상 (Charactrization of encephalomyocarditis virus isolated from Korea)

  • We have investigated the characteristics of encephalomyocarditis(EMC) virus isolated in Korea. The CPE, buoyant density, polypeptide profile and the size of RNA of EMC virus were examined. The granulation, pyknosis and necrosis were observed from 30 to 48 hour's post inoculation of the virus into baby hamster kidney and lung cells. The buoyant density was 1.30 and $1.35g/m{\ell}$. Three different polypeptides, 26Kd, 32Kd, and 34Kd in size, were observed and the size of viral RNA was 7.7Kb.

Decomposition of Biological Macromolecules by Plasma Generated with Helium and Oxygen

  • In this study, we attempted to characterize the biomolecular effects of an atmospheric-pressure cold plasma (APCP) system which utilizes helium/oxygen $(He/O_2)$. APCP using $He/O_2$ generates a low level of UV while generating reactive oxygen radicals which probably serve as the primary factor in sterilization; these reactive oxygen radicals have the advantage of being capable to access the interiors of the structures of microbial cells. The damaging effects of plasma exposure on polypeptides, DNA, and enzyme proteins in the cell were assessed using biochemical methods.

한국환자유래의 녹농균 90-2-2205로부터 S형 Pyocin의 정제 및 특성 (Purification and Characterization of the S-type Pyocin of Pseudomonas aeruginwa 90-2-2205 Isolated from Korean Patients)

  • The s-type pyocin was purified from the lysate of the mitomycin C-induced Pseudomonas aeruginosa 90-2-2205 cells by the other of ammonium sulfate precipitation, DEAE-Sephadex A-50 chromatography and Sephadex G-200 gel filtration. The purity was confirmed by the polyacry-lamide gel electrophoresis. The molecular weight of the purified pyocin was estimated 180, 000 by gel filtration. The pyocin was analyzed to be a complex of some polypeptides by the SDS-PAGE. The pyocin was stable by heat treatment and at pH 6-7.5 by adding 10-3% gelatin and 0.2M NaCl to the 10mM Tris-HCl buffer (pH7.5). Its killing action against the sensitive cells was assumably a single hit process.
